Monoclonal antibody against Interleukin-2 peptide was generated. One hundred four hybridomas which were survived in HAT media produced anti-IL2 antibody. Among 104 hybridomas, eleven clones has been identified to produce IgG1 type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgG1 monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weights are 55 kD and 25 kD, respectively. These IL-2 antibodies were confirmed using immunoblotting techniques. IgGl type IL-2 monoclonal antibody were bound to 133 amino acids IL-2 antigen whose M.W. is 15 kD. These monoclonal antibodies were used for purifying IL-2 peptides. IL-2 ELISA systems were generated using immuno-affinity purified IL-2 as antigens.

Low Intensity Exercise Suppresses Pathological Cardiac Hypertrophy Related Gene Expression in SHR

This study determined the effects of exercise training on cardiac function and myocardial gene expression, because we hypothesized that how exercise training prevents pathological hypertrophy in the left ventricle by modulation of myocardial genes. We used the spontaneously hypertensive rat (non-exercise SHR, n = 12) as a model of pathological hypertrophy and exercise-trained SHR rats (n = 12, treadmill exercise for 12 wk) as a model of physiological hypertrophy. We also used sedentary Wistar-Kyoto rats as the control group (n = 12). Exercise-trained SHR expressed clear adaptive changes such as reduced body weight, heart rate, blood pressures, and cardiac wall thickness. The mRNA expression of angiotensin converting enzyme (ACE), endothelin-1 (ET-1), brain natriuretic peptides (BNP) in the left ventricle was significantly higher in the non-exercise SHR than in the WKY and the exercise-trained SHR, whereas mRNA expression of caveolin-3 and eNOS in the left ventricle was significantly lower in the non-exercise SHR than in the WKY and the exercise-trained SHR. These findings suggest that exercise training prevents pathological hypertrophy in the left ventricle by modulation of myocardial genes. In conclusion, the exercise-trained SHR showed improved cardiac function, including reduced body weight, hemodynamic profiles, and cardiac wall thickness, as distinct from the pathological cardiac adaptation in the non-exercise SHR. Finally, exercise training prevents pathological cardiac hypertrophy in the left ventricle by altering myocardial genes.

이번 여행을 통해, 여러 각도에서 국제사회가 어떻게 변해가는 가를 주시하여야 할 것이라는 생각이 들었다. 특히 생명공학 분야에서 가까운 ㅇ리본 뿐 아니라, 중국 대만에서도 많은 연구가 이루어지고 있으니, 독불장군 style의 연구태도를 지양하여 국제화된 사회로서, 국제적인 새로운 지식의 교환으로서 좀 더 광범위한 빠른 지식의 장을 열 수 있으리라. 이런 면을 고려해 볼때, 이번 한.중 미생물학회는 서로의 지식을 교환할 수 있는 좋은 매개체 역할을 하였다 생각되며, 앞으로도 이런 기회가 자주 열렸으면 한다.

전국체육대회에 참가하는 재외동포의 세대 간 참가동기, 참가경험 및 정체성교섭에 관한 연구

This research aims to examine the difference in participation motivation, participation experience and identity negotiation between generations of overseas Koreans in the National Sports Festival(NSF). The subjects for this study were 360 overseas Koreans who took part in the 100th NSF held in Seoul in 2019. The results are as follows; First, while the first generation participated in the NSF because of visiting family and maintaining Korean identity, the second generation took part in the event for self-realization. Second, the second generation had more negative experiences than the second generation during the NSF. Third, while the first generation negotiated more identity with origin, the second generation had more identity with settlement. 본 연구는 전국체육대회에 참가하는 재외동포의 세대 간 참가동기, 참가경험 및 정체성교섭에 대한 차이를 실증적으로 분석하는데 그 목적이 있다. 이와 같은 연구의 목적을 달성하기 위해서 본 연구는 2019년 제 100회 전국체육대회에 참가한 재외동포를 모집단으로 설정하고, 실제 360명의 자료를 수집해 분석하였다. 이러한 연구의 목적과 연구방법을 토대로 얻어진 결과는 다음과 같다. 첫째, 전국체육대회에 참가하는 재외동포의 세대 간 참가동기 중 이주 1세대의 경우 가족방문과 정체성유지가 1.5세대나 2세대보다 높다. 하지만, 자아실현을 추구하기 위한 참가동기의 경우 이주 1.5세대나 2세대가 1세대보다 높다. 둘째, 전국체육대회에 참가하는 재외동포의 세대 간 참가경험은 두 집단 모두 긍정경험에서는 통계적으로 유의한 차이가 없었으나, 이주 1.5세대나 2세대가 1세대보다 전국체육대회 동안 부정적인 경험이 높다. 셋째, 전국체육대회에 참가하는 재외동포의 세대 간 정체성교섭은 이주 1세대의 경우 모국정체성이 1.5세대와 2세대는 거주국 정체성이 높다.

Effect of Urgency Symptoms on the Risk of Depression in Community-Dwelling Elderly Men

Purpose: To evaluate the association of a specific type of lower urinary tract symptom (LUTS) and the depression in community-dwelling elderly Korean men. Materials and Methods: A total of 392 men aged 65 years or older, who completed urological and psychiatric evaluations as a participant of the Korean Longitudinal Study on Health and Aging, were included. From each subject, an interview on the demographic characteristics and medical history, IPSS, and psychiatric questionnaire were taken. Subjects were divided into two groups; depression and euthymic. Subjects with IPSS subscore more than 3 points was considered ‘high’ subscore. IPSS subscores were compared between the two groups, and the relationship between depression and LUTS severity was assessed. Results: The mean age of the subjects was 75, and 6.4% of the subjects were diagnosed to have major depressive disorders. The depression group showed higher IPSS scores than the euthymic group (16.1±9.9 vs. 11.6±8.6, p=0.01). IPSS subscores of question 1 (incomplete empty), question 3 (intermittency), question 4 (urgency) and question 6 (straining to void) were higher in the depression group compared with the euthymic group. Chi-square test revealed subjects with high IPSS 1, 3, 4, and 6 score were associated with depression, but multivariate analysis identified only high IPSS question 4 as a significant prognostic factor for depression. Conclusions: Elderly population with depression is more likely to have more severe LUTS than population without depression. Among the urinary symptoms, urgency was strongly associated with depression. Patients with moderate to severe LUTS and especially urgency may need their mental health status evaluation.

Generation of Monoclonal Antibody Against Hepatitis B virus (HBV) Pre-S2 Peptide

대장균에서 재조합 발현된 Hepatitis B virus 표면항원 Pre-$S_2$ peptide로 면역시킨 Balb/c 마우스 비장세포와 SP2/O-Ag 14 미에로마 세포를 융합시켜 Pre-$S_2$에 대한 단일클론 항체를 생산하였다. HAT 선택배지에서 자란 70개의 융합세포군 중 Pre-$S_2$에 대한 항체를 생산하는 6개의 융합세포군을 제한 희석법으로 클로닝하였으며 ELISA 방법을 이용하여 이들 6클론을 isotype 한 결과 모두 kappa light chain을 갖는 IgM이었다. 이들 항체의 항원특이성은 western bolt를 이용하여 55 amino acid Pre-$S_2$ peptide와 결합함을 확인하였다. Monoclonal antibody against HBV pre-S2 peptide was generated. Six clones among 70 hybridomas which were survived in HAT media produced anti pre-S2 antibody. All six clones has been identified to produce IgM-type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgM monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weights are 79KD and 26.5KD, respectively. These pre-S2 antibodies were confirmed using immunoblotting techniques. IgM type pre-S2 monoclonal antibody were bound to 55 amino acid pre-S2 antigen whose M.W. is 55KD.

사회서비스 바우처 프로그램의 효과성 및 이용만족에 대한 영향요인: 스포츠바우처를 중심으로

The purpose of this study was to measure and find the determinants of the effectiveness and customer satisfaction of the sport voucher service in Korea. The data has been gathered from 20 basic local governments, 100 service providing institutions, and 400 voucher service customers. The study findings were synthesized as following. First, the effectiveness and the customer satisfaction were very high as compared to those of other service, therefore the voucher program can be evaluated as very successful. Second the market competitiveness and an important characteristic of voucher management greatly influenced the effectiveness and the customer satisfaction of the program. Third, the period of using the voucher significantly influenced the effectiveness of the program, but did not influence the customer satisfaction of the program. Fourth, profit organization showed higher effectiveness than non-profit organization, but it did not show any significant difference in terms of customer satisfaction. Fifth, the destitute group evaluated the effectiveness of the program lower than the other upper group. therefore the level of income influenced the program. Finally, the age did not influence the customer satisfaction and the service effectiveness.
