Rapid, sensitive and specific detection of Brucella canis by real-time PCR assay using HybProbe

We designed and evaluated a specific HybProbe set for identification of Brucella canis, which is based on a specific single nucleotide polymorphism (SNP) of the omp25 gene. Real-time PCR assay using this probe specifically identified B. canis reference and field strains, however, the other Brucella species and general bacteria strains were not amplified. This real-time PCR assay can re-confirm specific amplification in melting curve analysis. The average melting temperature values for generation of B. canis strains wan 67℃m, due to a single base-pair mismatch at the binding site of the probe. The detection limit appeared to be 30 fg of DNA and 100CFU by serial dilution of whole blood inoculated with B. canis. In addition, use of the buffy coat resulted in approximately 50 times higher sensitivity than whole blood directly. This technique is widely applicable for use in diagnostic laboratories and is readily discriminative with regard to small amounts of biomass.

The pantograph for Korean High Speed Train was developed by home-grown technology. In this study, test and evaluation of the current collection performance of the pantograph is conducted. For this purpose, a measuring system is developed and installed on the prototype high speed train. Measurement is conducted while the train runs on the test track. The measuring system is composed of video monitoring system and telemetry & data processing system. It monitors the hazard behavior of the pantograph and measures acceleration and vertical force of the pan head. By applying the measuring system, accurate evaluation of the performance of the pantograph and safety assessment of the interface system of pantograph and catenary is facilitated.
Fabrication of conductive polymer-based nanofiber scaffolds for tissue engineering applications.

<P>Natural and synthetic polymers, in particular those that are conductive, are of great interest in the field of tissue engineering and the pursuit of biomimetic extracellular matrix (ECM) structures for adhesion, proliferation, and differentiation of cells. In the present study, natural chitin and conductive polyaniline (PANi) blended solutions were electrospun to produce biodegradable and conductive biomimetic nanostructured scaffolds. The chitin/PANi (Chi-PANi) nanofibrous materials were characterized using field emission scanning electron microscopy, Fourier transform-infrared spectroscopy, wettability analysis, mechanical testing, and electrical conductivity measurements using a 4-point probe method. The calculated electrical conductivities of the PANi-containing nanofiber scaffolds significantly increased as the amount of PANi increased, reaching 5.21 0.28 x 10(-3) S/cm for 0.3 wt% content of the conducting polymer. In addition, the viability of human mesenchymal stem cells (hMSCs) cultured on the Chi-PANi nanofiber scaffolds in vitro was found to be excellent. These results suggest that the Chi-PANi nanofiber scaffolds have great potential for use in tissue engineering applications that involve electrical stimulation.</P>

There is a critical developmental period during which brain sexual differentiation proceeds irreversibly under the influence of gonadal hormone. Recently, kinesin superfamily-associated protein 3 (KAP3) gene expressed during the 'critical period' of rat brain differentiation was identified by us (Choi and Lee, 1999). KAP3 functions as a microtubule-based motor that transports membranous organelles anterogradely in cells, including neurons (Yamazaki et al., 1996). mRNA level of KAP3 gene markedly increased before the initiation of puberty. Neonatal treatment of estrogen clearly inhibited the prepubertal increase in KAP3 mRNA level (Choi and Lee, 1999). In the present study, we aimed to investigate the effects of polychlorinated biphenyls (PCBs), as endocrine disruptors (EDs) on the expression of KAP3 gene during the 'critical period' of rat brain development. In our data, PCBs significantly decreased the expression of KAP3 gene in the fetal (day 17) and the neonatal (day 6 after birth in) male and female rat brains. The body weight and the breeding ability were significantly decreased in the PCBs-exposed rats compared with the control. These results showed that PCBs affect the transcriptional level of brain sexual differentiation related gene, KAP3, in the fetal and the neonatal rat brains. The maternal exposure to the PCBs may lead to toxic response in embryonic brain sexual differentiation and breeding ability after sexual maturation. This study indicates that KAP3 gene may be useful as a gene marker to analyze the molecular mechanism of toxic response in the animal brain development and sexual maturation exposed to PCBs.

At present, the most widely accepted explanation for these changes is that they result from a slowing in central information process stage. The purpose of this study was to examine the differences in RT and MT according to age of programming process. The subjects were at the age of 7, 11, 20, the task was about simple choice reaction and the apparatus was multipurpose reaction timer. This study established the research hypothesis to be difference between age and programmingprocess on RT and MT. Analizing the affects of RT and MT according to age of programming process revealled that RT and MT decrease as the age becomes higher. The results can be summerized as follows. First, Reaction Time decresed as the increase of Age [F (2,21)=113.84, p<.0001] second, Movement Time decresed as the increase of Age.[F(2,21)=40.00, p<.0001]
Dimethylformamide(DMF)에 의한 전격성 간염 1예

Dimethylformamide (DMF) is widely used as an organic solvent in chemical industry. The effects of DMF exposure in human are not well documented. Complaints of abdominal pain, nausea, vomiting, headache, and hepatotoxicity have been reported in workers exposed to DMF. We report a patient with fulminant hepatitis due to the solvent DMF. DMF handling was his major job. He complained of general weakness and showed jaundice. He suffered from hepatic coma due to fulminant hepatitis after admission. The possibility of viral hepatitis, drug induced and alcoholic hepatitis were ruled out from his history and serological study data. We couldn't find any cause of death except fulminant hepatitis from DMF toxicity.
