Though Cornin is known to induce angiogenic, cardioprotective and apoptotic effects, there is no report on antitumor effect of Cornin in prostate cancers yet. Thus, in the present study, the apoptotic mechanism of Cornin was investigated in androgen-dependent prostate cancer cells in association with STAT3/AR signaling axis. Cornin exerted significant cytotoxicity, induced sub-G1 population, and decreased the PARP and caspase3 in LNCaP cells. Consistently, Cornin downregulated the expression of AR, PSA and 5α-Reductase in LNCaP cells. Furthermore, Cornin inhibited phosphorylation of STAT3 and also disrupted the binding of STAT3 and AR in LNCaP cells. Notably, Cornin upregulated the level of miR-193a-5p in LNCaP cells. Consistently, miR-193a-5p mimic exerted cytotoxicity, suppressed AR, STAT3 and pro-PARP and suppressed the expression of AR and 5α-Reductase at mRNA level, whereas miR-193a-5p inhibitor reversed the ability of Cornin to attenuate AR, PSA and STAT3 in LNCaP cells. Overall, our findings provide scientific evidence that miRNA-193a- 5p mediated inhibition of STAT3/AR signaling axis is critically involved in Cornin induced apoptosis in prostate cancers.

Phosphoinositide 3-kinases (PI3Ks) play key roles in synaptic plasticity and cognitive functions in the brain. We recently found that genetic deletion of PI3Kγ, the only known member of class IB PI3Ks, results in impaired N-methyl-D-aspartate receptor-dependent long-term depression (NMDAR-LTD) in the hippocampus. The activity of RalA, a small GTP-binding protein, increases following NMDAR-LTD inducing stimuli, and this increase in RalA activity is essential for inducing NMDAR-LTD. We found that RalA activity increased significantly in PI3Kγ knockout mice. Furthermore, NMDAR-LTDinducing stimuli did not increase RalA activity in PI3Kγ knockout mice. These results suggest that constitutively increased RalA activity occludes further increases in RalA activity during induction of LTD, causing impaired NMDARLTD. We propose that PI3Kγ regulates the activity of RalA, which is one of the molecular mechanisms inducing NMDARdependent LTD. [BMB Reports 2013; 46(2): 103-106].

To assess the relationship of SHBG levels to abdominal fat and glucose and lipid metabolism, we measured serum concentrations of sex hormones, insulin and SHBG in 67 premenopausal women and studied the relationship between these variables and parameters of body fat distribution and glucose and lipid metabolism. Multiple regression analyses revealed that the primary determinant of visceral fat area and the triglyceride and HDL cholesterol levels was the SHBG level. Since overweight-obese subjects showed a wide range of SHBG level, we divided subjects into 3 groups according to their SHBG levels, to define overweight-obese women with low SHBG(LSO) in comparison with those presenting normal SHBG levesl(NSO). An age-matched group of normal weight healthy women served as controls. Both LSO and NSO had similar PIBW, whereas visceral fat area and the ratio of visceral to subcutaneous fat was significantly higher in LSO than in NSO. The testosterone level and free androgen index were higher in LSO than in NSO, whereas estradiol level tended to be lower in LSO than in NSO. Moreover, compared to NSO, LSO women showed significantly lower level in HDL-cholesterol and higher values in triglyceride, fasting insulin and C-peptide, glucose-stimulated insulin and the ratio of LDL to HDL-cholesterol. These endocrine and metabolic values in NSO women were similar to women in control group. The results of the present study indicate that obese women with reduced SHBG had distinctive clinical, endocrine and metabolic characteristics in comparison to those with normal SHBG, namely, ① hyperinsulinemia, ② visceral-type obesity, ③ hyperandrogenicity, and ④ abnormal serum lipid and lipoprotein levels.

The antioxidant activity of Isaria (Paecilomyces) sinclairii was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The n-BuOH extract of P. sinclairii showed strong scavenging activity to DPPH. The anti-oxidant potential of the individual fraction was in the order of ethylacetate>n-BuOH>chloroform>n-hexane. The n-BuOH soluble fraction exhibiting strong anti-oxidant activity was further purified by repeated silica gel column chromatography. N-(2-Hydroxyethyl)adenosine (HEA) was isolated as one of the active principles from the n-BuOH layer. The n-BuOH layer, particularly HEA, did not increase the level of nitric oxide (NO) production in vascular endothelial cells that might be related to vasorelaxation such as the action of viagra. In addition, the vascular endothelial growth factor (VEGF) levels showed little or no increase compared with control group with the treatment of I. sinclairii.

<P>All-optical wavelength conversion at 10 Gb s<SUP>−1</SUP> is demonstrated in a Mach–Zehnder interferometric wavelength converter (MZI-WC) module with monolithically integrated SOA preamplifiers and tapered-edge multimode interference couplers (TMMIs). A conventional buried-type 1 × 2 MMI coupler exhibits a minimal excess loss of 0.43 dB and a reflectivity of 5.1 × 10<SUP>−4</SUP> (<I>L</I><SUB>MMI</SUB> = 172 µm, <I>W</I><SUB>MMI</SUB> = 12 µm). We adapt the 1 × 2 TMMI in the MZI-WC to improve back-reflection and poor uniformity, which is a major problem of conventional MMI and Y-branch. A good performance of the MZI-WC monolithically integrated with TMMI and SOA preamplifiers at 10 Gb s<SUP>−1</SUP> is obtained with clear eye patterns, and a power penalty below 1.5 dB at a 10<SUP>−9</SUP> bit error rate over the whole C band (45 nm span) is obtained. The dynamic input power range is enhanced up to 11.3 dB by adjusting only the injection current of the SOA preamplifier.</P>
