        쥐의 큰포식세포주에서 자가포식현상에 의한 Salmonella enterica serovar Typhimurium의 감염 조절

        자가포식현상(autophagy)은 세포 내 또는 세포 외의 스트레스나 영양분의 고갈, 그리고 병원체 감염에 의해 유도되는 기전으로, 병원균, 손상된 단백질이나 세포 소기관을 autophagosome으로 격리하여 리소좀(lysosome)과 융합하여 분해시키는 기전이다. Salmonella enterica serovar Typhimurium (S. Typhimurium)은 세포 내로 감염되는 세균으로 급성 위장염과 식중독을 야기한다. S. Typhimurium 감염 시 세포 내에서 자가포식현상이 유도되며 이는 감염을 제어하는데 중요하다는 연구 논문들을 통해 본 연구에서는 자가포식현상 유도제인 rapamycin으로 자가포식현상을 유도했을 때, S. Typhimurium의 감염을 조절할 수 있는지 알아보고자 하였다. 자가포식현상 유도제인 rapamycin과 저해제인 3-methyladenine(3-MA)를 각각 처리한 후 쥐의 큰포식세포주인 RAW 264.7 세포에 S. Typhimurium을 감염시켰다. rapamycin을 전처리한 후 S. Typhimurium을 감염시켰을 때, 세포 내에서 S. Typhimurium의 성장률이 감소한 반면 3-MA의 전처리는 S. Typhimurium의 성장을 촉진시켰다. 또한, RAW 264.7 세포에 rapamycin을 처리 후 감염시켰을 때, 자가포식현상 관련 단백질의 발현이 유의하게 증가하였다. Rapamycin에 의하여 유도된 자가포식현상이 활성산소종(reactive oxygen species, ROS)과 활성 산화질소종(nitric oxide, NO)의 생성을 통해 감염을 제어하는지를 확인하기 위하여 이 두 물질을 측정하였다. 감염 전 rapamycin 처리 시 RAW 264.7 세포에서 NO의 생성은 증가하였으나 ROS의 생성에는 별다른 차이가 없었다. 이상의 결과는 쥐의 큰포식세포주에서 rapamycin처리로 유도된 자가포식현상은 NO 생성을 통해 항박테리아능을 나타낸다고 할 수 있다. Autophagy is one of the lysosomal degradation pathways to maintain cellular homeostasis. The damaged proteins or organelles are uptaken through extra- and intra-cellular stress, starvation and infected pathogens, subsequently, autophagosomes are fused with lysosomes to break down the molecules. Salmonella enterica serovar Typhimurium (S. Typhimurium), intracellular bacteria, cause acute gastroenteritis and food poisoning. Given that autophagy induced by S. Typhimurium plays an important role in the cells to control the infection, we identify whether the induction of autophagy with rapamycin, chemical inducer of autophagy, before infection regulates S. Typhimurium infection. After treatment of rapamycin or 3-methyladenine (3-MA), autophagy inhibitor, RAW264.7 cells were infected with S. Typhimurium. Pretretment of rapamycin decreased the growth rate of S. Typhimurium in the cells; otherwise, pretreatment of 3-MA increased the growth rate of S. Typhimurium. The expression of autophagy-related genes was significantly increased in the S. Typhimurium-infected cells pretreated with rapamycin. To examine whether induced autophagy by rapamycin control the infection with increase the production of reactive oxygen species (ROS) and nitric oxide (NO), antibacterial radical substrates were measured in infected cells followed by the treatment with either rapamycin or 3-MA. NO production increased in RAW264.7 cells; otherwise, ROS production remained unchanged during the infection. These findings suggest that inducing autophagy with rapamycin reveals antimicrobial activity as producing NO against S. Typhimurium infection in mouse macrophages.

        우울증의 자기 관리 방법

        Objectives : Depression disturbs the individual life from the loss of productivity to suicide. Furthermore the whole society is also influenced by depression in social and economic areas. Many scientific treatment methods are known to be effective in depression. But in reality more than half of patients with depression prefer self help treatment rather than medically based treatment. For the patients with mild depression and primary physicians, it is reasonable to provide the evidence based self care including self help or alternative treatment. This study is supported by the Korean Health 21 R&D Project for the Depression Center to establish the Korean treatment guideline for depression. Methods : The members of Depression Center including the authors attended the workshops where the methods of evidence based medicine such as literature search, quality evaluation of the articles, making the level of evidence were educated. The professional librarians helped the authors for get the searched articles. Among the total of 354 abstracts of systemic review and 300 abstracts of random controlled trials, 3 systemic reviews and 2 random controlled trials were the main structure of evaluation. Result : Among the 37 self care methods known to be effective for depression, 11 kinds of methods were worth of evaluation. The first step for the self care of mild depression is to use St. John's wort, exercise, bibliotherapy, and light therapy in depression during winter season. Acupuncture, negative ion therapy, massage, relaxation technique are known to be efective with evidence. Music therapy, hypnotherapy, and aromatherapy are known to be effective but until now the evidences is insufficient and further evaluations are needed. Conclusion : Among 11 listed self-care methods, several methods need more evidence especially in Korean situation. Exercise is the most recommendable evidence based self care method in Korea nowadays. St. John's wort is effective with evidence, but it is not the OTC drug in Korea, so it is not recommendable now. For the bibliotherapy, there is no book such as "Feeling Good" in Korea. Only the introductory book about depression is available. Publishing the self help book based upon cognitive behavioral approach or psychodynamic approach is needed. Light therapy is practiced in a few clinics, but not self-help style. Exposure to the sunlight is recommended in the level of commen sense. These self care methods are for the mild patient. Moderate to severe patients with depression should seek the professional treatment.

        Clean technology of energy is increasingly becoming valid business propositions. Across the globe, growing demand, tight hydrocarbon supplies, high oil prices and environmental consciousness are rapidly driving solar technology into the energy of transportation energy. In the present study, to meet these requirements, eco friendly heater with heat pipes was designed and manufactured. Eco friendly heater consists of solar evacuated tube collector which a heat pipe installed inside and oscillating heat pipe in the condenser section. The solar thermal energy was collected in heat pipe installed into the evacuated tube. And the collected solar energy transferred to the oscillating heat pipe of condenser section through the conducting cube. In this study, experimental and theoretical study were carried out. Theoretical calculation showed that about 6.7  could be transferred to the room.

        폐절제술을 받은 폐암환자의 영양섭취 상태와 수술 후 폐합병증

        Purpose: The aim of this study was to examine the nutritional intake status of the lung cancer patients who underwent pulmonary resection and to analyze the relationship between the status of the nutritional intake and the occurrence of postoperative pulmonary complications. Methods: This study was a secondary analysis to determine whether the changes in the nutritional intake after surgery were related to pulmonary complications. Data of a total of 89 patients were included in the analysis and the nutritional intake status was confirmed using a 24-hour dietary recall method. The data were analyzed by descriptive statistics, chi-square or Fisher’s exact test, and ANOVA using the SPSS WIN 26.0 program and word clouds were generated using the R software program. Results: Overall, a decrease in the postoperative nutritional intake was observed in the patients who underwent pulmonary resection, except for the intake of fat. The pulmonary complications were identified to be associated with BMI and the presence of comorbidity. Twenty-three out of 74 patients with vitamin E levels below the Estimated Average Requirements developed pulmonary complications after surgery. Conclusion: Lung cancer patients who underwent pulmonary resection generally have difficulty in acquiring appropriate nutritional intake and need balanced nutritional management. Future investigations on the impact of increased vitamin E intake on postoperative pulmonary complications may provide better insight into the relationship between vitamin E intake and pulmonary complication among patients who underwent pulmonary resection.

      • Visual-FlaMap: PLS-DA 통계기법을 이용한 플라보노이드 프로파일의 웹기반 시각화 도구

        Databases related to polyphenols have been constructed and expressed on the website. These databases were provided valuable information such as phenolic compounds and their contents from food materials. Visual-FlaMap, a web-based application tool that visualized 3205 flavonoid contents from 268 of agro-foods was developed. The visualization of flavonoid profile was constructed as follow; 1) the metadata for systematic identification of flavonoids was collected from literatures 2) actual analytical data was produced by UPLC-DAD-QTOF/MS 3) the database was designed by Oracle software 4) both data were uploaded in the database using NutriManager. Results of this tool were expressed as a scatter plot using D3 chart, the PLS-DA algorithm designed using 'R' software for discriminant analysis on the basis of RDA flavonoids database. The designed algorithm clustered the sample set in to three primary groups; food type of ten classes (cereals, potatoes and starches, pules, nuts and seeds, vegetables, mushrooms, fruits, teas, wines, seasonings, and others), flavonoids type of seven classes (flavones, flavonols, flavanones, isoflavones, flavanols, chacones, and xanthones), and four levels of contents. Visual-FlaMap expected to be a powerful tool to provide considerable information on the distribution of flavonoids in diverse agro-food materials, to promote the utilization of these resources in food industry, and to support in developing competitive market.

      • UPLC-DAD-QToF/MS를 이용한 벚나무 속 과일의 페놀산 특성 비교

        Phenolic acids are widely distributed in plants. Especially, fruits may play a important role in preventing several diseases as phenolics rich source. In this study, phenolic composition and concentration of the Prunus genus were evaluated using ultra performance liquid chromatography with diode array detection and quadrupole time-of-flight mass spectrometric system (UPLC-DAD-QToF/MS). In positive ion mode, a total of 18 hydroxycinnamic acid derivatives (HCAs) and 1 hydroxybenzoic acid derivative were also identified and quantified from Prunus fruits. Among them, 15 phenolic acids were newly identified in the fruits of Prunus genus. One feruloyl acetylsucorse was also newly named from apricot: 3-O-feruloyl (2'-O-acetyl)sucrose (armenose A). Japanese apricot, apricot and cherry showed the highest amounts of HCAs. Overall, 3-O-caffeoylquinic acid was major phenolic acid in Prunus fruits. Various types of feruloyl acetylsucrose and p-coumaroyl acetylsucrose were identified in apricot and Japanese apricot, respectively. With previous studies, the catechin content was increased while flavonol content was reduced during the ripening stage of Japanese apricot, as harvest time late. It was also identified that HCAs content was also reduced from this research. It was considered that phenolic acids and flavonols were used for biosynthesis of catechins in maturing. On the basis of phenolic information, PLS-DA model showed different patterns by cluster in various genus and ripening stages.

      • 고온고압 처리에 따른 인진쑥(Artemisia capillaris Thunberg)과 쑥갓(Chrysanthemum coronarium L.) 내 Caffeoylquinic Acid 유도체의 변화

        Capillary wormwood (Artemisia capillaris Thunberg) and crown daisy (Chrysanthemum coronarium L.) belonging to Family Asteraceae have been regarded as traditional medicinal herbs for preventive effects against chronic diseases including vascular disease, diabetes mellitus, and obesity. These plants are known for major resource of diverse caffeoylquinic acid (CQA) derivatives and flavonoid glycosides. Thus, objective of this study was to observe effects on high thermal and pressure processing to their CQA derivatives from capillary wormwood and crown daisy. A total of twenty-six hydroxycinnamoylquinic acid (HCQA) derivatives including p-coumaroylquinic acids (21), CQAs (3), and feruloylquinic acids (2) were separated and identified using UPLC-DAD-QToF/MS. Among them, a total twelve of HCQA derivatives was newly reported. Capillary wormwood and crown daisy were presented great majority of 5-O-caffeoylquinic acid (chlorogenic acid) and 3,5-di-O-caffeoylquinic acid (isochlorogenic acid A). In addition, 3,5-di-O-caffeoyl-4-O-succinylquinic acid was predominant compound in crown daisy. The total contents of HCQA derivatives were tended to decrease slightly during high thermal and pressure processing. Especially, the CQA derivatives which were esterified at 5- position of quinic acid moiety including chlorogenic acid and isochlorogenic acid A were tended to convert 3- and 4- position of CQA derivatives during processing. Therefore, proper high thermal and pressure processing is possible to enhance active ingredients. Furthermore, it can give valuable information for future research of functional foods and drugs.

        내러티브 슈퍼비전의 다양한 방법과 국내 교육훈련 시사점

        본 연구의 목적은 내러티브접근의 교육훈련 방법에 관한 문헌고찰 연구를 통해 국내 내러티브 슈퍼비전 장면에 적용할 수 있는 여러 방안을 모색하는 것이다. 이를 위해 국내외 다양한 유형의 문헌 및 자료를 토대로 내러티브 치료자의 역량 개발에 초점을 둔 7가지 독특한 방법들(자가평가모형, 질문다시쓰기, 거꾸로 배우기, 상담치료문화 해체하기, 슈퍼바이저 면접, 외부증인실천, 내부증인실천)을 파악하고 본론을 통해 각각의 기대효과와 유용성에 대한 문헌검토 및 분석을 제시하였다. 결론에서는 내러티브 슈퍼비전의 핵심요소 및 국내 교육훈련 필요영역을 6가지(세계관의 전환, 내러티브 대화의 흐름 이해, 내러티브 대화 제어기술, 비지시적ㆍ비판단적 언어사용, 가능성 개방과 선택지 생성의 촉진, 다중 관점의 장려)로 설정하고, 본 연구에서 검토한 방법들의 국내 적용 시사점 및 관련 방안을 제안하였다. 한국 상담치료 임상 현장에서 내러티브접근에 대한 관심과 필요성이 많은 공감을 얻고 있는 상황에서 본 연구는 엄밀한 교육훈련과 효과적 슈퍼비전 방안을 적극 모색하는 데 의의가 있다. Purpose: The purpose of the study is to conduct a literature review on education and training of narrative therapy and discuss the implications for narrative supervision in Korea. Method: The author identified seven methods of training that can be used in enhancing narrative therapy competencies for practitioners: Self-evaluation model, reauthoring therapeutic questions, learning backwards, deconstruction of therapy culture, supervisor’s interview, and outsider and insider witness practices. Results: The analysis section illustrates each method in terms of purpose, background, and implementation. Sample practice questions or examples of work from the training are provided; ideas and challenges are also discussed regarding how to apply them in the Korean context. Conclusion: The author makes recommendations on how each of six core elements of narrative supervision can be addressed by the use of the seven training methods examined in this study.
