Zebrafish 동물모델에서 human HtrA2의 expression system 정립에 관한 연구

Mitochondrial serine protease로 알려진 human HtrA2 (hHtrA2)는 apoptosis 유도 과정에서 중요한 역할을 담당하고 있을 뿐만 아니라 hHtrA2가 motor neuron degeneration과 관련이 있다는 최근 연구 결과가 있으나, hHtrA2의 생리적 기능은 아직 명확하게 밝혀져 있지 않다. 이와 같이 생체내에서 필수적인 업무를 담당하는 hHtrA2의 기능을 심도 있게 연구하기 위해서는 적절한 동물모델 시스템이 필요하나 이에 대한 연구도 미흡한 실정이다. 따라서 본 연구에서는 hHtrA2의 기능 분석을 위한 기본적인 실험으로 zebrafish라는 동물모델을 선택하여 hHtrA2의 발현 시스템을 정립하였다. 먼저 zebrafish에 hHtrA2를 발현시키기 위하여 zebrafish에서 일반적으로 사용되는 발현 시스템인 pCS2+ vector에 hHtrA2와 GFP를 cloning하고 plasmid를 HEK293 cell에 transfection한 후, hHtrA2-GFP fusion 단백질의 발현을 immunoblot과 immunofluorescence staining assay로 확인한 바 약 64 kDa의 hHtrA2 단백질의 발현을 확인할 수 있었다. Zebrafish에서 hHtrA2-GFP fusion 단백질의 발현양상은 immunofluorescence microscope으로 확인하였다. hHtrA2-GFP DNA와 mRNA를 zebrafish embyro에 microinjection하여 두 가지 component의 발현을 비교 분석한 결과, DNA는 dot 형태로 mRNA는 몸 전체에 퍼져보이는 형태로 발현 양상의 차이는 있었으나 둘 다 zebrafish embryo에서 잘 발현되는 것을 알 수 있다. 다음 DNA를 주 component로 microinjection하여 zebrafish embryo에서 발현을 확인한 결과 hHtrA2는 72 hpf 까지 발현이 지속되는 것을 확인하였다. 본 연구에서 정립한 hHtrA2의 zebrafish 발현 조건은 앞으로 zebrafish에서 hHtrA2의 생리적 기능을 심도있고 정확하게 연구하는 데 있어 기본적인 자료로 활용 할 수 있을 것이다. HtrA2/Omi, a mitochondrial trypsin-like serine protease, is pivotal in regulating apoptotic cell death. Several lines of recent evidence suggest that HtrA2 is associated with the pathogenesis of neurodegenerative disorders; however, the physiological function of HtrA2 still remains elusive. For studying physiological function of HtrA2 in depth, it is necessary to develop a suitable expression system in the model animal. We therefore utilized the zebrafish as a model animal to establish expression of human HtrA2 (hHtrA2) in vivo. For expression of mature HtrA2 as GFP fusion in zebrafish embryos, the HtrA2 (WT) or (S306A) cDNAs with the C-terminal GFP tag were inserted into the pCS2+ plasmid. Expression patterns of HtrA2 in HEK293 cells were first monitored by immunofluorescence staining and immunoblot assays, showing approximately 64 kDa of the HtrA2-GFP fusion proteins. Subsequently, the hHtrA2 plasmid DNA or in vitro transcribed mRNA was microinjected into zebrafish embryos. The expression patterns of HtrA2 in Zebrafish embryos were monitored by GFP fluorescence in 24 hours-post-fertilization (hpf). Although expression patterns of HtrA2-GFP in developing embryos were different between the injected DNA and mRNA, both nucleic acids revealed good expression levels to further study the physiological role of HtrA2 in vivo. This study provides a suitable condition for expressing hHtrA2 in the zebrafish embryos as well as a method for generating useful system to investigate physiological properties of the specific human genes.

Some Chronic HBV Carriers have persistent HBV replication despite the presence of anti-HBe Ag antibodies. This atypical chronic HBV infection has recently been related to the presence, in the pre-core region of HBV genome, of a translational stop codon that prevents HBeAg synthesis. We analyzed nucleotide sequences of the pre-core region of HBV isolated from the serum of an anti-HBe, HBV DNA positive severe chronic active hepatitis patient. Nucleotide sequence revealed that there was a point mutation from G to A at nucleotide 1896 of the pre-core region, converting tryptophan (TGG) to a stop codon (TAG). Additional mutation of A-T substitution at position 1846 was found. In conclusion, mutant HBV is responsible for developing severe anti-HBe positive chronic hepatitis B in Korea.

Low frequency (400 kHz) Ferrite ICP has been proposed for large area processing. Because the coupling coefficient is close to 1, the transformer matching can be adapted to Ferrite ICP. The transformer matching system is simple. In this paper the new matching system by controlling the turns of transformer using relays is proposed for impedance matching. We developed a simple matching system and characterized it when the turns were changed. It was observed that the 2-channel relay is available for transformer matching.

N/A Gastric bezoars are movable intraluminal masses of congealed ingested materials that can be demonstrated in an upper gastrointestinal series by typical coating with barium. Ultrasonographic diagnosis of gastric bezoars may be relatively specific. A few cases of ultrasound appearance of gastric bezoars has been previously reported in the literature. Hyperechoic curvilinear dense strip can be seen superficially, with the clean nature of the acoustic shadow behind. But there area few pitfall that shou]d be differentiated from advanced stomach caocer, retained food materals in stomach etc. on u)trasound. In order to evaluate the diagnostic a vailability of ultrasonography in the detection of gastric bezoars, we analysed the ultrasonographic findings in gastric bezoar 6 cases, advanced gastric cancer 10 cases, and food particle in the stomach 3 cases. We took 4 ultrasonographic findings for landmarks of their differential diagnosis; 1) hyperechoic curvilinear reAective strip 2) the nature of the acoustic shadow 3) pseudokidney sign 4) gastric wall thickening. Ultrasonographic procedure of the removed bezoar was also performed in the water bath. Using these landmarks, there seemed to be distinct difference between gastric bezoar and other gastric conditions. Ultrasonographic feature of gastric bezoar, performed in the water bath, revealed some specific findings for gastric bezoar. We concluded that ultrasongraphic finding of gastric bezoars was sufficiently characteristic to make a diagnosis.

N/A Single-photon ernission computed tomography (SPECT) is a new image method that offers the advantage of cross-sectional imaging and improved contrast resolution as compared with conven- tional planar imaging. Ultrasound (US), conventional planar scintigraphy (PS), and single-photon emmission computed tomography (SPECT) were compared in 36 patients with suspected focal hepatic disease. Ultrasonography had slightly higher sensitivity (90%) and specificity (100%) than either PS or SPECT. A comparison of the diagnosis demonstrated that SPECT had greater sensitivity, specificity, and accuracy than conventional scans. Although SPECT was inferior to the US examination in detecting focal hepatic disease, especially less than 3 cm in size. The combination of SPECT and US was an excellent method for the earlv detection of focal hepatic disease.

N/A Ultrasongraphy has proven useful in evaluating the diagnosis of many hepatobiliary diseases. But a few reports about the ultrasonographic findings of biliary ascariasis appeared in the literature. Sonographic diagnosis of biliary ascariasis in the common bile duct was made in 2 patients, and was confirmed by endoscopic visualization and retrograde cholangiography. Ultrasonographic procedure of the removed worm was also performed in the water bath. The results was as follows: 1) In case 1, Ultrasonogram showed spaghetti apperance of 2 overlapping longitudinal interfaces without acoustic shadow in the logitudinal scanning of the common bile duct(spaghetti sign) and bull's eye echo pattern of annular hyperechoic configuration in the horizontal scanning of the common bile duct(bull's eye appearance). 2) In case 2, Ultrasonogram showed inner tube appearance of hyperechoic parallel stripes containing a central longitudinal anechoic tube(inner tube sign) and a hyperechoic strip without an inner tube(strip sign) in the longitudinal scanning of the common bile duct. Bull's eye appearance was also showed in the horizontal scanning of the common bile duct. 3) As for ultrasonographic configuration of ascaris lumbricoides, performed in the water bath, we got three aspects. In the longitudinal scanning, it showed hyperechoic parallel stripes containing a central anechic tube, probably representing its digestive tract, on the vertical sonobeam(inner tube sign) and hyperechoic string on the non-ventical sonobeam(strip sign). In the horizontal scanning, it showed annular hyperechoic configuration(bull's eye apperance). We now believe that ultrasonography, being safe, quick and noninvasive, should be the first imaging method for suspected biliary ascariasis.

N/A Hepatic cyst is a rare disease, and cases have been reported since first discription of Bristowe in 1885. However there are difficulties in classification, diagnosis and treatment about this disease. The diagnostic metbod have been developed, and the ultrasonography is important and accurate diagnostic tool. We have experienced 46 cases of hepatic cystic disease at the Soon Chun Hyang University from Jan. 1980 to Jun. 1986 and analysed the following results obtained. 1) Incidence of hepatic cyst is about 0.53% and all patients were over the age of 40. 2) Male and female ratio was 1:2.3. 3) Solitary liver cyst only were 32 cases, solitary liver cyst with renal cyst were 3 cases, polycystic liver only were 14 cases, and combined polycystic liver and polycystic kidneys were 4 cases. 4) Size of the hepatic cysts was from 0.9 cm to 26 cm and 65% of the cases were below 2 cm 5) The location of hepatic cysts showed 20 cases in right lobe and 18 cases in left lobe of the liver. 6) The ultrasonography was important and accurate diagnostic tool.
용매 최적화를 통한 페로브스카이트 태양전지의 전하수송층 제어 기술

태양전지의 효율을 증가시키기 위해서 계면에서의 전하수송 특성을 제어하는 것은 매우 중요하다. 우리 팀은 평면 구조의 페롭스카이트 태양전지 개발에 있어서 자주 이용되는 PCBM 소재의 모폴로지 제어를 위해서 유기 용매의 최적 조합에 대해 연구하였다. 실제적으로 클로로벤젠이라는 용매가 주로 많이 사용되고 있는데, 이 용매로 용액 공정을 진행할 경우 표면 모폴로지가 평탄화 공정이 잘 진행되지 않는 경우가 많다. 따라서 본 논문에서는 평탄화 공정의 최적화를 위해서 클로로벤젠에 추가적인 용매 Y2를 추가하여 끓는점과 증기압의 변화를 추구하였다. 그를 통해서 PCBM의 용액 공정 후의 열처리 공정에서 원활한 평탄화 작업을 지을 수 있음을 실험적으로 검증하였다. 이를 통해서, 페롭스카이트 박막 태양전지의 기본 효율이 16% 수준에서 19%에 가까운 변환 효율의 향상을 검증할 수 있었으며, 향후 추가적인 연구를 통해 본 연구분야의 전하수송층 용액 공정에서의 가이드라인을 제공할 수 있을 것으로 기대된다.
영유아 보육공간에서 미세먼지의 외부 유입 및 실내 발생 부하 추정에 관한 연구

Atmospheric particle concentration is increasing in Korea owing to industrialization in Korea and the influence of China. Outdoor particles can enter buildings through cracks or windows and affect the health of residents. Care rooms in childcare centers are meant for infants and children, who are more vulnerable to particles than adults; hence, indoor air quality management is necessary. Outdoor particles as well as the particles generated through children’s activities and indoor cooking should be considered for the assessment of indoor air quality of these places. Thus, particles that should be removed from such buildings comprise outdoor particles entering indoor and those generated indoor. The total concentration of these particles can be termed as particle load. Ventilation and air cleaning devices are effective in removing particle loads, and efficient indoor air quality management can be achieved by using devices of appropriate capacity and performance based on load estimation. Therefore, to estimate the particle load of care rooms, indoor air quality monitoring was conducted for one year, and the particle loads were estimated by analyzing the monitoring data.
