        발효콩 추출물의 항돌연변이원성 및 세포독성 효과

        This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TA100 strains showed 82.4% and 90.8% inhibition against 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-1), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 ㎎/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.

      • 가시오갈피 열매 추출물의 항돌연변이원성 및 세포독성 효과

        가시오갈피(Eleutherococcus senticous Maxim) 열매의 에탄올 추출물 및 분획물의 항돌연변이성 및 항암활성을 규명하였다. 항돌연별이원성 실험결과에서는 직접변이원인 MNNG, 4NQO 그리고 간접변이원인 B(α)P, Trp=P-1에 대해서 농도의존적인 돌연변이 억제활성을 나타내었다. MNNG (0.4㎍/plate)의 경우 S. typhimurium TA100 균주에서는 시료농도 200㎍/plate에서 클로로포름 분획물을 제외한 모든 분획물들에서 90%이상의 높은 억제효과를 나타내었다. 4NQO(0.15㎍/plate)에 대한 S. typhimurium TA98 균주의 경우 물 분획물의 경우 88.3%, TA100 균주에서는 에틸 아세테이트 분획물이 84.4%의 억제효과를 보였다. 간접변이원의 경우 B(α)P(10㎍/plate)에선느 에탄올 추출물과 핵산 분획물은 시료농도 200㎍/plate에서 각각 96.1%와 97.5%로 가장 높은 억제효과를 나타내었다. 그리고 Trp-P-1(0.15㎍/plate)에서는 TA100 균주의 경우 200㎍/plate 농도에서 에탄올 추출물과 물 분획물이 각각 95.5%와 90%로 높은 억제율을 보여주었고, TA98 균주에 대해서는 에틸아세티에트 분획물이 88.3%로 비교적 높은 억제효과를 나타내었다. 인간의 암세포인 폐암세포(A549), 위암세포(AGS), 간암세포(Hep3B), 유방암세포(MCF-7)에 대한 세포독성 억제 효과를 검토한 결과 모든 암세포에서 각분획물들은 1㎎/mL 농도에서 60% 이상의 비교적 높은 암세포 성장 효과를 나타내었다. 특히 MCF-7에서 높은 암세포 성장억제 효과를 나타내었는데 핵산 분획물(1㎎/mL)에서 92.7%의 높은 암세포 성장 억제 효과를 나타내었고 Hep3B에서는 부탄을 분획물(1㎎/mL)에서 82%의 비교적 높은 성장 억제효과를 나타내었다. This study was carried out to investigate the antimutagenic and cytotoxic effects of Eleutherococcus senticosus Maxim fruits ethanol extract on Salmonella typhimurium TA98, TA 100 and cancer cell lines using Ames test and SRB assay, respectively. They were extracted with ethanol and then fractionated with hexane, chloroform, ethyl acetate, butanol and water to get active fractions. In the Ames test, most of the extract had strong antimutagenic effects against the mutagenesis induced by MNNG, 4NQO, B(α)P Trp-P-1. The ethanol extract (200 ㎍/plate) of Eleutherococcus senticosus fruits showed 87.2% inhibitory effect on the mutagenesis induced by MNNG against TA100. And also, The suppression ratio against B(α)P and AGS, MCF-7, Hep3B), the value value of inhibition were mostly above 60% for each fraction (1 ㎎/mL). Hexane fraction (1 ㎎/mL) against showed the strongest cytotoxic effects of 92.7% compared to those of other fraction and butano fraetion against Hep3B was relatively high growth inhibitory effect of 82%.

      • A New Dithienopyridine-Based Polymer for an Organic Electronics

        <P>A new dithienopyridine-based copolymer (PDT-TBT) was designed and synthesized. The copolymer showed good solubility in chloroform, chlorobenzene, and dichlorobenzene. The thermogravimetric analysis curve showed that the copolymer had good thermal stability with weight loss of less than 5% at 341 degrees C. The UV absorption maximum of the polymer appeared at 550 nm in solution and 600 nm in the film. The HOMO and LUMO of the copolymer were -5.50 eV and -3.63 eV, respectively. The copolymer exhibited the best performance when the PC61BM ratio was 1: 2 (w/w %), with a J(SC) of 3.56 mA/cm(2), a VOC of 0.45 V, an FF of 0.27%, and a PCE of 0.43%. A thin film transistor with PDT-TBT as the active layer showed mobility of 5.02x10(-3) cm(2)/Vs, an on/off ratio of 1.19x10(5) and a threshold voltage of -22.75 V.</P>

        rTMS (repetitive Transcranial Magnetic Stimulation) 및 침도치료를 병행하여 호전된 이명 환자 23례

        Objectives: The aim of this report is to report the effect of Acupotomy and rTMS on tinnitus. Methods: A total of 23 patients who visited the clinic for tinnitus symptoms received treatment, which included rTMS stimulation on the temporoparietal cortex and acupotomy treatment on the transverse processes of the cervical vertebrae. Rectus capitis posterior major, Rectus capitis posterior minor and TE17 for 10 times. Results: The average NRS of patients before receiving rTMS and acupuncture treatment was 7.61±1.12 points, which decreased to an average of 3.09±1.53 points after treatment, with statistical significance. Conclusion: This research suggests that rTMS and acupotomy treatment may be effective therapeutic methods for alleviating tinnitus symptoms.

        Relaxation Effect of Schisandra Chinensis Lignans on the Isolated Tracheal Smooth Muscle in Rats and Its Mechanism

        Schisandra chinensis (S. chinensis) is one of the core drugs used for relieving cough and asthma in traditional Chinese medicine. However, there are few basic studies on the treatment of respiratory diseases with S. chinensis in modern pharmacology, and the material basis and mechanism of its antiasthmatic effect are still unclear. Lignans are the main active components of S. chinensis. The aim of this study was to observe the relaxation effect of S. chinensis lignans (SCL) on the tracheal smooth muscle of rats by in vitro tracheal perfusion experiments, and to explore the mechanism by preincubation with L-type calcium channel blocker verapamil, four potassium channel blockers glibenclamide, tetraethylamine, 4-aminopyridine and barium chloride (BaCl2), β-adrenoceptor blocker propranolol, nitric oxide synthase inhibitor Nω-nitro-L-arginine methyl ester (L-NAME), and the cyclooxygenase inhibitor indomethacin, respectively. The results showed that SCL (0.25–1.75 mg/mL) reduced the contraction of isolated tracheal smooth muscle induced by acetylcholine, the preincubation with verapamil and glibenclamide could attenuate the relaxation effect, whereas propranolol, 4-aminopyridine, BaCl2, tetraethylamine, L-NAME, and indomethacin had no such effect. These results suggest that SCL has a significant relaxation effect on the isolated tracheal smooth muscle of rats, and the mechanism may be related to the inhibition of extracellular calcium influx and intracellular calcium release from the sarcoplasmic reticulum, as well as the activation of ATP-sensitive potassium channels. These findings may provide a pharmacological basis for the traditional use of S. chinensis to treat asthma.

        Effect of Electrolyte Concentration Difference on Hydrogen Production during PEM Electrolysis

        Proton exchange membrane (PEM) water electrolysis systems offer several advantages over traditional technologies including higher energy efficiency, higher production rates, and more compact design. In this study, all the experiments were performed with a self-designed PEM electrolyser operated at 1 atm and $25^{\circ}C$. Two types of electrolyte were used: (i) potassium hydroxide (KOH), and (ii) sulfuric acid ($H_2SO_4$). In the experiments, the voltage, current, and time were measured. The concentration of the electrolyte significantly affected the electrolyser performance. Overall the best case was with 15 wt% $H_2SO_4$ at the anode channel and 20 wt% at the cathode channel with. In addition, increasing the difference in concentration of the sulfuric acid had an effect on the diffusion. The diffusion flux became larger when the difference in concentration became larger, increasing electrolyser efficiency without the addition of extra energy.

        Synthesis of Thiophene-based Polymeric Semiconductor with High Aromatic Density and Its Application in Organic Thin-Film Transistors

        A new p-type polymer, poly(3,3'''''-didodecyl sexythiophene) (PST), was synthesized via <TEX>$FeCl_3$</TEX> oxidative coupling reaction. The mole ratio of <TEX>$FeCl_3$</TEX> and monomer (4:1), and keeping room temperature for polymerization were very important to obtain soluble polymer with regulated molecular weight. The photoluminescence (PL) maxima of the polymer are found at 500 and 562 nm for the solution and at 656 nm for the film. The low density of side chains enables substantial torsional deviations of the thienylene units, thereby inducing a relatively low highest occupied molecular orbital (HOMO) level (~ -5.22 eV). X-Ray diffraction analysis showed that PST forms a highly interdigitated structure due to the low density of side chains. PST were introduced as the active layers of organic thin-film transistors, and the resulting device showed field-effect mobilities of <TEX>$0.12cm^2/Vs$</TEX> (on/off <TEX>$ratio=1.7{\times}10^5$</TEX>) in air. The intrinsic mobility of PST measured by using the time of flight technique also supports a high conductance of <TEX>$4{\times}10^{-4}cm^2/Vs$</TEX>.
