        Protein Profile in Corpus Luteum during Pregnancy in Korean Native Cows

        Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism, but the profile of proteins associated with progesterone synthesis in cyclic and pregnant corpus luteum (CL) is not well-known in cattle. In Experiment 1, plasma progesterone level was monitored in cyclic cows (n = 5) and pregnant cows (n = 6; until d-90). A significant decline in the plasma progesterone level occurred at d-19 of cyclic cows. Progesterone level in abbatoir-derived luteal tissues was also determined at d 1 to 5, 6 to 13 and 14 to 20 of cyclic cows, and d-60 and -90 of pregnant cows (n = 5 each). Progesterone level in d-60 CL was not different from those in d 6 to 13 CL and d-90 CL, although the difference between d 6 to 13 and d-90 was significant. In Experiment 2, protein expression pattern in CL at d-90 (n = 4) was compared with that in CL of cyclic cows at d 6 to 13 (n = 5). Significant changes in the level of protein expression were detected in 32 protein spots by two-dimensional polyacrylamide gel electrophoresis (2-DE), and 23 of them were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Six proteins were found only in pregnant CL, while the other 17 proteins were found only in cyclic CL. Among the above 6 proteins, vimentin which is involved in the regulation of post-implantation development was included. Thus, the protein expression pattern in CL was disorientated from cyclic luteal phase to mid pregnancy, and alterations in specific CL protein expression may contribute to the maintenance of pregnancy in Korean native cows.

      • d-pinitol regulates Th1/Th2 balance via suppressing Th2 immune response in ovalbumin-induced asthma

        d-pinitol has been demonstrated to exert insulin-like and anti-inflammatory activities. However, its anti-allergic effect in the Th1/Th2 immune response is poorly understood. Recently, it was shown that T-bet and GATA-3 are master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether d-pinitol regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in OVA-induced asthma model mice. We also examined to ascertain whether d-pinitol could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, and the development of airway hyper-responsiveness (AHR). The administration of d-pinitol before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that d-pinitol plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of d-pinitol in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of d-pinitol.

      • Combination of metformin and VSL#3 additively suppresses western-style diet induced colon cancer in mice

        <P>Western-style diet (WD) and dysbiosis are known to be associated with colonic inflammation, which contributes to carcinogenesis. Metformin (Met) exerts anti-inflammatory effects to induce AMP-activated protein kinase (AMPK), resulting in suppressed protein synthesis and reduced cell proliferation. Probiotic VSL#3 (V) modifies microbial composition. We investigated the chemopreventive mechanisms of Met and V in WD-induced colitis-associated colon carcinogenesis. Male BALB/c mice were randomly divided into five groups: a control diet (CD) group, WD group, WD+ Met (250 mg/kg/day) group, WD+V (1.3 million bacteria/day) group, and WD+ Met+V group. All mice were exposed to azoxymethane (10 mg/kg) followed by 2% dextran sodium sulfate (DSS) for 7 days. Using HCT-116 human colon cancer cell line, expression of AMPK, extracellular signal-regulated kinase (ERK), cyclin D1, and Bcl-2 was investigated and cell cycle arrest was assessed. WD enhanced the severity of colitis and tumor growth compared with CD. The combination of Met and V significantly ameliorated colitis and tumor growth by inhibiting macrophage infiltration and maintaining epithelial integrity. In vitro assays showed that the combination therapy promoted late apoptosis by inhibiting cyclin D1 and Bcl-2 and activating proapoptotic ERK. A combination therapy with Met and V attenuates tumor growth in a mouse model of WD-induced colitic cancer, suggesting that this strategy could be useful for the chemoprevention of colon cancer.</P>

        15-Deoxy-Δ<sup>12,14</sup>-prostaglandin J<sub>2</sub> induces p53 expression through Nrf2-mediated upregulation of heme oxygenase-1 in human breast cancer cells

        <P>Heme oxygenase-1 (HO-1) is a stress-responsive enzyme that has antioxidant and cytoprotective functions. However, HO-1 has oncogenic functions in cancerous or transformed cells. In the present work, we investigated the effects of HO-1 on the expression of p53 induced by 15-deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> (15d-PGJ<SUB>2</SUB>) in human breast cancer (MCF-7) cells. Treatment of MCF-7 cells with 15d-PGJ<SUB>2</SUB> led to time-dependent increases in the expression of p53 as well as HO-1. Upregulation of p53 expression by 15d-PGJ<SUB>2</SUB> was abrogated by si-RNA knock-down of HO-1. In MCF-7 cells transfected with HO-1 si-RNA, 15d-PGJ<SUB>2</SUB> failed to induce expression of p53 as well as HO-1. In addition, HO-1 inducers enhanced the p53 expression. We speculated that iron, a by-product of HO-1-catalyzed reactions, could mediate 15d-PGJ<SUB>2</SUB>-induced p53 expression. Upregulation of p53 expression by 15d-PGJ<SUB>2</SUB> was abrogated by the iron chelator desferrioxamine in MCF-7 cells. Iron released from heme by HO-1 activity is mostly in the Fe<SUP>2+</SUP> form. When MCF-7 cells were treated with the Fe<SUP>2+</SUP>-specific chelator phenanthroline, 15d-PGJ<SUB>2</SUB>-induced p53 expression was attenuated. In addition, levels of the Fe-sequestering protein H-ferritin were elevated in 15d-PGJ<SUB>2</SUB>-treated MCF-7 cells. In conclusion, upregulation of p53 and p21 via HO-1 induction and subsequent release of iron with accumulation of H-ferritin may confer resistance to oxidative damage in cancer cells frequently challenged by redox-cycling anticancer drugs.</P>

      • OGLE-2015-BLG-1482L: The First Isolated Low-mass Microlens in the Galactic Bulge

        <P>We analyze the single microlensing event OGLE-2015-BLG-1482 simultaneously observed from two ground-based surveys and from Spitzer. The Spitzer data exhibit finite-source effects that are. due to the passage of the lens close to or directly over. the surface of the source star as seen from Spitzer. Such finite-source effects generally yield measurements of the angular Einstein radius, which when combined with the microlens parallax derived from a comparison between the ground-based and the Spitzer light curves. yields the lens mass and lens-source relative parallax. From this analysis, we find that the lens of OGLE-2015-BLG-1482 is a very low-mass star with a. mass 0.10 +/- 0.02 M-circle dot or a brown dwarf with a. mass 55 +/- 9MJ, which are. located at D-LS = 0.80 +/- 0.19 kpc and D-LS = 0.54 +/- 0.08 kpc, respectively,. where DLS is the distance between the lens and the source, and thus it is the first isolated low-mass microlens that has been decisively located in the Galactic bulge. The degeneracy between the two solutions is severe ( Delta chi(2) = 0.3). The fundamental reason for the degeneracy is that the finite-source effect is seen only in a single data point from Spitzer, and this single data point gives rise to two solutions for rho, the angular size of the source in units of the angular Einstein ring radius. Because the rho degeneracy can be resolved only by relatively high-cadence observations around the peak, while the Spitzer cadence is typically similar to 1 day(-1), we expect that events for which the finite-source effect is seen only in the Spitzer data may frequently exhibit this rho degeneracy. For OGLE-2015-BLG-1482, the relative proper motion of the lens and source for the low-mass star is mu(rel) = 9.0 +/- 1.9 mas yr(-1), while for the brown dwarf it is 5.5 +/- 0.5 mas yr(-1). Hence, the degeneracy can be resolved within similar to 10 years from direct-lens imaging by using next-generation instruments with high spatial resolution.</P>

        Circulating hepatocyte growth factor as an independent prognostic factor of disseminated intravascular coagulation

        Background: Hepatocyte growth factor (HGF), a pleiotropic factor regulating development and wound healing, is secreted as inactive pro-HGF and is converted into active HGF by coagulation serine proteases. HGF receptor overexpression can cause massive venous thrombi, and factor Xa is reported to release soluble HGF from granulocytes. We hypothesized that a hypercoagulable condition, such as disseminated intravascular coagulation (DIC), may increase circulating HGF through active cleavage by coagulation serine proteases. Methods: In 172 DIC-suspected patients, plasma levels of total and active HGF, thrombin-antithrombin complex (TAT), plasmin-antiplasmin complex (PAP), and interleukin (IL)-6 were measured by ELISA. Active HGF release in granulocytes was examined in patients with and without overt-DIC. HGF-induced tissue factor expression in peripheral monocytes was measured by flow cytometry. Results: Circulating levels of total and active HGF correlated well with coagulopathy severity, including DIC score, D-dimer, TAT and PAP levels. HGF positively correlated with IL-6 and absolute neutrophil count. In contrast to the cancer group, HGF levels were significantly increased in accordance with increased DIC scores in non-cancer group. Elevated circulating HGF was an independent prognostic marker in the non-cancer group, while HGF level failed to predict mortality in the cancer group. Amounts of HGF released from stimulated granulocytes were not significantly different between overt-DIC and no overt-DIC patients. HGF potentiated endotoxin-induced tissue factor expression of monocytes in vitro. Conclusion: These findings suggest that circulating HGF is a potential laboratory marker reflecting coagulation activity and DIC prognosis in non-cancer patients and that HGF may play a role in a vicious cycle of hypercoagulability.

        Use of gamma irradiation for inactivation of pathogens inoculated into Kimbab, steamed rice rolled by dried laver

        The effect of gamma irradiation for inactivating the pathogens inoculated into the ready-to-eat Kimbab, steamed rice rolled by dried laver, was investigated. The pathogens used were Salmonella Typhimurium, Escherichia coli, Staphylococcus aureus and Listeria ivanovii which are important for public health. The growth of four test organisms inoculated (about 10<SUP>6</SUP>-10<SUP>7</SUP>CFU/g) into the Kimbab were sustained by an irradiation treatment during 24h of storage regardless of the temperature at 10, 20 and 30<SUP>o</SUP>C. Four pathogen inoculated into Kimbab decreased 2-3log CFU/g by 1kGy treatment and was not detected after 3kGy. The D<SUB>10</SUB> value of pathogens inoculated into the Kimbab were 0.31-0.44kGy among the four organisms. This study indicated that a low dose irradiation can maintain microbial safety for ready-to-eat Kimbab, steamed rice rolled by dried laver.
