      • Synthesis and biological evaluation of the mimics of cis ligand for CD22

        Yuki Sugamuna,Naoko Matsubara,Yuki Iwayama,Akiharu Ueki,Akihiro Imamura,Hiromune Ando,Takeshi Tsubata,Hideharu Ishida,Makoto Kiso 한국당과학회 2012 한국당과학회 학술대회 Vol.2012 No.1

CD22 (siglec-2) is an accessory molecule of the B-cell receptor complex (BCR) that exertsnegative effects on receptor signaling. It is also well-documented that CD22 is a regulatoryprotein that sets a threshold for immune responses. The carbohydrate ligand recognized byCD22 is the sequence Neuα(2,6)Galβ(1,4)GlcNAc found on both neighboringglycoconjugate of the same cell (cis ligand) and on other cells that interact with B cells (transligands). Recently, we have reported that the C-9 amido derivative of sialic acid (GSC718;9-(4’-hydroxy-4-biphenyl)acetamido-9-deoxy-Neu5Gc-OBn) show a potent affinity andselectivity for CD22 than other siglecs such as MAG [1]. Moreover, the compoundpromoted the proliferation of B cells in vitro. As next step of our investigation, we intend toreinforce the promoting activity of GSC718 for B cell growth by chemical modification.Herein, we report the efficient synthesis of GSC718 analogs which have varied aglyconmoieties. To achieve the comprehensive synthesis of GSC718 analogs having varied aglycons, wereexamined every synthetic process to obtain a fine target compound. In case of thesialoside synthesis, the most time-consuming and troublesome process is thechromatographic separation of α-sialoside from other byproducts such as β−isomer, 2,3-enederivative etc. after glycosylation reaction with aglycon part. To improve this process, weemployed 1,5-lactam formation as the key step for separation because the lactam formation isknown to proceed only in α-sialoside [2]. At the beginning of the synthesis of targetmolecules, we synthesized a suitably modified sialic acid donor in good yields. Then, thesialyl donor was reacted with various 2-substituted-ethanols to give the mixtures of α- and β-glycosides and 2,3-ene derivative, which were subsequently advanced to 1,5-lactamformation. As we anticipated, 1,5-lactamized α-sialosides became isolable from themixtures due to its different polarity from the other byproducts. Finally, the obtained 1,5-lactamized silaosides were successfully converted into target structures via reaction sequenceincluding C9-midification with biphenyl amide group, lactam opening and globaldeprotection. The synthesized analogs were advanced to biological assay using B cells. In this poster presentation, we will also discuss the structure-activity relationships of thesynthesized analogs. [1] H. H. M. • Recognition of Surrounding Environment for Electric Wheelchair Based on WideSeg

        Yuki SAKAI,Yoshiki NAKAYAMA,Huimin LU,Yujie LI,Hyoungseop KIM 제어로봇시스템학회 2019 제어로봇시스템학회 국제학술대회 논문집 Vol.2019 No.10

At present, the aging population is growing in Japan. Along with that, the expectation for the utilization of welfare equipment is increasing. Electric wheelchair, a convenient transportation tool, is popularized rapidly. On the other hand, accidents have occurred, and the dangers for driving are pointed out. Therefore, it needs to improve accident factors, reduce accidents and improve the convenience of electric wheelchair by automation. Environmental recognition is necessary for the development of autonomous electric wheelchair. Environmental recognition includes self-position estimation, recognition of sidewalks, crosswalks and traffic lights, moving object prediction, etc. In order to solve these various problems, this paper examines the segmentation of sidewalks, crosswalks and traffic lights. We develop the WideSeg that is one of semantic segmentation algorithms applying convolutional neural networks (CNN).

      • Equivalency of Series-Parallel Hybrid System Using Planetary Gear and Hybrid System Using EVT

        Yuki Kubo,Satoshi Ogasawara,Masatsugu Takemoto 전력전자학회 2011 ICPE(ISPE)논문집 Vol.2011 No.5

The series-parallel hybrid system using a planetary gear realizes low-fuel consumption and restraining of the exhaust gas by performing an optimal control of the internal combustion engine (ICE) with various driving modes. On the other hand, the hybrid system using electric variable transmission (EVT) has been proposed. This paper presents a theoretical consideration on equivalency of the series-parallel hybrid system and the hybrid system using the EVT. Additionally, some experimental results and simulation results are compared to confirm equivalency of the two systems.

Running exercise and food restriction affect bone chemical properties in young female rats

        Yuki Aikawa,Yuich Noma,Umon Agata,Yuya Kakutani,Satoshi Hattori,Hitomi Ogata,Ken Kiyono,Naomi Omi 한국운동영양학회 2023 Physical Activity and Nutrition (Phys Act Nutr) Vol.27 No.2

[Purpose] To investigate the effects of a combination of running and food restriction on the chemical properties of the bone in young female rats using Raman spectroscopy. Furthermore, we investigated whether the chemical property parameters correlated with the bone-breaking strength. [Methods] Female Sprague–Dawley rats (7 weeks old) were randomly divided into four groups: sedentary and ad libitum feeding (SED, n = 8), voluntary running exercise and ad libitum feeding (EX, n = 8), sedentary and 30% food-restricted (SED-FR, n = 8), and voluntary running exercise and 30% food-restricted (EX- FR, n = 8). The experiment was conducted for a period of 12 weeks. Four parameters measured by Raman spectroscopy were used to evaluate the bone chemical quality. [Results] Exercise and restriction had significant interactions on the mineral to matrix ratio. The mineral-to-matrix ratio in the SED-FR group was significantly higher than that in the SED group and significantly lower in the EX-FR group than that in the SED-FR group. Running exercise had significant effects on increasing the crystallinity and carbonate-to-phosphate ratio. In the ad libitum intake condition, there were significant positive correlations between breaking energy and crystallinity (r = 0.593) and between breaking energy and carbonate-to-phosphate ratio (r = 0.854). [Conclusion] Our findings show that running exercise and food restriction, alone or in combination, affect the chemical properties of bone. Furthermore, under ad libitum intake conditions, positive correlations were found between the breaking energy and crystallinity, or carbonate-to-phosphate ratio.

      • Outlier/Noise-Robust Partition of Unity Implicit Surface Reconstruction

        Yukie Nagai,Yutaka Ohtake,Hiromasa Suzuki,Hideo Yokota (사)한국CDE학회 2010 한국CAD/CAM학회 국제학술발표 논문집 Vol.2010 No.8

In this paper, we propose an algorithm for outlier/noise-robust surface reconstruction based on a partition of unity (PU) approach. PU based surface reconstruction is a local method that covers an area including sampling points with spherical supports of local approximations, and then generates an approximation function whose zero-level sets approximate the surface. This algorithm has many advantages including representation of fine details, and fast and memory efficient computation. Many of these advantages are realized with the locality of PU however, it is also the reason of outlier/noise-instabilities. Unfortunately, scanned data generally contain much amount of noise, and hence improving the robustness of PU based algorithm is required. We achieve an outlier/noise-robust algorithm with integrating Graph-cut and diffusion of local approximations. Since the characteristics of outliers and noise are fundamentally different, overcoming these two with different approaches is reasonable. In our algorithm, first a spherical cover of an area containing input points is generated following the PU manner. And then Graph-cut is performed in order to determine spherical supports which are considered wrongly approximating affected by outliers. Finally, the PU approximation function is updated so that its gradient field smoothed. This smoothing is based on a diffusion of the local approximations. In this paper we show the effects of this integration approach for several scanned data sets.

Prognostic significance of tumor laterality in advanced ovarian cancer

        ( Yuki Yamada ),( Seiji Mabuchi ),( Naoki Kawahara ),( Ryuji Kawaguchi ) 대한산부인과학회 2021 Obstetrics & Gynecology Science Vol.64 No.6

Objective This study aimed to investigate the effect of incorporating tumor laterality into the International Federation of Gynecology and Obstetrics (FIGO) staging system for advanced ovarian cancer. Methods The clinical data of 131 patients with advanced ovarian cancer treated between 2008 and 2018 were retrospectively reviewed. To investigate the prognostic significance of tumor laterality, we divided the patients into unilateral and bilateral groups. The prognostic significance of tumor laterality (bilateral vs. unilateral) was evaluated using univariate and multivariate analyses. The effect of incorporating tumor laterality into the FIGO staging system to predict survival outcomes was evaluated using the Kaplan-Meier method. Results Both overall survival (OS) and progression-free survival (PFS) were longer in the unilateral group than in the bilateral group. Multivariate analysis demonstrated that tumor laterality was an independent predictor of OS (hazard ratio, 1.75; confidence interval, 1.05-2.92; P=0.032). In patients with stage III disease, the bilateral group had a shorter OS than the unilateral group, but it was comparable to the OS in stage IV patients (P=0.354). The incorporation of tumor laterality into the FIGO staging system improved the stratification of survival probabilities. Conclusion Tumor laterality can be an independent prognostic factor in patients with advanced ovarian cancer. The incorporation of tumor laterality may improve the predictive performance of the FIGO staging system in patients with advanced ovarian cancer.

Fabrication and evaluation of various types of micro one-way valves through micro rubber molding process

Yuki UOHASHI,Koichi SUZUMORI,Hironari TANIGUCHI 대한기계학회 2010 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.24 No.1

In recent years, research and development on micro fluid systems has become active in the fields of chemical technology and biotechnology. For the realization of these micro fluid systems, micro fluid devices such as micro-valves and micro pumps are very important. This paper reports the fabrication and experimental evaluation of several micro one-way valves. The valves are fabricated through a micro molding process, which makes the fabrication process simple and suitable for mass production. The experimental results indicate that the developed valve has great performance with a normal flow rate of 130.2 ml/min and a leak flow of 2.2 ml/min. The normal flow rate is 1.4 times higher than that of the previous model.

      • SYNTHESIS AND SPECTROSCOPIC CHARACTERIZATION OF SALMON DNA-WRAPPED SINGLE-WALL CARBON NANOTUBES

        YUKI ASADA,HIROFUMI DOHI,SHOTA KUWAHARA,TOSHIKI SUGAI,RYO KITAURA,HISANORI SHINOHARA 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2007 NANO Vol.2 No.5

Single-wall carbon nanotubes (SWNTs) can be well-dispersed in water by wrapping with short segments of natural DNA from salmon sperm. We report here the isolated DNA-wrapped SWNT hybrids. Measurements were carried out using UV-vis-NIR, near-infrared photoluminescence (PL) spectrum and atomic force microscopy (AFM). A possible charge transport between SWNTs and salmon-DNA is discussed in terms of observed spectral shifts in the photoluminescence spectra.
