A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation. Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 ㎎/㎏. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation. Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 ㎎/㎏. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 ㎍/㎖. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Marasmius maximus, Hirschioporus abietinus, and Russula sanguinea and the the ether extracts of M. maximus and H. abietinus showed antibiotic activities against Sarcina lutea; and the methanol and ethanol extract of M. maximus showed antibiotic activity against Staphylococcus aureus. None of the extracts, however, showed any activity against gram negative bacteria, Serratia marcescens and Proteus vulgaris.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon and Keyryoung Mountain Area in Chungcheungnamdo, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Inocybe trechispora, Lactarius velumis, and Laccaria laccata and the ether extracts of I.trechispora showed antibiotic activities against Sarcina lutea; and the methanol extracts of I. trechispora and L. velumis and physiological saline extract of Coriolus versicolor showed antibiotic activities against Staphylococcus aureus; and the methanol extract of L. velumis showed antibiotic activity against Escherichia coli. None of the extracts, however, showed any activity against Serratia marcescens and Proteusvulgaris.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetes, which were collected from several locations of Korea, were subjected to a screening test for antibiotic activity by disc-plate method using Sarcina lutea, Bacillus subtilis, Escherichia coli, and Serratia marcescens as target organisms. As the result, the methanol extract of five basidiomycetes-Amanita rebescens, Amanita solitaria, Lactarius vellereus, Latarius subzonarius and Scleroderma verrucosum-were found te have antibiotic activity. Among them, the methanol extracts of A. rubescens and L. vellereus exerted strong activity against both of the gram positive and negative bacteria. The ether extracts of A. rubescens and S. verrucosum also showed activity against S. lutea. The physiological saline extract, however, showed no activity at all.

To develop new potent antitumor substancces with low toxicity from natural products, especially from higher fungi of Korea, the carpophores of some wild basidiomycetes were collected and the antitimor test for their polysaccharides was done. The dried carpophores were extracted with hot water, and from the extracts, crude polysaccharides were obtained by alcohol precipitation. The alcohol precipitated crude polysaccharides were partially purified by dialysis and then used as the samples for antitumor tests. Among tested, the polysaccharide of Laccaria laccata, Trametes sanguineus, Armlliariella mellea, Clitocybe in fundibuliformis, and Xeromphalina campanella respectively showed the inhibition ratio of 75%, 72%, 94% 55%, and 47% when administered i. p. once dailly for ten days at the dose level of 20mg/kg/day into the mice implanted with $10^{6}$ cells of sarcoma 180/mouse. However, those of Craterellus cornucopiodes, Daedaleopsis confrogosa, and Coriolus sp. showed almost no activities.

The carpophores of three Korean mushrooms, Coriolus versicolor, Pleurotus ostreatus and Lentinus edodes were respectively extracted with hot water and the extracts were dialyzed through Visking tube. They were found to exert an antitumor activity against sarcoma-180 implanted in mice. Especially, the inhibition ratio of the extract of Coriolus versicolor (100 mg/ Kg.i.p) was almost 100% . But all the extracts did not affect the growth of leukemia L517Y cells in vitro. Therefore these facts indicate that the extracts appear to stimulate cell-mediated immunity.
Antitumor Constituents of Polyporus giganteus

To investigate antitumor constituents of higher fungi, the carpophores of Polyporus giganteus Pers. ex. Fr. (81 g, dry weight) which were collected in Indiana, U. S. A. were examined for antitumor activity. Two protein-bound-polysaccharide fractions (I and II) were prepared from the hot water extract and one fraction (III) from the 0.1 N NaOH extract of the carpophores. The antitumor effect of each fraction was tested against sarcoma 180 implanted subcutaneously in female ICR mice. Of three fractions, Fraction II showed 85.2% inhibition ratio at the dose of 20 mg/kg/day for 10 days and was named gigantan. Gigantan was found to contain 59% polysaccharide and 27% protein. Its polysaccharide moity was a heteroglycan that consisted of mainly glucose (89.3%), galactose (7.7%), minaose (2.0%) and fructose (1.0%).

Of 450 strains isolated from the soil microbes collected in various locations in Korea, a strain had a strong inhibitory activity against bacterial α-amylase and was named strain DMC-72 of the genus Streptomyces. The amylase inhibitory metabolite produced by this strain was purified by means of acetone precipitation, adsorption on Amberlite IRC-50 and SP-Sephadex C-25. The inhibitor was found to be a derivative of oligosaccharides by spectral and chemical data. The inhibitor was stable at the pH range of 1∼13 and at 100℃ for half an hour, also inhibited other amylases such as salivary α-amylase, pancreatic α-amylase, fungal α-amylase and glucoamylase. However, it showed no inhibitory activity against α-glucosidase, β-glucosidase, dextranase, and β-amylase. The kinetic studies of the inhibitor showed that its inhibitory effects on starch hydrolysis by α-amylase were noncompetitive.
C-2. Production of Monoclonal Antibodies Reactive with Human T - Cell Leukemia

To develop hybridomas secreting monoclonal antibodies to be used as unlimited sources of reagents indispensable for the diagnosis and treatment of leukemic malignancy, a monoclonal antibody was generated to human pre-T leukemia cells (Jurkat). Hybridomas were produced against Jurkat cell line by fusing spleen cells from hyperimmunzed mice with murine plasmacytoma cells (P3×63Ag 8. V653). One monoclonal antibody derived from this fusion, designated DMJ-2 was reactive with T-cell lines(Jurkat, Mol t4 and RPMI 8402) and normal peripheral Erosette forming T cells, but unreactive with B-cell lines (Daudi, Nalm-6) and non-T, non-B cell line (K562). Conclusively DMJ-2 was reactive with mature and immature T-lineage lymphoid cells.

To find antitumor components in the hot water extract of the cultured mycelia of Ganoderma lucidum, protein-bound polysaccharides were purified and fractionated (Fr. I-V) by DEAE-cellulose ion exchange column chromatography and Sepbarose CL-4B gel filtration. When a dose of 20 mg/kg/day of each was, i.p., injected into ICR mice, the inhibition ratios against the solid form of sarcoma 180 were $64.2{\sim}75.8%$. The antitumor component was examined for immunological activity. It increased the amount of superoxide anion released by induced macrophages in peritoneal cavity to 1.8 times and the count of hemolytic plaque-forming cells (PFC) was increased to 4.4 times as compared with those of the control group. It contained 68.6% polysaccharide which consisted of mannose, glucose, galactose, fucose and xylose and 5.1% protein consisting of 17 amino acids. The contents of hexosamine were 0.78%. The molecular weight of Fr. V that showed the highest antitumor activity was $5.8{\times}10^4$ dalton by Sepharose CL-4B gel filtration. It was named lucidan.
