유전자 재조합 사람 IFN-beta인 제네베타와 베네세린의 생물학적 활성의 비교

Background: Recombinant IFN-beta products have been approved for the treatment of several diseases. Recently, Inbionet, Co. has successfully developed rhIFN-beta product, Genebeta. This study was performed in a purpose of confirming the biological activity of Genebeta by comparison with another recombinant IFN-beta, Beneserin which was already approved. Methods: In vitro antiporliferative activity, regulation of MHC class I expression, antiviral activity, regulation of natural killer cell cytotoxicity were studied for functional comparison of these two rhIFN-beta products. Results: Proliferation assay data showed that two recombinant IFN-beta inhibited T cell and Daudi cell proliferation to a similar extent. When antiviral activity was compared, there was no significant difference between them. The treatment of these two rhIFN-beta also resulted in a similar level of upregulation of class I expression on the surface of A549 cells. Cytotoxicity experiment against tumor cells revealed that both of rhIFN-beta could enhance NK cell cytotoxicity to comparable level. Conclusion: All comparison data indicated that rhIFN-beta products, Genebeta and Beneserin have comparable potentials as a biological response modifier.

Thiamine-HCI, Thiamine Propyl Disulfide (TPD) and Thiamine Tetrahydrofur furyl Disulfide (TTFD) were studied to stability in artifcial gastric juice and artificial pancreatic juice. And in the in vitro isolated intestine of rabbit, absorption rate of this compounds were examined. The results were as follows. 1. The stability of this compounds in artificial gastric juice and artificial panereatic juice were stable. 2. In the in vitro isolated intestine of rabbit, absorption rate of TPD was increased than that of thiamine-HCI about 20% after 120 minutes. And absorption rate of TTFD was much the same in the case of thiamine-HCI.
상황버섯(Phellinus linteus)으로부터 Ergosterol의 분리

상황버섯을 80% MeOH 용액으로 추출하고, 추출물을 EtOAc, n-BuOH 및 물로 분배, 추출하였다. 이 중 EtOAc 분획을 silica gel 및 octadecylsilica gel(ODS) column chromatography로 정제하여 1종의 sterol 화합물을 분리하였다. 화합물의 화학구조는 NMR, MS 및 IR 등의 스펙트럼 데이터를 해석하여, ergosta-7,22E-diene-3β,5α,6β-triol(cerevisterol, 1)로 동정하였다. 이 화합물은 상황버섯에서 처음 분리 보고 되었다. Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and H_(2)O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to the isolation of one sterol. From the result of spectral data including NMR, MS and IR, the chemical structure of the sterol was determined as ergosta-7,22E-diene-3β,5α,6β-triol (cerevisterol, 1). Compound 1 was isolated from the Phellinus linteus for the first time in this study.

Objectives : To evaluate the toxic effect of methylmercuric chloride(MMC) and the protective effect of Sophorae Radix(SR) in cultured mouse myocardial cells, cytotoxicity was measured by MTT assay Methods : Cultured myocardial cells were incubated for 72 hours in the media containing 5~40 ㎛ concentrations of MMC. And also, the protective effect of SR was determined by sulforhodamine B(SRB) for protein sythesis and radioactive assay for DNA sythesis in these cultures. Results : Cell viability was significantly decreased in a dose-dependent manner after cultured myocardial cells were exposed to 20 ㎛ MMC for 72 hours. In the neuroprotective effect of SR on MMC-induced cytotoxicity, SR blocked the MMC-induced myotoxocity in these cultures. Conclusion : It suggests that MMC is toxic against cultured mouse myocardial cells and SR is effective in blocking the meurotoxicity induced by MMC.

Background: Proper education regarding inhaler usage and optimal management of chronic obstructive pulmonary disease (COPD) is essential for effectively treating patients with COPD. This study was conducted to evaluate the effects of a comprehensive education program including inhaler training and COPD management. Methods: We enlisted 127 patients with COPD on an outpatient basis at 43 private clinics in Korea. The patients were educated on inhaler usage and disease management for three visits across 2 weeks. Physicians and patients were administered a COPD assessment test (CAT) and questionnaires about the correct usage of inhalers and management of COPD before commencement of this program and after their third visit. Results: The outcomes of 127 COPD patients were analyzed. CAT scores ($19.6{\pm}12.5$ vs. $15.1{\pm}12.3$) improved significantly after this program (p<0.05). Patients with improved CAT scores of 4 points or more had a better understanding of COPD management and the correct technique for using inhalers than those who did not have improved CAT scores (p<0.05). Conclusion: A comprehensive education program including inhaler training and COPD management at a primary care setting improved CAT scores and led to patients' better understanding of COPD management.

The hippocampus makes new memories and is involved in mental cognition, and the hippocampal dentate gyrus (DG) is critical because neurogenesis, which occurs throughout life, occurs in the DG. We observed the differentiation of neuroblasts into mature neurons (granule cells) in the DG of C57BL/6 mice at various early postnatal (P) ages: P1, P7, P14, and P21 using doublecortin (DCX) immunohistochemistry (IHC) for neuroblasts and calbindin D-28k (CB) IHC for granule cells. DCX-positive cells decreased in the DG with age; however, CB? cells increased over time. At P1, DCX and CB double-labeled (DCX?CB?) cells were scattered throughout the DG. At P7, DCX?CB? cells (about 92% of CB+ cells) were seen only in the granule cell layer (GCL) of the dorsal blade. At P14, DCX?CB? cells (about 66% of CB? cells) were found in the lower half of the GCL of both blades. In contrast, at P21, about 18% of CB? cells were DCX?CB? cells, and they were mainly located only in the subgranular zone of the DG. These results suggest that the developmental pattern of DCX?CB? cells changes with time in the early postnatal stages.

<P>In the present study, we used immunohistochemistry and western blot analysis to examine changes in the levels and cellular localization of iron, heavy chain ferritin (ferritin-H), and transferrin in the gerbil hippocampal CA1 region from 30 minutes to 7 days following transient forebrain ischemia. Relative to sham controls, iron reactivity increased significantly in the stratum pyramidale and stratum oriens at 12 hours following ischemic insult, transiently decreased at 1–2 days and then increased once again within the CA1 region at 4–7 days after ischemia. One day after ischemia, ferritin-H immunoreactivity increased significantly in the stratum pyramidale and decreased at 2 days. At 4–7 days after ischemia, ferritin-H immunoreactivity in the glial components in the CA1 region was significantly increased. Transferrin immunoreactivity was increased significantly in the stratum pyramidale at 12 hours, peaked at 1 day, and then decreased significantly at 2 days after ischemia. Seven days after ischemia, Transferrin immunoreactivity in the glial cells of the stratum oriens and radiatum was significantly increased. Western blot analyses supported these results, demonstrating that compared to sham controls, ferritin H and transferrin protein levels in hippocampal homogenates significantly increased at 1 day after ischemia, peaked at 4 days and then decreased. These results suggest that iron overload-induced oxidative stress is most prominent at 12 hours after ischemia in the stratum pyramidale, suggesting that this time window may be the optimal period for therapeutic intervention to protect neurons from ischemia-induced death.</P>
