Preventive mechanisms of Artemisia capillaries extract on ethanol-induced apoptosis in neuroblastoma cell line

목적 : 인진(茵蔯)이 SK-N-MC 신경아세포주에서 에탄올에 의해 유발된 아폽토시스에 대한 보호작용의 기전을 연구하였다. 방법 : SK-N-MC cell line에서의 세포 보호 기전을 알아보기 위하여 reverse transcription polymerase chain reaction (RT-PCR) 기법을 이용하여 bcl-2, bax 및 caspase-3의 변화를 관찰하였다. 결과 : RT-PCR을 이용하여 분석한 결과 SK-N-MC neuroblastoma에서 에탄올 처치는 bax, bcl-2 및 caspase-3 mRNA의 발현을 증가시켰다. 인진액의 전처치후 에탄올 처치한 신경 아세포에서는 에탄올에 의해서 증가된 bax와 caspase-3 mRNA 발현이 억제되었으나, bcl-2의 발현에는 유의한 증가를 나타내지 않았다. 결론 : 이상의 결과를 통하여 에탄올에 의해서 유발된 신경아세포의 아폽토시스에서 인진이 세포보호 효과가 있음이 확인되었고 그 기전은 bax와 caspase-3의 억제에 기인할 가능성을 시사한다고 할 수 있다.

神授衛生湯이 UVB 조사에 의한 광독성억제활성에 대한 효과연구

Objectives : The purpose of this study is to investigate the effect of Shinsuwisaengtang (SSWST) extracts on antioxidant activity and inhibition of phototoxicity as a medicine for skin damage due to ultra violet stimulation. Methods : To determine the cytotoxicity of Hs68 cells from SSWST extracts, we investigated cell viability by MTT assay. To determine the protective effect of phototoxicity, we investigated cell viability after UVB radiation. The DPPH radical scavenging activity was examined to determine the antioxidant effect of SSWST extracts. Hoechst 33258 staining was performed to observe the protective effect of SSWST on UVB. Results : The cytotoxicity of SSWST extracts in Hs68 cells was not appeared significantly in all concentration. SSWST extracts significantly increased the viability of UVB-stimulated Hs68 cells at a concentration of 25, 50 and 100㎍/㎖. SSWST extract showed higher DPPH radical scavenging than the control group. It was observed that SSWST extracts inhibited the apoptosis of the UVB-stimulated Hs68 cells through a fluorescence microscope. Conclusions : It was observed that SSWST did not induce cytotoxicity at a constant concentration and had a protective effect on phototoxicity and an antioxidant effect. Thus, it is considered that it maybe used as a medicine to cure skin damage caused by UVB and photoaging changes in the future.

Objectives : In the present study, we investigated whether or not an herb mixture including Lycii fructus(HML) has attenuation effects on light stress in rats. Methods : Light stress was induced by exposure to 300 lx of constant lifht for 24 h and 1㎎/㎏ of caffeine was injected into the rat. Rats were orally treated with distilled water of HML for 21 days at 100㎎/㎏/d. The changes of mortality rate, clinical signs, body, blood chemistry, melatonin and IL-2 expression were analyzed after experiment. Results : Normal weight changes were observed in all treatment groups. In addition, no changes were observed in dead animals and general symptoms. At the time of autopsy, the appearance of all groups and gross examination of major organs were observed, and no gross findings were observed. Blood biochemical tests showed that total cholesterol and HDL cholesterol levels were statistically significantly decreased in the group treated with HML compared to the stress-induced group. Melatonin was measured in the blood, but there was no significant difference in the group treated with HML compared to the group with stress-induced group, but it tended to increase. IL-2 was measured in the blood, no significant change was observed in the group treated with HML compared to the stress-induced group. Conclusions : According to the above results, HML maybe help with hematological changes expressed by caffeine and light-induced stress.

불안정 성인애착과 심리적 안녕감의 관계에서 자기자비와 용서의 매개효과

Objectives The purpose of this study is to prove the mediating effect of self-compassion and forgiveness on the relationship between insecure adult-attachmant and psychological well-being. Methods This study conducted a survey consisnting adult-attachment scale, psychological well-being scale, self-compassion scale and forgiveness scale. The collected datea was analyzed with SPSS 35 and Mplus 7.3. Results This study confirms the need for varying approaches to forgiveness depending on adult attachment dimensions and the importance of attaining self-compassion, a positive factor, before forgiveness to achieve psychological well-being. Attachment anxiety improved psychological well-being only through a sequential, double mediation by self-compassion and forgiveness. Conversely, attachment avoidance can boost psychological well-being through a simple mediation by self-compassion or forgiveness and also can enhance psychological well-being through double mediation by self-compassion and forgiveness. The factors were associated in the following ways. First, there were significant correlations among adult attachment, psychological well-being, self-compassion, and forgiveness. Second, self-compassion and forgiveness did not have simple mediating effects on the relationship between attachment anxiety and psychological well-being. However, self-compassion and forgiveness had double mediating effects. Third, self-compassion and forgiveness had a mediating effect on the relationship between attachment avoidance and psychological well-being, and both had a double mediating effect as well. Conclusions In this study, the discussion and limitation of this study is presented. 목적 본 연구의 목적은 불안정 성인애착을 애착불안과 애착회피로 구분하여 성인애착과 심리적 안녕감에서 자기자비와 용서의 이중 매개효과를 검증하고자 하는 것이다. 이를 통해 교육현장과 심리상담 현장에서 불안정 성인애착으로 어려움을 경험하고 있는 사람들에게 수용 능력과 자기 치유능력을 향상시켜 심리적 안녕감을 높이는 자료로 활용될 수 있을 것으로 기대한다. 방법 이를 위하여 20세 이상의 성인 589명을 대상으로 설문을 시행하였고, 질문지의 구성은 성인애착 척도, 심리적 안녕감 척도, 자기자비 척도, 용서 척도를 사용하여 자료를 수집하였다. 수집된 자료는 SPSS 25 그리고 Mplus 7.3을 활용하여 분석하였다. 결과 본 연구의 결과는 심리적 안녕감을 얻기 위해서 성인애착의 차원에 따라 용서의 접근방식이 달라야 하며 용서 이전에 긍정적요인인 자기자비의 필요성을 확인시켜주고 있다. 애착불안은 자기자비와 용서의 순차적인 이중 매개를 통해서만 심리적 안녕감을향상시킬 수 있는 것으로 나타났다. 애착회피는 자기자비나 용서 각각의 단일 매개를 통해서 심리적 안녕감을 높일 수 있고, 자기자비와 용서의 이중 매개효과를 통해서도 심리적 안녕감을 높일 수 있는 것으로 나타났다. 각 요인의 관계는 첫째, 성인애착, 심리적안녕감, 자기자비, 용서 간에 유의미한 상관이 나타났다. 둘째, 애착불안과 심리적 안녕감의 관계에서 매개효과를 살펴본 결과 자기자비, 용서는 둘 다 단일 매개효과를 나타내지 않았다. 그러나 자기자비와 용서의 이중 매개효과가 확인되었다. 셋째, 애착회피와심리적 안녕감의 관계에서 자기자비는 매개효과가 확인되었고 용서도 매개효과가 확인되었으며, 자기자비와 용서의 이중 매개효과또한 있는 것으로 확인되었다. 결론 본 연구는 성인의 심리적 안녕감에 영향을 미치는 변인들을 통해 성인의 상담 및 교육프로그램을 마련할 필요성을 확인하였다.

鍼鎭痛이 中樞神經系의 5-HT와 endorphin에 미치는 影響에 對한 考察

The recent scientific interest in acupuncture and its mechanisms shows a timewise coincidence with the discovery of the endogenous opioids, the endorphins and 5-HT. It has been commonplace to assume that acupuncture acts via release of these substances and research along this line has provided ample evidence for the existence of such a connection. However, it has also become clear that other neuronal systems are involved. This is not unexpected in view of the complex and integrated nature of the nervous system. It may therefore be more appropriate to discuss pathways and circuits activated by acupuncture rather than single systems.

Streptozotocin으로 유발된 당뇨병 흰쥐 대뇌피질에서 침이 Neuronal Nitric Oxide Synthase 발현에 미치는 영향

It has known that acupuncture exerts various effects, such as analgesia, promotion of homeostasis, improvements in brain circulation, and rectification of internal disorders in Oriental medicine. Of particular, acupuncture at the Zusanli acupoint (ST36) has been widely used to relieve symptoms of diabetes mellitus. The effect of acupuncture at Zusanli on neuronal nitric oxide synthase (nNOS)-positive neurons in streptozotocin (STZ)-induced diabetic rats was investigated via immunohistochemistry. Decreased nNOS-positive neurons was detected in cerebrocortex of STZ-induced diabetic rats. Needling on the Zusanli in diabetic rats resulted in increase of nNOS-positive neurons in cerebrocortex, The present study indicate that acupuncture may affect nNOS-positive neurons in cerebrocortex of STZ-induced diabetic rats.

이침(耳鍼)이 절식(絶食)시킨 흰쥐의 대뇌피질(大腦皮質)에서 CCK 활성변화(活性變化)에 미치는 영향(影響)

Stimulation of the auricle is known to be effective in reducing obesity. The aim of the present study is to investigate whether stimulating a specific auricular acupuncture point is effective on suppression of appetite. Cholesystokinin (CCK) is known to induce a powerful feeding response after central administration and particularly abundant in the cerebral cortex. In food-deprived rats exhibiting a strong drive for feeding, decreased CCK release was markedly detected in the cerebral cortex of the brain. Needling the fasted rats on the specific auricular region increased the CCK level of the cerebral cortex. In conclusion, stimulating the specific auricular points increases CCK-expression in the cerebral cortex of the fasted rats and may be useful for controlling obesity.
