        Modulation of the Self-Assembly of Collagen by Phytic Acid: An In Vitro Study

        Phytic acid, containing a myoinositol ring coupled with six phosphate groups, can react with the amino groups of collagen to regulate their self-assembly behavior. The aim of this research is to evaluate the effects of phytic acid on the selfassembly behavior of collagen, the structures and properties of the resulting fibrils and hydrogels. Turbidity and chloramine T assay suggested that phytic acid could improve the self-assembly kinetics and degree of collagen, and the optimal ratio of phytic acid/collagen was 1/1 (w/w). Scanning electron microscopy (SEM) analysis indicated that co-fibrils of collagen with phytic acid are more slender than that of pure collagen, and transmission electron microscopy (TEM) reveals that the characteristic D-periodicity of collagen fibrils is not affected by phytic acid. Besides, differential scanning calorimetry (DSC) and rheology revealed that the thermal stability of collagen fibrils and the viscoelasticity of collagen hydrogels could be improved by phytic acid and the optimal ratio of phytic acid/collagen is 1/1 (w/w).

        A Two-Dimensional Risk Assessment Method for Oil and Gas Pipelines Based on “Accident Probability–Reputation Loss”

        Oil and gas pipeline accidents not only caused personnel casualties, economic losses and environmental pollution but also brought great panic to the public. To provide accurate accident prevention strategies, risk assessment (RA) has been widely used in the oil and gas pipeline industry. It aims to quantify the potential risk due to unexpected eventualities. However, the current RA methods mainly pay attention to the tangible risk (such as the human safety risk, economic risk and environmental risk), and the intangible risk (such as reputation loss risk) is often neglected. This cannot refl ect the real risk for the oil and gas pipeline. To settle existing research limitation, a two-dimensional risk assessment method for oil and gas pipelines based on “accident probability–reputation loss” is proposed. The method mainly involves four steps, namely, risk identifi cation, probability calculation, consequence assessment and risk assessment. The proposed method is used to quantitatively assess a natural gas pipeline in central China. Its results indicate that pipeline risk is unacceptable due to the unacceptability of accident probability and various dimensions of risk. Therefore, this study can enrich and develop the current oil and gas pipeline RA method to refl ect the real risk of oil and gas pipeline accidents.

        The Predictive Value of Magnetic Resonance Imaging-based Texture Analysis in Evaluating Histopathological Grades of Breast Phyllodes Tumor

        Purpose: Knowing the distinction between benign and borderline/malignant phyllodes tumors (PTs) can help in the surgical treatment course. Herein, we investigated the value of magnetic resonance imaging-based texture analysis (MRI-TA) in differentiating between benign and borderline/malignant PTs. Methods: Forty-three women with 44 histologically proven PTs underwent breast MRI before surgery and were classified into benign (n = 26) and borderline/malignant groups (n = 18 [15 borderline, 3 malignant]). Clinical and routine MRI parameters (CRMP) and MRI-TA were used to distinguish benign from borderline/malignant PT. In total, 298 texture parameters were extracted from fat-suppression (FS) T2-weighted, FS unenhanced T1-weighted, and FS first-enhanced T1-weighted sequences. To evaluate the diagnostic performance, receiver operating characteristic curve analysis was performed for the K-nearest neighbor classifier trained with significantly different parameters of CRMP, MRI sequence-based TA, and the combination strategy. Results: Compared with benign PTs, borderline/malignant ones presented a higher local recurrence (p = 0.045); larger size (p < 0.001); different time-intensity curve pattern (p = 0.010); and higher frequency of strong lobulation (p = 0.024), septation enhancement (p = 0.048), cystic component (p = 0.023), and irregular cystic wall (p = 0.045). TA of FS T2- weighted images (0.86) showed a significantly higher area under the curve (AUC) than that of FS unenhanced T1-weighted (0.65, p = 0.010) or first-enhanced phase (0.72, p = 0.049) images. The texture parameters of FS T2-weighted sequences tended to have a higher AUC than CRMP (0.79, p = 0.404). Additionally, the combination strategy exhibited a similar AUC (0.89, p = 0.622) in comparison with the texture parameters of FS T2-weighted sequences. Conclusion: MRI-TA demonstrated good predictive performance for breast PT pathological grading and could provide surgical planning guidance. Clinical data and routine MRI features were also valuable for grading PTs.

        Ziwuliuzhu Acupuncture Modulates Clock mRNA, Bmal1 mRNA and Melatonin in Insomnia Rats

        Background: In clinics, Ziwuliuzhu acupuncture is widely considered an effective method of treating insomnia; however, there is currently limited information available regarding its possible mechanisms. Although the method of Ziwuliuzhu acupuncture possesses a unique rhythmic pattern. Objectives: In this study, we have creatively combined the traditional Chinese medicine of Ziwuliuzhu with a modern biological rhythm to investigate the internal mechanism of insomnia. Methods: Pathological tissue from the hypothalamus was analyzed using hematoxylin– eosin staining. The level of TNF (tumor necrosis factor)-α in the SCN (suprachiasmatic nucleus) area of the hypothalamus was detected in situ using the TUNEL fluorescence staining assay. The concentration of hypothalamic melatonin was detected using the enzyme-linked immunosorbent assay (ELISA). The mRNA expression of Clock and Bmal1 was measured using RT-qPCR. Results: In the Ziwuliuzhu acupuncture groups, the structural damage in the hypothalamic neurons was alleviated compared to the model group and the expression of inflammatory factors was reduced. The mRNA expression levels of Clock and Bmal1 were significantly increased (p < 0.05). The concentration of melatonin was significantly increased (p < 0.001). Although there were no significant differences between the treatment groups (diazepam group, Nazi group, Najia group, and routine group) (p > 0.05). Conclusion: Ziwuliuzhu acupuncture alleviated neuronal damage and modulated the inflammatory reaction in the hypothalamus of rats with insomnia.
