Inhibitory Effects of α-Pinene on Hepatoma Carcinoma Cell Proliferation

Background: Pine needle oil from crude extract of pine needles has anti-tumor effects, but the effective component is not known. Methods: In the present study, compounds from a steam distillation extract of pine needles were isolated and characterized. Alpha-pinene was identified as an active anti-proliferative compound on hepatoma carcinoma BEL-7402 cells using the MTT assay. Results: Further experiments showed that ${\alpha}$-pinene inhibited BEL-7402 cells by arresting cell growth in the G2/M phase of the cell cycle, downregulating Cdc25C mRNA and protein expression, and reducing cycle dependence on kinase 1(CDK1) activity. Conclusion: Taken together, these findings indicate that ${\alpha}$-pinene may be useful as a potential anti-tumor drug.
The Optimum Dietary Essential Amino Acid Pattern for Male Taiwan Country Chicks

The objective of this study was to estimate the optimum dietary essential amino acid pattern for male Taiwan country chicks. A series of experiments was conducted with chicks, 14 d of age, for 2 wks. A basal synthetic diet was established using a dose response test for all essential amino acids referring to the broiler requirements recommended by NRC (1994). Twelve chicks were sacrificed at the beginning to provide initial body nitrogen data, and every group of six birds received the basal diet or a diet with a deficiency in a single essential amino acid in twenty one treatments by intubation according to their daily metabolic body weight (MBW). Deposited body nitrogen was determined using comparative slaughtering. According to the daily intake from the limiting amino acid per unit of MBW and the body nitrogen accretion rates for every two deficient groups with the same limiting amino acid but at different levels, a corresponding straight line was computed for each essential amino acid to intersect with a horizontal line made by the body nitrogen accretion rate of the control group. The x coordinate of the intersection represented the daily requirement for growth plus maintenance based on MBW corresponding to the essential amino acid. The amino acid ratios can be considered as the optimum pattern of dietary essential amino acids. The results, expressed with respect to lysine = 100, were arginine 105, methionine 81, histidine 34, tryptophan 18, leucine 103, phenylalanine 135, isoleucine 69, threonine 65 and valine 79. This pattern could be utilized to compute the dietary requirements (g/kg feed) for all essential amino acids by multiplying by the requirement of a single essential amino acid cited from the literature.
Cloning of Phospholipase D from Grape Berry and Its Expression under Heat Acclimation

To investigate whether phospholipase D (PLD, EC 3.1.4.4) plays a role in adaptive response of post-harvest fruit to environment, a PLD gene was firstly cloned from grape berry (Vitis Vinifera L. cv. Chardonnay) using RT-PCR and 3'- and 5'-RACE. The deduced amino acid sequence (809 residues) showed 84.7% identity with that of PLD from Ricinus communis. The secondary structures of this protein showed the characteristic C2 domain and two active sites of a phospholipid-metabolizing enzyme. The PLD activity and its expression in response to heat acclimation were then assayed. The results indicated PLD was significantly activated at enzyme activity, as well as accumulation of PLD mRNA and synthesis of new PLD protein during the early of heat acclimation, primary suggesting that the grape berry PLD may be involved in the heat response in post-harvest grape berry. This work offers an important basis for further investigating the mechanism of post-harvest fruit adaptation to environmental stresses.
A Substation-Oriented Approach to Optimal Phasor Measurement Units Placement

State Estimation (SE) is the basis of a variety of advanced applications used in most modern power systems. An SE problem formed with enough phasor measurement units (PMUs) data is simply a linear weighted least squares problem requiring no iterations. Thus, designing a minimum-cost placement of PMUs that guarantees observability of a power system becomes a worthy challenge. This paper proposes an equivalent integer linear programming method for substation-oriented optimal PMU placement (SOOPP). The proposed method uses an exhaustive search to determine a globally optimal solution representing the best PMU placement for that particular power system. To obtain a more comprehensive model, contingencies and the limitation of the number of PMU measurement channels are considered and embodied in the model as changes to the original constraints and as additional constraints. The proposed method is examined for applicability using the IEEE 14-bus, 118-bus and 300-bus test systems. The comparison between SOOPP results and results obtained by other methods reveals the excellence of SOOPP. Furthermore, practical large-scale power systems are also successfully analyzed using SOOPP.
Derivation of hypermethylated pluripotent embryonic stem cells with high potency

<P>Naive hypomethylated embryonic pluripotent stem cells (ESCs) are developmentally closest to the preimplantation epiblast of blastocysts, with the potential to contribute to all embryonic tissues and the germline, excepting the extra-embryonic tissues in chimeric embryos. By contrast, epiblast stem cells (EpiSCs) resembling postimplantation epiblast are relatively more methylated and show a limited potential for chimerism. Here, for the first time, we reveal advanced pluripotent stem cells (ASCs), which are developmentally beyond the pluripotent cells in the inner cell mass but with higher potency than EpiSCs. Accordingly, a single ASC contributes very efficiently to the fetus, germline, yolk sac and the placental labyrinth in chimeras. Since they are developmentally more advanced, ASCs do not contribute to the trophoblast. ASCs were derived from blastocysts in two steps in a chemically defined medium supplemented with Activin A and basic fibroblast growth factor, followed by culturing in ABCL medium containing ActA, BMP4, CHIR99021 and leukemia inhibitory factor. Notably, ASCs exhibit a distinct transcriptome with the expression of both naive pluripotency genes, as well as mesodermal somatic genes; <I>Eomes, Eras, Tdgf1, Evx1, hand1, Wnt5a</I> and distinct repetitive elements. Conversion of established ESCs to ASCs is also achievable. Importantly, ASCs exhibit a stable hypermethylated epigenome and mostly intact imprints as compared to the hypomethylated inner cell mass of blastocysts and naive ESCs. Properties of ASCs suggest that they represent cells at an intermediate cellular state between the naive and primed states of pluripotency.</P>

A new synthetic route to N-Cbz-cis-(3R,4R)-3-methylamino-4-methylpiperidine, key intermediate for CP- 690,550, was disclosed with L-malic acid as the chiral pool starting material. The title compound was obtained in 16 steps with a total yield of 26% and more than 98% ee.
Mesh Generation Based on Virtual Geometry

Finite element mesh generation algorithm often requires the ability to abstract and modify the topology of a boundary representation model. A method based on virtual geometry for mesh generation is proposed to modify the model topology without changing the geometry of the model. By this method, a tool is given to adjust the model topology for optimal meshing, while leaving the geometry or shape of the model, and the accuracy of the resulting mesh in representing the model, unchanged. Our practical examples show that the proposed method is effective and reliable for mesh generation in molding analysis field.
