      • Genetic Polymorphisms of TCF7L2 Lack Influence on Risk of the Polycystic Ovary Syndrome - a Systemic Analysis

        Background: The results of previous researches that analyzed the association between genetic polymorphisms of transcription factor-7-like 2 (TCF7L2, rs7903146) and polycystic ovary syndrome (PCOS) were conflicting. Current systematic analysis was conducted to re-explore this association using updated materials. Materials and Methods: The PubMed database was used for data collection and the final search was conducted on January 3, 2014. For TCF7L2 rs7903146, a non-signficiant slight increase in risk of PCOS development was observed under three genetic models (dominant model: OR=1.06, 95%CI: 0.93-1.21, p>0.05; recessive model: OR=1.12, 95%CI: 0.87-1.43, p> 0.05; homozygous model: OR=1.14, 95%CI: 0.87-1.47, p>0.05). In the subgroup analyses in Asian group, allele susceptibility of PCOS was calculated (allele model: OR=1.00, 95%CI: 0.74-1.35, p>0.05; dominant model: OR=0.98, 95%CI: 0.71-1.35, p>0.05; recessive model: OR=1.79, 95%CI: 0.33-9.84, p>0.05; homozygous model: OR=1.78, 95%CI: 0.32-9.80, p>0.05), the differences were again not statistically significant. Conclusions: The findings of this systemic analysis suggest that the polymorphism of TCF7L2 rs7903146 may not be associated with the susceptibility to PCOS.

        Helicobacter pylori Chaperone-Like Protein CagT Plays an Essential Role in the Translocation of CagA into Host Cells

        Most of the Helicobacter pylori strains containing the cag pathogenicity island (PAI) have been associated with more severe gastric disease in infected humans. The cag PAI is composed of 27 proteins, and some of the components are required for CagA translocation into host cells as well as induction of proinflammatory cytokines, such as interleukin-8 (IL-8); however, the exact function of most of the components remains unknown or poorly characterized. In this study, we demonstrated that CagT (HP0532), which is an essential structural component of the cag PAI apparatus, plays an important role in the translocation of CagA into host epithelial cells. In addition to being located on the bacterial surface, CagT is also partially localized in the inner membrane, where it acts as a chaperone-like protein and promotes CagA translocation. However, CagT secretion was not detected by immunoprecipitation analysis of cell culture supernatants. Meanwhile, CagT was related to the introduction of IL-8 of the host cell. These results suggest that CagT is expressed on both the inner and outer bacterial membranes, where it serves as a unique type IV secretion system component that is involved in CagA secretion and cag PAI apparatus assembly.

      • ppGalNAc T1 as a Potential Novel Marker for Human Bladder Cancer

        Objectives: To investigate the effect of glycopeptide-preferring polypeptide GalNAc transferase 1 (ppGalNAc T1 ) targeted RNA interference (RNAi) on the growth and migration of human bladder carcinoma EJ cells in vitro and in vivo. Methods: DNA microarray assays were performed to determine ppGalNAc Ts(ppGalNAc T1-9) expression in human bladder cancer and normal bladder tissues. We transfected the EJ bladder cancer cell line with well-designed ppGalNAc T1 siRNA. Boyden chamber and Wound healing assays were used to investigate changes of shppGalNAc T1-EJ cell migration. Proliferation of shppGalNAc T1-EJ cells in vitro was assessed using [3H]-thymidine incorporation assay and soft agar colony formation assays. Subcutaneous bladder tumors in BALB/c nude mice were induced by inoculation of shppGalNAc T1-EJ cells and after inoculation diameters of tumors were measured every 5 days to determine gross tumor volumes. Results: ppGalNAc T1 mRNA in bladder cancer tissues was 11.2-fold higher than in normal bladder tissues. When ppGalNAc T1 expression in EJ cells was knocked down through transfection by pSUPER-shppGalNAc T1 vector, markedly reduced incorporation of [3H]-thymidine into DNA of EJ cells was observed at all time points compared with the empty vector transfected control cells. However, ppGalNAc T1 knockdown did not significantly inhibited cell migration (only 12.3%). Silenced ppGalNAc T1 expression significantly inhibited subcutaneous tumor growth compared with the control groups injected with empty vector transfected control cells. At the end of observation course (40 days), the inhibitory rate of cancerous growth for ppGalNAc T1 knockdown was 52.5%. Conclusion: ppGalNAc T1 might be a potential novel marker for human bladder cancer. Although ppGalNAc T1 knockdown caused no remarkable change in cell migration, silenced expression significantly inhibited proliferation and tumor growth of the bladder cancer EJ cell line.

        A Wireless Power Transfer System Based on a Hybrid Transmitting Coil for Targeted Therapy Microrobots in the Intestine

        A wireless power transfer system for targeted therapy microrobots has been received more attention recently. However, it usually fails to work due to weak coupling caused by misalignment in position and angle between the transmitting coil and the receiving coil. It will not be tolerated even if it occurs at certain angles. To address this issue, a three-dimensional hybrid transmitting coil, combining a fixed Helmholtz coil pair and an adjustable curved rectangular coil pair, is proposed. Based on the novel structure, the proposed hybrid transmitting coil could produce a three-dimensional magnetic field, realizing well coupled with the receiving coil embedded on the microrobot at any posture. To verify the efficiency and practical applicability of the transmitting coil proposed in this paper, we build the coil model via both analytical calculation and simulation analysis. Finally, the designed hybrid transmitting coil is also implemented in the wireless power transfer system with a receiving coil. The magnetic field distribution indicates that a large and uniform magnetic field could be obtained. The experimental results demonstrate that in the central zone of the transmitting coil, 300 × 200 mm, the magnetic field distribution is uniform, which can meet the requirements of the microrobot system working area. And the maximum output efficiency and power can be reached at 5% and 1001 mW, respectively. What's more, the proposed hybrid transmitting coil has solved the weak coupling problem due to misalignment in position and angle with which the electromagnetic energy decays quickly.

      • Prognostic Value of MAC30 Expression in Human Pure Squamous Cell Carcinomas of the Lung

        Recent evidence haas indicated that meningioma-associate protein (MAC30) exhibits different expression patterns in various tumors. However, little is known about the value of MAC30 in human squamous cell carcinoma of lung (SQCLC). The purpose of our study was to investigate the expression of MAC30 and to explore its clinical significance in SQCLC patients. A total of 156 Chinese patients diagnosed with SQCLC were selected for this study. The expression of MAC30 in all tissues was confirmed by immunohistochemical staining. Quantitative real-time PCR was performed to analyze MAC30 mRNA expression in 32 cases of SQCLC patients with corresponding non-tumor lung tissues. We observed enhanced mRNA expression of MAC30 in SQCLC as compared to control samples. Further, elevated MAC30 protein expression was strongly associated with poor tumor differentiation, TNM stage, and lymph node metastasis. In addition, we observed that patients with increased MAC30 expression demonstrated poor overall survival. Multivariate analysis explicated that increased MAC30 expression was a valuable independent predictable factor for poor tumor differentiation and short survival in SQCLC patients. Our present study suggests that MAC30 may serve as a biomarker for poor tumor differentiation and outcomes of patients with SQCLC.

        Bipolar resistive switching characteristics of atomic layer deposited Nb_2O_5 thin films for nonvolatile memory application

        Resistive switching behavior of Nb_2O_5 prepared by atomic layer deposition was investigated as a promising candidate for next generation nonvolatile memory technology. The crystalline structure of deposited film at 300 ℃ was found to be polycrystalline by X-ray diffraction (XRD) and the film was estimated to be oxygen deficient by X-ray photoelectron spectroscopy (XPS). The low resistance ON state and high resistance OFF state can be reversibly altered under low voltage about ±1 V. More than 1000reproducible switching cycles by DC voltage sweep were observed with a resistance ratio above 10,which was large enough to read out for memory applications. Moreover, the HRS and LRS of the devices are stable for more than 5 × 10^4 s and does not show any degradation during the test.

        Constructing mixed matrix membranes for CO2 separation based on light lanthanide fluoride nanosheets with mesoporous structure

        The incorporation of porous nano-fillers into mixed matrix membranes (MMMs) has a great impact onCO2 separation. In this study, a series of F-Ln (Ln is the Light Lanthanide, Ln = La, Ce, Pr, Nd) nanosheetswith mesoporous structure was fabricated as a filler in Pebax1657 matrix to improve the performance ofCO2 separation. F-Ln nanosheets are uniformly distributed in the membrane and the mesoporous structurecan construct multiple channels for CO2 fast transport in Pebax/F-Ln MMMs. The pore diameter ofnanosheet is in order: F-La > F-Ce > F-Pr > F-Nd, and the smaller pore diameter of nanosheets is beneficialto improve the CO2 separation performance. In humidified conditions, Pebax/F-Nd-6 MMMs show optimumseparation performance among MMMs, and the CO2 permeability is up to 1265 Barrer and CO2/CH4selectivity is 36.7, which are 2.3 times and 1.9 times higher than pure Pebax and approaches 2019 upperbound. Meanwhile, to demonstrate the potential universal applicability of F-Ln nanosheets in CO2 separation,XLPEO/F-Ce MMMs were fabricated to separate CO2/N2 and the performance exceeds the 2019upper bound. The efficient CO2 separation performance of 2D F-Ln nanosheets with mesoporous structurein membranes reveals the potential application of these nanosheets in industrial CO2 separation.

        Ultrafast liquid chromatographytandem mass spectrometry determination of donepezil in human plasma: application to a bioequivalence study

        A liquid chromatography equipped with tandem mass spectrometric method using multistage flow rates was developed for the determination of donepezil in human plasma to support a randomized, crossover bioequivalence (BE) study in which healthy volunteers each received a single oral dose of the reference and test formulations of 10 mg donepezil hydrochloride. This integrated liquid chromatography with tandem mass spectrometry (LC-MS/MS) system with electrospray ionization and a deuterium-labeled internal standard (IS) were employed for the positive multiple-reaction-monitoring (MRM) analyses. The baseline separation using a high-resolution monolithic column under gradient and flexible flowrate conditions between donepezil and multiple interfering peaks from the extracted quality control, calibration standard and study plasma samples following simple protein precipitation extraction procedures was accomplished within 1.5 minutes. The ultrafast monolithic column performance in terms of chromatographic separation efficiency, peak asymmetry and resolution and retention time reproducibility was found to be sustainable. The linear dynamic range was detected over a concentration range of 0.2–50 ng/mL. The intra- and inter-day assay accuracy and precision were within 15% for the analyte in individual biological fluids. A positive correlation coefficient (r) greater than 0.995 for donepezil concentrations in study plasma samplers measured by the proposed and the other validated LC-MS/MS methods in support of a bioequivalence study was observed.

        The effect of a parabolic potential on the properties of a strongly coupled polaron in an asymmetric Gaussian quantum well

        In this work, the effect of parabolic potential on the properties of strongly coupled polaron in an asymmetric Gaussian quantum well (QW) has been studied. Relations of the frequency vibration and the strongly coupled polaron’s ground-state energy in an asymmetric Gaussian QW for RbCl crystals at different confinement strengths along the x and the y directions, the asymmetric Gaussian QW barrier heights, and the Gaussian confinement potential widths and temperatures have been studied using the Lee–Low–Pines unitary transformation and the linear combination operator methods. Calculations show that the absolute value of ground-state energy increased with increasing confinement strengths along the x and the y directions and asymmetric Gaussian QW barrier height, and derease with increeasing Gaussian confinement potential width, and temperature. In addition, vibrational frequency was found to decrease with increasing Gaussian confinement potential width and to increase with increasing confinement strengths along the x and the y directions, asymmetric Gaussian QW barrier height, and temperature. The five parameters involved in this paper are important physical quantities for studying the properties of Gaussian QWs, and have important physical significance for the study of nanomaterials.
