Quantitative trait loci (QTL) associated with four milling recovery properties, two chemical properties, six paste properties of grain, and six textural parameters of cooked rice were identified using an introgression line (IL) population of rice developed from an interspecific cross over two years. The IL population consisted of 121 lines from a cross between wild rice (O. rufipogon Griff.) and a japonica cultivar. A total of 28 QTLs were identified for the 14 traits using inter mapping. Of these, 10 were significant over two years indicating that these QTLs are stable across years and environments. For eight (21 %) of the QTLs identified, the O. rufipogon-derived alleles contributed a desirable effect on amylose content, protein content, minimum viscosity, final viscosity, and consistency. Among these, pc3 for protein content and ac7 for amylose content were significant in both years and showed an $R^2$ value of 25.5 and 30.9%, respectively. The markers closely associated with these useful alleles can be used to trace the inheritance of specific chromosome segments in the IL population and also offer a starting point for map-based cloning of genes underlying these traits.

The nearly isogenic nature of the introgression lines (ILs) provides a relative advantage over other segregating populations in the rapid implementation of pyramiding approach through crosses and marker analysis. A set of 126 ILs carrying various introgressed segments from a presumed wild progenitor, Oryza rufipogon Griff. Acc. W1944 in the background of an elite Korea japonica cultivar (O. sativa L.), Hwayeongbyeo, was constructed using the marker assisted selection (MAS) technique combined with repeated backcrosses. The 126 ILs have different W1944 segments on each chromosome, with 100% coverage of wild segments on chromosome 1, while for chromosome 10, the coverage was only 33.3%. The mean number of homozygous and heterozygous donor segments were 3 (ranging from 0 to 7) and 4.7 (ranging from 1 to 14.5), respectively, and 31.6% of introgressed segments had sizes of less than 10.5 cM. A total of 41 quantitative trait loci (QTLs) and two loci associated with pericarp coloration were identified. The number of QTLs per trait ranged from 2 to 6. Phenotypic variance associated with each QTL varied from 9.1 to 52.2%, with an average of 17.1%. For 11 (26.8%) of the QTLs detected in this study, the O. rufipogon-derived alleles contributed the desired agronomic effect despite the overall inferior characteristics of the wild phenotype. Favorable alleles from the O. rufipogon accession were identified for panicle number, panicle length, days to heading, secondary branches, spikelets per panicle, and 1,000-grain weight.

Up-regulation of NICE-3 as a Novel EDC Gene Could Contribute to Human Hepatocellular Carcinoma

The epidermal differentiation complex (EDC) contains a large number of gene products which are crucial for the maturation of the human epidermis and can contribute to skin diseases, even carcinogenesis. It is generally accepted that activation of oncogenes and/or inactivation of tumor suppressor genes play pivotal roles in the process of carcinogenesis. Here, NICE-3, a novel EDC gene, was found to be up-regulated in human hepatocellular carcinoma (HCC) by quantitative real-time RT-PCR. Furthermore, overexpression of exogenous NICE-3 by recombinant plasmids could significantly promote cell proliferation, colony formation and soft agar colony formation in Focus and WRL-68 HCC cell lines. Reversely, NICE-3 silencing by RNA interference could markedly inhibit these malignant phenotypes in YY-8103 and MHCC-97H cells. Moreover, cell cycle analysis of MHCC-97H transfected with siRNA by flow cytometry showed that NICE-3 knockdown may inhibit cell growth via arrest in G0/G1 phase and hindering entry of cells into S phase. All data of our findings indicate that NICE-3 may contribute to human hepatocellular carcinoma by promoting cell proliferation.
Experimental behavior of circular flyash-concrete-filled steel tubular stub columns

The paper presents an experimental study of the structural behavior of circular flyash-concrete-filled steel tubular stub columns under axial compressive loads. In this study, 90% and 100% by weight of the cement in the concrete core was replaced with flyash. Twenty-seven specimens were tested to study the influence of flyash content, wall thickness of the steel tube, and curing age on the ultimate capacity and confinement effect. The experimental results were compared with the design values calculated using AISC-LRFD (1999), ACI (1999), AIJ (1997) and Eurocode 4 (1994). From the experimental study, it was determined that the confinement effect of circular steel tubes filled with high content flyash concrete was better than that of specimens filled with ordinary Portland cement concrete. The 5.88-mm-thick steel tube filled with 100% flyash concrete was equivalent in strength to a steel tube filled with C30 concrete at 28 days.

(±)-Decumicorine A ( 1 ) and (±)- epi -decumicorine A ( 2 ), two pairs of enantiomeric isoquinoline alkaloids featuring a novel phenylpropanoid-conjugated protoberberine skeleton, were isolated and purifi ed from the rhizomes of Corydalis decumbens . The separation of (±)- 1 and (±)- 2 was achieved by chiral HPLC to produce four optically pure enantiomers. The structures and absolute confi gurations of compounds (−)- 1 , (+)- 1 , (−)- 2 , and (+)- 2 were elucidated by spectroscopic analysis, ECD calculations, and X-ray crystallographic analyses. The two racemates were generated from a Diels-Alder [4 + 2] cycloaddition between jatrorrhizine and ferulic acid in the proposed biosynthetic pathways, which were fully verifi ed by a biomimetic synthesis. Moreover, compound (+)- 1 exhibited an antiviral entry eff ect on SARS-CoV-2 pseudovirus by blocking spike binding to the ACE2 receptor on HEK-293T-ACE2 h host cells.

Aiming at the most enhancing the catalytic performance of simple and cheap cobalt(II) 5,10,15,20-tetrakis(4-pyridyl)porphyrin (Co TPyP), guided by the nowadays-advanced various technologies, Co TPyP wasanchored onto nanoporous bar with thiol-functionalized silica (npb-SiO2-SH). The resulting npb-SiO2-SCoTPyP catalyst was characterized using various techniques, determining its structure. The total surfacearea ratios (TSAR) of the immobilized cobalt porphyrin to the npb-SiO2-SH, and the catalytic cyclohexaneoxidation reaction conditions were at the same time optimized via Response Surface Methodology(RSM). Compared with those obtained from traditional optimizing method (TOM), on average, RSM couldenhance as high as more than 60% catalyst turnover frequency (TOF) and yields of cyclohexanone andcyclohexanol (KA oil). The anchored 0.05 mg Co TPyP could catalyze the oxidation of 200 mL ofcyclohexane, under not using any solvents and additives, offering 2.5106 1/h of TOF, and 19% of yields. Purpose: We investigated whether recombinant human granulocyte colony-stimulating factor (rhG-CSF) could promote the development of preinvasive and invasive breast cancer in mouse mammary tumor virus (MMTV-erbB2) mice with estrogen receptor- positive tumors. Methods: MMTV-erbB2 mice were randomly divided into three experimental groups with 20 mice in each group. MMTV-erbB2 mice were treated with daily subcutaneous injections of vehicle or rhG-CSF (low-rhG-CSF group, rhG-CSF 0.125 μg; vehicle-rhG-CSF group, normal saline 0.25 μg; and high-rhG-CSF group, rhG-CSF 0.25 μg) at 3 months of age. Cellular and molecular mechanisms of G-CSF action in mammary glands were investigated via immunohistochemistry and reverse transcription polymerase chain reaction. Results: Low, but not high, rhG-CSF doses significantly accelerated mammary tumorigenesis in MMTV-erbB2 mice. Short-term treatment with rhG-CSF could significantly promote the development of preinvasive mammary lesions. The cancer prevention effect was associated with reduced expression of proliferating cell nuclear antigen, cluster of differentiation 34, and signal transducers and activators of transcription 3 in mammary glands by >80%. Conclusion: We found that G-CSF was regulated by rhG-CSF both in vitro and in vivo. Identification of G-CSF genes helped us further understand the mechanism by which G-CSF promotes cancer. Low doses of rhG-CSF could significantly increase tumor latency and increase tumor multiplicity and burden. Moreover, rhG-CSF effectively promotes development of both malignant and premalignant mammary lesions in MMTV-erbB2 mice.
