      • Role of Nucleation-Promoting Factors in Mouse Early Embryo Development

        <B>Abstract</B><P>During mitosis nucleation-promoting factors (NPFs) bind to the Arp2/3 complex and activate actin assembly. JMY and WAVE2 are two critical members of the NPFs. Previous studies have demonstrated that NPFs promote multiple processes such as cell migration and cytokinesis. However, the role of NPFs in development of mammalian embryos is still unknown. Results of the present study show that the NPFs JMY and WAVE2 are critical for cytokinesis during development of mouse embryos. Both JMY and WAVE2 are expressed in mouse embryos. After injection of JMY or WAVE2 siRNA, all embryos failed to develop to the morula or blastocyst stages. Moreover, using fluorescence intensity analysis, we found that the expression of actin decreased, and multiple nuclei were observed within a single cell indicating that NPFs-induced actin reduction caused the failure of cell division. In addition, injection of JMY and WAVE2 siRNA also caused ARP2 degradation, indicating that involvement of NPFs in development of mouse embryos is mainly through regulation of ARP2/3-induced actin assembly. Taken together, these data suggested that WAVE2 and JMY are involved in development of mouse embryos, and their regulation may be through a NPFs-Arp2/3-actin pathway.</P>

        Intestinal Anti-Inflammatory Effects of Selenized Ulva pertusa Polysaccharides in a Dextran Sulfate Sodium-Induced Inflammatory Bowel Disease Model

        The purpose of this study was to examine the alleviative effects of selenized polysaccharides from Ulva pertusa (ulvan-Se) on inflammatory bowel disease (IBD) in mice. The dextran sulfate sodium (DSS)-induced IBD mouse model was used to explore the protective effects of ulvan-Se on the intestinal mechanical and immune barrier. At doses less than 1208 mg/kg·bw ulvan-Se showed no significant damage to Institute of Cancer Research (ICR) mice in an acute toxicity test. The results showed that DSS destroyed the mechanical barrier, which includes epithelial cells, while ulvan-Se promoted mRNA expression of tight junction proteins (zonula occludens protein 1, occludin, and claudin-1) and inhibited the infiltration of white blood cells into the intestines. At 100 mg/kg·bw, ulvan-Se enhanced the antioxidant capacity of mice more effectively than the 50 mg/kg·bw ulvan-Se. Furthermore, ulvan-Se improved the intestinal immune barrier by increasing immunoglobulin A and immunoglobulin M, while regulating the levels of interleukin (IL)-1β, interferon-γ, and IL-4. Oral administration of ulvan-Se also suppressed tumor necrosis factor-α, IL-1β, IL-6, and cyclooxygenase-2 mRNA expression mediated by the nuclear factor kappa B pathway. Taken together, our findings reveal that ulvan-Se could be used as a potential alternative supplement for reducing intestinal inflammation in IBD.

        Adaptive Sliding Mode Control with Enhanced Optimal Reaching Law for Boost Converter Based Hybrid Power Sources in Electric Vehicles

        This paper proposes an adaptive sliding mode control (ASMC) strategy with an enhanced optimal reaching law (EORL) for the robust current tracking control of the boost converter based hybrid power source (HPS) in an electric vehicle (EV). A conventional ASMC strategy based on state observers and the hysteresis control method is used to realize the current tracking control for the boost converter based HPS. Then a novel enhanced exponential reaching law is proposed to improve the ASMC. Moreover, an enhanced exponential reaching law is optimized by particle swarm optimization. Finally, the adaptive control factor is redesigned based on the EORL. Simulations and experiments are established to validate the ASMC strategy with the EORL. Results show that the ASMC strategy with the EORL has an excellent current tracking control effect for the boost converter based HPS. When compared with the conventional ASMC strategy, the convergence time of the ASMC strategy with the EORL can be effectively improved. In EV applications, the ASMC strategy with the EORL can achieve robust current tracking control of the boost converter based HPS. It can guarantee the active and stable power distribution for boost converter based HPS.

        Highly Ordered Mesoporous Cobalt Oxide Nanostructures: Synthesis, Characterisation, Magnetic Properties, and Applications for Electrochemical Energy Devices

        <P>Highly ordered mesoporous Co<SUB>3</SUB>O<SUB>4</SUB> nanostructures were prepared using KIT-6 and SBA-15 silica as hard templates. The structures were confirmed by small angle X-ray diffraction, high resolution transmission electron microscopy, and N<SUB>2</SUB> adsorption–desorption isotherm analysis. Both KIT-6 cubic and SBA-15 hexagonal mesoporous Co<SUB>3</SUB>O<SUB>4</SUB> samples exhibited a low Néel temperature and bulk antiferromagnetic coupling due to geometric confinement of antiferromagnetic order within the nanoparticles. Mesoporous Co<SUB>3</SUB>O<SUB>4</SUB> electrode materials have demonstrated the high lithium storage capacity of more than 1200 mAh g<SUP>−1</SUP> with an excellent cycle life. They also exhibited a high specific capacitance of 370 F g<SUP>−1</SUP> as electrodes in supercapacitors.</P> <B>Graphic Abstract</B> <P>Highly ordered mesoporous Co<SUB>3</SUB>O<SUB>4</SUB> nanostructures have been prepared by using a hard-template method. Mesoporous Co<SUB>3</SUB>O<SUB>4</SUB> (general TEM image shown) exhibits a low Néel temperature and bulk antiferromagnetic coupling.         Synchronization for Fractional Order Chaotic Systems with Uncertain Parameters

        This paper deals with the synchronization of fractional order chaotic systems with uncertain parameters. An adaptive control law consisting of fractional order feedback and sliding mode control is proposed. The twofractional order chaotic systems can be synchronized and the uncertain parameters can be identified under thecontroller and the update rule proposed. Illustrative example and numerical simulation results are provided todemonstrate the effectiveness of the control method proposed.

      • Identification of Biomarkers for Diagnosis of Gastric Cancer by Bioinformatics

        Background: We aimed to discover potential gene biomarkers for gastric cancer (GC) diagnosis. Materials and Methods: Genechips of 10 GC tissues and 10 gastric mucosa (GM, para-carcinoma tissue, normal control) tissues were generated using an exon array of Affymetrix containing 30,000 genes. The differentially expressed genes (DEGs) between GC tissues and normal control were identified by the Limma package and analyzed by hierarchical clustering analysis. Gene ontology (GO) and pathway enrichment analyses were performed for investigating the functions of DEGs. Receiver operating characteristics (ROC) analysis was performed to measure the effects of biomarker candidates for diagnosis of GC. Results: Totals of 896 up-regulated and 60 down-regulated DEGs were identified to be differentially expressed between GC samples and normal control. Hierarchical clustering analysis showed that DEGs were highly differentially expressed and most DEGs were up-regulated. The most significantly enriched GO-BP term was revealed to be mitotic cell cycle and the most significantly enriched pathway was cell cycle. The intersection analysis showed that most significant DEGs were cyclin B1 (CCNB1) and cyclin B2 (CCNB2). The sensitivities and specificities of CCNB1 and CCNB2 were both high (p<0.0001). Areas under the ROC curve for CCNB1 and CCNB2 were both greater than 0.9 (p<0.0001). Conclusions: CCNB1 and CCNB2, which were involved in cell cycle, played significant roles in the progression and development of GC and these genes may be potential biomarkers for diagnosis and prognosis of GC.

        Transient Expression of the GUS Gene in a Unicellular Marine Green Alga, Chlorella sp. MACC/C95, via Electroporation

        A transient expression system for a unicellular marine green alga, Chlorella sp. MACC/C95, was developed using a reporter GUS gene coded for by plasmid pBI121. The results demonstrated a high transformation efficiency could be achieved by using electroporation to deliver DNA into intact cells and the CaMV35S promoter to drive the foreign gene expression in Chlorella sp. MACC/C95. The use of a carrier DNA coupled with osmosis treatment improved the transformation efficiency, while linearization of the plasmid had minor effects. Investigation of the effects of DNA concentration and growth phases of Chlorella sp. MACC/C95 on transformation efficiency indicated that the highest level of transient expression was observed when $6{\mu}g\;mL^{-1}$ of plasmid DNA and cells $2{\sim}6$ days old were used.

      • Associations Between Three Polymorphisms in the Interleukin-4 Receptor Gene and Risk of Cancer: a Meta-analysis

        Interleukin-4 receptor (IL-4R) gene single nucleotide polymorphisms (SNPs) are implicated in cancer development. However, results from the published reports have remained inconclusive. The objective of this study was to conduct a meta-analysis investigating the association between polymorphisms in IL-4R gene and cancer risk. Pubmed, EMBASE and China National Knowledge Infrastructure (CNKI) were searched for case-control studies published up to October 30, 2012 that investigated IL-4R polymorphisms and cancer risk. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to evaluate the strength of any associations. Three IL-4R polymorphisms (Q576R, rs1801275; I75V, rs1805010; S503P, rs1805015) in 21 case-control studies were analyzed. Our meta-analysis indicated that these three polymorphisms are not associated with cancer risk when all studies were pooled together. In the subgroup analysis by tumor site, the results showed that Q576R G allele carriers were associated with a significantly decreased cervical cancer risk (recessive model: OR = 0.77, 95%CI = 0.60-0.98; homozygote comparison: OR = 0.76, 95%CI = 0.58-0.98). I75V G allele carriers were associated with a decreased risk of renal cancer (dominant model = 0.71, 95%CI = 0.57-0.89, heterozygote comparison: OR = 0.69, 95%CI = 0.55-0.87). When stratified by ethnicity, Q576R G allele carriers were associated with a decreased cancer risk in Caucasians (dominant model: OR = 0.90, 95%CI = 0.83-0.98; heterozygote comparison: OR = 0.89, 95%CI = 0.82-0.98). I75V G allele carriers were associated with a decreased cancer risk in Asians (heterozygote comparison: OR = 0.76, 95%CI = 0.62-0.94). S503P C allele carriers were also associated with a decreased cancer risk in Asians (CC VS TT: OR = 0.29, 95%CI = 0.08-0.99). Our results suggest that Q576R, I75V and S503P may be associated with a decreased cancer risk for certain types of cancers and in some specific ethnic groups. Future case-control studies with large sample size are needed to evaluate these associations in detail.
