        Effects of 3,5,3'-triiodo-L-thyronine () on Sex Steroid Levels and Gonadal Development in Black Porgy, Acanthopagrus schlegeli

        웅성 선숙형 해산 어류인 감성돔에게 를 경구 투여하여 성스테로이드인 testosterone(T), 및 cortisol 수준의 변화와 생식소 발달을 조사하였다. 외인성 는 사육 60일째 감성돔의 혈장 T의 수준을 유의하게 상승시켰으나, 혈장 및 난모세포의 크기에는 영향을 미치지 못하였다. 또한 처리는 생식소 내 정소 발달과 정자 형성과정을 촉진시켰으며, 정자 방출 기간을 연장하는 것으로 나타났다. 를 처리한 실험구에서 60일째에 혈장 cortisol 수준 The objective of the present study was to investigate changes of sex steroid(testosterone: T and ), cortisol levels and gonadal development following treatment to protandrous black porgy, Acanthopagrus schlegeli. Exogenous was found to significantly stimulate the increase of T levels in plasma of black porgy after 60 days of treatment. However no effects of on levels and oocyte size were found. treatment resulted in stimulated spermatogenesis and testicular development in gonad and prolonged spermiation. Also, the levels of cortisol were significantly increased in the fish treated with as compared to control fish at 60 days. The results showed that exogenous had direct effect on the release of T and cortisol, thus seems to play, either directly or indirectly, an important role in the testis development of functional male black porgy.

        저염분 노출에 따른 감성돔( Acanthopagrus schlegelii ) 아가미의 Na+/K+-ATPase 활성 및 발현

        We investigated the branchial osmoregulatory response of black sea bream Acanthopagrus schlegelii to short-term (3-48 h) exposure to a hyposaline environment (5 psu). Gill Na+/K+-ATPase (NKA) activity was decreased after 3 h in fish transferred to 5 psu compared to salt water-acclimated (control) fish, but the level of activity returned to that observed in the control fish at 6 h after transfer. NKA activity increased significantly at 24 h after transfer, but it returned to the level observed in the control fish at 48 h after transfer. Immunohistochemical staining revealed that gill NKA was localized to chloride cells. The number of chloride cells tended to change in parallel with NKA activity. Substantial decreases in plasma Na+, Cl-, and osmolality were observed after 12 h of exposure to 5 psu; however, these parameters began to recover to the values detected in the controls at 24 h after transfer. In conclusion, our results suggest that black sea bream are able to adjust their osmoregulatory mechanisms to shift from hypo- to hyperosmoregulation within 6 h of exposure to a hypoosmotic environment.

        저산소 노출에 따른 감성돔(Acanthopagrus schlegelii)의생리학적 스트레스 반응

        The black seabream Acanthopagrus schlegelii is an important species for aquaculture in Korea. There are, however, no reports on the physiological responses to hypoxia in this species. The objective of this study was to evaluate the effects of acute hypoxia on the physiological stress responses (plasma cortisol as the primary response, and plasma glucose, hematocrit (Ht), hemoglobin (Hb), sodium, chloride, osmolality, AST, ALT and total protein as the secondary responses) of the fish. The experimental fish were exposed to 0.5 ppm dissolved oxygen (DO) in the seawater via two methods (progressive stepwise decline (Exp.Ⅰ), and direct decline (Exp.Ⅱ)). A highly significant increase was detected in plasma cortisol levels due to the hypoxia treatments, and fish in Exp.Ⅰ had hormone levels that were significantly higher than the fish in Exp.Ⅱ. For plasma glucose, there was no significant difference between the Exp.Ⅰ fish and the control fish, whereas Exp.Ⅱ fish showed significantly higher plasma glucose levels than Exp.Ⅰ fish and control fish. Ht values increased in both hypoxia treatments; however, Hb concentrations increased only in Exp.Ⅰ. Although plasma chloride levels were unaffected by acute hypoxia, plasma sodium and osmolality levels increased in Exp.Ⅰ. Progressive hypoxia (Exp.Ⅰ fish) increased plasma AST, ALT and total protein. These results suggest that the value and direction of changes to the investigated parameters can be used to determine the resistance of black seabream to acute hypoxia

        산천어 (Oncorhynchus masou masou) 정자의 냉동보존에 미치는 희석액과 동해방지제의 영향

        We experimentally determined the physico-chemical properties of seminal plasma as well as the sperm cryopreservation techniques of masou salmon, Oncorhynchus masou masou. Seminal plasma contained 18±1 mmol/L potassium, 144±4mmol/L sodium, 116±3mmol/L chloride, 83.2±3.1mg/L calcium, 14.8±0.7mg/L magnesium, 45±9mg/L glucose, and 1.0±0.0g/L total protein. The osmolality and pH of seminal plasma were 287±7 and 7.7±0.1mmol/kg, respectively, and the spermatocrit was 28±2. The rate of embryonic survival at the eyed-stage and the hatching rate were highest in 10% methanol with 300mM glucose. Compared to DMSO or glycerol, methanol served as a better cryoprotectant of masou salmon sperm.

        선발 육종넙치 Paralichthys olivaceus 및 일반넙치의 성장비교

        To estimate the effect of selective breeding on the improvement of growth in olive flounder Paralichthys olivaceus, we compared the growth of a strain selected for rapid growth to that of an unselected strain from a commercial hatchery. The fish strains were fed with either moist pellets (MP) (dry matter, 59.8% crude protein; 14.1% lipid) or extruded pellets (EP) (dry matter, 50.4% crude protein; 13.8% lipid) for 190 days and were reared under similar conditions. The mortality rates were less than 2% and were not significantly different among the experimental groups. The growth rate of the selected fish was significantly greater than that of the unselected fish regardless of the diet type, and both strains fed MP grew significantly faster than those fed EP. The selected fish consumed more feed than the unselected fish. However, there was no significant difference in the feed efficiency between the selected and unselected fish. These results demonstrate that the selected fish exhibited superior growth rates, and that this was associated with a greater intake of food. Thus, selective breeding may be useful for improving the growth of commercial olive flounder.

        감성돔, Acanthopagrus schlegeli 의 담수순화 과정에서 나타나는 생리학적 반응

        해산어류의 담수순화 양식을 위한 기초 자료를 얻고자, 단계적인 염분변화의 방법을 이용하여 감성돔을 해수에서 담수로 순화시켰을 때 여러 생리학적 반응을 조사하였다. Cortisol 농도는 실험개시시에 13.0±14.8ng/㎖로 유의하게 증가하였지만 실험개시 10일째에 19.7±1.6ng/㎖로 실험개시시 수준으로 회복되었다. Glucose 농도는 cortisol과 동반 상승하는 경향이 나타났다. Na^(+) 및 Cl^(-)는 실험개시시에 각각 176.7±3.8mEq/L, 152.7±1.5mEq/L였던 것이 실험개시 5일째 111.8±38.0mEq/L, 73.5±26.5mEq/L로 유의하게 감소하였으나, 실험종료시에는 실험개시시 수준으로 회복되었다. 삼투질 농도 또한 이와 비슷한 경향을 나타내었다. AST는 실험개시 후 12시간째에 증가하였지만 그 이후 실험개시시 수준으로 감소하였다. Het, RBC 및 Hb는 실험기간동안 유의한 변화가 관찰되지 않았다. 생존율을 실험구에서 실험개시 4일째에 90%로 감소하기 시작하여 실험개시 5일째에는 86.7%, 실험개시 20일째에는 83.3%로 하락하였으나, 대조구에서는 실험종료시인 20일째에 96.7%의 생존율을 보였다. 이상의 결과로 종합해 볼 때 본 연구는 단계적인 염분변화를 통한 감성돔 담수순화 양식의 가능성이 있을을 알 수 있었다. Physiological responses (cortisol, glucose, Na^(+), Cl^(-), osmolality, aspartate aminotransferase (AST), alanine aminotransferase (ALT), hematological factors, and survival rate) were investigated during freshwater acclimation of seawater-cultured black porgy (Acanthopagrus schlegeli). As control, in seawater, there were no significant differences in the investigated items during the experimental period. In step-salinity change, plasma cortisol level was significantly increased from 13.02 1.4 ng/mL at the beginning to 29.52 14.8 ng/mL at 5 days, and recovered to the basal level (19.7±1.6 ng/mL) at 10 days. Glucose showed a tendency to co-increase with cortisol. Plasma Na^(+) and C1^(-) concentrations decreased from 176.7±3.8 and 152.7±1.5 mEq/L to 111.8f38.0 and 73.5k26.5 mEq/L, respectively, at 5 days. But these recovered as completely as the basal levels at the end of the experiment (20 days). Also, changes of osmolality levels showed a similar tendency with Na^(+) and C1^(-) levels. Although AST was markedly elevated at 12 hours, its level soon recovered to the basal level. No significant differences in Hct, RBC and Hb were found during the experimental period. Survival rate was 86.7% at 5 days and 83.3% at 20 days. The results presented in this paper suggest that it may be possible to culture black porgy in freshwater.
