Embryonic stem cells are known to renew themselves through cell division and can be induced to develop into whole different specialized cell types. These characteristics make stem cells worthy of investigation as a source of cells for tissue engineering and cell therapy, but technical and ethical considerations impede the therapeutic application of this technology. Reprogramming of fibroblasts to a pluripotent state can be induced in vitro through ectopic expression of the four transcription factors Oct4, Sox2, c-Myc and Klf4. Many investigators showed that reprogrammed induced pluripotent stem cells are indistinguishable from those of ES cells in DNA methylation, gene expression and chromatin state. Advances in reprogramming cellular mechanisms are transforming biomedical research and offer new expectations on development and potential treatment of various diseases. This review focused on recent researches and developments of iPS and is to stimulate the research in an area of tissue engineering.

The effects of prostaglandin El [PGE1], testosterone, 17β-estradiol and indomethacin on the zinc flux rate across the jejunal segements isolated from rats of each sex were determined using the Ussing chamber technique. Addition of PGE1 [5.0 μM] to the segment bathing medium significantly stimulated the zinc flux rate from mucosa-to-serosa [J_(ms)] of the jejunal segements isolated from male rats and inhibited it in those from female rats. 17β-estradiol [10 nM] inhibited J_(ms) of jujunal segments isolated from male rats, but testosterone stimulated those from female rats. To confirm that testosterone stimulates and 17β-estradiol inhibits J_(ms), the effects of testosterone on the zinc flux rates of segments isolated from male rats and 17β-estradiol on those from female rats were determined. In those experiments, both testosterone and 17β-estradiol inhibited J_(ms) without affecting the zinc flux rate from serosa-to-mucosa [J_(sm)]. However, when rats were ovariectomized, both of these steroid hormones stimulated J_(ms). Interestingly neither PGE1 nor steroid sex hormones produced any effect on the J_(sm), although indomethacin stimulated the J_(sm) of segments from male rats. These results suggest that steroid sex hormones interact with PGs in influencing the intestinal zinc transport and that endogenous PGs and steroid sex hormones augment the effects of exogenous hormones and PGs.

Based on the finding arachidonic acid (AA) stimulates, and prostaglandin (PGE₂) inhibits calcium absorption in female rats in vivo, we examined whether or not arachidonic acid also stimulates calcium absorption in male rats in vivo as well as in vitro. When 1 ㎎ of AA was given to male rats two times via gastric intubation, 24 and 4 h before sacrifice, and 20 μCi of ^(45)Ca (10 μCi/㎎) 1 h before sacrifice, organ ^(45)Ca contents significantly decreased compared with control rats. When the same amount of AA and ^(45)Ca were given to female rats, organ ^(45)Ca contents significantly increased compared with control rats. When duodenal segments from the same experimental rat groups studied in vivo were mounted to Membrane Ion Transport Chambers to measure the net ^(45)Ca absorption rates in the male rats, there was no significant difference between the control rats and those treated with AA, while the net ^(45)Ca absorption rate was significantly higher for AA-treated female rats compared with control rats. In contrast, when 10 μM AA was added to the duodenal segment bathing medium in the Membrane Ion Transport Chamber experiments, the net ^(45)Ca absorption rate significantly decreased for male rats, but there was no discernible effect on female rats. These data demonstrate that the modulation of intestinal calcium absorption with AA added exogenously in vivo and in vitro is dependent on the sex of the animals. They also suggest that AA inhibits and AA metabolites other than PGE₂ stimulate the absorption of calcium, and these reactions are mediated by female sex hormones.

Calcium sulfate is widely used for bone grafts as binder and expander. It was also reported that carboxymethylcellulose(CMC) can improve chemical characteristics for clinical applications when it is used as binder of calcium sulfate and demineralized bone matrix(DBM) for particulate bone grafts. DBM is an osteoinductive material which has been used to reconstruct bone tissue in maxillocraniofacial surgery, periodontics, and orthopedics. In this study, the effects of CMC-based putties containing DBM and/or calcium sulfate hemihydrate were evaluated in rat skull defects. Radiographs and histological evaluation of defect area after a period of time showed healing progress and new bone appeared greater for the treatment of CMC-based calcium sulfate hemihydrate putty compared to the untreated defect. When DBM was added to the putty, the most prominent bone regeneration effect was observed. We conclude that fast-resorbing CMC-based calcium sulfate hemihydrate putty containing DBM is effective for healing from the defective bone. CMC-based calcium sulfate may be used as a carrier and binder of bone graft materials for the restoration of bony defects.

Cyclo(his-pro) is a metabolite of thyrotrophin releasing hormone(TRH). It may be involved in the regulation of insulin metabolism and stimulates intestinal zinc absorption. In order to test the real function of cyclo(his-pro), a large amount of enantiomeric pure cyclo(his-pro) is needed. Cis-(L,L)-cyclo(his-pro) and trans-(L,D) -cyclo(his-pro) were successfully synthesized.

The role of protein phosphorylation in UV induced apoptosis in cultured human preerythrocyte leukemia cells was examined. It showed that the UV induced cell death is occured by apoptosis. Cells with apoptotic body were observed by UV treatment. In addition to the morphological changes, UV treatment caused a rapid increase of cytoplasmic Ca^(++) concentration followed by DNA fragmentation. The increased digestion of DNA in apoptosis process is believed to be the results of increased nuclear endonuclease of which the activity is regulated by the protein phosphorylation.

The risk of coronary heart disease is linked to cholesterol levels. Previous studies have reported that the level of human plasma cholesteryl esters is influenced more by genetic factors than the level of free cholesterol. To assess the genetic nature and population differences, a serum cholesterol with free and esterified choleterol were measured from 22 pairs of twins who ranged from 12 to 26 in age. Thin layer chromatography was used to separate cholestaryl ester and free cholesterol from the rest of serum lipids. No signfcant contributions of genetic effects were found for any of cholesterol variables. Heterogeneity in variance for the total cholesterol was found in comparison of monozygotic versus dizygotic twins. This might indicated an association of cholesterol variations with twin types as reported previously.
