유방암세포주에서 고농도 5-fluorouracil의 세포주기 조절효과

Background: Chemotherapy with 5-fluorouracil (5-FU) has been one of the mainstay in breast cancer treatment. The effects of high dose 5-FU on cell cycle regulation were studied in breast caner cells. Methods: A breast cancer cell line MCF-7 was used. Protein expressions of G1/S cyclins, $p21^{Waf1/Cip1}$, cdk2, E2F1 and retinoblastoma were tested by western blot analysis. Immunoprecipitation and immune complex kinase assay were done for the assessment of E2F1/RB interacton and the activity of cdk2 respectively. Results: $p21^{Waf1/Cip1}$ expression was barely detectable in control cells. With addition of 5-FU level of $p21^{Waf1/Cip1}$ were induced and cyclin D3 level was decreased as cell growth decreases. In accordance with increased expression of $p21^{Waf1/Cip1}$, cyclin E-associated cdk2 kinase activity was reduced. Retinoblastoma protein (RB) became dephosphorylated and E2F-1 binding activity with RB was increased. Conclusion: In this situation of high concentration of 5-FU breast cancer cells tend to be G1/S cell cycle arrested. Overexpression of $p21^{Waf1/Cip1}$ and dephosphorylation of RB may mediate the effectss of 5-FU by inhibiting E2F-1 activity, which contributes to G1/S cell cycle arrest. These results could be an indicating landmark for further study of high dose chemotherapy with 5-FU.

이연구의 목적은 미국의 사회적 기업의 자금조달 방식을 검토하여 우리나라의 사회적 기업의 창출에 필수적인 자원동원 계획을 수립하는데 기초적인 자료를 제공하고자 하는데 있다. 미국 사회적 기업의 재원조달방식을 검토하기 위해서 먼저 미국의 비영리조직의 발달과 사회적 기업의 성장에 대해 고찰하였다. 미국은 역사적으로 비영리조직이 발달해왔으며 특히 1990년대 이후에는 비영리조직들이 사회적 기업을 운영하기 시작했다. 미국의 비영리조직은 영리조직과 경쟁하면서 사회서비스 영역에서 급성장을 이루어왔다. 미국은 최근 사회적 기업과 사회적 투자에 대한 관심이 높아지고 있는 가운데 다양한 형태의 자금이 이 영역에 투자되고 있다. 이러한 사회적기업의 재원은 정부와 민간부문에서 증가되고 있다. 정부는 1990년대 지역재투자법(CRA)을 제정하고 지역개발금융기관(CDFIs)에 자금을 지원함으로써 사회적 기업을 활성화하는데 일조하고 있다. 또한 민간차원에서도 민간재단이나 기업, 제도 금융권, 사회적 벤쳐투자자들의 사회적 기업에 대한 지원이 급증하고 있다. 이렇게 사회적 기업에 대한 지원이 증가하고 있는 것은 정부가 일정 규모의 기금을 조성하고 이러한 기금을 중간대출기관에 제공하는 한편 중간대출기관들도 정부 이외의 민간기관의 재원을 유입하는 매칭펀드 형식을 취하게 함으로써 민간부문의 자원을 유입하는 장치를 마련했기 때문이다. 이러한 관점에서 우리 정부도 사회적 기업을 직접적으로 지원해야 할 뿐 아니라 일정 규모의 재원을 조성하고 이를 기반으로 민간자본을 유입하는 방안들이 강구되어야 한다. 그리고 지역사회나 국민이 사회적기업에 대한 관심을 제고시켜서 기업이나 민간자본의 사회적 책임투자나 기업 파트너십을 형성할 수 있는 여건을 마련해야 할 것이다. 또한 우리사회에 기부문화가 활성화되어야하고 이러한 자금을 효과적으로 운용할 수 있는 지역재단이 설립되어 사회적 기업을 효과적으로 지원할 수 있도록 해야 한다. The purpose of the study is to provide fundamental data needed to setup a necessary financial resources provision plan to the creation of Korean social enterprises by examining funds supply methods of American social enterprises. First of all, there are several characteristics of the third sectors of the United States to review in order to examine funds supply methods of American social enterprises. Historically in the U.S, NPOs have been developed, especially since 1990, NPOs have run social enterprises. In the U. S. NPOs have developed in their social areas, competing with profit enterprises. They are interested in social enterprise and social venture investment increasingly and a variety of funds are invested in these areas. Financial resources of these social enterprises have been increased by the U. S. government set up CRA in 1990’s and supported financial resources, and promoted the activation of social enterprises, Also private foundation and enterprise, public banks and venture investor supporting are increasing rapidly. The government established systematical devices; making funds and providing to CDFIs, promoting a type of matching fund with private financial resources. Moreover, the government supports the non-profit organizations through tax policies. In conclusion, the governmental support is essential to an activation of social enterprises of Korea. Furthermore, contributory culture also must be vitalized, and there should be local foundations which can manage the funds efficiently so that effective support of social enterprises can be possible. Finally, systematical devices are necessary in order to activate social contribution projects of enterprises, and loans of nongovernmental financial agencies must be expanded.

여성의 지역사회활동 참여욕구에 영향을 미치는 요인 - 서울시 서초구를 중심으로 -

This research aims at contributing to activation of local social welfare by studying the factors which affect women’s participation needs for community social activities. This research involves a survey conducted during a month from August 1st to August 31st, 2016 for 242 Seocho-Gu residents. Questionnaires were distributed to members of community committees as well as residents through community centers. The survey focuses on effects of variables including socio-demographic factors, community satisfaction, cognition degree of social problems, and community attachment on participation needs of community social activities. As a result, the average number of participation in community social activities of the respondents is 2.44(SD = .095). For the effects of socio-demographic factors on participation needs of community social activities, all the variables are not statistically meaningful. For the correlations among main variables, participation needs of community social activities is positively related to cognition degree of social problems(r = .244, p < .01) and community attachment(r = .174, p < .01). Based on these analyses on correlations, the effects on participation needs of community social activities by community satisfaction, cognition degree of social problems, and community attachment are the followings. Firstly, as a result of including community satisfaction variable in the model, its effect on participation needs of community social activities is not statistically meaningful. Secondly, as a result of including cognition degree of social problems variable in the first model, it explains about 6.8% factors of participation needs of community social activities. Thirdly, as a result of including community attachment variable in the second model, it explains about 10.4% factors of participation needs of community social activities thus, additionally explaining about 3.6% statistically meaningfully.

N/A Dry mouth is a commnon symptom in rheumatoid arthritis patients and an important component of sjoren's syndrome. The symptoms of dry mouth are very sultle difficult to assess. So, different procedures have been employed to assess, alivary gland function. One of them, parotid sialography, is a useful diagnostic procedure for sialadenitis. But this kind of investigation can cause pain, swallowing of the parotid gland, or exceptional allergic reaction to radiologic contrast media. In addition, it is a relatively complicated procedure to implement. On the other hand, despite the fact that the salivary gland can readily be evaluated by ultrasonography (US), there have been few reports describing the value of US in salivary gland diseases. We investigated the discriminative role of US versus parotid sialography in 31 patients with complaints of dry mouth and took minor salivary gland biopsies prospectively. All the patients in our study were women, and the age distribution was 28 to 66. The results were as follows: 1) In parotid sialography, 16 of the 20 patients with sialadenitis in minor salivary gland biopsy had abnormal findings over stage 2, and nine of the 11 without sigladenitis had normal findings (p=0.003). 2) In parotid US, 15 of the 20 patients with sialadenitis in minor salivary gland biopsy had decreased, namely abnormal, parotid gland echogenecity, and eighe of the 11 without sialadenitis had normal findings (p=0.028). 3) Comparing paratid US with parotid sialography, a good correlation between US parotid gland echogenecity and sialographic abnormalities was found. Ten of the 13 patients with abnormal US findings had abnormal sialograms, whereas three of the 15 with normal parotid gland echogenecity had abnormal sialo-grams (p=0.003). 4) No significant difference in submandibular echogenecity on US could be demonstrated. ESR tends to increase for patients with decreased parotid gland echogenecity (p=0.03). In summary, our study demonstrated that US, although not perfectly sensitive or specific, is a valuable noninvasive imaging techinque in screening for salivary gland involvement in patients with complants of dry mouth. Together with other studies it could be used as a first step in the diagnostic investigation of sialadenitis.
