        Feasibility of Using a One-temperature Model for Determining Plasma’s Temperature by Measuring Its Refractive Index

        In this paper, the feasibility of using a one-temperature model to determine plasma’s temperatureby measuring its refractive index is studied based on the consideration that many plasma flow fieldsare not in local thermodynamic equilibrium (LTE). We compare the refractive indices between theone- and the two-temperature models of an arc plasma, in order to discuss the applicability ofthe one-temperature model in optical diagnosis. We found that the applicability is determined bythe pressure, the temperature ratio between electrons and heavy particles, the probe wavelength,and the chosen optical method’s refractive index measurement precision. Consequently, a commonmodel is deduced to describe the refractive index of plasma whether it be in LTE or not. In a word,this study is crucial to better apply optical methods based on measurements of the refractive indexto diagnose plasma flow fields.

        Reference Gene Screening for Analyzing Gene Expression Across Goat Tissue

        Real-time quantitative PCR (qRT-PCR) is one of the important methods for investigating the changes in mRNA expression levels in cells and tissues. Selection of the proper reference genes is very important when calibrating the results of real-time quantitative PCR. Studies on the selection of reference genes in goat tissues are limited, despite the economic importance of their meat and dairy products. We used real-time quantitative PCR to detect the expression levels of eight reference gene candidates (18S, TBP, HMBS, YWHAZ, ACTB, HPRT1, GAPDH and EEF1A2) in ten tissues types sourced from Boer goats. The optimal reference gene combination was selected according to the results determined by geNorm, NormFinder and Bestkeeper software packages. The analyses showed that tissue is an important variability factor in genes expression stability. When all tissues were considered, 18S, TBP and HMBS is the optimal reference combination for calibrating quantitative PCR analysis of gene expression from goat tissues. Dividing data set by tissues, ACTB was the most stable in stomach, small intestine and ovary, 18S in heart and spleen, HMBS in uterus and lung, TBP in liver, HPRT1 in kidney and GAPDH in muscle. Overall, this study provided valuable information about the goat reference genes that can be used in order to perform a proper normalisation when relative quantification by qRT-PCR studies is undertaken.

        Effectiveness and Safety of Dabrafenib in the Treatment of 20 Chinese Children with BRAFV600E-Mutated Langerhans Cell Histiocytosis

        Purpose We sought to investigate the effectiveness and safety of dabrafenib in children with BRAFV600E-mutated Langerhans cell histiocytosis (LCH). Materials and Methods A retrospective analysis was performed on 20 children with BRAFV600E-mutated LCH who were treated with dabrafenib. Results The median age at which the patients started taking dabrafenib was 2.3 years old (range, 0.6 to 6.5 years). The ratio of boys to girls was 2.3:1. The median follow-up time was 30.8 months (range, 18.9 to 43.6 months). There were 14 patients (70%) in the risk organ (RO)+ group and six patients (30%) in the RO– group. All patients were initially treated with traditional chemotherapy and then shifted to targeted therapy due to poor control of LCH or intolerance to chemotherapy. The overall objective response rate and the overall disease control rate were 65% and 75%, respectively. During treatment, circulating levels of cell-free BRAFV600E (cfBRAFV600E) became negative in 60% of the patients within a median period of 3.0 months (range, 1.0 to 9.0 months). Grade 2 or 3 adverse effects occurred in five patients. Conclusion Some children with BRAFV600E-mutated LCH may benefit from monotherapy with dabrafenib, especially high-risk patients with concomitant hemophagocytic lymphohistiocytosis and intolerance to chemotherapy. The safety of dabrafenib is notable. A prospective study with a larger sample size is required to determine the optimal dosage and treatment duration.

      • CYP1A1 Genetic Polymorphisms and Risk for Esophageal Cancer: a Case-control Study in Central China

        The purpose of this study was to evaluate the associations of CYP1A1 genetic polymorphisms with the risk of developing esophageal cancer (EC). A case-control study was carried out in a Chinese population in which 157 hospital based EC cases and 157 population based healthy controls with 1:1 match by age and sex were included. PCR based restriction fragment length polymorphisms (PCR-RFLP) were used to detect genotypes in case and control groups. For the CYP1A1 Ile/Val polymorphism, comparing with wild genotype Ile/Ile, both the heterozygote genotype Ile/Val and the combined variant genotype Ile/Val+Val/Val increased the risk of esophageal cancer (OR: 2.05, 95%CI: 1.19-3.54, OR: 1.86, 95%CI: 1.11-3.12). No significant association was found between the CYP1A1 MspI polymorphism and EC. According to analysis of combined genotypes, the TC/AG combined genotype which contained both variant alleles of these two polymorphisms increased the risk of developing EC (OR: 2.12, 95%CI: 1.16-3.85). Our results suggested that genetic polymorphisms of CYP1A1 may increase the susceptibility to EC.

        High Ratio Bidirectional DC-DC Converter with a Synchronous Rectification H-Bridge for Hybrid Energy Sources Electric Vehicles

        In order to match the voltages between high voltage battery stacks and low voltage super-capacitors with a high conversion efficiency in hybrid energy sources electric vehicles (HESEVs), a high ratio bidirectional DC-DC converter with a synchronous rectification H-Bridge is proposed in this paper. The principles of high ratio step-down and step-up operations are analyzed. In terms of the bidirectional characteristic of the H-Bridge, the bidirectional synchronous rectification (SR) operation is presented without any extra hardware. Then the SR power switches can achieve zero voltage switching (ZVS) turn-on and turn-off during dead time, and the power conversion efficiency is improved compared to that of the diode rectification (DR) operation, as well as the utilization of power switches. Experimental results show that the proposed converter can operate bidirectionally in the wide ratio range of 3~10, when the low voltage continuously varies between 15V and 50V. The maximum efficiencies are 94.1% in the Buck mode, and 93.6% in the Boost mode. In addition, the corresponding largest efficiency variations between SR and DR operations are 4.8% and 3.4%. This converter is suitable for use as a power interface between the battery stacks and super-capacitors in HESEVs.

        A Bidirectional Three-level DC-DC Converter with a Wide Voltage Conversion Range for Hybrid Energy Source Electric Vehicles

        In order to meet the increasing needs of the hybrid energy source system for electric vehicles, which demand bidirectional power flow capability with a wide-voltage-conversion range, a bidirectional three-level DC-DC converter and some control strategies for hybrid energy source electric vehicles are proposed. The proposed topology is synthesized from Buck and Boost three-level DC-DC topologies with a high voltage-gain and non-extreme duty cycles, and the bidirectional operation principle is analyzed. In addition, the inductor current ripple can be effectively reduced within the permitted duty cycle range by the coordinated control between the current fluctuation reduction and the non-extreme duty cycles. Furthermore, benefitting from duty cycle disturbance control, series-connected capacitor voltages can also be well balanced, even with the discrepant rise and fall time of power switches and the somewhat unequal capacitances of series-connected capacitors. Finally, experiment results of the bidirectional operations are given to verify the validity and feasibility of the proposed converter and control strategies. It is shown to be suitable for hybrid energy source electric vehicles.

        Research on the Model, Structure and Characteristics of a New Vibration Generator

        The vibrational energy is prevalent in the natural environment, which is studied by energy researchers as a new energy resource in recent years. Vibration generation utilizes electromagnetic induction technology, piezoelectric technology and certain characteristics of smart materials to convert mechanical energy into electrical energy. In this paper, a new method of using MSMA (magnetic shape memory alloy) to generate electricity is proposed and the principle of generating electricity is demonstrated. Martensitic variants and magnetic domain characteristics of MSMA are analyzed. Combining with Gibbs free energy function thermal theory, the mathematics model of MSMA vibration generator is established. The basic structure of MSMA vibration generator is designed and simulation is done to analyze that the effects of generator output voltage when the input amplitude and frequency of vibration stress change. The simulation experiments verify the feasibility of using MSMA to make the micro vibration generators and the correctness of the mathematical model, which lays a good foundation for the further research and application of MSMA vibration generator.

        Calcineurin-Responsive Transcription Factor CgCrzA Is Required for Cell Wall Integrity and Infection-Related Morphogenesis in Colletotrichum gloeosporioides

        The ascomycete fungus Colletotrichum gloeosporioides infects a wide range of plant hosts and causes enormous economic losses in the world. The transcription factors (TFs) play an important role in development and pathogenicity of many organisms. In this study, we found that the C2H2 TF CgCrzA is localized in both cytoplasm and nucleus under standard condition, and it translocated from cytoplasm to nucleus in a calcineurin- dependent manner. Moreover, the ΔCgCrzA was hypersensitive to cell wall perturbing agents and showed severe cell wall integrity defects. Deletion of the CgCRZA inhibited the development of invasive structures and lost pathogenicity to plant hosts. Our results suggested that calcineurin-responsive TF CgCrzA was not only involved in regulating cell wall integrity, but also in morphogenesis and virulence in C. gloeosporioides.

      • Efficacy and Safety of Endostar<sup>®</sup> Combined with Chemotherapy in Patients with Advanced Soft Tissue Sarcomas

        Background: Soft tissue sarcomas (STS) are a heterogeneous group of tumors, and approximately 40-50% of patients with STS develop metastatic disease. The median overall survival of those patients was 12 months and their 5-year survival rate was 8%. Therefore, study on more effective treatment, especially the targeting therapies, is urgently needed. Objective: To evaluate the efficacy and safety of Endostar$^{(R)}$ combined with chemotherapy in patients with advanced STS. Methods: A retrospective case-series study was conducted in Cancer Institute of PLA, Xinqiao Hospital. A total of 71 patients suffering from advanced STS (IIB - IV) were included, of whom 49 cases treated with chemotherapy alone were defined as the control group and the rest 22 cases treated with the traditional chemotherapy combined with Endostar$^{(R)}$ were defined as the test group. The short-term therapeutic effects including objective response rate (ORR), disease control rate (DCR) and safety were evaluated in the two groups. In the follow-up, progression-free survival (PFS) and overall survival (OS) were also observed. Results: In the test and control groups, the ORR was 18.2% and 12.2%, respectively (P=0.767), and the DCR was 86.4% and 61.2%, respectively (P=0.034). The median time to progression in the test and control groups was 120 days and 70 days with significant difference (P = 0.017), while the median overall survival was 452 days and 286 days without significant difference (P=0.503). The one-year survival rate in the test group and control group was 56.2% and 35.4%, respectively, while the two-year survival rate was 30.2% and 26.5%, respectively. No significant difference in the side effects was found between the two groups. Conclusions: Endostar$^{(R)}$ combined with chemotherapy resulted in a higher DCR and longer PFS in the patients with advanced STS, and the toxicity was tolerable.
