소 모색관련 MC1R 유전자의 SNP와 관련한 MGB probe에 기초한 real-time PCR을 이용한 한우육과 Holstein육의 판별

The melanocortin 1 receptor (MC1R) plays an important role in regulation of melanin pigment synthesis within mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat color variations within several mammalian species including cattle. To develope a rapid and accurate method for the identification of Hanwoo meat, we performed a single nucleotide polymorphism (SNP) analysis in Melanocortin 1 receptor (MC1R) gene using TaqMan$^{(R)}$ MGB probe-based real-time PCR. Two specific probes (one for Hanwoo and the other for Holstein and Black angus) were designed. At the 5' end of 2 TaqMan$^{(R)}$ MGB probes, 6-carboxyfluorescein (FAM) was labeled for Hanwoo, and VIC for Holstein and Black angus. As a result, Hanwoo samples showed FAM-positive signal only, whereas other samples showed VIC-positive. This result suggests that the TaqMan$^{(R)}$ MGB probe based real-time PCR technique would be very accurate, easy and reproducible method to discriminate between Hanwoo meat and Holstein/Black angus meat.

u-헬스케어 서비스는 언제, 어디서, 어떠한 상황에서도 사용자에게 적절한 서비스를 제공할 수 있어야 한다. 그러한 u-헬스케어 서비스를 제공하기 위해서는 센서로부터 수집되는 데이터들에서 사용자의 상황에 적절한 정보인지, 그리고 데이터들의 유효성과 중복성을 검사해야 한다. 또한 이렇게 수집된 정보들을 u-헬스케어 서비스를 제공하는 시스템에서 적절하고 효과적인 관리 및 분석이 되어야만 사용자에게 적절한 헬스케어 서비스를 제공할 수 있게 된다. 따라서 본 논문에서는 사용자에게 적절한 의료 서비스를 제공하는 u-헬스케어 시스템을 위하여 각종 상황정보를 검사하여 데이터를 추출하고, 이러한 데이터를 정형화하기 위한 모델링 기법을 제안한다.

In the previous experiment, we have transiently analyzed a 307 bp cis-acting regulatory element of HOXA-7 in transgenic mice (TG). Since it was sufficient to direct a reporter gene to be expressed at the specific region of day 12.5 post coitum (p.c.) embryo with a sharp anterior boundary, we generated transgenic mice line using the NM307 (307 bp/tk promoter/LacZ/SV40 poly A) construct in order to see the temporal expression pattern following embryonic development. Through polymerase chain reaction (PCR) with LacZ specific primers and genomic DNAs isolated from ear tissue, 4 transgenic mice (founder mice, F0) were obtained and 3 exhibited characteristic expression pattern. The expression detected as early as day 8.5 p.c. in the neuroectoderm and epiblast at the posterior region; from the closed neural tube around the region S10-12 to the adjacent posterior neural fold, neural groove and to the posterior epiblast along the anteroposterior axis. At day 7.5 p.c., however, LacZ expression was not detected. As embryo develops, the expression was restricted in the neural tube from S10 area to the end of tail of day 9.5 p.c. embryo, among which rather low level of expression was detected particularly in the region of S10-13, and the overall expression pattern continued through the following day. From day 11.5 to 12.5 p.c. weak spreading of the expression was detected to the anterior of S10 and a repression at the tail region. These results altogether indicate that the 307 bp is a temporal as well as a spatial regulatory element of HOXA-7 initiating the function at around day 7.5.~8.5 p.c. and continuing through mid gestation.

To provide such u-healthcare service must check that gathering the sensing data whether or not proper information in context of user, and that are inspected of validation and redundant. Also these data should manage and analyze through the u-healthcare service providing system. And then, can provide appropriate service to user. So, in this paper propose modeling method for well-formed data that various context-information check and then, extraction for u-healthcare system provide proper medical service to user.

Mycobacterium (M.) bovis, a member of the M. tuberculosis complex (MTC), is a re-emerging, zoonotic agent of bovine tuberculosis whose prevalence probably depends on variations in direct exposure to cattle and ingestion of raw milk. Accurate species differentiation of M. bovis and M. tuberculosis is needed to distinguish between human and zoonotic tuberculosis. This study successfully developed a loop-mediated isothermal amplification (LAMP) assay for rapid detection and differentiation of M. bovis and M. tuberculosis, however showed negative reactions in eight non-tuberculous mycobacteria (NTM) samples and ten other bacterial species. Sensitivity of this assay for detection of genomic M. bovis DNA was 10 fg/μl. And this assay successfully detected M. bovis in bovine clinical specimens. In conclusion, the LAMP assay is a simple and powerful tool for rapid detection of M. bovis in both pure bacterial culture and in clinical samples.

The purpose of this study was to determine the zoonotic diseases of stray dog and cat in Gwangju, Korea. We chose randomly 300 samples which is 265, dogs and 35, cats in the public animal shelter from March to August of 2013. The animals’ blood biochemistry values measured, and the out of normal values were 49.7% GOT, 36.3% GPT, and 78.0% GGT. The tested items were Dirofilaria immitis, Toxoplasma gondii, Brucella canis, Rabies virus. The positive results were 10% Dirofilaria immitis, 6.3% Toxoplasma gondii (antibody detected), 10% Rabies (antibody detected) but 0.0% in B. canis. The stray animals’ antibodyㆍantigen positivity take effect high from Mar.-May. Therefore, we propose that those diseases should be monitering and vaccinating in Korea.

The melanocortin 1 receptor(MC1R) encoded by the coat color extension gene(E) plays a key role in the signaling pathway of melanin synthesis. The primers for the amplification of bovine MC1R gene were designed based on a bovine MC1R gene sequence(GenBank accession no. Y19103). A size of 483bp(482bp for Hanwoo) was amplified by PCR, digested with HpaII restriction enzyme and electrophoresed in 1.5% agarose gel. When the amplified DNA product(483bp) was digested with HpaII restriction enzyme, Hanwoo meat showed a single band of 482bp, whereas two fragments of 325bp and 158bp were detected in Holstein, Angus and meat of Hanwoo/Holstein cross cow having back coat color phenotype, respectively. The results of this experiment indicate that new designed primers of bovine MC1R gene may be useful for identification of Hanwoo meat from Holstein, Black Angus and Hanwoo/Holstein cross cow meat.
