        Extracellular Vesicles-Encapsulated miR-153-3p Potentiate the Survival and Invasion of Lung Adenocarcinoma

        Extracellular vesicles (EVs) play an essential role in the communication between cells and the tumor micro­environment. However, the effect of tumor-derived EVs on the growth and metastasis of lung adenocarcinoma (LUAD) remains to be explored. This study aimed to elucidate the role of miR-153-3p-EVs in the invasion and migration capabilities of LUAD cells and explore its mechanism through in vivo and in vitro experiments. We found that miR-153-3p was specifically and highly expressed in LUAD and its secreted EVs. Furthermore, the expression of BANCR was negatively regulated by miR-153-3p and identified as a target gene of miR-153-3p using luciferase reporter assays. Through further investigation, we found that the downregulation of BANCR activates the PI3K/AKT pathway and accelerates the process of epithelial-mesenchymal transition (EMT), which ultimately leads to the aggravation of LUAD. The orthotopic xenograft mouse model was established to illustrate the effect of miR-153-3p-EVs on LUAD. Animal studies showed that miR-153-3p-EVs accelerated tumor growth in mice. Besides, we found that miR-153-3p-EVs could damage the respiratory ability of mice and produce a mass of inflammatory cells around the lung tissue of mice. Nevertheless, antagomir-153-3p treatment could inhibit the deterioration of respiratory function and inhibit the growth of lung tumors in mice. In conclusion, our study reveals the potential molecular mechanism of miR-153-3p-EVs in the development of LUAD and provides a potential strategy for the treatment of LUAD.

        Sensitive and Extraction-Free Detection of Methicillin-Resistant Staphylococcus aureus through Ag + Aptamer-Based Color Reaction

        Refractory infections, such as hospital-acquired pneumonia, can be better diagnosed with the assistance of precise methicillin-resistant Staphylococcus aureus (MRSA) testing. However, traditional methods necessitate high-tech tools, rigorous temperature cycling, and the extraction of genetic material from MRSA cells. Herein, we propose a sensitive, specific, and extraction-free strategy for MRSA detection by integrating allosteric probe-based target recognition and exonuclease-III (ExoIII)-enhanced color reaction. The penicillin-binding protein 2a (PBP2a) aptamer in the allosteric probe binds with MRSA to convert protein signals to nucleic acid signals. This is followed by the DNA polymerase-assisted target recycle and the production of numerous single-strand DNA (ssDNA) chains which bind with silver ion (Ag+ ) aptamer to form a blunt terminus that can be identified by Exo-III. As a result, the Ag+ aptamer pre-coupled to magnetic nanoparticles is digested. After magnetic separation, the Ag+ in liquid supernatant catalyzes 3,3’,5,5’-tetramethylbenzidine (TMB) for a color reaction. In addition, a concentration of 54 cfu/mL is predicted to be the lowest detectable value. Based on this, our assay has a wide linear detection range, covering 5 orders of magnitude and demonstrating a high specificity, which allows it to accurately distinguish the target MRSA from other microorganisms.

        Designing an atmosphere controlling hollow fiber membrane system for mango preservation

        We devised an atmosphere controlling facility to gain a longer life span for mango. A membrane module made of polyethersulfone/polydimethylsiloxane (PES/PDMS) composite membrane was applied to selectively permeate CO2 from the gas mixture of the fruit container. To design the membrane separation system, two models were introduced into our mathematical simulations: (1) an equilibrium model giving the optimal membrane area, the compositions of CO2 and O2 in the fruit container, feed flow rate and pressures on both the feed and permeate sides of the module, and (2) a dynamic model simulating the change in the gas composition of the fruit container with time. The pressure build-up in the bore side of the hollow fiber was also discussed using the Hagen-Poiseuille equation. The best membrane module configuration was obtained based on the pressure build-up analysis. That was (1) the vacuum pressure should be set at 0.1 bar, (2) the hollow fiber inner diameter should be 0.45mm, and (3) the vacuum should be applied at both ends of the hollow fiber bore sides.

        Expression and Preliminary Functional Profiling of the let-7 Family during Porcine Ovary Follicle Atresia

        Most follicles in the mammalian ovary undergo atresia. Granulosa cell apoptosis is a hallmark of follicle atresia. Our previous study using a microRNA (miRNA) microarray showed that the let-7 microRNA family was differentially expressed during follicular atresia. However, whether the let-7 miRNA family members are related to porcine (Sus scrofa) ovary follicular apoptosis is unclear. In the current study, real-time quantitative polymerase chain reaction showed that the expression levels of let-7 family members in follicles and granulosa cells were similar to our microarray data, in which miRNAs let-7a, let-7b, let-7c, and let-7i were significantly decreased in early atretic and progressively atretic porcine ovary follicles compared with healthy follicles, while let-7g was highly expressed during follicle atresia. Furthermore, flow cytometric analysis and Hoechst33342 staining demonstrated that let-7g increased the apoptotic rate of cultured granulosa cells. In addition, let-7 target genes were predicted and annotated by TargetScan, PicTar, gene ontology and Kyoto encyclopedia of genes and genomes pathways. Our data provide new insight into the association between the let-7 miRNA family in granulosa cell programmed death.

        EFTG: Efficient and Flexible Top-K Geo-textual Publish/Subscribe

        With the popularity of mobile networks and smartphones, geo-textual publish/subscribe messaging has attracted wide attention. Different from the traditional publish/subscribe format, geo-textual data is published and subscribed in the form of dynamic data flow in the mobile network. The difference creates more requirements for efficiency and flexibility. However, most of the existing Top-k geo-textual publish/subscribe schemes have the following deficiencies: (1) All publications have to be scored for each subscription, which is not efficient enough. (2) A user should take time to set a threshold for each subscription, which is not flexible enough. Therefore, we propose an efficient and flexible Top-k geo-textual publish/subscribe scheme. First, our scheme groups publish and subscribe based on text classification. Thus, only a few parts of related publications should be scored for each subscription, which significantly enhances efficiency. Second, our scheme proposes an adaptive publish/subscribe matching algorithm. The algorithm does not require the user to set a threshold. It can adaptively return Top-k results to the user for each subscription, which significantly enhances flexibility. Finally, theoretical analysis and experimental evaluation verify the efficiency and effectiveness of our scheme.

        Synchronization of Nonlinear Coupled Delayed Networks via Intermittent Pinning Control: An Average Technique

        This paper is concerned with the aperiodically intermittent pinning control (AIPC) for the synchronization of nonlinear coupled networks, where both the internal time delay and the coupling time delay are considered. We consider the aperiodicity of AIPC from the average sense. Then by extending the Halanay’s inequality to the case with two kinds different delays, we obtain two kinds of synchronization criteria: one is the upper bound of time delay must be less than the average control width but can be larger than certain control widths; the other is that the upper bound of time delay has no relationship with the average control width. We show that the two criteria lead to complementary results. The effectiveness of our results is confirmed by numerical simulations.

      • Development of Multidirectional Nonlinear Numerical Wave Tank by Naoe-FOAM-SJTU Solver

        A three-dimensional multidirectional nonlinear numerical wave tank (NWT) based on the Navier-Stokes equations and the Finite Volume Method (FVM) is developed by using the two-phase hydrodynamic flow solver naoe-FOAM-SJTU based on the open source toolbox OpenFOAM. The free surface is capturing with the Volume Of Fluids (VOF). The directional wave including Stokes wave, solitary wave and nonlinear wave are simulated and verified. The multi-directional waves are also simulated with particular wave spectral such as JONSWAP and wave directional spreading function. The obtained numerical results show the capability of the solver to generate different type of multidirectional nonlinear waves accurately. Meanwhile, it implies that the presented NWT can easily extend to model the wave-structures interactions, which will be great help to the offshore structures design.

        Composition and abundance of microbiota in the pharynx in patients with laryngeal carcinoma and vocal cord polyps

        The pharynx is an important site of microbiota colonization, but the bacterial populations at this site have been relatively unexplored by culture-independent approaches. The aim of this study was to characterize the microbiota structure of the pharynx. Pyrosequencing of 16S rRNA gene libraries was used to characterize the pharyngeal microbiota using swab samples from 68 subjects with laryngeal cancer and 28 subjects with vocal cord polyps. Overall, the major phylum was Firmicutes, with Streptococcus as the predominant genus in the pharyngeal communities. Nine core operational taxonomic units detected from Streptococcus, Fusobacterium, Prevotella, Granulicatella, and Veillonella accounted for 21.3% of the total sequences detected. However, there was no difference in bacterial communities in the pharynx from patients with laryngeal cancer and vocal cord polyps. The relative abundance of Firmicutes was inversely correlated with Fusobacteria, Proteobacteria, Actinobacteria, and Bacteroidetes. The correlation was evident at the genus level, and the relative abundance of Streptococcus was inversely associated with Fusobacterium, Leptotrichia, Neisseria, Actinomyces, and Prevotella. This study presented a profile for the overall structure of the microbiota in pharyngeal swab samples. Inverse correlations were found between Streptococcus and other bacterial communities, suggesting that potential antagonism may exist among pharyngeal microbiota.
