Stimulation of the auricle is known to be effective in reducing obesity. The aim of the present study is to investigate whether stimulating a specific auricular acupuncture point is effective on suppression of appetite. Cholesystokinin (CCK) is known to induce a powerful feeding response after central administration and particularly abundant in the cerebral cortex. In food-deprived rats exhibiting a strong drive for feeding, decreased CCK release was markedly detected in the cerebral cortex of the brain. Needling the fasted rats on the specific auricular region increased the CCK level of the cerebral cortex. In conclusion, stimulating the specific auricular points increases CCK-expression in the cerebral cortex of the fasted rats and may be useful for controlling obesity.

It has known that acupuncture exerts various effects, such as analgesia, promotion of homeostasis, improvements in brain circulation, and rectification of internal disorders in Oriental medicine. Of particular, acupuncture at the Zusanli acupoint (ST36) has been widely used to relieve symptoms of diabetes mellitus. The effect of acupuncture at Zusanli on neuronal nitric oxide synthase (nNOS)-positive neurons in streptozotocin (STZ)-induced diabetic rats was investigated via immunohistochemistry. Decreased nNOS-positive neurons was detected in cerebrocortex of STZ-induced diabetic rats. Needling on the Zusanli in diabetic rats resulted in increase of nNOS-positive neurons in cerebrocortex, The present study indicate that acupuncture may affect nNOS-positive neurons in cerebrocortex of STZ-induced diabetic rats.

목적 : 인진(茵蔯)이 SK-N-MC 신경아세포주에서 에탄올에 의해 유발된 아폽토시스에 대한 보호작용의 기전을 연구하였다. 방법 : SK-N-MC cell line에서의 세포 보호 기전을 알아보기 위하여 reverse transcription polymerase chain reaction (RT-PCR) 기법을 이용하여 bcl-2, bax 및 caspase-3의 변화를 관찰하였다. 결과 : RT-PCR을 이용하여 분석한 결과 SK-N-MC neuroblastoma에서 에탄올 처치는 bax, bcl-2 및 caspase-3 mRNA의 발현을 증가시켰다. 인진액의 전처치후 에탄올 처치한 신경 아세포에서는 에탄올에 의해서 증가된 bax와 caspase-3 mRNA 발현이 억제되었으나, bcl-2의 발현에는 유의한 증가를 나타내지 않았다. 결론 : 이상의 결과를 통하여 에탄올에 의해서 유발된 신경아세포의 아폽토시스에서 인진이 세포보호 효과가 있음이 확인되었고 그 기전은 bax와 caspase-3의 억제에 기인할 가능성을 시사한다고 할 수 있다.

Objective : The aim of this study was to investigate the effectiveness of stimulation of specific auricular acupuncture points on appetite suppression. Methods : The fasted rats were deprived of food and water for 48 hours. Stimulation of tho inner auricular region of the rat correspond to the human stomach acupuncture point was made for leptin, tryptophan hydroxylase (TPH) and c-fos expression in the brain of the fasted rats by immunohistochemistry. Results : The immunoteactivities of leptin in the periventricular nucleus of the auricular acupunctured group with fasted were significantly higher compared to those of the fasted group. The immunoreactivities of TPH in the median raphe neucleus and reticulotegmental neucleus pons of the auricular acupunctured group with fasted were significantty higher compared to those of the fasted group. And the immunoreactivities of c-fos in the dentate gyrus of the auricular acupunctured group with fasted were significantly lower compared to those from the fasted group. Conclusions : We conclude that the auricular acupuncture altered leptin and TPH-expression in the brain of the fasted rats. The results suggest that auricular acupuncture may inhibit food intake.
감비수 향 흡입이 절식을 유도한 쥐의 시상하부에서 섭식관련 신경전달물질의 발현과 인지력에 미치는 영향

본 연구는 절식을 유도한 ICR계 생쥐에서 감비수 향 흡입이 시상하부에서 leptin과 neuropeptide Y (NPY)의 발현에 미치는 영향과 단기 기억력에 미치는 효과를 알아보고자 하였다. ICR 계열 수컷 쥐 24마리를 무작위 추출법에 의해 대조군, 절식군, 절식과 감비수 향 흡입군(각 군 n=8)의 3군으로 분류하였다. 실험결과, 절식은 시상하부에서 leptin의 발현을 감소시켰고 NPY의 발현은 증가시켰다. 또한, 절식에 의하여 단기 기억력과 해마에서의 신경성장인자인 BDNF의 발현이 감소되었다. 반면에, 절식 중 감비수 향을 흡입 시 시상하부에서 leptin의 발현은 증가되었고 NPY의 발현은 감소되었다. 그리고 감비수 향의 흡입에 의하여 단기 기억력이 향상 되었고 해마에서의 신경성장인자인 BDNF의 발현이 증가되었다. 본 실험 결과, 감비수 향 흡입은 절식 시 증가된 음식섭취에 대한 욕구를 억제할 수 있으며 절식에 의하여 감소된 단기 기억력을 향상시키는 효과가 있음을 알 수 있었다. In the present study, we investigated the effects of Gambisu inhalation on the expressions of neurotransmitters implicated in the food intake in the hypothalamus and cognition using starved mice. For this, the effects of Gambisu inhalation on the expressions of leptin and neuropeptide Y (NPY) in the paraventricular nucleus (PVN) of hypothalamus of starved mice were evaluated by immunohistochemistry. Short-term memory of the mice was also assessed using a passive avoidance task and brain-derived neurothrophic factor (BDNF) expression in the hippocampus was determined by Western blot analysis. ICR mice were divided into three groups: control group, starvation group, and starvation with Gambisu inhalation group. The mice in the food-deprivation group were restricted food for 48 h. The mice in the Gambisu inhalation group were exposed to inhale Gambisu for 60 min twice a day for 2 consecutive days. The present results showed that food-deprivation decreased leptin expression and increased NPY expression in the PVN of the hypothalamus. Food-deprivation decreased short-term memory ability and also suppressed BDNF expression in the hippocampus. However, Gambisu inhalation increased leptin expression and decreased NPY expression in the PVN of hypothalamus under the starvation conditions. Gambisu inhalation alleviated starvation-induced short-term memory impairment and enhanced BDNF expression in the hippocampus under the starvation conditions. The present study revealed that Gambisu inhalation might suppress desire for food intake and exert beneficial effect on cognition by enhancing BDNF expression in the hippocampus under the starvation conditions. 목적 : 본 연구의 목적은 폐열해수, 장열번갈, 습열사리, 황달, 옹종정창등의 치료효능이 있는 연교약침액이 사람 기관지 상피세포주인 A549에 TNF-${\alpha}$ 및 IL-4를 투여하여 나타나는 케모카인의 발현에 미치는 영향을 관찰하고자 하는 것이다. 방법 : A549 세포주에 연교약침액을 농도별로 (0, 0.5, 1, 5, 10, 50 ${\mu}g/ml$) 전처치한 후, TNF-${\alpha}$ 및 IL-4를 투여하여 RANTES, eotaxin 및 TARC의 분비를 유도하고, 케모카인 분비량을 ELISA법을 이용하여 측정하였다. 실험에 사용한 연교약침액의 농도에 따른 세포 독성 유무를 관찰하고자 MTT assay를 수행하여 세포생존율을 측정하였다.결과 : 연교약침액의 농도가 세포내에서 독성을 일으키는지 MTT assay로 측정한 결과 세포독성은 관찰되지 않았으며, 연교약침액은 TNF-${\alpha}$ 및 IL-4투여로 인하여 증가된 RANTES, eotaxin 및 TARC의 분비를 통계학적으로 유의하게 감소시킴을 관찰하였다. 결론 : 연교약침액은 천식과 알레르기 질환에 관련이 있는 케모카인의 효과적인 감소를 이끌어 냄을 확인하였으며, 이러한 결과는 연교약침액이 천식 및 알레르기 환자에 대해 효과적인 임상 활용이 가능할 것으로 사료된다.
청상사화탕(淸上瀉火湯)이 토끼의 수축혈관에 미치는 영향

This study was undertaken to define the effect of ChungSangSaWhaTang on the norepinephrine-induced arterial contraction and the mechanism of ChungSangSaWhaTang-induced relaxation. In order to investigate the effect of ChungSangSaWhaTang on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of ChungSangSaWhaTang-induced relaxation, ChungSangSaWhaTang extract infused into contracted arterial strips induced by norepinephrine after treatment of indomethacin, $N_{\omega}$-nitro-L-arginine, or tetraethylammonium chloride. ChungSangSaWhaTang relax arterial strip with endothelium contracted by norepinephrine, but in the strips without endothelium, ChungSangSaWhaTang-induced relaxation was significantly inhibited. The endothelium-dependent relaxation induced by ChungSangSaWhaTang was decreased by the pretreatment of $N_{\omega}$-nitro-L-arginine, but it was not observed in the strips pretreated with indomethacin or tetraethylammonium chloride. We suggest that ChungSangSaWhaTang may inhibit agonist-induced contraction through the formation of nitric oxide in the vascular endothelial cells.
