      • Chondrogenic potential and anti-senescence effect of hypoxia on canine adipose mesenchymal stem cells

        <P>Mesenchymal stem cells (MSCs) have the ability to differentiate into multi-lineage cells, which confers great promise for use in regenerative medicine. In this study, canine adipose MSCs (cAD-MSCs) were isolated from canine adipose tissue. These cells clearly represented stemness (Oct4, Sox2, and Nanog) and differentiation potential into the mesoderm (adipocytes, chondrocytes, and osteoblasts) at early passages. The aim of this study was to evaluate the effects of hypoxia on the differentiation potential into mesoderm, and the expression of anti-apoptotic genes associated with cell survival for the optimal culturing of MSCs. We observed that the proliferation of the cAD-MSCs meaningfully increased when cultured under hypoxic condition than in normoxic condition, during 7 consecutive passages. Also, we found that hypoxia strongly expressed anti-senescence related genes such as HDAC1 (histone deacetylase 1), DNMT1 (DNA (cytosine-5)-methyltransferase 1), Bcl-2 (inhibitor of apoptosis), TERT (telomerase reverse transcriptase), LDHA (lactate dehydrogenase A), SLC2A1 (glucose transporter), and DKC1 (telomere holoenzyme complex) and differentiation potential of cAD-MSCs into chondrocytes, than seen under the normoxic culture conditions. We also examined the multipotency of hypoxic conditioned MSCs using quantitative real-time RT-PCR. We found that the expression levels of stemness genes such as Oct-4, Nanog, and Sox-2 were increased in hypoxic condition when compared to the normoxic condition. Collectively, these results suggest that hypoxic conditions have the ability to induce proliferation of MSCs and augment their chondrogenic potential. This study suggests that cell proliferation of cAD-MSC under hypoxia could be beneficial, when considering these cells for cell therapies of canine bone diseases.</P>

        <P>Transition metal oxide-based memristors have widely been proposed for applications toward artificial synapses. In general, memristors have two or more electrically switchable stable resistance states that device researchers see as an analogue to the ion channels found in biological synapses. The mechanism behind resistive switching in metal oxides has been divided into electrochemical metallization models and valence change models. The stability of the resistance states in the memristor vary widely depending on: oxide material, electrode material, deposition conditions, film thickness, and programming conditions. So far, it has been extremely challenging to obtain reliable memristors with more than two stable multivalued states along with endurances greater than ∼1000 cycles for each of those states. Using an oxygen plasma-assisted sputter deposition method of noble metal electrodes, we found that the metal-oxide interface could be deposited with substantially lower interface roughness observable at the nanometer scale. This markedly improved device reliability and function, allowing for a demonstration of memristors with four completely distinct levels from ∼6 × 10<SUP>-6</SUP> to ∼4 × 10<SUP>-8</SUP> S that were tested up to 10<SUP>4</SUP> cycles per level. Furthermore through a unique in situ transmission electron microscopy study, we were able to verify a redox reaction-type model to be dominant in our samples, leading to the higher degree of electrical state controllability. For solid-state synapse applications, the improvements to electrical properties will lead to simple device structures, with an overall power and area reduction of at least 1000 times when compared to SRAM.</P> [FIG OMISSION]</BR>

        Original articles : Anti-inflammatory effects of equine adipose-derived mesenchymal stem cells for bone fracture in thoroughbred racehorses

        Bone fractures are most often seen in racetrack horses because of the high level of intensity in racing. These issues are the main cause of decreased performance in racehorses. Mesenchymal stem cells (MSCs) have been explored to improve intra-articular therapy in racehorses. MSCs are essential for the repair and regeneration of damaged tissues. In this study, the effect of intra-articular injection of MSCs in racehorses was investigated. Before accessing the MSC therapy, synovial fluids were obtained from the fracture site of racehorses, and adipose tissue was collected for MSC isolation. Using the MSC specific marker, adipose tissue-derived MSCs were identified. The racehorses received intra-articular injection of autologous MSCs (or allogeneic) (3 × 107 cells/3 mL). After 1 or 2 weeks, synovial fluids were collected from racehorses. To test the effect of MSC injection using ELISA, we analyzed inflammatory factors from the untreated samples compared to MSC-treated samples of racehorses. The level of pro-inflammatory factors (interleukin-1β and prostaglandin E2) was significantly decreased in synovial fluids of MSC-injected racehorses, compared to before accessing the MSC therapy, whereas, the level of anti-inflammatory factor (interleukin-10) was higher than prior to accessing the MSC therapy. Further studies are needed to investigate the anti-inflammatory mechanism of MSC in racehorses.

        The Derived Neutrophil-to-Lymphocyte Ratio Is an Independent Prognostic Factor in Transplantation Ineligible Patients with Multiple Myeloma

        <P><B><I>Background:</I></B> The neutrophil-to-lymphocyte ratio (NLR) is an independent prognostic marker in solid and hematological cancers. While the derived NLR (dNLR) was shown to be non-inferior to the NLR in large cohorts of patients with different cancer types, it has not been validated as a prognostic marker for multiple myeloma (MM) to date. <B><I>Methods:</I></B> Between May 22, 2011 and May 29, 2014, 176 patients with MM from 38 centers who were ineligible for autologous stem cell transplantation were analyzed. The dNLR was calculated using complete blood count differential data. The optimal dNLR cut-off value according to receiver operating characteristic analysis of overall survival (OS) was 1.51. All patients were treated with melphalan and prednisone combined with bortezomib. <B><I>Results:</I></B> The complete response rate was lower in the high dNLR group compared to the low dNLR group (7 vs. 26.1%, respectively; <I>p</I> = 0.0148); the corresponding 2-year OS rates were 72.2 and 84.7%, respectively (<I>p</I> = 0.0354). A high dNLR was an independent poor prognostic factor for OS (hazard ratio 2.217, 95% CI 1.015–4.842; <I>p</I> = 0.0458). <B><I>Conclusion:</I></B> The dNLR is a readily available and cheaply obtained parameter in clinical studies, and shows considerable potential as a new prognostic marker for transplantation-ineligible patients with MM.</P>

        Screening of Selected Korean Sweetpotato (Ipomoea batatas) Varieties for Fusarium Storage Root Rot (Fusarium solani) Resistance

        A common post-harvest disease of sweetpotato tuber is root rot caused by Fusarium solani in Korea as well as the other countries. Storage root rot disease was monitored earlier on sweetpotato (Ipomoea batatas) in storehouses of different locations in Korea. In the present study, an isolate SPL16124 was choosen and collected from Sweetpotato Research Lab., Bioenergy Crop Research Institute, NICS, Muan, Korea, and confirmed the identification as Fusarium solani by conidial and molecular phylogenetic analysis (internal transcribed spacer ITS and translation elongation factor EF 1-α gene sequences). The isolate was cultured on potato dextrose agar, and conidiation was induced. The fungus was screened for Fusarium root rot on tuber of 14 different varieties. Among the tested variety, Yenjami, Singeonmi, Daeyumi, and Sinjami showed resistant to root rot disease. Additionally, the pathogen was tested for pathogenicity on stalks of these varieties. No symptom was observed on the stalk, and it was confirmed that the disease is tissue specific.

        Nilotinib is a second-generation tyrosine kinase inhibitor active in patients with chronic myeloid leukemia (CML) resistant to imatinib, and has been recently approved for newly diagnosed patients. We present a case of nilotinib-induced interstitial lung disease (ILD). A 67-year-old female patient was initially treated with imatinib for chronic-phase Philadelphia chromosomepositive (Ph+) CML. Imatinib was replaced by nilotinib because of hematological toxicity. The patient had received nilotinib for about 3 years without significant adverse effects. She visited the clinic due to chronic cough; chest X-ray revealed consolidations in both lung fields. Nilotinib-induced ILD was diagnosed based on intensive workup, including lung biopsy. She responded dramatically to corticosteroid therapy. To our knowledge, this is the first reported case of nilotinib-induced ILD in a patient with Ph+ CML. We emphasize that if unexplained lung abnormalities progress in patients receiving nilotinib, physicians should consider this potentially fatal complication in their differential diagnoses.

        Usefulness of neutrophil to lymphocyte ratio in patients with chronic obstructive pulmonary disease: a prospective observational study

        Background/Aims: Neutrophil to lymphocyte ratio (NLR) in peripheral blood is a useful systemic inflammatory response biomarker. However, NLR has not been studied in patients with chronic obstructive pulmonary disease (COPD). This study was aimed to evaluate the usefulness of NLR in patients with COPD. Methods: NLR was prospectively measured and compared in patients with COPD exacerbation (n = 59), patients with stable COPD (n = 61), and healthy controls (n = 28). NLR in patients with COPD exacerbation was repeatedly measured in the convalescent period. The correlation between NLR and clinical parameters was evaluated, and the predictors for respiratory hospitalization were analyzed by multivariate logistic regression. Results: NLR values were significantly higher in patients with COPD exacerba­tion compared with stable COPD patients and controls (12.4 ± 10.6, 2.4 ± 0.7, 1.4 ± 0.5, respectively; p < 0.001). NLR was significantly decreased during the con­valescent period in patients with COPD exacerbation (4.5 ± 4.6 vs. 11.5 ± 8.8, p < 0.001). NLR exhibited a significant correlation with the body mass index, degree of airway obstruction, dyspnea, and exercise capacity (BODE) index, the 6-minute walk test, and the modified Medical Research Council scale. NLR ≥ 2.8 was an in­dependent predictor with a borderline significance for respiratory hospitalization (odds ratio, 2.083; p = 0.079). Body mass index and forced expiratory volume in 1 second were independent predictors for respiratory hospitalization. Conclusions: NLR is a straightforward and effective biomarker of COPD exac­erbation that may serve as a predictor for respiratory hospitalization in patients with COPD.

        지속가능한 농업을 위한 저탄소 인증 농산물 지불의사 분석: 20대를 중심으로

        This study estimates the consumer value provided by low-carbon agrifood with a focus on people in their 20s, who are current and future consumers. Information was collected regarding the respondents experiences, environmental awareness, perceptions of climate change and low-carbon agrifood, and their willingness to pay for it. Consumer preference for low-carbon agrifood was analyzed using the Contingent Valuation Method, which is commonly employed to estimate consumers willingness to pay for non-market goods. To closely align with real market conditions and to facilitate respondents answers to questions, a double-bounded dichotomous choice format was utilized. The empirical results show that for people in their 20s, the willingness-to-pay ratio for low-carbon agrifood was approximately 27% relative to conventional food. Environmental interest shown by such consumers had a statistically significant impact on the willingness-to-pay decision for these products, while treatment information, climate change awareness, and prior purchasing experiences contributed to numerical differences in the willingness-to-pay ratio. The analysis indicates a noticeable shift in willingness to pay based on the level of environmental concern. Therefore, prioritizing enhancement of individuals awareness and interest in their environment should be considered a primary focus. However, while the awareness of climate change and the environment among individuals in their 20s appears to be high, their recognition of low-carbon agrifood seems comparatively low.

      • Solitary Abdominal Tuberculous Lymphadenopathy Misdiagnosed as the Recurrence of Ovarian Cancer

        Monitoring cancer antigen 125 (CA-125) serum levels is very useful for determining how epithelial ovarian cancer is responding to treatment and for predicting a patient`s prognosis after treatment. An increase in CA-125 levels within patients in a remission is a strong predictor of the recurrence of ovarian cancer. However, while CA-125 is best known as a tumor marker for ovarian cancer, it may also be elevated in other various malignancies and in some benign conditions such as peritonitis, pancreatitis, chronic liver diseases, pelvic inflammatory diseases, or endometriosis. We describe herein a case of solitary intra-abdominal tuberculous lymphadenopathy mimicking a local relapse of epithelial ovarian cancer. A 51-year-old woman presented with progressive rise in serum CA-125. She had been diagnosed with advanced epithelial ovarian malignancy and undergone debulking surgery about 4 years ago. She had been given platinum-based adjuvant chemotherapy and achieved complete remission (CR). During regular follow-up in CR, a progressive rise in serum CA-125 was detected. The abdominal computed tomography (CT) showed a cystic mass, 3.6 cm in diameter, located adjacent to the right common iliac artery, suggesting a metastatic lymph node. We had planned second-look operation in settings of suspected focal recurrence of ovarian cancer, supported by evidence of rising CA-125 levels. However, the suspected tuberculous lesion was detected incidentally on preoperative chest CT. The patient had no symptoms and signs associated with tuberculosis (TB) and chest X-ray was also normal. We postponed the planned second-look operation and anti-TB medication was started. Mycobacterium TB was identified on culture of sputum specimens later. During anti-TB medication, the suspected metastatic isolated intra-abdominal lymphadenopathy improved and serum CA-125 level was also normalized. We emphasize here to consider not only the recurrence or progression of ovarian cancer itself but also benign conditions including TB in their differential diagnoses, when serum CA-125 level increases during the follow-up of epithelial ovarian cancer patients.
