        나노 입자화 한 복분자 저온 추출물의 면역 활성 증진

        This study was performed to improve immune activities of Rubus coreanus Miquel by encapsulation of nanoparticles. Immuno-activities of R. coreanus were investigated through aqueous extracts associated with process of water at 60℃. It showed high promotion of human B and T cells growth about 50%, compared to the case of other conditions. The secretion of IL-6 and TNF-α was also enhanced as 2.44×10-4pg/cell and 1.94×10-4pg/cell, results by adding nano samples. NK cell activation was improved up to 29% higher than the conventional extraction process. The secretion of NO from macrophage showed 14.9 μm on the nano-encapsulation process extracts, which was higher than others. The size of nanoparticles was in the range of 50~300 nm, which can effect the penetration into the cells. It was clearly observed by real time confocal microscope.

        고압액화공정을 이용한 구멍갈파래의 발효용 알코올 당화수율 증진

        Green alga, Ulva pertusa kjelmann has been known to be one of the largest pollutants in Korea. Therefore, the efficient pretreatment processes have been required to improve the yields of fermentable sugar. The optimal pretreatment conditions were determined to be 195℃ for 15 min. The sugar yield of glucose and xylose were estimated as 20.5%, and 5.0% respectively, based on theoretical yields. However solid residues were estimated enzymatic digestibility of 90-95% with cellulase loading of 15 FPU/g glucan. This process was proved to generate the low concentration of Hydroxy-Methyl-Furfural (51 ppm), which resulted in ethanol production with 95% of the maximum conversion yield from glucose in the culture of Saccharomyces cerevisiae (ATCC, 24858). This study showed that Ulva pertusa kjellmann can be used as a bioetahnol resource using the high temperature liquefaction process.

        유채대의 이단 고온 처리에 의한 알콜 발효용 당화물 생산

        우리 연구팀은, 농산부산물인 유채대의 알콜 발효용 당 화물 생산 가능성을 연구하였다. 이를 위해 농산부산물인 유채대를 연속적으로 5 mL/min의 속도로 산을 이용하지 않고 증류수만을 이용해 이단 고온 처리 (200℃ and 15 Mpa, 375℃ and 23 Mpa)하였다. 본 전처리 공정을 통한 가수수분해물의 당화물 생성은 최종적으로 자일로스와 글루코스의 경우 25.6 g/L, 5.5 g/L가 생성되었다. 이는 유채대에 존재하는 글루코스와 자일로스의 초기 양 대비 각각 18%와 59%의 전환 수율을 나타낸다. 또한 이 공정은 타 공정들에 비하여 대표발효 저해 산물인 HMF의 생성량이 0.2 ppm으로 극히 낮은 수치를 보였으며, 가수분해물의 에탄올 생산시, 글루코스의 발효를 통한 에탄올 생성 전환수율이 90% 이상으로 높은 생성율을 보였다. 따라서 본 공정을 통해 다른 농산부산물이나 해조류 전체에 응용된다면, 고 수율의 에탄올 생산용 당화물을 생산할 것으로 예상한다. Two-step pretreatment process was investigated to efficiently hydrolyzed rape stems for obtaining fermentable sugars. The process was consisted of two consecutive steps as 200℃ and 15 MPa and 374℃ and 24 MPa with the flow rate of 5 mL/min. Under this condition, 5.5 (g/L) of glucose and 25.6 (g/L) of xylose were obtained from rape stems, showing 18% of glucose yield based on 25% cellulose in the rape stems. It was also found that this process could generate less amounts of toxic residues, such as HMF (Hydroxy- Methyl-Furfural) and other fulfural components during hydrolysis process. It could reaction maintain relatively high ethanol production yield as 90% of theoretical conversion yield from glucose. Therefore, this pretreatment process could be applied to hydrolyze other cellulosic and marine resources such as woods, stem and algae for bioethanol production.

        구멍갈파래의 면역활성 증진을 위한 추출방법 비교

        본 연구는 제주도의 녹조현상의 원인인 구멍갈파래의 활용을 위해 고압추출 공정을 통한 추출물의 면역활성 확인 하였다. 150MPa, 80℃ 조건에서 Superoxide Radical 소거효과에서는 90% 로 활성도가 제일 높았으며, 면역세포 생육도면에서도 B&T cell, 각각 14.2×10sup{4} cells/mL 14.5×10sup{4} cells/mL 나타나 제일 높았다. 반면, NK cell과 NO 면역능 실험에서는 100MPa, 30℃ 조건에서 각각 11.8×10sup{2} cells/mL, 30.0 μM을 보여 주었다. 결과를 통해 구멍갈파래는 면역활성을 가지며 고압 공정을 통한 추출물의 활성이 일반추출에 비해 높은 것을 알 수 있다. 따라서 해조류 및 다른 유용 자원에서도 고압 공정이 기존의 추출방법보다 높은 활성을 나타낼 것으로 판단되며 기능성 식품이나 기능성 소재로서 사용가능성이 있는 것으로 사료된다. The purpose of this study was to investigate the immunomodulatory effect of Ulva pertusa kjellman extract after undergoing a low temperature and high-pressure extraction process. First, the extracts obtained under the extraction conditions of 150 MPa and 80℃ showed a relatively high antioxidant activity, with 90% super oxide radical activity compared to the extracts from conventional extraction process with water at 100℃. This extract also improved the growth of both human immune B and T cells up to 14.5×10sub{4} cells/mL and 14.2×10sub{4} cells/mL compared to 9.1×10sub{4} cells/mL in adding the extracts from conventional processes. It was found that the extracts obtained at 100 MPa and 60℃ showed better activities in NK cell growth and NO production from macrophage as 11.8×10sub{2} cells/mL and 30.0 μM. Overall, the extracts from high pressure and low temperature extraction process had relatively higher immune activation activity, possibly because the low temperature and high pressure extraction process may have higher yields of active compounds and have less damage to useful ingredients from relatively weak marine natural resources, such as Ulva pertusa kjellmann than that from the conventional extraction system.

        초음파 복합처리를 통한 참굴 펩타이드의 피부미백 및 피부면역 활성

        초음파를 병행한 참굴 시료로부터 조단백질의 수율은 34% 이상으로 높은 수율을 나타내었다. 분리 단백질을 SDS-PAGE로 검정한 결과 70 kDa와 50 kDa 사이에 특정 피크를 나타내었다. 이를 sephadex G-75 컬럼을 이용하여 분리하였으며 MALDI-TOF MS를 이용하여 분자량을 검정한 결과 66 kDa으로 판단하였다. 굴 추출물 및 펩타이드의 세포독성을 측정한 결과 최고농도인 1.0㎎/mL에서 모두 22.8%를 나타내었으며 형태학적 차이를 나타내지 않아 굴의 저온 추출물 및 펩타이드가 식품 또는 향장 소재로서 독성을 나타내지 않는 것으로 사료된다. Melanocyte를 이용한 멜라닌 생성량 측정을 통해 굴 펩타이드의 미백활성을 탐색한 결과 최고 62.7%의 멜라닌 생성 저해 효과를 나타내었으며, 1.0 ㎎/mL의 농도에서 tyrosinase의 활성을 35%까지 저해하는 것을 확인할 수 있었다. 피부 염증과의 관계를 알아보기 위하여 실행한 PGE₂ 생성량 측정의 경우 특정 수용체(EP3)와 결합하면 알레르기 증상이 억제되는 것으로 밝혀졌는데, 연구의 결과를 바탕으로 특정 펩타이드와 결합으로 인한 PGE₂의 생성 억제 가능성이 있으리라 사료된다. 이상의 결과에서 시료의 활성은 농도 의존적으로 경향을 나타남에 따라 특정 물질의 작용으로 불 수 있으며 melanocyte의 멜라닌 생성량 저해능과 더불어 향후, 보다 면밀한 정제를 통한 물질 검정의 단계를 거치면 천연 피부미백용 향장소재로서 피부미백 및 피부 면역 활성을 안전하게 향상시킬 수 있는 기능성 소재로 활용이 가능할 것으로 사료된다. In this study, peptides were isolated from Crassostrea gigas using an ultrasonification process at 40℃. The yield of the peptides was greater than 34%, and their cytotoxicity was found to be less than 22.8% against several cell lines that were treated with the extracts at a dose of 1.0 ㎎/mL. In addition, the tyrosinase inhibitory and melanin synthesis of the peptides isolated from Crassostrea gigas were also evaluated to determine if they could be used as a potential cosmetic agent. The peptides were found to significantly inhibit the melanin synthesis of the clone M-3 cell line by up to 62.7%. The inhibitory activities of the tyrosinase were observed 34.51% in ascorbic acid, 42.49% in extract with the ultrasonification at 40℃ and 35.37% in 40℃ extract at 1.0 ㎎/mL concentration, respectively. Finally, when samples were treated with the peptide extracts at a concentration of 0.6 ㎎/mL, PGE2 expression was significantly decreased. Taken together, these results indicate that Crassostrea gigas may be a source of cosmetic agents capable of improving physiological hyperpigmenting and immunomodulating skin disorders.

        제주 손바닥 선인장의 초음파 추출을 통한 면역활성 증진

        Immune activities of Opuntia ficus indica L. Miller were investigated through aqueous extracts associated with ultrasonification process at 60℃, and showed the highest promotion of human B and T cell growth, up to 52% and 41%, respectively, compared to the control. The secretion of TNF-α and IL-60 was also enhanced by the addition (0.5mg/ml) of the extracts, up to 48%. NK cell activation was significantly improved up to 1.3 times higher than the case of adding other extracts. It was also found that extracts from O. ficus indica could yield higher nitric oxide production from macrophage than Lipopolysaccaharides (LPS). It can be concluded that, in general, the extracts treated with ultrasonification has higher Immune acitivities than others, possibly by higher yielding immune-modulatory than conventional extraction process. The optimum condition for the extraction of O. ficus-indicais water extraction associated with ultrasonification.

The secretion of TNF-α and IL-6 was also enhanced by the addition (0.5mg/ml) of the extracts. NK cell activation was Improved up to 1.37 times higher than the control, through adding extracts. It was also found that extracts from C. endivia L. could yield higher nitric oxide production from macrophage than Lipopolysaccaharides (LPS). It can be concluded that, in general, the extracts treated with ultrasonification has higher immune activity than others, possibly by higher yielding immune-modulatory activity than conventional extraction process. The optimum condition for the extraction of C. endivia L. is ethanol extraction at 60~100℃ associated with ultrasonification.

        초고압 추출 처리에 의한 마황과 당귀의 항암 활성 증진

        This study was performed to enhance anticancer activities of E. sinica, and A. gigas by ultra high pressure extraction process. The cytotoxicity of E. sinica and A. gigas on human kidney cell (HEK293) was as low as 24.94% and 25.3% in adding 1.0 mg/ml of the sample extracted at 500 Mpa for 15 minute. Generally, the inhibition of cancer cell growth on A549 and MCF-7 was increased over 20% in the ultra high pressure samples, compared to the conventional extraction process. Under the extracts from ultra high pressure process showed not only the strongest anticancer activities, but also had better stability than normal extracts. It was also found that the extracts of A. gigas reduced the hypertrophy of the internal organs, such as adrenal and spleen caused stresses in several mouse models.

        초고압 저온처리에 의한 매자나무의 면역 활성

        This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

        초음파를 이용한 무증자 저 상품용 감자와 고구마의 알코올 발효 비교

        This study was performed to compare alcohol fermentation ability and saccharification of potatoes and sweet potatoes. Cooperated with ultrasonification process is not contain pass through cook process, but contain process using ulrasonification instead cook process. In result of sugar contents measurements sweet potato and potato was highest of a family at 6 hours fermentation, and it showed the highest sugar contents as each 11.5 brix, 10.4 brix. In result of alcohol contents measurements of sweet potato and potato, highest of a family in 4 days, and it showed th highest alcohol contents as each 8.2, 6.0%. Finally complex enzymes II process revealed similar activities like cook process.
