Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.

Pragmatic clinical studies, an emerging clinical research discipline, include a wide range of studies that are largely embedded with routine clinical practice and aim to evaluate the comparative effectiveness and safety of different clinical intervention strategies. Increased availability and quality of electronic medical/health records drives the development of pragmatic clinical studies. In this review, we describe evolution of the conceptual framework of pragmatic clinical studies and share perspectives on the importance of pragmatic clinical studies in evidence-based practice for cardiovascular diseases, as a complement to conventional randomized controlled trials. We also highlight specific needs of pragmatic clinical studies in improving evidence-based practice for cardiovascular disease in Asian countries. The main challenges of pragmatic clinical studies are discussed briefly in this review.

This paper addresses the exponential stability for an interconnected system of an nth-order ODE systemwith the input governed by the Dirichlet boundary of a heat equation, and conversely, the output of the ODE isfluxed into the heat equation. The semigroup approach is adopted to show that the system operator is well-posed. We establish the exponential stability of the system by Riesz basis method. Furthermore, with MATLAB software,some numerical simulations are presented to show the effectiveness of the interconnection between the heat PDEand ODE systems.

High sugar concentration culturing was commonly used in modern fermentation industry. However, it leads to the reduction of the foam stability in beer brewing due to the excess secretion of proteinase A. To better understand the characterization of proteinase A excretion from Saccharomyces cerevisiae in high sugar stress conditions, the cultures were grown by using YNB medium with a high concentration of glucose. Pro-PrA isolated from the medium was purified by gel exclusion chromatography, and the PrA activity was detected using fluorescent substrate analysis. The relative molecular weight of the purified PrA and pro-PrA was estimated at 42 and 54 kDa, respectively, by SDS-PAGE. It indicated that the metabolic behavior of PrA in the high glucose culturing was quite different from the normal conditions, and glucose concentration may have a big influence on its secreted process. Further study showed that PrA was released at the logarithmic growth phase of the culturing, and the amount of PrA was 11 times higher compared with the normal culturing. PrA was considered to be activated by itself under acidic conditions. And it was also confirmed in this work that the step-wise pathway for the autoactivation known as a pseudo-PrA has a major contribution to the autoactivation process of PrA zymogen outside the cell.
Effects of Chlorine Addition to TiO2 Nanorods-Based Perovskite Solar Cells

Titanium dioxide (TiO2) nanorods (NR) structure and Chlorine substitution is beneficial to the extraction and diffusion of electrons in perovskite solar cells (PSCs). In this work, different concentrations of PbCl2 in perovskite are integrated with TiO2 (NR) to fabricate PSCs. The total thickness of perovskite absorber layer and electron transport layer (ETL) is about 850 nm, and the PSCs exhibit excellent performance. Scanning electron microscope (SEM) and X-ray diffraction (XRD) test reveal that moderate PbCl2 additive improves the perovskite film morphology and crystal quality of perovskite films. Optimal perovskite films with high crystallinity and uniform surface were prepared by adding 3 mol% PbCl2 into perovskite precursor solution, the crystal boundary and defect states are greatly reduced, thus reducing the electrons and holes recombination. The power conversion efficiency (PCE) enhancement of the device with this optimal molar ratio of PbCl2 is over 24% compared with the device without PbCl2.

In this paper, the well-posedness and stability of a 1-d 2×2 hyperbolic system under proportional feedback control is addressed. The semigroup approach and norm equivalence theorem is adopted to obtain the explicit dissipative condition for control parameters. The asymptotic spectral analysis of the system is presented. It is shown that the real part of the eigenvalues goes to some negative constant as the modulus of eigenvalues goes to +∞. Moreover, we find that there is a group of generalized eigenfunctions which forms a Riesz basis of the Hilbert state space. Hence, the spectrum determined growth condition holds, and then the exponential stability is established.

Ginger, one of worldwide consumed dietary spice, is not only famous as food supplements, but also believed to exert a variety of remarkable pharmacological activity as herbal remedies. In this study, a ginger constituent, 12-dehydrogingerdione (DHGD) was proven that has comparable anti-inflammatory activity with positive control 6-shogaol in inhibiting LPS-induced interleukin (IL)-6, tumor necrosis factor (TNF)-α, prostaglandin (PG) E2, nitric oxide (NO), inducible NO synthase (iNOS) and cyclooxygenase (COX)-2, without interfering with COX-1 in cultured microglial cells. Subsequent mechanistic studies indicate that 12-DHGD may inhibit neuro-inflammation through suppressing the LPS-activated Akt/IKK/NF-κB pathway. Furthermore, 12-DHGD markedly promoted the activation of NF-E2-related factor (Nrf)-2 and heme oxygenase (HO)-1, and we demonstrated that the involvement of HO-1 on the production of pro-inflammatory mediators such as NO and TNF-α by using a HO-1 inhibitor, Zinc protoporphyrin (Znpp). These results indicate that 12-DHGD may protect against neuro-inflammation by inhibiting Akt/IKK/IκB/NF-κB pathway and promoting Nrf-2/HO-1 pathway.
Effects of Zinc on the Activity and Conformational Changes of Arginine Kinase and Its Intermediate

The effects of zinc on arginine kinase and its collapsed-state intermediate were studied. Both arginine kinase and the collapsed-state intermediate were inactivated in the presence of zinc, following a biphasic kinetic course. The corresponding apparent rate constants of inactivation at different zinc concentrations and conformational changes in the presence of 0.5 mM zinc were obtained. The conformational changes of arginine kinase and the collapsed-state intermediate were followed by fluorescence spectra and circular dichroism spectra. Comparison of the results for arginine kinase and the collapsed-state intermediate showed that the collapsed-state intermediate was more susceptible to zinc, which indicated that the collapsed-state intermediate was more flexible and unstable than arginine kinase. The special structure of arginine kinase might explain these diverse phenomena.
Fluorescent Nanoparticles for Observing Primo Vascular System Along Sciatic Nerve

The primo vascular system was found in the epineurium along the rat sciatic nerve following subcutaneous injection of fluorescent nanoparticles at the Zusanli acupoint (ST-36). Nanoparticles were injected into the primo-vessel near ST-36 and flowed along the sciatic nerve. Fluorescence revealed a structure in the epineurium that was hardly detectable. Images of the isolated sample stained with 4’,6-diamidino-2-phenylindole were captured using confocal microscopy. These images showed the distinctive nuclei distribution and multi-lumen structure of primo-vessels that differentiate them from lymphatic vessels, blood capillaries and nerves. This study demonstrates a new use for nanoparticles in fluorescence reflectance imaging techniques during in vivo imaging of primo-vessels.

The effects of aqueous extract of Schizandra Chinensis Fruit (AESC) on cadmium-induced changes of monoamine neurotransmitters in the different brain regions of adult rats were investigated. Male rats were received intraperitoneal (i.p.) administration of CdCl2 (0.6 mg/kg/d) for 21 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA) in striatum and serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in cortex were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Cd intoxicated rats (P < 0.05), while pretreatment with AESC (20 mg/kg/d or 60 mg/kg/d, p.o., 30 min before $CdCl_2$) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC significantly increased the reduction of glutathione contents and superoxide dismutase activities in cortex induced by $CdCl_2$. These results suggest that AESC ameliorates Cd-induced depletion of monoamine neurotransmitters in brain through its antioxidant activity.
