Shiga toxin 2 (Stx2) is a major virulence factor for enterohemorrhagic Escherichia coli (EHEC), which is encoded by λ lysogenic phage integrated into EHEC chromosome. Stx2A1, A1 subunit of Stx2 toxin has gathered extensive concerns due to its potential of being developed into a vaccine candidate. However, the substantial progress is hampered in part for the lack of a suitable in vitro expression system. Here we report use of the prokaryotic system pET-28a::espA-Stx2A1/BL21 to carry out the fusion expression of Stx2A1 which is linked to E. coli secreted protein A (EspA) at its N-terminus. Under the IPTG induction, EspA- Stx2A1 fusion protein in the form of inclusion body was obtained successfully, whose expression level can reach about 40% of total bacterial protein at 25°C, much higher than that at 37°C. Western blot test suggested the refolded fusion protein is of excellent immuno-reactivity with both monoclonal antibodies, which are specific to EspA and Stx2A1, respectively. Anti-sera from Balb/c mice immunized with the EspA-Stx2A1 fusion protein were found to exhibit strong neutralization activity and protection capability in vitro and in vivo. These data have provided a novel feasible method to produce Stx2A1 in large scale in vitro, which is implicated for the development of multivalent subunit vaccines candidate against EHEC O157:H7 infections.

This paper proposes a novel zero-voltage-switching (ZVS) Push-pull DC-DC Converter for high input voltage and high power applications. This topology utilizes two switches in series to replace one switch in conventional push-pull converter, and two clamping diodes are introduced. The voltage stress of the switches is the input voltage, and the switches can realize ZVS with the use of the leakage inductance of the transformer. Furthermore, secondary full-wave rectifier with a clamping capacitor is used to eliminate the voltage oscillation and spike of the rectifier diodes due to the reverse recovery. Therefore, the electromagnetic interference is reduced effectively. The operation principle of the proposed converter is analyzed theoretically. The output characteristic, ZVS condition and design principle of the clamping capacitor are discussed. Experimental results obtained from a 270V input 2kW prototype with $95.8\%$ high efficiency confirms the design.

Objective: This study was conducted to investigate the effects of weaning times on the growth performance, rumen fermentation and microbial communities of yellow cattle calves. Methods: Eighteen calves were assigned to a conventional management group that was normally weaned (NW, n = 3) or to early weaned (EW) group where calves were weaned when the feed intake of solid feed (starter) reached 500 g ($EW_{500}$, n = 5), 750 g ($EW_{750}$, n = 5), or 1,000 g ($EW_{1,000}$, n = 5). Results: Compared with NW, the EW treatments increased average daily gain (p<0.05). The calves in $EW_{750}$ had a higher (p<0.05) starter intake than those in $EW_{1,000}$ from wk 9 to the end of the trial. The concentrations of total volatile fatty acids in $EW_{750}$ were greater than in NW and $EW_{1,000}$ (p<0.05). The EW treatments decreased the percentage of acetate (p<0.05). The endogenous enzyme activities of the rumen were increased by EW (p<0.05). EW had no effect on the number of total bacteria (p>0.05), but changes in bacterial composition were found. Conclusion: From the present study, it is inferred that EW is beneficial for rumen fermentation, and weaning when the feed intake of the starter reached 750 g showed much better results.

There are a large number of concrete bonding interfaces in assembled concrete structures. Moreover, the problems of deformation and coordination at the interfaces lead to weak bonding under shrinkage and creep. The theoretical methods and analytical solutions for the long-term performance of composite structures in recent literatures are relatively complicated for the review or optimization in the design stage. Hence, a method for calculating the mid-span deflection of and the cross-sectional stress on composite beams under shrinkage and creep was developed based on the average curvature method. The calculation method was then verified by the experiments and the numerical simulations. Further, factors affecting the deflection of and the stress on the assembled concrete composite beams, including the loading age, the reinforcement ratio, the section dimensions, and the surface area in contact with the atmosphere were analyzed by using the developed method. The results revealed that the adjustment of the section dimensions and the loading time is the effective optimization approach. A framework for review or optimization of composite concrete beams based on the sensitivity analysis is proposed and a case study is carried out. The results of this work can helpfully be used to control the risk of additional deflections or cracking during the long-term operation of buildings.

A typical three-dimensional finite element model for a fuel assembly is established, which is composed of 16 monolithic U-10Mo fuel plates and Al alloy frame. The distribution and evolution results of temperature, displacement and stresses/strains in all the parts are numerically obtained and analyzed with a self-developed code of FUELTM. The simulation results indicate that (1) the out-of-plane displacements of Al alloy side plates are mainly attributed to the bending deformations; (2) enhanced out-of-plane displacements appear in fuel plates adjacent to the outside Al plates, which results from the occurred bending deformations due to the applied constraints of outside Al plates; (3) an intense interaction of fuel foil with the cladding occurs near the foil edge, which appears more heavily in the fuel plates adjacent to the outside Al plates. The maximum first principal stresses in the fuel foil are similar for all the fuel plates and appear near the fuel foil edge; while, the through-thickness creep strains of fuel foil in the fuel plate near the central region of fuel assembly are larger, and the induced creep damage might weaken the fuel skeleton strength and raise the fuel failure risk.

This paper deals with the model reference tracking control problem of continuous-time periodic linearsystems when the actuator occurs jumping fault. The main contribution is to formulate the parametric designalgorithm for the systems by utilizing the parametric solution of the generalized Sylvester matrix equations. Theexistence condition of the controller is deduced based on the Lyapunov stability theory. The controller consistsof the additive contribution of two terms: a feedback term and a feedforward term. The feedback term is thefeedback control law which can stabilize the system with finite expected cost. The feedforward term is the completeparametric feedforward tracking compensator. The simulation for flying around mission is carried out about twospacecrafts in elliptical orbit. The simulation results show the effectiveness of the proposed approach.

Aim: Tanscatheter arterial embolization irrespective of with or without an anticancer agent and lipiodol has been controversial with regard to survival benefit. Therefore, we conducted a prospective study to analyze the effect of transcatheter arterial lipiodol chemoembolization (TACE) on the survival of HCC. Methods: A prospective study was conducted, and a total of 326 patients with primary liver cancer who were newly diagnosed were collected from January 2004 to January 2005 in Zhejiang Provincial People's Hospital of China. A univariate Cox's regression analysis was used to assess the survival of the HCC cases receiving TACE. Results: The duration of follow-up for the HCC patients treated with TACE ranged from 3 months to 60 months. For the overall patients, survival rate at 5 years was 42%. Both HBV Ag and HCV Ab positive patients showed significantly low survival rate at 5 years. The multivariate analysis revealed The IV TNM stage was related to an heavy increased risk of death of HCC patients, and Child C grade group showed a significant moderate increased risk. Conclusion: Our study showed TACE is associated with a better prognosis of HCC patients, and the HBV infection, TNM stage, Child-Pugh grade and number of TACE may influence the survival probability. Further TACE studies should be assess the quality of life of HCC patients, so as to provide more information for treatment of HCC.

The chigger mite Leptotrombidium sialkotense is one of the 6 main vectors of scrub typhus in China. Before present study, L. sialkotense was found in some parts of Hunan province, China with a narrow geographical distribution. During field investigation 2016-2017, we found L. sialkotense in Jingha, southern Yunnan, China. Of 15 small mammal host species, L. sialkotense were collected from 6 species of the hosts. Rattus brunneusculus was a dominant host of L. sialkotense, from which 98.3% of the mites were collected. The chigger mite showed a relatively high infestation prevalence (PM=11.7%) and mean abundance (MA=0.5) in comparison with the rest 5 host species. These results reveal a certain host specificity of L. sialkotense to a rat R. brunneusculus. The mite L. sialkotense showed an aggregated distribution on the host (P<0.05). A positive correlation observed between L. sialkotense and the body length of hosts. There was a positive interspecific association between L. sialkotense and 2 other dominant vectors, L. deliense and L. scutellare.
