Penicillium oxalicum(PENOX) has shown the potential as a biocontrol agent(BCA) for control of a weed, clover(Trifolium repens L.) in grass plots. The bioherbicidal activity may be due to germinative and growth capacities and substrate availability of the agent over a range of environmental factors. The influences of different water activities(0.94-0.995 aw) and temperatures(18~30℃) on mycelial growth, conidial germination, sporulation on 2% MEA(malt extract agar) adjusted to different water activities with glycerol, and carbon source utilization using BIOLOG GN MicroPlate were determined in vitro. Decreases in aw on MEA caused a reduction in mycelial growth and conidial germination depending on temperature. The mycelial growth of PENOX was greatest at 30℃/0.995 a<SUB>w</SUB>. At some lowered water activity(0.97 a<SUB>w</SUB>), the growth was similar between 25 and 30℃, and considerably decreased at lowered temperature(20℃). The germination rate was also greatest at 30℃/0.995 a<SUB>w</SUB>. Lag phase times for PENOX at 18℃ on MEA were>6hrs at the whole a<SUB>w</SUB> level tested, and at 18 and 25℃ they were >18hrs and >12hrs at 0.94 a<SUB>w</SUB>, respectively. However, its sporulation was some better at 0.97 a<SUB>w</SUB> than 0.995 a<SUB>w</SUB> or 0.94 a<SUB>w</SUB>, and better at 20℃ than 30℃. In contrast, the number of carbon sources(niche size) utilized by PENOX varied with aw and temperature. Under some water stress condition(0.95 a<SUB>w</SUB>), the agent utilized smaller number of carbon sources than 0.995 a<SUB>w</SUB> depending on temperature. The niche size at 0.995 and 0.95 aw were highest at 25℃, and showed 86 and 65, respectively. At 30℃, the niche size at 0.995 and 0.95 a<SUB>w</SUB> showed 84 and 50, respectively. There was no carbon source utilized by PENOX at 0.90 a<SUB>w</SUB> regardless of temperature. These information of the fungal ecophysiology will be useful for the effective development of BCA.
Caeoma makinoi에 의한 매실나무 꽃눈녹병의 발생 보고

Abnormal chloranthy was observed on Prunus mume trees grown in an orchard in Uiseong, Gyeongbuk province, Korea in 2015. Flower buds showing chloranthy were covered with numerous orange-colored sori, which were confirmed to contain spermogonia and aeciospores by stereo microscopy and light microscopy. When observed using scanning electron microscope, the aeciospores were globose, ellipsoid, or ovoid, and measured 22.8~35.6 × 19.8~36.8 μm, with a verrucae wall ornamentation. To construct a phylogeny, the large and small subunit regions of rDNA were sequenced and compared with those of allied species by the maximum likelihood method. The isolated fungus species showed high similarities with Caeoma makinoi (Teleomorph: Blastospora smilacis), which was previously isolated from P. mume in Japan. Therefore, we initially report this fungal agent parasitic on P. mume as C. makinoi in Korea and name the disease it causes as “flower bud chloranthy disease”.
항 Pseudomonas tolaasii 및 Pseudomonas agarici 항체의 면역측정법을 위한 평가와 그 이용

느타리버섯에 발생하여 갈색부패병을 일으키는 P. tolaasii 및 P.agarici 를 면역화학적 방법으로 신속하게 검출하기 위한 항체를 생산하였고 면역확산법과 ELISA 로 본 항체의 특이성을 조사하였다. $\alpha$-P tolaasii 항체는 P. tolaasii와 반응하여 선명한 침강선을 형성하지 않아 P. tolaasii 에 대하여만 특이적으로 반응하는 항체임을 알 수 있었다. 한편 $\alpha$-P. agarici 항체는 P.agarici뿐만 아니라 다른 세균에 대해서도 침강선을 형성하지 않는 특징을 나타내었다. 비경합 간접 ELISA의 표준곡선으로부터 P.agarici의 검출에 대하여 최고 발색치의 50% 갑을 나타내는 균의 밀도는 $2{\times}10^4$ cfu/ml로 나타났으며, 검출한계농도는 $2{\times}10^3$ cfu/ml 이었다.${\alpha}$-P. agarici 항체는 P. tolaasii의 특이적인 검출에도 효과를 나타냈다. ${\alpha}$-P. tolaasii, ${\alpha}$-P. agarici 항체를 이용하여 확립한 ELISA법은 직접 균체나 버섯 자실체내 P. tolaasii 와 P. agarici의 신속한 검출을 가능하게 하였고, P. tolaasii 와 P. agarici에 오염된 시료를 간단하게 진단하는데 아주 유용하게 사용될 수 있을 것으로 판단되었다. To produce antibodies against Pseudomonas tolaasii and P. agarici, lyophilized P. tolaasii and P. agarici ($5{\times}10^7$ cfu/ml) and Freund, s adjuvant were immunized into rabbits 4 times. The specificity and sensitivity of the antibodies were evaluated by immunodiffusion test and indirect enzyme-linked immunosorbent assay (id ELISA). The ${\alpha}$-P. tolaasii antibody was very specific only against P. tolaasii, while ${\alpha}$-P. agarici antibody was not specific and showed a high cross reactivity toward P. tolaasii with detection limit concentration of $2{\times}10^3$ cfu/ml. However, the cross reactivities of ${\alpha}$-P. agarici antibody toward the related species including P. reactans were very low. Our results showed that ${\alpha}$-P. tolaasii and ${\alpha}$-P. agarici antibodies against P. tolaasii and P. agarici, respectively, might be useful for rapid and simple detection of the causal agents of bacterial brown and yellow blotches in cultivated oyster mushrooms.
고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究)

고추 종자에서 Colletotrichum acutatum, C. coccodes, C.dematium 및 C. gloeosporioides 등 4종의 탄저병균(炭疽病菌)이 검출(檢出)되었다. 그중 C.dematium과 C.gloeosporioides가 높은 비율(比率)로 검출(檢出)되었으며, 감염율(感染率)이 높은 종자시료(種子試料)는 각각(各各) 최고 84% 및 28%의 감염율(感染率)을 나타내었다. 반면에 C.acutatum과 C.coccodes는 각각 1개의 종자 시료(試料)에서 1% 및 2%의 낮은 비율(比率)로 검출(檢出)되었다. 표준습지법(標準濕紙法)이 냉동습지법(冷凍濕紙法)이나 한천배지배양법(培地培養法)에 비하여 종자상의 탄저병균(炭疽病菌) 검출(檢出)에 효과적(效果的)이었다. C.dematium과 C. gloeosporioides의 종자내 감염부위(感染部位)를 조사(調査)하였던 바, 종피(種皮)뿐 아니라 종자내 깊숙이 배유(胚乳)와 자엽(子葉)에 까지 감염(感染)되어 있음이 확인되었으나 종피(種皮) 감염율(感染率)이 높았다. C.dematium과 C.gloeosporioides 감염종자(感染種子)를 한천배지와 살균토양에 옮겨 발아율과 유묘생육상황(幼苗生育狀況)을 조사한 결과, 감염종자(感染種子)는 종자부패(種子腐敗), 모잘록, 유묘고사(幼苗枯死) 및 자엽(子葉)과 하배축(下胚軸)의 변색(變色)을 초래하였다. 접종실험(接種實驗) 결과 C.acutatum은 고추의 적과(赤果)에 병원성(病原性)이 있었고 C.coccodes는 고추 적과(赤果)에 강한 병원성(病原性)이, 고추 잎에 약한 병원성(病原性)이 있었다. C.dematium은 고추 잎과 청과(靑果)에 강한 병원성(病原性)이 있었고, C.gloeosporioides 는 고추 잎과 청과(靑果) 및 적과(赤果)에 병원성이 있었다. 이들 4종의 종자전염성(種子傳染性) 탄저병균(炭疽病菌)의 고추 이외의 다른 작물(作物)에 대한 병원성도 조사하였다. Colletotrichum acutatum, C. coccodes, C. dematium and C. gloeosporioides were detected in seed samples of red pepper (Capsicum annuum). C. dematium and C. gloeosporioides were the predominant species, maximum seed infection of the species in some samples were 84% and 28%, respectively. C. acutatum and C. coccodes were recorded only in low percentages of 1-2. The blotter method proved more suitable for detecting Colletotricum spp. than the deep freezing blotter or agar plate methods. Plating of seed components showed that C. dematium and C. gloeosporioides were recovered more frequently from seed coat, and decreasing amounts of infection were observed in the endosperm and cotyledon. Seed-borne C. dematium and C. gloeosporioides caused seed rot, damping-off, seedling blight and brown discoloration of cotyledon and hypocotyl when infected seeds were sown in agar of test tube or in soil. Inoculation experiments showed that C. acutatum was pathogenic to red fruit of red pepper and C. coccodes was highly pathogenic to red fruit and weakly pathogenic to leaf of the plant. C. dematium was highly pathogenic to leaf and green fruit and C. gloeosporioides was pathogenic to not only leaf but also green and red fruits. Host range of the four seed-borne species of Colletotrichum was also investigated.
제주도와 남해 장사도 해변 등으로부터 분리, 선별된 국내 미기록 야생효모들의 균학적 특성

The present study aimed to screen unrecorded wild yeasts from Jeju lsland and Jangsado on the southern coast of Korea, and to investigate their microbiological characteristics. To date, Coniozyma leucospermi JJD37-2, Hanseniaspora thailandica JJD44-1, Kluyveromyces nonfermentans JJD15-1, Kockovaella fuzhouensis JJD47-3, Vishniacozyma heimaeyensis JJD8-4, Candida carpophila JSDH24-1, Wickerhamomyces strasburgensis JSDH34-2, Candida savonica HJD6-4, and Candida karawaiewii YP23-3 have not been previously recorded in Korea. In the present study, we investigated the microbiological characteristics of these previously unrecorded yeasts. Except for W. strasburgensis JSDH34-2 strain, none of the strains formed spores, and only the C. leucospermi JJD37-2 strain formed pseudomycelia. Almost all strains grew well in yeast extract-peptone-dextrose (YPD) medium, potato dextrose (PD) medium and yeast extract-malt extract (YM) media. C. carpophila JSDH24-1 and W. strasburgensis JSDH34-2 also grew well in YPD medium containing 10% NaCl. H. thailandica JJD44-1 is fermented to produce glucose, fructose and mannose.
