      • Roles of AKT1 and AKT2 in non-small cell lung cancer cell survival, growth, and migration.

        <P>Although AKT ? protein kinase B is constitutively active in nonsmall cell lung cancer (NSCLC) cells and is an attractive target for enhancing the cytotoxicity of therapeutic agents, the distinct roles of the AKT isoforms in NSCLC are largely unknown. In the present study, we investigated the roles of AKT1 and AKT2 in NSCLC cells using RNAi. The siRNA targeting of AKT1 or AKT2 effectively decreased protein levels of AKT1 and AKT2, respectively, in A549 and H460 cells. Cisplatin treatment of these cells increased apoptotic cell death compared with control. The siRNA-induced knockdown of AKT1 in H460 cells significantly decreased basal MEK? ERK1 ? 2 activity, resulting in nuclear factor-κB activation, whereas knockdown of AKT2 resulted in anti-apoptotic Bcl-2 family protein MCL-1 (MCL-1) cleavage, the collapse of mitochondrial membrane potential, cytochrome c release, and activation of the caspase cascade. Consequently, both siRNA treatments enhanced the chemosensitivity of H460 cells to cisplatin. However, neither AKT1 nor AKT2 siRNA treatment had any effect of p27 expression, and although both treatments tended to induced G2 ?M phase arrest, the effect was not statistically significant. Treatment with AKT1 siRNA markedly decreased colony formation growth and migration, but AKT2 siRNA had no significant effects on these parameters. These data suggest that AKT1 and AKT2 both contribute to cell survival, albeit via different mechanisms, and that the effects on cell growth and migration are predominantly regulated by AKT1. These findings may aid in refining targeted strategies for the inhibition of AKT isoforms towards the sensitization of NSCLC cells to therapeutic agents.</P>

      • Forecast Driven Simulation Model for Service Quality Improvement of the Emergency Department in the Moses H. Cone Memorial Hospital

        Patient satisfaction with the Emergency Department(ED) in a hospital is related to the length of stay, and especially to the amount of waiting time for medical treatments. ED over-crowding decreases quality and efficiency, therefore affecting hospitals` profitability. This paper presents a forecasting and simulation model for resource management of the ED at Moses H. Cone Memorial Hospital. A linear regression forecasting model is proposed to predict the number of ED patient arrivals, and then a simulation model is provided to estimate the length of stay of ED patients, system throughput, and the utilization of resources such as triage nurses, patient beds, registered nurses, and medical doctors. The near future load level of each resource is presented using the proposed models.

        In this work, we explore the feasibility of roll-pitch-yaw integrated autopilots for high angle-of-attack missiles. An investigation of the aerodynamic characteristics of a surface-to-air missile is presented, which reveals the strong effects of cross coupling between the longitudinal and lateral dynamics. Robust control techniques based on H∞ synthesis are employed to design roll-pitch-yaw integrated autopilots. The performance of the proposed roll-pitch-yaw integrated controller is tested in high-fidelity nonlinear five-degree-of-freedom simulations accounting for kinematic cross-coupling effects between the lateral and longitudinal channels. Against nonlinearity and cross-coupling effects of the missile dynamics, the integrated controller demonstrates superior performance when compared with the controller designed in a decoupled manner.

      • Development of Strip Casting Process for Fabrication of Wrought Mg Alloys

        <P>AZ31 and experimental ZMA611 alloys were strip cast into 2 mm thick strips. The as-cast AZ31 alloy strip consists of columnar dendrites. On the other hand, as-cast ZMA611 alloy strip shows equiaxed dendritic structure through the thickness of strip. Strip cast AZ31 alloy in H24 condition has equivalent yield and tensile strengths with similar ductility compared to commercial ingot cast AZ31-H24 alloy, indicating that strip casting is a viable process for the fabrication of Mg alloy strips. The ZMA611 alloy has a large volume fraction of fine dispersoid particles in the microstructure, resulting from the beneficial effect of strip casting on microstructural refinement. It has been shown that the ZMA611 alloy has superior tensile properties compared to commercial ingot cast AZ31-H24 alloy, suggesting the possibility of the development of new wrought Mg alloy sheets by strip casting.</P>

        레지오넬라 폐렴의 진단용 바이오마커의 발굴 : A/J 마우스 감염 모델에서 Legionella pneumophila의 독력 유전자들의 발현양상 분석

        Background: Legionella pneumophila is the causative agent of Legionnaires’ disease, a severe form of pneumonia. After L. pneumophila is inhaled through contaminated aerosols, it is phagocytized by alveolar macrophages, multiplies in a specialized phagosome approximately 10 h postinfection, and eventually leads to the death of host cells. Currently available diagnostic tests for Legionella pneumonia have some limitations. This study was conducted to find diagnostic biomarkers for Legionella pneumonia using virulence gene expression profiling in a murine experimental model. Materials and Methods: A/J mice were intranasally inoculated with L. pneumophila serogroup 1, and lungs were harvested 4, 8, 24, and 48 h postinfection. The strain grown in buffered yeast extract broth was used as reference samples. Cy-dye labeled cDNA samples were prepared with total RNA from lungs or broth culture, and hybridized on the oligo-microarray slide containing 2,895 genes of L. pneumophila serogroup 1. Virulence gene expression patterns were analyzed using a MIDAS software from TIGR (www.tigr.org). Results: Among a total of 332 virulence genes examined, 17 genes including sidA, lepB, the genes related to flagella assembly (fliR and fliP), LPS lipid A biosynthesis, and the enhanced entry protein EnhA were up-regulated at all four time points. We further confirmed by quantitative real-time reverse transcription PCR that the expression of fliP gene was highly expressed in lung tissue as well as in bronchoalveolar lavage fluids from the mouse infected with L. pneumophila serogroup 1. Conclusions: Through gene expression analysis of L. pneumophila in a mouse model, several candidate biomarkers for diagnosing Legionnaires’ disease could be identified.

      • Microstructure and Mechanical Properties of Twin-Roll Strip Cast Mg Alloys

        <P>Development of wrought Mg alloys, particularly in sheet form, is essential to support the growing interest for lightweight components in the automotive industry. However, development of Mg alloy sheets has been quite slow due to the complexity of sheet production originated from limited deformability of Mg. In this respect, twin-roll strip casting, a one-step processing of flat rolled products, can be an alternative for the production of Mg alloy sheets. In this study, AZ31 and experimental ZM series alloys are twin-roll strip cast into 2 mm thick sheets. The microstructure of the as-cast AZ31 alloy sheet consists of columnar zones near the roll side and equiaxed zones in the mid-thickness region. On the other hand, as-cast ZM series alloy sheets show equiaxed dendritic structure through the thickness of sheet. These alloys were subjected to various thermo-mechanical treatments and their tensile properties were evaluated.
