        miR-101 Inhibiting Cell Proliferation, Migration and Invasion in Hepatocellular Carcinoma through Downregulating Girdin

        miR-101 is considered to play an important role in hepatocellular carcinoma (HCC), but the underlying molecular mechanism remains to be elucidated. Here, we aimed to confirm whether Girdin is a target gene of miR-101 and determine the tumor suppressor of miR-101 through Girdin pathway. In our previous studies, we firstly found Girdin protein was overexpressed in HCC tissues, and it closely correlated to tumor size, T stage, TNM stage and Edmondson-Steiner stage of HCC patients. After specific small interfering RNA of Girdin was transfected into HepG2 and Huh7.5.1 cells, the proliferation and invasion ability of tumor cells were significantly inhibited. In this study, we further explored the detailed molecular mechanism of Girdin in HCC. Interestingly, we found that miR-101 significantly low-expressed in HCC tissues compared with that in matched normal tissues while Girdin had a relative higher expression, and miR-101 was inversely correlated with Girdin expression. In addition, after miR-101 transfection, the proliferation, migration and invasion abilities of HepG2 cells were weakened. Furthermore, we confirmed that Girdin is a direct target gene of miR-101. Finally we confirmed Talen-mediated Girdin knockout markedly suppressed cell proliferation, migration and invasion in HCC while downregulation of miR-101 significantly restored the inhibitory effect. Our findings suggested that miR-101/Girdin axis could be a potential application of HCC treatment.

        Intracellular Localization and Sustained Prodrug Cell Killing Activity of TAT-HSVTK Fusion Protein in Hepatocelullar Carcinoma Cells

        Gene therapy with nonviral vectors using the suicide gene/prodrug activating system of herpes simplex virus type-1 thymidine kinase (HSV1-TK)/ganciclovir (GCV) is inefficient in killing malignant tumor cells due to two major factors: (a) an unsatisfactory bystander effect; (b) short-lived expression of the protein. To study the capacity of the protein transduction domain (PTD) of HIV-1 TAT protein to enhance HSV1-TK/GCV cancer gene therapy, we constructed three fusion proteins TAT-TK, TK-TAT and TK. TATTK retained as much enzyme activity as TK, whereas that of TK-TAT was much lower. TAT-TK can enter HepG2 cells and much of it is translocated to the nucleus. The transduced HepG2 cells are killed by exogenously added GCV and have bystander effects on untransduced HepG2 cells. Most importantly, the introduced recombinant protein is stable and remains functional for several days at least, probably because nuclear localization protects it from the cytoplasmic degradation machinery and provides access to the nuclear transcription machinery. Our results indicate that TAT fusion proteins traffic intercellularly and have enhanced stability and prodrug cell killing activity. We conclude that TAT has potential for enhancing enzyme prodrug treatment of liver cancers.

        Cuprum Metal-Organic-Framework and Polyacrylonitrile-Derived Cu-N-C Electrocatalyst for Application in Zinc-Air Batteries

        Cu-N-C electrocatalyst was successfully prepared with Cu-BTC and polyacrylonitrile as templates by electrospinning and in-situ growth method. The effect of various N-doped content on the electrochemical performance of the catalyst were investigated and the Cu-N-C-0.25 exhibited the best OER/ORRcatalytic activities. When it was applied to the cathode of the zinc-air batteries, the power density of Cu-N-C-0.25 was 209.1 mW cm -2, which exceeded that of the commercial Pt/C + IrO2 hybrid catalyst (157 mW cm -2). The excellent electrochemical performance may be attributed to large number of active sites and high specific surface area on Cu-N-C electrocatalyst. It will be a promising alternative to precious metal catalysts containing Pt, Ir or Ru and their oxides.

      • Improved Switching Ripple Suppression Strategy for Multi-Paralleled Grid-Connected Inverters System

        The switching operations of inverters in the distributed system inevitably introduce high-frequency harmonics into grid, which may make influences on electrical devices. To address it, this paper proposes a ripple suppression strategy based on active cancellation combined with passive filtering. The active cancellation is realized by carrier-phase-shift (CPS) technique and the passive filtering is achieved by filters. Furthermore, two component-shared LCL-type filters improved by traditional LCL filters are studied from the points of resonance and accuracy of gridinjected current. To valid the effectiveness of the proposed strategy, simulations and experimental test are carried out. The results show that the proposed strategy can effectively suppress high-frequency harmonics and two componentshared LCL-type filters can become promising filters for the multi-paralleled grid-connected inverters.

        Effects of Antioxidant Tempol on Systematic Inflammation and Endothelial Apoptosis in Emphysematous Rats Exposed to Intermittent Hypoxia

        Purpose: Obstructive sleep apnea and chronic obstructive pulmonary disease are independent risk factors of cardiovascular disease(CVD), and their coexistence is known as overlap syndrome (OS). Endothelial dysfunction is the initial stage of CVD; however,underlying mechanisms linking OS and CVD are not well understood. The aim of this study was to explore whether OS canlead to more severe inflammation and endothelial apoptosis by promoting endothelial dysfunction, and to assess the interventioneffects of antioxidant tempol. Materials and Methods: Male Wistar rats (n=66) were exposed to normal oxygen [normal control (NC) group], intermittent hypoxia(IH group), cigarette smoke (CH group), as well as cigarette smoke and IH (OS group). Tempol intervention was assessed inOS group treated with tempol (OST group) or NaCl (OSN group). After an 8-week challenge, lung tissues, serum, and fresh bloodwere harvested for analysis of endothelial markers and apoptosis. Results: The levels of intracellular adhesion molecule-1, vascular cellular adhesion molecule-1, and apoptosis in circulating epithelialcells were the highest in OS group and the lowest in NC group. These levels were all greater in IH group than in CH group,and were lower in OST group than in OS and OSN groups (all p<0.001). Conclusion: Synergistic effects of IH with cigarette smoke-induced emphysema produce a greater inflammatory status and endothelialapoptosis. OS-related inflammation and endothelial cell apoptosis may play important roles in promoting cardiovasculardysfunction, and antioxidant tempol could achieve a partial protective effect.

        Dendritic porous SnO2/SiO2@polymer nanospheres for pH-controlled styptic drug release

        Dendritic porous tin oxide/silica (DTS) nanospheres (NSs) were prepared by emulsion-condensationroute. DTS NSs have an average diameter of 80 nm and possess accessible center-radial large porechannels, which endow them high surface area, less drug diffusion resistance and more mass transport. SnO2 quantum dots (QDs) embedded in silica skeleton can be used to track the drug release behavior. DTS NSs were coated by chitosan/polymethacrylic acid and polymer shell would make them achievegreat loading capacity and pH-sensitive release for aminomethylbenzoic acid (AMBA).

        Enhanced Performance of Near-Infrared-Absorption CdSeTe Quantum Dot-Sensitized Solar Cells Via Octa-Aminopropyl Polyhedral Oligomeric Silsesquioxane Modification

        The charge recombination caused by surface defects limits photovoltaic properties of quantum dot-sensitized solar cells (QDSSCs), which can be suppressed by modifying organic or inorganic molecules and atomic ligands. In this paper, octa-aminopropyl polyhedral oligomeric silsesquioxane (OA-POSS) connected and modified near-infrared absorption CdSeTe quantum dots (QDs) through coupling agent (1-ethyl-3-3-dimethylaminopropyl carbodiimide hydrochloride). The results suggest that OA-POSS reduces the surface defects of CdSeTe QDs and suppresses charge recombination. Therefore, the power conversion efficiency improves nearly 41%, which increases from 2.00% to 2.82%.

        Secondary Short-Lasting Unilateral Neuralgiform Headache with Conjunctival Injection and Tearing: A New Case and a Literature Review

        Short-lasting unilateral neuralgiform headache attacks with conjunctival injection and tearing (SUNCT) is a primary headache syndrome with an unclear pathogenesis. However, there is increasing evidence in the literature for secondary SUNCT being attributable to certain known lesions. We explored the possible neurobiological mechanism underlying SUNCT based on all reported cases of secondary SUNCT for which detailed information is available. Here we report a case of neuromyelitis optica spectrum disorders that had typical symptoms of SUNCT that might have been attributable to involvement of the spinal nucleus of the trigeminal nerve. We also review cases of secondary SUNCT reported in the English-language literature and analyze them for demographic characteristics, clinical features, response to treatment, and imaging findings. The literature review shows that secondary SUNCT can derive from a neoplasm, vascular disease, trauma, infection, inflammation, or congenital malformation. The pons with involvement of the trigeminal root entry zone was the most commonly affected region for inducing secondary SUNCT. In conclusion, the neurobiology of secondary SUNCT includes structures such as the nucleus and the trigeminal nerve with its branches, suggesting that some cases of primary SUNCT have underlying mechanisms that are related to existing focal damage that cannot be visualized.
