A method for the determination of anionic drug, mefenamate with ion-selective electrode using Fe(II)-1,10-phenanthroline chelate as a counter ion was developed. Benzyl nitrophenyl ether (BNPE) plasticized membrane was more selective and sensitive than the other tested membranes. This membrane electrode exhibits a linear response for 10-2 M~5 ×10-5 M of mefenamic acid with a slope of - 61.4mV/dec. in borate buffer sloutions (Ph 9.0). Potentiometric selectivity measurements revealed negligible interferences from various organic and ionorganic anions. Direct potentiometry and potentiometric titration method of mefenamic acid in capsule preparations are presented and compared.

The $H^+$-selective membrane consists of tridodecylamine, 2-nitrophenyl octyl ether and a little amount of additive in carboxylated PVC matrix, where penicillinase(Pcase ) and glutaraldehyde may be covalently attached to the matrix. When the $H^+$-selective electrode was used as a detector of biosensor, calibration curve calculated from Nernst equation was not linear. So we investigated the characteristic responses of the $H^+$-selective electrode to the product H+ from hydrolysis of penicillin-G(Pc-G) and plotted calibration curve correlating potential to concentration of Pc-G linearly. The optimal concentration of buffer solution was theoretically calculated and was also experimented. We tried to explain the linear curve of potential to concentration of Pc-G by using Henderson-Hasselbach equation. This method is more effective in calibration curve plotting than any other previous methods. The results obtained may help in further developing pH electrodes with improved analytical preformance.

A method for the determination of basic drugs based on their reaction with picric acid to form an ion-association complex extractable into several plasticizers was developed. Basic drugs-picric acid complexes in acid medium could be extracted quantitatively into several plasticizers except phosphates. For example, the chlorpromazine-picric acid complex showed maximum absorbance at near 410nm and was applied to extraction spectrophotometric determination of chlorpromazine. The calibration curves are linear (r>0.998) within a range from 10-6 to 5 X 10-4M and the precision of the method was acceptable because RSD was less than 2.6% (n=7). The factors affecting the extraction system was discussed.

Many poly (vinyl chloride: PVC) membrane electrodes were investigated for the determination of basic drugs, chlorpromazine, amitriptyline, nortriptyline, etc. These electrodes are based on the use of the ion-association complexes of the basic drugs with eriochrome cyanine R, chromoxane cyanine, chrome azurol S and picric acid as ion-exchange sites in a plasticized PVC matrix. All ion-exchangers except picrate complex were not proper for use, because those complexes in plasticized membrane were excluded into aqueous working solution. These drug electodes show excellent Nernstian responses in the concentration range 10-2~10-6. Their selectivity with respect to each other, as well as their workable pH range have been investigated. The major advantages of the proposed methods are their simplicity and speed.

Ion-selective poly(vinyl chloride)(PVC) membrane electrodes for the determination of the calcium antagonist verapamil and its pharmaceutical preparations were described. Verapamil-superchrome garnet Y(SGY), lumogallion(LG), acid red 97(AR97), Dragendorff(DD) and Meyer reagent ion pairs were inverstigated as an electroactive compound for membrane electrode. Stable potentiometric response was obtained with azo dye at pH 6.5-4.0 and with DD, and Meyer reagent at pH 6.5-3.0. The best plasticizer was 49w/w% 2-nitrophenyl octyl ether for azo dye, and 65.3w/w% tri(n-butyl) citrate for DD and Meyer reagent. Potentiometric response slopes of optimized membrane electrodes based on SGY, LG, AR97, DD, and Meyer complex for verapamil were 52.49, 54.88, 50.81, 54.13 and 49.31 mV/dec., respectively. Lower limits of linear range were 1.0X106M for SGY, LG, and AR97, while those for DD and Meyer reagent were 4X10-6M. Detection limits for all these electrodes were 1X10-5M, 4X10-6M, 1.8X10-6M, 8X10-7M, and 1X10-6M with relative standard deviation of 2.56, 3.6, 3.96, 2.56, 3.20%, respectively.

Penicillin sensor was manufactured by immobillizing penicillinase with glutaraldehyde on the H+-selective membrane based on PVC-COOH-TDDA. This membrane was not interfered by K+ ion in Pc-G potassium salt. When enzyme was immobilized with glutaraldehyde, the PVC-COOH matrix was more effective than PVC matrix. Calibration curve calculated from Nernst equation was not linear. But potential was relative to concentration of Pc-G. And maximal potentiometric velocity was also relative to concentration of Pc-G. Therefore, it may be applied to Michaelis-Menten equation. The penicillin sensor was useful for determination of Pc-G at concentration of 0.1~10mM level.

Garlic tissue cells are employed for the conversion of L-asparagine into ammonia. An ammonia gas electrode is used as a detector. The effect of pH, buffer solution, temperature and life time of electrode to have used were investigated in order to optimize the electrode response. The combination of L-asparaginase in garlic tissue cells and the gas electrode response linearly to L-asparagine over the concentration range 1.0X10-4 ~ 1.0X10-1M with a slope of 72.0mV/decade and is selective with respect to other L-amino acids.

PVC membrane electrodes based on the lipophilic neutral carrier, dibenzo-18-crown-6, cyclic oligomers of tert-butylphenol-formaldehyde condensates, calix[4]arenes as the active sensors for verapamil have been prepared and tested in a variety of plasticizers. At pH 5.0, the electrode exhibits a Nernstian response in the range of 1X10-2 ~ 5X10-5M verapamil with a slope of 49.1 +/- 0.5mV per concentration decade. The electrode constructed in this work can be used continuously for at least 1 month before any damage to the membrane occurs. And the analysis of the local anesthetic amine, which are good to select a specific compound in a mixed solution, were also accomplished by using of another neutral carrier, a DB18C6, for comparing with calix[4]arene.
