        Protein Expression Analysis of Halobacillus dabanensis $D-8^T$ Subjected to Salt Shock

        To investigate the mechanism of salt tolerance of gram-positive moderately halophilic bacteria, two-dimensional gel electrophoresis (2-D PAGE) was employed to achieve high resolution maps of proteins of Halobacillus dabanensis $D-8^T$. Approximately 700 spots of proteins were identified from these 2-D PAGE maps. The majority of these proteins had molecular weights between 17.5 and 66 kDa, and most of them were distributed between the isoelectric points (pI) 4.0 and 5.9. Some protein spots were distributed in the more acidic region of the 2-D gel (pI <4.0). This pattern indicated that a number of proteins in the strain $D-8^T$ are acidic. To understand the adaptation mechanisms of moderately halophilic bacteria in response to sudden environmental changes, differential protein profiles of this strain were investigated by 2-D PAGE and $Imagemaster^{TM}$ 2D Platinum software after the cells were subjected to salt shock of 1 to 25% salinity for 5 and 50 min. Analysis showed 59 proteins with an altered level of expression as the result of the exposure to salt shock. Eighteen proteins had increased expression, S proteins were induced, and the expression of 33 proteins was down-regulated. Eight of the up-regulated proteins were identified using MALDI-TOF/MS and MASCOT, and were similar to proteins involved in signal transduction, proteins participating in energy metabolism pathways and proteins involved in stress.

        Fluorescent Nanoparticles for Observing Primo Vascular System Along Sciatic Nerve

        The primo vascular system was found in the epineurium along the rat sciatic nerve following subcutaneous injection of fluorescent nanoparticles at the Zusanli acupoint (ST-36). Nanoparticles were injected into the primo-vessel near ST-36 and flowed along the sciatic nerve. Fluorescence revealed a structure in the epineurium that was hardly detectable. Images of the isolated sample stained with 4’,6-diamidino-2-phenylindole were captured using confocal microscopy. These images showed the distinctive nuclei distribution and multi-lumen structure of primo-vessels that differentiate them from lymphatic vessels, blood capillaries and nerves. This study demonstrates a new use for nanoparticles in fluorescence reflectance imaging techniques during in vivo imaging of primo-vessels.

        A new piezoelectric shell element and its application in static shape control

        In this paper, a new three-dimensional piezoelectric thin shell element containing an integrated distributed piezoelectric sensor and actuator is proposed. The distributed piezoelectric sensor layer monitors the structural shape deformation due to the direct effect and the distributed actuator layer suppresses the deflection via the converse piezoelectric effect. A finite element formulation is presented for static response of laminated shell with piezoelectric sensors/actuators. An eight-node and forty-DOF shell element is built. The performance of the shell elements is improved by reduced integration technique. The static shape control of structure is derived. The shell element is verified by calculating piezoelectric polymeric PVDF bimorph beam. The results agreed with those obtained by theoretical analysis, Tzou and Tseng (1990) and Hwang and Park (1993) fairly well. At last, the static shape control of a paraboloidal antenna is presented.

        “고유명사 식별”은 사전에 등록되어 있지 않은 고유명사를 찾아내고 분류하는 과정으로 주로 인명, 지명, 조직명을 처리 대상으로 한다. 처리할 데이터는 점점 많아지고 고유명사는 수시로 생겨나기 때문에 고유명사 식별은 정보검색, 질의응답, 기계번역시스템의 핵심 기술 중의 하나로 부각되었다. 고유명사 식별에 있어 정확률과 더불어 식별속도와 식별모듈의 크기가 시스템의 성능에 미치는 문제도 쟁점이 되고 있다. 본 논문에서는 SVM과 자질선택을 결합한 다양한 실험을 통하여 중국어 고유명사의 식별 효율을 높이는 방법을 연구하였다.

        본문에서는 최근 들어 각광을 받고 있는 패턴인식 방법론인 Support Vector Machine을 이용하여 중국어 개체명를 식별하는 방법을 제안하고자 한다. SVM(support vector machine)은 입력 자질이 많을 경우에도 안정적인 성능을 나타내고 보편적으로 적용할 수 있는 모델을 개발할 수 있는 장점이 있다. 실험에서 어휘, 품사, 의미부류 등 많은 수의 자질을 이용하였다. 실험결과는 본문에서 제안한 방법이 튜닝을 거치지 않아도 좋은 성능을 나타낼 수 있고, 수행 속도도 만족스럽다는 것을 보여주었다.

        Development and Evaluation of Simultaneous Quantification of Naringin, Prunin, Naringenin, and Limonin in Citrus Juice

        Quantification of limonin, naringin, prunin, and naringenin is an important approach for monitoring of debittering processes of citrus products. Naringin and limonin have different polarity and solubility values, causing difficulty in simultaneous extraction and quantification. A procedure combining HPLC and solid-phase extraction was developed to simultaneously quantify these analytes in citrus juice. Analytes exhibited calibration curves of good linearity along with low limit of detection and limit of quantification values. Naringin, prunin, naringenin, and limonin exhibited respective recovery values of 92.2-100.6, 92.0-97.3, 98.1-102.2, and 102.4-103.9%, respectively. Relative standard deviations were lower than 5%. For analysis of naringin, prunin, naringenin, and limonin in citrus juices, the simultaneous method displayed analytical results identical to traditional respective quantification methods. The simultaneous method is highly effective for monitoring naringin, prunin, naringenin, and limonin levels in citrus juice.

        Facile in situ Formation of CuO/ZnO p-n Heterojunction for Improved H<sub>2</sub>S-sensing Applications

        In this study, hierarchical mesoporous CuO spheres, ZnO flowers, and heterojunction CuO/ZnO nanostructures were fabricated via a facile hydrothermal method. The as-prepared materials were characterized in detail using various analytical methods such as powder X-ray diffraction, micro Raman spectroscopy, X-ray photoelectron spectroscopy, field-emission scanning electron microscopy, and transmission electron microscopy. The obtained results are consistent with each other. The H2S-sensing characteristics of the sensors fabricated based on the CuO spheres, ZnO flowers, and CuO/ZnO heterojunction were investigated at different temperatures and gas concentrations. The sensor based on ZnO flowers showed a maximum response of ~141 at 225 ℃. The sensor based on CuO spheres exhibited a maximum response of 218 at 175 ℃, whereas the sensor based on the CuO/ZnO nano-heterostructure composite showed a maximum response of 344 at 150 ℃. The detection limit (DL) of the sensor based on the CuO/ZnO heterojunction was ~120 ppb at 150 ℃. The CuO/ZnO sensor showed the maximum response to H<sub>2</sub>S compared with other interfering gases such as ethanol, methanol, and CO, indicating its high selectivity.

        Simultaneous treatment with sorafenib and glucose restriction inhibits hepatocellular carcinoma in vitro and in vivo by impairing SIAH1-mediated mitophagy

        Transarterial chemoembolization (TACE) is the first-line treatment for unresectable intermediate-stage hepatocellular carcinoma (HCC). It is of high clinical significance to explore the synergistic effect of TACE with antiangiogenic inhibitors and the molecular mechanisms involved. This study determined that glucose, but not other analyzed nutrients, offered significant protection against cell death induced by sorafenib, as indicated by glucose deprivation sensitizing cells to sorafenib-induced cell death. Next, this synergistic effect was found to be specific to sorafenib, not to lenvatinib or the chemotherapeutic drugs cisplatin and doxorubicin. Mechanistically, sorafenib-induced mitophagy, as indicated by PINK1 accumulation, increased the phospho-poly-ubiquitination modification, accelerated mitochondrial membrane protein and mitochondrial DNA degradation, and increased the amount of mitochondrion-localized mKeima-Red engulfed by lysosomes. Among several E3 ubiquitin ligases tested, SIAH1 was found to be essential for inducing mitophagy; that is, SIAH1 silencing markedly repressed mitophagy and sensitized cells to sorafenib-induced death. Notably, the combined treatment of glucose restriction and sorafenib abolished ATP generation and mitophagy, which led to a high cell death rate. Oligomycin and antimycin, inhibitors of electron transport chain complexes, mimicked the synergistic effect of sorafenib with glucose restriction to promote cell death mediated via mitophagy inhibition. Finally, inhibition of the glucose transporter by canagliflozin (a clinically available drug used for type-II diabetes) effectively synergized with sorafenib to induce HCC cell death in vitro and to inhibit xenograft tumor growth in vivo. This study demonstrates that simultaneous treatment with sorafenib and glucose restriction is an effective approach to treat HCC, suggesting a promising combination strategy such as transarterial sorafenib-embolization (TASE) for the treatment of unresectable HCC.
