        자외선 B를 조사한 각질형성세포의 조건배양액내 Endothelin - 1 이 멜라닌세포의 증식과 기능에 미치는 영향

        Background: The regulatory rnechanisrns of melanoc:ytes underlying ultraviolet(UV) melanogenesis have been an interest tu many investigators. They have shown that several materials produced and secreted frorn norm il human keratinocytes play roles as mitogens of human melanocytes, and demonstrated that UVB exposure stirnulated highly the paracrine linkage of endothelin(ET) be tween keratinocytes and melanocytes. It suggest that ET is one of keratinocytes-derived intrinsic mit.ogens in UVB induced hyperpigmentation. Objective : To evaluate whether ET secreted from keratinocytes under t.he UV irradiation works as paracrine effects such as melanocyte pro)iferation and function, the present, study was under taken. Methpds : Primary kevatinocytes and rnelanocytes cultures from neonatal foreskins were grown in complete MCDB 154 medium. Cultured human keratinocytes were irradiated with UVB 50mJ/ cm2. Twenty fours hour later, conditioned medium of keratinocytes was added into the growth medium of melanocytes of concentration in 10%, 20%, and 30%, respectively. Four days later, in order to detect of melanocytes proliferat.ion and function, number of melanocytes, melanin granules and tyrosinase activity v ere measured. Results : J. The number of me anocytes were higher increase in groups of incubation with conditioned medium of irradiated keratinocytes than that of incubation with conditioned mediurn of irradiated keratinocytes and treatment of anti-ET-1(5ug/ml)(p>0.05). 2. The melanin contents were significant.ly higher increase in groups of incubation with condi t.ioned medium(20%, 30%) of irradiated keratinocyt.es than that of incubation with conditioned medium of irradiated keiatinocytes and treatment of anti-ET-1(5ug/ml)(p<0.05). 3. The tyrosinase acti ity of melanocyte incubated with 30% COllcentration of conditioned medium from cultured keratinocyte irradiated with UVB was significantly higher increase than that of melanocyte incubated with 30% concentration of conditioned mediurn from cultured keratinocytes irradiated with UVB and treated with anti-ET-1(p<0.05). Conclusion : This stud provided an important confirmation of the proposal that ET-1 is intrin sic factor for proliferation and differentiation of human melanocytes. These findings suggest that keratinocyte derived ET-1 make a considerable effect on human melanocyte proliferation and function in UV melanog nesis. (Korean J Dermatol 1997;35(6): 1184-1192)

        전경골 점액수종

        Pretibial myxedema, which consists of localized cutaneous accumulations of acid mucopolysaccharides, oceurs in a few patients with hyperthyroidi';m after its correction by surgery or radioactive iodine therapy. We report a case of pretibial myxedema with exophthalmos in a 36 year old male. He has suffered from hyperthyroidism with bilateral pretibial myxedema for a period of 4 years. He was treated with a subtotal thyroidectomy and excision of both pletibia lesions 1 year ago. The lesions recurred on t,he left pretibial area, as a 11 X 16cm sized, non pitting, non tender, swollen, brownish plaque. The histopathologic findings showed epidermal hyperkeratosis and a considerable arnount of mucin in the mid-dermis. We treat.ed him with intralesional injection of triamcinolone acetonide with marked improvement. (Kor J Dermetol 1992;30(2): 239-243)

        유전성 망막색소변성 한방 치험 1례

        대전대학교 부속 한방병원 안이비인후피부과에서 유전성 망막색소변성 진단받은 63세 남자환자에 대학 기혈량휴(氣血兩虧), 간신부족(肝腎不足)로 변증하여 보간신치료법(補肝腎治療法)으로 9개월간의 팔물탕가미방(八物湯加味方)과 안구주의 전침술을 시행하여 임상 증상 호전을 경험한 예가 있어 문헌 고찰과 함께 보고하는 바이다. Retinitis pigmentosa(RP) is characterized by night-blindness, contraction of the visual field, distinct hereditary nature, wide-spread pigmentary in the midperiphery. We have experienced a case of retinitis pigmentosa treated by oriental medicine (Herbal medicaiton, Acupuncture & Electroacupuncture therapy). In the point of Differentiation of syndrome, this subject was diagnosed deficiency of both Gi & blood and Eum deficiency of Liver and Kidney. So we was administrated with Palmultang-Gamibang. After treatment of orient medicine, symptoms of patient were improved.

        한련초 에탄올 추출물의 멜라닌 합성 경로에 대한 연구

        Objectives : This Study was conducted to investigate the melanin synthesis effect of Eclipta prostrata and to determine the relationship between melanin synthesis effect of Eclipta prostrata and cAMP/PKA melanin synthesis pathway. Methods : We measured melanin contents, tyrosinase activity, expression of TRPs, p-CREB in B16F10 cells cultured with Eclipta prostrata ethanol extracts(EEP). And after treatment with H89 and dibutyryl cAMP, which inhibit or promote the activation of PKA, we observed changes in melanin synthesis and tyrosinase activity stimulated by EEP. Results : EEP increased melanin synthesis by promoting the expression of tyrosinase and TRP-1. It also promoted expression of p-CREB. H89 suppresses melanogenic effect and expression of tyrosinase, TRP-1 in B16F10 stimulated by EEP. Dibutyryl cAMP promotes melanogenic effect of EEP. Conclusions : The results of this study suggest that melanin synthesis effect of EEP is related to induction of tyrosinase and TRP-1 expression through the cAMP/PKA pathway.

        온폐탕(溫肺湯)이 알레르기 비염 유발 흰쥐 모델에 미치는 영향

        Objectives : Allergic rhinitis is an inflammation or irritation of the mucous membranes that in the nose. Common symptoms include sneezing; a stuffy or runny nose; itch eyes, nose and throat; and watery eyes. We aimed to determine therapeutic effect of Onpye-tang by observing changes in blood cells and the nasal mucosal tissue. Methods : Twenty-four Sprague-Dawley rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitoneal and intranasal sensitization with 0.l% Ovalbumin solution. Then Onpye-tang was orally administered only to the sample group for 28days, while the rats in the control group was given normal saline. We observed changes in the number of RBC and WBC; changes of neutrophils, lymphocyte, monocyte, and eosinophil proportions; total IgE level and AST & ALT change; also, changes in the nasal mucosal tissue. We used Student T-test statistically(p<0.05). Results : Onpye-tang showed significantly decreased IgE level on the serum of the rat model. Onpye-tang showed significantly decreased eosinophil level on the blood of the rat model. Onpye-tang inhibited the inflammatory reaction on the nasal mucosal tissue, according to nasal mucosal biopsy. Onpye-tang showed an inhibitory effect on the process of allergic rhinitis, according to IgE level, Eosinophil level, nasal mucosal biopsy. Onpye-tang had no hepatoxicity, according to AST and ALT on the serum. Conclusion : According to the above results, it is considered that Onpye-tang has an inhibitory effect on the process of allergic rhinitis and it can be used in relieving symptoms of allergic rhinitis.

        노회(蘆?)(알로에), 자화지정(紫花地丁)의 항산화, 항염증, 주름, 미백에 미치는 영향

        Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.

        황련해독탕 및 발효황련해독탕의 항염증 효과

        Objective : Recently interest of fermented herb medicine has increased in Oriental medicine. Herb medicine fermented in a proper condition resultingly has better or new virtue by microbe. This study was designed to compare the anti-inflammatory effects of herbal decoction and fermented herbal decoction in Hwangnyeonhaedok-tang (Huanglian fiedu-tang; HHT). Methods : The effects of HHT and FHHT on anti-inflammation were measured by production of NO and pro-inflammatory cytokine productions including TNF-${\alpha}$ and IL-1${\beta}$, IL-6 in LPS induced RAW 264.7 cells. Results : 1. Cell viability in three doses of HHT extract (0.01, 0.05 and 0.10 mg/ml) and two doses of FHHT extract (0.01, 0.05 mg/ml) was more than 70% and 80% each. 2. In RAW 264.7 cells, LPS displayed significant stimulatory effects on the production of NO. However, both HHT, FHHT showed significant inhibitory effects on the production of NO. 3. Pre-treatment by HHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$, IL-1${\beta}$, IL-1${\beta}$ mRNA, IL-6 mRNA. More than 0.05mg/ml concentrations of HHT reduced the production of IL-6. And TNF-${\alpha}$, IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). 4. Pre-treatment by FHHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$. More than 0.05mg/ml concentrations of FHHT reduced the expression of TNF-${\alpha}$ mRNA. 0.1, 0.50mg/ml concentrations of FHHT reduced the production of IL-6. And IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). Conclusions : These results indicated that both HHT and FHHT could have anti-inflammatory capacity by inhibiting the production of NO and cytokines in vitro. But HHT has more strong anti-inflammatory effects than that of FHHT in broad outlines.

        연구논문 : 기업규모에 따른 CSR활동과 기업가치에 대한 실증분석

        본 연구는 CSR(Corporate Social Responsibility)활동과 기업가치의 관련성을 심도있게 검증하기 위해 기존 연구결과에서 더 나아가 개별 CSR활동과 기업가치의 관련성에 대해 살펴보았다. 이를 위해 CSR활동의 대용치로 경실련 산하 경제정의연구소의 경제정의지수(Korean Economic Justice Institute Index:이하 KEJI지수)를 사용하였으며, KEJI지수에 따라 CSR활동을 기업활동의 건전성과 공정성, 사회봉사활동, 소비자보호활동, 환경보호활동, 종업원만족, 경제발전기여로 구분하였다. 또한 시차모형(Time-Lag Model)을 사용하여 전기와 당기의 CSR활동과 기업가치(Tobin`s Q)의 관련성을 동시에 양방향으로 분석하였다. 여기에 특정 CSR활동이 기업가치에 미치는 기업규모별 차이를 검정하고자, 기업가치에 유의한 특정 CSR활동이 기업규모에 따라 기업가치에 미치는 영향을 비교?분석하였다. 연구결과를 요약하면 다음과 같다. 첫째, CSR활동 중 종업원만족과 경제발전기여는 기업가치와 정(+)의 관련성을 보였다. 종업원의 만족이 높을수록 경제발전에 더 기여할수록 기업가치상승을 초래하는 것으로 나타났다. 또한 기업가치가 높은 기업일수록 지속적 성장을 위해 종업원만족과 경제발전기여에 힘을 기울이는 것으로 나타났다. 반면, 환경보호활동과 기업가치는 부(-)의 관련성을 보였다. 환경보호활동은 기업가치상승에 직접적으로 기여하지 못하는 것으로 나타났으며, 기업가치가 높은 기업이 환경보호활동에 적극적이지 않는 것으로 나타났다. 둘째, 기업가치에 유의한 환경보호활동, 종업원만족과 경제발전기여는 기업규모에 따라 기업가치에 미치는 영향에 차이가 있는 것으로 나타났다. 즉, 대규모 상장기업의 환경보호활동, 종업원만족, 경제발전기여가 기업가치에 미치는 영향은 소규모 상장기업의 환경보호활동, 종업원만족, 경제발전기여가 기업가치에 미치는 영향과 차이가 있음을 보여주었다. 본 연구는 지속적인 CSR활동과 기업가치간 관련성을 분석함으로써 상장기업의 적극적인 사회적 활동을 유도하고 기업가치 향상을 위한 CSR활동의 중요성을 제고한다는 점에서 가치있는 연구이다. 나아가, 기업의 이해관계자들이 CSR활동의 어떤 면을 중요시하고, 어떻게 인식해야 하는지 분석함으로써 CSR투자활동의 의사결정에 도움을 줄 수 있다. 또한 본 연구결과를 기초로 기업규모에 따라 어느 활동을 강화할 것인지 전략적 판단이 가능하다는 한 점에서 실무적으로 의미가 있다고 하겠다. This study investigates in depth what individual activities of CSR(Corporate Social responsibility) has affirmatively correlated with firm value to expand prior studies` result that CSR activity itself had positively correlated with firm value. For this analysis, an index published by Korean Economic Justice Institute(KEJI) is used as the measure of CSR activities. KEJI Index has been measured with seven factors such as soundness and fairness of firm`s activity, social service activity, customer protection activity, environmental protection activity, employee satisfaction and economic development contribution. With these measures, relations between the prior and current year`s CSR activities and the prior and current year`s firm values(Tobin`s Q) are simultaneously bi-directionally analyzed by using time-lag model. To test different influence of firm size, additional tests are conducted whether there has been different effects on firm values that showed statistically significant specific CSR activity by firm size. The results of the analysis are as follows: First, employee satisfaction and economic development contribution of CSR activities are positively correlated to the firm value. In other words, firms with higher employee satisfaction and economic development contribution activities have higher firm value and the firm with higher firm value has higher employee satisfaction and economic development contribution activities for continuous growth. But environmental protection activity is negatively related to the firm value and vice versa. In other words, environmental protection activity showed that it didn`t contribute to the increase of firm value. and the firm with high firm value wasn`t active to environmental protection activities. Second, environmental protection, employee satisfaction and economic development contribution activities have different effects on firm values by firm size. In other words, environmental protection, employee satisfaction and economic development contribution activities`s value relevance of large-sized firm are different from those of small and medium-sized firm. This study induced Korean firm`s positive social activities through correlation analysis between continuous CSR activities and firm value. and it is a worthy pursuit to improve CSR activities`s importance to enhance firm value. And this study can provide useful insight into firms` decision-makers as follows: First, what kinds of CSR activities have been recognised as significance for shareholders and thus, how to and what to consider priority on CSR activities. Second, by their firm size, different strategic decisions can be made on what specific CSR activities should be strengthened.
