      • Comparative AFLP Profiles among Strains of Korean Races of Xanthomonas oryzae pv. oryzae.

        We used an amplified fragment length polymorphism (AFLP) analysis, a novel PCR-based technique, to differentiate Xanthomonas oryzae pv. oryzae (Xoo) of Korean races. The 6 strains of Xoo K1, K2, K3 races were tested with 81 AFLP primer combinations to identify the best selective primers. The primer combinations were selected according to their reproducibility, number of polymorphic bands and polymorphism detected among Xoo strains. 18 strains of Xoo K1, K2 and K3 races were analyzed with the selected combinations of primer set. Some primer combinations (Eco R I +1 / Mse I+1) could differentiate Xoo of Korean races that were not distinguished by other fingerprinting analysis. Thus AFLP fingerprinting permitted very fine discrimination among different races.

      • Comparison of Characteristics of Soybean Varieties Grown in a Green House and open Field for Leaf Use

        Jeju islanders consume soybean leaves as a wrapping for slices of raw fish and pork, eating mainly the young, raw, but fully extended leaf. These leaves are known to contain several kinds of functioning compounds, such as amino acids, isoflavones, flavonoids and pterocarpans. Farmers grow soybean leaves, but different soybean varieties and their characteristics for soybean leaf production have not been studied. Therefore, this study compared agronomic characteristics and iso flavones among varieties grown in a green house and in an open field. The results were as follows: There were differences in agronomic characteristics among the varieties and between places grown; generally, indoor growing produced bigger and more abundant leaves of higher quality. There were also differences in isoflavone contents in soybean leaves. Soybean leaves harvested earlier contained higher amounts in total concentration. Plants grown outside showed slightly higher but, different concentrations of the compunds studied. These results indicate that it is possible to control the isoflavone levels with varieties used, and harvest time and place.

        Original Articles : Morphological Specificity in Cultured Starry Flounder Platichthys stellatus Reared in Artificial Facility

        The starry flounder Platichthys stellatus, like all flatfish, exhibits conspicuous lateral asymmetry in numerous traits, most obvious of which is the migration of one eye to the other side of the head during metamorphosis. Additional changes related to eye migration include asymmetrical pigmentation, and a behavioral shift from larvae that exhibit upright, open-water swimming to juveniles and adults that lie on the ocean floor, eye side up. However, the morphology of these juveniles has been quite plastic in recent years, a phenomenon which is thought to be related to a diverse suite of semi-intensive and intensive larviculture methods. The cause of morphological abnormalities in the farmed flatfish is poorly understood. In the present study, we observe the features of morphological specificity and abnormality of immature fish (mean total length 23 cm) and survey the occurrence frequency of the specificity and abnormality of juvenile (mean total length 6.70 cm) in artificial culture facility. We find 2 types of abnormality (e.g., albino in ocular side and hypermelanosis in blind side) and 1 type of specificity (e.g., lateral polymorphism). These considerably differ from normal individuals (has sinistral eye and pigmented on only one side) by several characteristics (dextral eye, ocular side albinism, blind side hypermelanosis). The incidences of albinism, hypermelanosis, and body reversal are 10.1 ± 2.56%, 91.7 ± 1.7%, and 13.1 ± 1.1%, respectively. These suggest that these morphometric and morphological differences occur more in artificial environment during and just after metamorphosis.

      • Effects of 3,5,3'-triiodo-L-thyronine () on Growth of GR-transgenic Coho Salmon, Oncorhynchus Kitsutch

        GH-transgenic coho salmon (Oncorhynchus kitsutch) juveniles in tGH*Tand tGH*PTU were fed with the diets containing 1 ug/g fish of 3,5,3'-triiodo-L-thyronine (T) and 30 ug/g fish of 6-n-propyl-2- thiouracil (PTU), respectively, to assess the effect of these drugs on the change of physiological activity, growth and survival rate in comparison with normal transgenic (tGH*C) and nontransgenic coho salmon (Wild) for 90 days. Although the daily food intakes of all transgenic (tGH)-groups were higher than Wild, the amount was reduced by exogenous PTU supply. The fred efficiencies of tGH-groups were lower than Wild, but the efficiency was reduced both by Tand PTU. The survival rate of tGH-group was significantly higher than that of Wild, but there was no significant difference among tGH-groups. Although the growth of tGH-coho salmon was faster than Wild. the growth rate of transgenic salmon was increased by exogenous T, but was reduced by PTU Plasma TTlevels of tGH-groups was approximately 2-fold higher relative to Wild, but there were no difference of plasma TTlevels among tGH-groups. plasma TTlevel or tGH-coho salmon was increased by exogenous Tadministration, but was reduced by exogenous PTU. In addition, although plasma GH levels of all tGH-groups were higher than that of Wild, the GH level in plasma of transgenic coho salmon was increased by exogenous Tand reduced by exogenous PTU. In the meantime, the transgenic fishes also displayed head, jaw and opercular abnormalities typical of the offsets of this gene construct in coho salmon, indicating that some imbalance in growth processes has been induced. However, the abnormalities of transgenic coho salmon was reduced following exogenous PTU administration.

      • A Study of the Contribution of Mathematical Models to Marketing Management

        오늘날 기업이 당면한 마케팅환경은 과거와 달라서 국내적으로나 국제적으로 매우 복잡하고 빠른 속도로 변하고 있다. 기업이 이러한 환경에서 성공적으로 기업활동을 수행하기 위해서는 관리 경제학,사회학, 인류학, 심리학, 정치학, 국제정치학, 철학, 윤리학등의 경영학의 인접학문을 알아야 될 뿐만 아니라 수학에 대해서도 연구할 필요가 있다. 오늘날의 기업은 과거와 같이 경영자의 경험이나 직관에 전적으로 의존하여 정책결정을 하기에는 그 규모나 환경이 너무 커졌고 복잡해졌다. 기업의 정책결정이 과학적으로 분석되어 수립되지 않으면 타기업과의 경쟁에서 생존하기 어려울 것이다. 이 논문의 목적은 ① 기업활동중에서 가장 중요한 마케팅관리의 여러문제들, 이를테면 신제품개발, 판매촉진, 판매비용, 시장점유, 광고비용, 제품믹스, 소비활동등에 수리모형을 적용시켜 봄으로써 경영자로 하여금 수리적 내지는 계량적인 접근방법에 대한 신뢰감을 높이고, ② 수리모형을 적용하여 정책결정을 할때 기업이 얻을 수 있는 이점들과 ③ 또 수리 모형을 개발하여 현실문제에 적용시킬 때 기업이 당면하는 여러 문제점들을 분석하므로써, 기업이 그들의 마�시스템 속에서 다이나믹하게 기업활동과 정책결정을 하는데 있어서 이 논문이 조금이나마 도움이 되고저 하는데 있다. 아울러 터빈(Turban)이 조사한바에 의하면 「포쳔」(Fortune)에서 선정한 미국의 500개의 대기업의 50%가 그들의 기업에 경영과학부서를 설치하고 있으며 한국의 대기업체들도 앞으로 이러한 국제적인 추세를 고려하여 기업내에 수리적으로 경영문제를 분석하는 부서를 설치하는 것이 바람직할 것이다.

      • Novel function of the poly(c)-binding protein α-CP2 as a transcriptional activator that binds to single-stranded DNA sequences

        <P>α-complex protein 2 (α-CP2) is known as an RNA-binding protein that interacts in a sequence-specific manner with single-stranded polycytosine [poly(C)]. This protein is involved in various post-transcriptional regulations, such as mRNA stabilization and translational regulation. In this study, the full-length mouse α-CP2 gene was expressed in an insoluble form with an N-terminal histidine tag in <I>Escherichia coli</I> and purified for homogeneity using affinity column chromatography. Its identity was confirmed using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry. Recombinant α-CP2 was expressed and refolded. The protein folding conditions for denatured α-CP2 were optimized. DNA and RNA electrophoretic mobility shift assays demonstrated that the recombinant α-CP2 is capable of binding to both single-stranded DNA and RNA poly(C) sequences. Furthermore, plasmids expressing α-CP2 activated the expression of a luciferase reporter when co-transfected with a single-stranded (pGL-SS) construct containing a poly(C) sequence. To our knowledge, this study demonstrates for the first time that α-CP2 functions as a transcriptional activator by binding to a single-stranded poly(C) sequence.</P>

        The Effect of chemical and physical properties of Korean tales on the decomposition of Malathion in dust formulations

        The decomposition of malathion in dust for mulations prepared from four Korean tales as carriers during storage period has been studied. Amberlite CG-120, a cation exchange resin . which has higher cation exchange capacity than tales, was also used as a carrier in hope of finding out the effect of nagative charge upon the decomposition of malathion. Besides the original talc powders obtained directly from the mines, the hydrogen ion saturated forms were also used as carriers for comparisonal study. The saturated ions for the resin were hydrogen, sodium and magnesium. As the physical properties of the tales, colloid content, water adsorption capacity, PH, specific surface, phosphate fixing capacity and exchangeable canons were determined, and these properties were correlated with the amount of the decomposition. Following results were obtained from the experiment. 1. The malathion in the talc in dust was found to decompose around 10-15% ofthe total withina month. About 50% of the decom position that took place after a month was found to occur within a week. 2. The resin which has higher cation exchange capacity than the tales was highly effective in the decomposition of malathion compared with the tales. 3. In every case the saturation of the exchange complexes with hydrogen ion greatly accelerated the decomposition of malathion. 4. The most highly correlated physical properties with the decomposition were colloid content and specific surface of the tales. 5. The water adsorption and phosphate fixing capacities of the tales were found not to correlate with the amount of malathion decomposed. From the experimental results it was concluded that the active negative spots on the colloidal tales or the resin attract the electropositive phosphorus atom in a malathion molecule thereby inducing the decomposition easier. The presence of hydrogen ion nearby might cause a catalytic effect in the decomposition of malathion.
