      • Illudins C_2 and C_3, New Illudin C Derivatives from Coprinus atramentarius ASI20013

        Ganoderma lucidum 균주는 인공 배지 상에서 광에 의하여 자실체 원기가 형성되었으며, 광질이 자실체 원기 형성에 미치는 영향을 조사한 결과, 자실체 원기 형성능을 가지고 있는 5균주중 3균주는 공시한 모든 형광등의 아래에서 즉, BLB, 순청색, 순녹색, 순황색, 순적색 형광등의 아래에서 자실체 원기가 형성되었고, 2균주는 BLB 형광등을 제외한 가시광선 영역에서 형성되었다. 그러나 암상태에서는 공시한 모든 균주가 자실체를 형성하지 않았다. 자실체 원기는 광도 0.05에서 10.0 μmol m^-2 s^-1의 범위에서 형성되었고, 광도가 0.5 μmol m^-2 s^-1 이상에서는 자실체 원기의 수가 감소하였다. 주기적인 광조사가 자실체 원기의 형성에 미치는 영향을 조사한 결과, G1-009 균주는 자실체 원기의 수와 무게가 연속광을 조사한 경우보다 증가하였다. 자실체 원기를 형성하기 위해서는 최소 4일간의 광조사가 필요하였다. 단색광의 조사가 자실체 원기의 형성에 미치는 영향을 조사한 결과 G1-003 균주는 400에서 800 nm의 모든 단색광 처리구에서 자실체 원기가 형성되었고, 그 외의 4균주는 400에서 500 nm, 그리고 700에서 750 nm 범위에서 형성되었다. The objective of this research was to determine the effect of light quality on formation of fruit body primordia (FBPs) of Ganoderma lucidum. To achieve this 5 isolates of the fungus that develops fruit body primordia on nutrient agar media were incubated with or without continuous irradiation. The fluorescent lamps used different colors such as black light blue (BLB), pure blue (P-B), pure green (P-G), pure yellow (P-Y) and pure red (P-R). Effect of periodic light and dark exposures on FBP formation of isolate G1-009 was investigated. The FBP formation in G. lucidum isolates was also tested under nomochromatic light produced by the combination of interference filters and colored glass filters. Three isolates produced FBPs under all kinds of fluorescent lamps, whereas two induced FBPs only under visible light except for BLB fluorescent lamp. However, these isolated did not form FBPs in the dark. The FBP was formed at light intensity from 0.05 to 10.0 μmol m^-2 s^-1, and begun to reduce its number as light intensity increase over 0.5 μmol m^-2 s^-1. When the isolate was incubated under periodic light and dark exposures, the number and weight of FBP increased as compared with those under continuous light. Initiation of FBP requires at least 4 days of light illumination. Although isolate G1-003 produced FBPs in a wide range of 400 to 800 nm, other four isolates had two effective regions 400 to 500 nm and 700 to 750 nm in FBP formation.

        現代國語의 客體尊待 問題

        The honorific system of Korean is very complicated. Not only according to time but also to social levels it shows many different shapes. So to give a clear description of the system is not easy. The honorification described here is no exception. Here I review a kind of Korean honorification usually called ‘object-honorification’. By ‘object-honorification’ we mean an honorification which can be expressed by limited verbs like cwu-(give), turi-(give), mosi -(escort), j??c'u-(tell), etc. The object here includes sentence elements which have object or dative case. All of these verbs are transitive verbs. So they have to have an object. Many persons think the verbs themselves are honorific words. However I think they are not honorific words but modest words. Of course, we can say that the use of modest words results in giving respect to someone. There must be at least four persons in expressing the so-called object-honorification: the speaker, the hearer, the actor of the modest verb and the object. I think the performative theory is very desirable to show the structure of the Korean honorific system, especially, of the one concerned here. The structure of the honorifics we need is generally something like NP_1+NP_2+[NP_3+NP_4+VP_1]_s+VP_2. The verb concerned is VP1. Here we reach the most difficult questions, which I tried to solve: Who expresses himself modestly? Whom does he want to respect by doing so? I think the answer to the first question is the speaker, and the answer to the second is the object. But the speaker can not express his respect to the object directly, because the actor of the modest action (VP_1) is not the speaker but the subject of the verb. Therefore, the speaker has to express himself indirectly. To express his respect to the object, the speaker maker the actor of VP_1 express himself modestly to the object. The speaker uses the modest verb not because of the interrelation between the subject and the object but the interrelation between the object and the speaker himself.

      • 인체동작분석시 Wavelet변환을 이용한 영상자료의 보간 및 분석

        Inputting data of human motion analysis means signal processing of changes in linear time dependent spatial coordinates of anatomical landmarks. Commercial CCD camera appears to be sampling 30 fps imposing limitation for human motion analysis which needs at least 60 fps. For the interpolation 60 fps fast algorithm of inverse wavelet transformation (IWT) was utilized producing 60 fps out of normal 30 fps. The original data from free fall and cycling motion of lower extremity were used to test the applicabiility of wavelet interpolation to human motion analysis. The test results showed linear relationship between two data sets. In the test of free fall, data curves of two sets, theoretical 30 frame data and 60 frame data after inverse wavelet transformation, appeared to be almost identical. Regression analysis of free fall test showed R^2 = 0.99 meaning Y = IWT. In the test with cycling motion of lower extremity % cycle of maximum flexion and extension at the joints was compared between two data sets showing same result, as in free fall test.

        Metarizium anisopliae (Metschn.) Sorok이 생산하는 Lactobacillus plantarum 용균효소의 분리, 정제 및 특성

        김치저장성을 향상시키기 위하여, Lactobacillus plantarum 용균효소 생산균주를 토양으로부터 분리한 후 그 효소를 정제하고 특성을 살펴보았다. 본 균주의 배양학적 및 형태학적 특성 등을 검토한 결과 Metarrisium anisopliae (Metschn). Sorok으로 동정되었으며, 본 효소는 affinity adsorption, CM-Sephadex C-50 column chromatography를 통하여 40%의 수율과 75배 정제도로 정제되었다. 효소활성의 최적 pH와 온도는 각각 4.0, 40℃였으며, pH 3.0~9.0, 60℃까지 효소의 안정성이 유지되었다. SDS-PAGE 및 gel filtration으로부터 본 효소의 분자량은 40,000 daltons인 monomeric protein임을 확인하였다. 본 효소는 Lactobacillus plantarum 세포벽 peptidoglycan의 peptide linkage를 절단 하는 endopeptidase였으며, 용균 spectrum으로부터 김치발효시 유용균인 Leuconostoc mesenteroides에 대해서는 활성을 거의 나타내지 않음을 확인할 수 있었다. 본 효소의 활성은 Ag^+, N-bromosuccinimide (NBS)에 의해서 완전히 저해되었으며, NBS에 의해서 활성이 저해된다는 사실로부터 본 효소는 활성에 tryptophan residue가 관여하는 것으로 확인되었다. 본 효소는 산성 아미노산의 함량이 염기성 아미노산의 함량에 비하여 높았으며, alanine, glycine, proline, tyrosine의 함량 또한 매우 높았다. To improve the preservation of Kimchi, we isolated Lactobacillus plantarum lytic enzyme-producing strain from soil, and the enzyme was purified and characterized. From the observation of cultural and morphological characteristics, the isolated strain was identified as Metarrisium anisopliae (Metschn). Sorok. The enzyme was purified to 75-folds with 40% yields through affinity adsorption and CM-Sephadex C-50 column chromatography. The optimum pH and temperature for lytic activity are 4.0 and 40℃, respectively, and the enzyme acitivity is stable between pH 3.0 and 9.0 and up to 50℃. The enzyme is a monomeric protein with molecular weight of 40,000 daltons by SDS-FAGE and gel filtration. The enzyme is endopeptidase which breaks the peptide linkage of Lactobacillus plantarum peptidoglycan. The lytic action spectra confirmed that Leuconostoc mesenteroides, a useful strain for the fermentation of Kimchi, is not lysed by the enzyme. The enzyme activity is inhibited by N-bromosuccinimide (NBS), which probably indicates the involvement of tryptophan residue in active site of the enzyme, and also inhibited by Ag^+. The amino acid composition analysis showed that the enzyme contains more acidic amino acids than basic ones, and composition of alanine, glycine, proline and tyrosines was very high.
