Effects of the Schisandra Fructus Water Extract on Cytokine Release from a Human Mast Cell Line

Schisandra fructus has been used for treatment of cough and thirst in Korea. However, its therapeutic mech-anisms remain largely unclear. To investigate the biological effect of Schisandra fructus water extract (SFWE), we examinedthe effect of SFWE on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced pro-inflammatorycytokine secretion in the human mast cell line HMC-1. HMC-1 cells were stimulated with PMA plus A23187 in the presenceor absence of SFWE. Tumor necrosis factor (TNF)-., interleukin 6 (IL-6), and granulocyte-macrophage colony-stimulatingfactor (GM-CSF) productions were measured by enzyme-linked immunosorbent assay and reverse transcription-polymerasechain reaction. Inhibitory I.B/nuclear factor-.B (NF-.B) expression was assessed by western blot. SFWE suppressed PMAplus A23187-induced TNF-., IL-6, and GM-CSF production in dose-dependent manners. Furthermore, SFWE inhibited I.Bdegradation and NF-.B nuclear translocation. These results suggest that SFWE inhibits the secretion of pro-inflammatory cy-tokines in HMC-1 cells through blockade of I.B degradation and NF-.B activation. Taken together, these findings may helpelucidate the mechanism of action of this medicine in the modulation of mast cell activation in inflammatory conditions.

Methicillin-resistant Staphylococcus aureus (MRSA) is a substantial contributor to morbidity and mortality. In search of a natural products capable of inhibiting this multidrug resistant bacteria, we have investigated the antimicrobial activity of brazilein (BRZ) isolated from Caesalpinia sappan L. (Leguminosae) against 8 different strains of Staphylococcus aureus (S. aureus). New antimicrobial activity was found using the minimum inhibitory concentrations (MICs), broth dilution as well as checkerboard method. Against the 8 strains, the minimum inhibitory concentrations of BRZ were in the range of 62.5-500μg/mL. From those results we performed the checkerboard test to determine the synergism of BRZ in combination with Hygromycin-b (HgB) against 4 strains. The combined activity of BRZ and HgB against 4 strains resulted in a fractional inhibitory concentrations index (FICI) ranging from 0.18-0.5. The effect of BRZ with HgB was found to be synergistic. We found that BRZ reduced the MICs of HgB. BRZ and HgB could lead to the development of new combination antibiotics against MRSA infection.

The span of life of human beings has lengthened because of medical progress and so on. Consequently, the old have begun to remark that quality of life (QOL) is as important as extension of the life expectancy. However, some diseases such as stroke, cancer induce decrease of QOL. In case of the stroke, it is followed by chronic headache and dementia as an aftereffect, causing serious decrease of QOL. Jodeungsan, traditional medicine, have been prescribed for headache and hypertension. Many clinical trials and laboratory experiments about pharmacological effects of Jodeungsan have been reported. Jodeungsan ameliorated chronic headache and improved recognitive deficit. Moreover, Jodeungsan lowered blood pressure in hypertensive condition which is one of a major cause of stroke. These effects of Jodeungsan on several diseases are partly attributed to antioxidant effect. This report reviews the pharmacological effect of Jodeungsan in the view of stroke-related diseases.

Objectives : The purpose of this research was to examine the oriental medical term 'Yukjin(in chinese, liùchén)' i.e. 'good long-term storage of the six oriental medicinal herbs'. Yukjin has meant the six sorts of drugs which were well known for the older they stored, the better efficacious they were. This research was carried out to obtain more conclusive information about the time and the reason of the giving a naming Yukjin, and to arrange Yukjin's properties, toxicities and processes. Methods : We investigated the documents recorded in the medical classics of Yukjin and did the current issues and recent experimental researches of these medicines. Results :1. The properties and flavors of Yukjin are pungent and bitter. When they were stored for a long time after gathering, their toxicities and stimuli were growing thinner, regarded as positive clinically with age and they should be used after processing. 2. The efficacies of Yukjin are not always in proportion to storage period. In case, they are stored too long, their efficacies and qualities may be deteriorated though volatility or pungent flavors are enfeebled. Conclusions : Consequently, toxicities and strong flavors of Yukjin can be reduced in various processes. So the traditional theory of long-term preservation is not necessarily retained. It may be desirable to store them for 1 to 3 years after harvest, to safen their toxicities and adverse reactions in the way of processing. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the various diseases.
