상심자(Morus alba)의 운동능력 향상과 스트레스 개선효과

상심자 추출물이 운동에 의한 체내 monoamine oxidase (MAO) 활성 변화에 미치는 영향을 연구하여. 상심자 추출물을 경구투여(0.3g/kg body weight) 한 흰쥐의 뇌와 간에서 MAO-A와 MAO-B의 활성에 중요한 영향을 미치는 사실을 확인하였다. 각 효소활성은 serotonin과 benzylamine을 기질로 이용하여 측정하였다. 운동 전 후 운동의 유형에 따라 효소활성의 변화경향이 서로 다른 경향을 나타내었다. 뇌에서 측정한 MAO-A 활성은 운동에 의해 효소활성이 현저히 감소하였으며 반면, 간에서 측정한 MAO-B의 활성은 운동이 끝나고 60분이 경과할 때까지 증가된 상태를 유지하고 있었다. 운동 시 체내 변화의 지표효소인 혈중 LDH의 활성 변화와 혈중 lactate의 농도변화를 함께 관찰함으로서 MAO 활성과의 상관관계를 비교하였다. 상심자 추출물을 경구투여 하고 운동을 한 동물의 MAO-A 활성은 증가하였고 MAO-B, LDH 활성과 lactate level은 감소하는 것을 확인하였다. 결과적으로 모은 지표들이 운동 전의 정상상태로 회복되는 것으로 확인되었다. 이 연구의 결과들로부터 상심자 추출물이 운동 전후의 MAO 활성을 조절함으로써 운동능력을 향상시키고 피로를 회복하는 효능을 갖는 것으로 추정되며 이러한 기능성을 갖는 건강기능식품의 소재로 활용이 가능할 것으로 생각한다. Effects of Morus alba fruit extracts on monoamine oxidase (MAO) activity were examined in rats during and after physical exercise. Oral administration of M. alba extract (0.3 g/kg body weight) significantly increased brain MAO-A activity but decreased liver MAO-B activity when they were measured using serotonin and benzylamine as substrates. Type of physical exercises had significant effect on MAO activity. Brain MAO-A activity markedly decreased with physical activity-related stress compared to normal group, whereas Liver MAO-B activity increased up to 60 min after exercise. Lactate dehydrogenase (LDH) activity and lactate concentration in blood, clinical indices of physical exercise activities, were also determined for correlation to MAO activities. MAO-A activity of rats subjected to oral administration of M. alba extract and physical exercise increased whereas MAO-B and LDH activities, and lactate level decreased, All indices eventually recovered normal levels, These results suggest M. alba may increase capability of physical activities by modulating MAO activities during exercise.
데치기(Blanching)로 조리된 22가지 채소류의 베타카로틴 함량의 변화와 영양소 보존율(True Retention)

본 연구는 채소류 22종을 대상으로 데치기 후의 베타카로틴 함량의 변화를 조사하였다. 또한, 조리 중 발생하는 시료의 중량 변화와 베타카로틴 함량 변화를 고려하여 영양소 보존율(true retention, TR)을 나타내었다. 실험대상 채소류 중에서 시금치를 예로 들면, 데친 후 품종이나 재배방식에 따라 베타카로틴 함량이 8.78에서 24.65%까지 증가하였다. 따라서 데친 후 베타카로틴 함량의 증가 때문에 데치기 전보다 높은 인체 이용률을 기대할 수 있다. 이를 데치기 과정 중 변화된 시금치 중량을 함께 고려하여 영양소 보존율로 나타내었을 때 시설 재배된 일반 품종의 시금치는 100.52%를 나타냈지만 다른 시금치(포항초, 섬초, 노지 재배된 일반품종)는 80.13~86.69%를 나타내었다. 다른 채소들의 경우 %TR이 100% 이상인 경우는 아욱(109.24%), 고사리(160.59%), 양배추(164.46%), 냉이(153.80%), 미나리(100.02%) 그리고 도라지(123.02%)에서 나타났는데 이는 데치기 후에 베타카로틴 함량이 증가하였거나 중량이 상대적으로 크게 감소하였기 때문이다. 이들 중 고사리와 양배추의 경우 중량의 감소보다는 베타카로틴 함량의 증가가 주된 요인이었고, 냉이의 경우 중량이 상대적으로 크게 감소하였지만 베타카로틴의 함량이 크게 증가하였기 때문이었다. 나머지 다른 시료들의 TR은 59.35%(곰취)에서 96.42%(유채 어린 잎)의 범위로 나타났다. In the present study, reversed-phase HPLC was utilized to quantify the β-carotene content of 22 kinds of raw and blanched vegetables consumed in Korea. In addition, true retention (TR) of β-carotene in samples was obtained. For quantification of β-carotene, external standard curve was obtained with limit of detection and limit of quantitation. The β-carotene contents in 22 raw vegetables ranged from 6.29 (bellflower root) to 7,050.73 μg/100 g (spinach, field culture). After blanching, β-carotene contents of 13 vegetables increased up to 103.05% while nine vegetables resulted in reduced content, ranging from -2.17 to -29.16%. However, even though increased β-carotene content was observed after blanching, TR of some vegetables was lower than 100% due to their weight reduction. The highest TR of β-carotene was found from blanched cabbage (164.46%) while the lowest TR was found from Turcz (Gomchwi) at 59.35%. TR is an effective method to evaluate retention of nutrients in cooked foods, considering changes of nutrient content and weight.
한약재의 절단, 수치, 전탕법에 관한 연구II : 숙지황

To improve the quality control of frequently used oriental medicinal plants, the storage, cutting, processing and decokting methods of Rehmannia glutinosa Libosch. var. purpurea Makino(熟地黃) were examined. The contents of 5-hydroxymethyl-2- furaldehyde(5-HMF) of Rehmannia glutinosa Libosch. var. purpurea Makino were analysed by HPLC at various conditions as the standard component. Raw materials were classified into 3 groups with their specific gravity(天 地 人黃) and determined the ding weights and the contents of water, the solid component of Ji-Hwang(地黃) was the most. Rehmannia glutinosa Libosch. var purpurea Makino was steamed and dried nine times and analysed the content of 5-HMF on every time. In the case of Ji-Hwang, the standard component of the ninth sample was produced the most. But which of the tenth samples of Chun-Hwang and Yin-Hwang(天黃 人黃) were the most. The best decokting conditions which were determined by the yield of 5-HMF were as follows : non cutting, soaking time was 0 minutes, decokting time was 75 minutes after boiling. The contents of sugar on each of samples were determined. The contents of standard component increased by steaming number and the contents of sugar decreased.
한성 및 열성한약재가 모노아민 산화효소의 활성에 미치는 영향

To explain the theory of KIMI which is the theory of therapeutics in oriental medicine, monoamine oxidase(MAO) activities were measured in the brain and liver of mice which were orally administered oriental medicinal herbs which were classified into cold and hot drugs. Rheum palmatum, Anemarrhena asphodeloides, Gardenia jasminoides, Scutellaria baicalensis and Coptis japonica were considered as the cold drugs and Zingiber officinale, Aconitum carmichaeli, Asiasarum sieboldi, Evodia officinalis and Cinnamomum cassia were included in the hot drugs. The effects of cold and hot drugs on in vitro enzyme activities were measured and compared with the in vivo effects. Serotonin is important neurotransmetter involved in the control of body temperature. The MAO plays a central role in the metabolism of many neurotransmetter monoamines including serotonin. MAO is a flavoprotein found exclusively in the mitochondrial outer membrane, occuring in the MAO-A and MAO-B subtypes. MAO-A deaminates serotonin and noradrenaline, whereas MAO-B prefers phenylethylamine and benzylamine as substrates. Coptis japonica and Aconitum carmichaeli elevated the in vivo MAO activities and especialy, in vivo MAO-B activities were significantly increased. In vitro MAO-A activities were increased by hot drugs, whereas the in vitro MAO-B activities were inhibited. Cold drugs inhibited both enzyme activities in vitro.

An antioxidant activity of Gardenia Fruit (Gardenia jasminoides Ellis) which has been used for food coloring was studied. The antioxidant activity was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA). Both water and methanol extracts of Gardenia Fruit showed the antioxidant activity. On solvent fractionation, the antioxidant activity was removed into the ethyl acetate and butanol soluble fractions. And the final water soluble fraction also showed the antioxidant activity in the low concentration, but it promoted the lipid peroxidation in the high concentration. Two compounds (I and II) having the antioxidant activity were isolated from the butanol fraction, and compound I also occurred in the ethyl acetate fraction. The antioxidant activity of compound II was more potent than that of I. By analyzing data for UV, IR and $^1H-NMR$, compounds I and II were identified as geniposide and crocin, respectively.
