Eleven compounds, including a new sesquiterpene, were isolated from the culture medium of Aspergillus oryzae incubated with capsaicin. The structure of the new compound was determined to be 1,3,5a,6,7,8,9,9a-octahydro-9-hydroxy-9-(hydroxymethyl)-6-isopropyl-1-oxobenzo[c]oxepine-4-carboxylic acid, a heptelidic acid derivative. In addition, 10 known compounds were identified, namely 5-(hydroxymethyl)-3-furancarboxylic acid (flufuran), 3-hydroxypropanoic acid, 5-(hydroxymethyl)-2-furancarboxylic acid, 2-(4-hydroxyphenyl)-ethanol, 4-hydroxybenzoic acid, vanillic acid, 3,4-dihydroxybenzoic acid, 2-furanol, hydroheptelidic acid, and trichoderonic acid A, using spectroscopic data from nuclear magnetic resonance and electrospray ionization-mass spectroscopy.

<P>We report extremely small-sized drug-carrying globular poly(ethylene glycol) particles. These particles were prepared using fullerene (C-60) as a backbone structure and poly(ethylene glycol) as a hydrophilic shell. All - carbon bonds in C-60 were combined with poly(ethylene glycol), which form a globular nano-cage with a hollow core (originating from the soccer-ball-shaped truncated icosahedron of C-60) and the poly(ethylene glycol) shell. Subsequently, we constructed chlorin e6-conjugated globular poly(ethylene glycol). The obtained globular poly(ethylene glycol)-chlorin e6 (average 3.6nm in diameter) was soluble in aqueous solution and enabled improved singlet oxygen generation. The preferential cellular uptake of globular poly(ethylene glycol)-chlorin e6 resulted in significant enhancement of in vitro or in vivo photodynamic tumor cell ablation under light illumination. Our approach offers a versatile strategy to create extremely small-sized drug carriers using a biocompatible polymer for various biomedical applications.</P>

<P>A protein synthesis enzyme, leucyl-tRNA synthetase (LRS), serves as a leucine sensor for the mechanistic target of rapamycin complex 1 (mTORC1), which is a central effector for protein synthesis, metabolism, autophagy, and cell growth. However, its significance in mTORC1 signaling and cancer growth and its functional relationship with other suggested leucine signal mediators are not well-understood. Here we show the kinetics of the Rag GTPase cycle during leucine signaling and that LRS serves as an initiating 'ON' switch via GTP hydrolysis of RagD that drives the entire Rag GTPase cycle, whereas Sestrin2 functions as an 'OFF' switch by controlling GTP hydrolysis of RagB in the Rag GTPase-mTORC1 axis. The LRS-RagD axis showed a positive correlation with mTORC1 activity in cancer tissues and cells. The GTP-GDP cycle of the RagD-RagB pair, rather than the RagC-RagA pair, is critical for leucine-induced mTORC1 activation. The active RagD-RagB pair can overcome the absence of the RagC-RagA pair, but the opposite is not the case. This work suggests that the GTPase cycle of RagD-RagB coordinated by LRS and Sestrin2 is critical for controlling mTORC1 activation, and thus will extend the current understanding of the amino acidsensing mechanism.</P>

Purpose: Primary ciliary dyskinesia (PCD) is a genetically heterogeneous disorder that leads to secondary ciliary dysfunction. PCD is a rare disease, and data on it are limited in Korea. This study systematically evaluated the clinical symptoms, diagnostic characteristics, and treatment modalities of pediatric PCD in Korea. Methods: This Korean nationwide, multicenter study, conducted between January 2000 and August 2022, reviewed the medical records of pediatric patients diagnosed with PCD. Prospective studies have been added to determine whether additional genetic testing is warranted in some patients. Results: Overall, 41 patients were diagnosed with PCD in 15 medical institutions. The mean age at diagnosis was 11.8 ± 5.4 years (range: 0.5 months-18.9 years). Most patients (40/41) were born full term, 15 (36.6%) had neonatal respiratory symptoms, and 12 (29.3%) had a history of admission to the neonatal intensive care unit. The most common complaint (58.5%) was chronic nasal symptoms. Thirty-three patients were diagnosed with transmission electron microscopy (TEM) and 12 patients by genetic studies. TEM mostly identified outer dynein arm defects (alone or combined with inner dynein arm defects, n = 17). The genes with the highest mutation rates were DNAH5 (3 cases) and DNAAF1 (3 cases). Rare genotypes (RPGR, HYDIN, NME5) were found as well. Chest computed tomography revealed bronchiectasis in 33 out of 41 patients. Among them, 15 patients had a PrImary CiliAry DyskinesiA Rule score of over 5 points. Conclusions: To our knowledge, this is the first multicenter study to report the clinical characteristics, diagnostic methods, and genotypes of PCD in Korea. These results can be used as basic data for further PCD research.

Poor embryo quality and low blastocyst formation have been major limitations in establishment of cloned embryonic stem cells and production of cloned animals through somatic cell nuclear transfer (SCNT). Aggregation of embryos is a promising method for improving developmental competence of blastocysts. The aim of this study was to improve the blastocyst formation and the quality of parthenogenetic (PA) pig embryos by the aggregation of blastomeres at the 4-cell stage that were cultured in various type of culture dishes with or without phytohemagglutinin (PHA). The PA embryos were produced by the general method of our laboratory. On Day 2 after PA, the zona pellucida of 4 cell-stage embryos were removed by treatment with 0.5% (wt/vol) pronase solution. The 3x zona-free blastomere (ZFB) were randomly distributed in each of the following treatments for aggregation. ZFB were cultured for 5 days at 39℃ in an atmosphere 5% CO2, 5% O2, and 90% N2. In Experiment 1, effect of culture dishes on the aggregation efficiency and developmental competence of PA embryos were investigated. ZFB were cultured on non-coated (control) culture dish or dishes coated with 1% (wt/vol) agarose substrate (AS) or Well of the Well in dishes coated with 1% (wt/vol) agarose substrate (WAS). The ZFB cultured in WAS showed significantly higher (P<0.05) aggregation (81.2%) than AS and control (21.6-45.5%). The mean cell number in blastocysts derived from AS and WAS (81.4-89.3 cells/blastocyst) was significantly higher (P<0.05) than that of control (63.8 cells/blastocyst). In Experiment 2, effects of 150 ug/ml PHA treatment on the aggregation efficiency and developmental competence of embryos were investigated. The ZFB cultured in AS with PHA showed a higher (P<0.05) aggregation rate (90.0%) than that in AS without PHA, control with PHA, and control (39.2%, 57.9% and 17.5%, respectively). In conclusion, aggregation of porcine ZFB treated with PHA and agarose substrate could be a useful technique for producing improving blastocyst development with increased mean cell number of blastocysts in pigs.

AI World Cup 환경은 다수 가상 에이전트들이 팀을 이뤄서 서로 상호작용하며 대전이 가능한 가상 축구 환경이다. 본 논문에서는 AI World Cup 환경에서 멀티 에이전트기반 학습/추론 기술을 사용하여 다양한 전략과 전술을 구사하는 가상 축구 에이전트 구현과 시뮬레이션 결과를 소개한다. 먼저, 역할을 바탕으로 협동하여 상대방과 대전할 수 있는 논리 기반 추론형 멀티 에이전트 기술이 적용된 Dynamic planning 축구 에이전트 9 세트를 구현하였다. 이후, 강화학습 에이전트 기반, 단일 에이전트를 조합한 Independent QLearning 방식의 학습형 축구 에이전트를 구현한 후, 이를 멀티 에이전트 강화학습으로 확장하여 역할 기반 전략 학습이 가능한 가상 축구 에이전트를 구현하고 시뮬레이션 하였다. 구현된 가상 축구 에이전트들 간 대전을 통해 승률을 확인하고, 전략의 우수성을 분석하였다. 시뮬레이션 예제는 다음에서 확인할 수 있다(https://github.com/I-hate-Soccer/Simulation).

A 9-year-old, intact female, Pungsan dog was referred for purulent vaginal discharge and depression. Abdominal radiographs revealed gas-filled and soft tissue opacity tubular structures in the mid to caudal abdomen. On ultrasonography, fluid-filled uterus and cervix accompanied by hyperechoic foci and reverberation artifacts were identified. Multiple hyperechoic foci were found within the uterine wall, indicating gas content. Escherichia coli was isolated from aerobic and anaerobic bacterial cultures. Radiological differential diagnosis of tubular shaped, dilated gas-filled structure, and gas in the wall of the structure should consider emphysematous pyometra with endometrial pneumatosis in intact female dogs with vulvar discharge.
