        Improvement of Lipid and Terpenoid Yield in Thraustochytrids Using Chemical Regulators: A Review

        For the increasing demand for lipids and terpenoids in humans, biological fermentation has become an attractive choice due to the safety and sustainability. Thraustochytrids have been identified as promising producers of polyunsaturated fatty acids because of their high lipid content and simple fatty acid composition. In addition, thraustochytrids are also potential producers of terpenoids for their completed mevalonate pathway. Chemical regulators can be used to stimulate or inhibit metabolic pathways, which are equivalent to effects of overexpression and suppression approaches. The application of chemical regulators is potentially an easy and practical approach to improve the lipid and terpenoid yield in thraustochytrids, which has become a research focus. In this review, the mechanisms of chemical regulators promoting lipid and terpenoid biosynthesis in thraustochytrids are elucidated. Various chemical regulators which can directly enhance the biosynthesis of lipids and terpenoids are summarized depending on the type of chemicals. In addition, some chemical regulators are demonstrated to indirectly improve lipid and terpenoid yield by reducing the conversion of metabolites and reducing oxidative stress. Therefore, the addition of chemical regulators can be a useful alternative strategy for improving lipid and terpenoid accumulation in large-scale cultivation of thraustochytrids.

        A modified SOM method based on nonlinear neural weight updating for bearing fault identification in variable speed condition

        Fault identification for bearings of special electromechanical equipment is significant to avoid catastrophic accidents. However, spectral aliasing and nonstationarity resulted from variable speed condition make this task difficult. In this paper, a modified self-organizing maps (SOM) based on nonlinear neural weight updating way is proposed to solve the problem of bearing fault severity identification in variable speed condition. Firstly, a multi-domain features extraction method based on angular re-sampling technique is introduced. Then considering the nonlinear relationship between fault severity and fault features, the traditional Euclidian distance of SOM is substituted with the geodesic distance when update the neural weight and select the best-matching cell, which can improve the nonlinear identification ability of proposed method. Finally, two cases are performed and the results show that the method can identify bearing fault with different severities effectively and have practical significance when considering both accuracy and time cost compared with other methods.

        Migraine Susceptibility Genes in Han Chinese of Fujian Province

        Background and Purpose Five single-nucleotide polymorphisms (SNPs) (rs4379368, rs10504861, rs10915437, rs12134493 and rs13208321) were recently identified in a Western population with migraine. These migraine-associated SNPs have not been evaluated in a Han Chinese population. This study investigated the associations of specific SNPs with migraine in a Han population. Methods This was a case-control study of Han Chinese residing in Fujian Province. Polymerase chain reaction—restriction-fragment-length polymorphism analysis and direct sequencing were used to characterize the relationships of SNPs in a control group of 200 subjects and in a migraine group of 201 patients. Results The frequencies of the five SNPs did not differ between patients with migraine and healthy non migraine controls. However, subgroup analysis indicated certain SNPs were more strongly associated with migraine with aura or migraine without aura than with controls. The CT genotype of rs4379368 was more common in migraine patients with aura (75%) than in migraine patients without aura (47.9%) and controls (48.5%) (p<0.05), and the TT genotype of rs10504861 was more common in migraine patients with aura than in controls (8.3% vs. 0.5%) (p<0.05). Meanwhile, the CC genotype of rs12134493 was less common in migraine patients without aura than in controls (80.6% vs. 88%) (p<0.05). Conclusions Our findings suggest that the rs4379368 and rs10504861 SNPs are markers for susceptibility to migraine with aura and that rs12134493 is a marker for the risk of migraine without aura in this Han population. Future studies should further explore if these associations vary by ethnicity.

        Synchronous tele-ultrasonography is helpful for a naive operator to perform high-quality thyroid ultrasound examinations

        Purpose: This study investigated the value of synchronous tele-ultrasonography (TUS) for naive operators in thyroid ultrasonography (US) examinations.Methods: Ninety-seven patients were included in this prospective, parallel-controlled trial. Thyroid scanning and diagnosis were completed by resident A independently, resident B with guidance from a US expert through synchronous TUS, and an on-site US expert. The on-site expert’s findings constituted the reference standard. Two other off-site US experts analyzed all data in a blind manner. Inter-operator consistency between the two residents and the on-site US expert for thyroid size measurements, nodule measurements, nodule features, American College of Radiology (ACR) Thyroid Imaging Reporting and Data System (TI-RADS) categories, and image quality was compared. Two questionnaires were completed to evaluate the clinical benefit.Results: Resident B detected more nodules consistent with the on-site expert than resident A did (89.4% vs. 56.5%, P<0.001). Resident B achieved excellent consistency with the on-site expert in terms of ACR TI-RADS categories, nodule composition, shape, echogenic foci, and vascularity (all intra-class correlation coefficients [ICCs] >0.75), while resident A achieved lower consistency in ACR TI-RADS categories, composition, echogenicity, margin, echogenic foci, and vascularity (all ICCs 0.40-0.75). Residents A and B had excellent consistency in target nodule measurements (all ICCs >0.75). Resident B achieved better performance than resident A for gray values, time gain compensation, depth, color Doppler adjustment, and the visibility of key information (all P<0.05). Furthermore, 61.9% (60/97) of patients accepted synchronous TUS, and 59.8% (58/97) patients were willing to pay for it.Conclusion: Synchronous TUS can help inexperienced residents achieve comparable thyroid diagnostic capability to a US expert.

        Mechanisms of Electroacupuncture Pretreatment in Alleviating Myocardial Ischemia Reperfusion Injury: Interactions between the Cerebellar Fastigial Nucleus and Lateral Hypothalamic Area

        Background: Myocardial ischemia reperfusion injury (MIRI) is an important mechanism of post-myocardial infarction injury and a main cause of death in patients with ischemic heart disease. Electroacupuncture (EA) pretreatment is effective for the prevention and treatment of MIRI, but mechanisms mediating the effects of cardiovascular disease EA treatments remain unclear. Objectives: To determine whether the lateral hypothalamus (LHA) and the cerebellar fastigial nucleus (FN) are involved in the protective effects of EA stimulation on MIRI. Methods: EA pretreatment was performed for 7 days before the establishment of the MIRI model. ST-segment changes on electrocardiograms were recorded and the Curtis–Walker arrhythmia score was used to evaluate changes in reperfusion injury. Hematoxylin–eosin staining was applied to evaluate the pathological and morphological changes in myocardial tissue. c-fos expression in the LHA and FN was determined by immunofluorescence staining. Glutamic (Glu) and γ-Aminobutyric acid (GABA) levels were measured using a high-performance liquid chromatography-electrochemical method. Results: EA pretreatment reduced ST-segment elevation, arrhythmia scores, and morphological changes in MIRI myocardial cells in rats, and decreased the c-fos protein expression in LHA/FN nuclei. MIRI was associated with an imbalance between GABA and Glu levels, whereas EA pretreatment increased GABA levels and decreased Glu levels in the LHA/FN. Conclusion: FN and LHA are involved in the EA-mediated attenuation of MIRI. Pretreatment with EA plays a protective role in the myocardium by regulating Glu and GABA release in the LHA and FN.
