        Establishment of a model to evaluate the nutritional quality of Bombyx mori Linnaeus (Lepidoptera, Bombycidae) pupae lipid based on principal components

        Silkworm pupae are a source of edible lipid with a high content of unsaturated fatty acids (UFAs) (70% of total lipid content) and are becoming a focus of pharmaceutical and dietary research in the functional oil field. To study the nutritional value of different silkworm strains, principal component analysis (PCA) was used to establish a mathematical model and cluster analysis on silkworm pupae lipid (SPL) in 90 strains based on Z-score. Single-factor correlation analysis indicated that the PUFAs content was significantly positively correlated with the α-linolenic acid (ALA) (ρ=0.98) and negatively correlated with the oleic acid (OA) (ρ=−0.73) contents, and comprehensive of principal components mathematical model (PC =0.148ZX1 +0.00 2ZX2 −0.197ZX3 −0.338ZX4 +0.220ZX5 +0.304ZX6 +0.314ZX7, ZXi is the standard score (Z-score) of X1, which was calculated as follow: ZXi = (Xi −−X )/S, where X1: palmitic acid, X2: palmitoleic acid, X3: stearic acid, X4: OA, X5: linoleic acid, X6: ALA, X7: PUFAs.−X is the arithmetic mean content of variable i, and S is the standard deviation of variable i for all samples included in the analysis.) showed the amount of ALA and PUFAs were the key factors determining the nutritional value of the pupae lipid. Cluster analysis on the composite scores calculated by the model of the principal component divided the 90 strains into three groups. A cluster of 6 strains (XHR, XIFF, ZX, YT, YO and BTN) with the best nutritional value was characterized by high contents of ALA and PUFAs and a low content of oleic acid.

        Facile Self-Assembly of Fe3O4 Nanoparticles@WS2 Nanosheets: A Promising Candidate for Supercapacitor Electrode

        Graphene-like dichalcogenides with huge surface area and nanostructuredtransition metal oxides with extraordinarily high theoretical capacities could becomposited as promising electrode candidates for supercapacitors. In this work,monolayer and few-layers WS2 nanosheets were exfoliated by combination ofball-milling and sonication. A facile strategy for the hierarchical self-assembly ofFe3O4 nanoparticles (Fe3O4NPs) on WS2 nanosheets was developed to synthesizeFe3O4NPs@WS2 nanocomposites via hydrothermal method. Fe3O4NPs areuniformly dispersed on the WS2 nanosheets without aggregation. The particlesize of Fe3O4NPs is about 3 nm. The nanocomposite shows strong enhancementsof electrochemical behaviors. This self-assembly synthesis strategy may havegreat prospects for other 0D/2D nanocomposites in supercapacitors and otherenergy devices.

        Effects of a traditional Chinese medicine formula and its extraction on muscle fiber characteristics in finishing pigs, porcine cell proliferation and isoforms of myosin heavy chain gene expression in myocytes

        Objective: This study evaluated the effects of a traditional Chinese medicine formula (TCMF) on muscle fiber characteristics in finishing pigs and the effects of the formula's extract (distilled water, ethyl acetate and petroleum ether extraction) on porcine cell proliferation and isoforms of myosin heavy chain (MyHC) gene expression in myocytes. Methods: In a completely randomized design, ninety pigs were assigned to three diets with five replications per treatment and six pigs per pen. The diets included the basal diet (control group), TCMF1 (basal diet+2.5 g/kg TCMF) and TCMF2 (basal diet+5 g/kg TCMF). The psoas major muscle was obtained from pigs at the end of the experiment. Muscle fiber characteristics in the psoas major muscle were analyzed using myosin ATPase staining. Cell proliferation was measured using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye and cytometry. Isoforms of MyHC gene expression were detected by real-time quantitative polymerase chain reaction. Results: The final body weight and carcass weight of finishing pigs were increased by TCMF1 (p<0.05), while the psoas major muscle cross-sectional area was increased by TCMF (p<0.05). The cross-sectional area and diameter of psoas major muscle fiber Ι, IIA, and IIB were increased by TCMF2 (p<0.05). The cross-sectional area and fiber diameter of psoas major muscle fiber IIA and IIB were increased by diet supplementation with TCMF1 (p<0.05). Psoas major muscle fiber IIA and IIB fiber density from the pigs fed the TCMF1 diet and the type IIB fiber density from the pigs fed the TCMF2 diet were lower compared to pigs fed the control diet (p<0.05). Pigs fed TCMF2 had a higher composition of type Ι fiber and a lower percentage of type IIB fiber in the psoas major muscle (p<0.05). The expression levels of MyHC Ι, MyHC IIa, and MyHC IIx mRNA increased and the amount of MyHC IIb mRNA decreased in the psoas major muscle from TCMF2, whereas MyHC Ι and MyHC IIx mRNA increased in the psoas major muscle from TCMF1 (p<0.05). Peroxisome proliferator-activated receptor ${\gamma}$ $coactivator-1{\alpha}$ and CaN mRNA expression in the psoas major muscle were up-regulated by TCMF (p<0.05). Porcine skeletal muscle satellite cell proliferation was promoted by $4{\mu}g/mL$ and $20{\mu}g/mL$ TCMF water extraction (p<0.05). Both $1{\mu}g/mL$ and $5{\mu}g/mL$ of TCMF water extraction increased MyHC IIa, MyHC IIb, and MyHC IIx mRNA expression in porcine myocytes (p<0.05), while MyHC Ι mRNA expression in porcine myocytes was decreased by $5{\mu}g/mL$ TCMF water extraction (p<0.05). Porcine myocyte MyHC Ι and MyHC IIx mRNA expression were increased, and MyHC IIa and MyHC IIb mRNA expression were down-regulated by $5{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). MyHC Ι and MyHC IIa mRNA expression in porcine myocytes were increased, and the MyHC IIb mRNA expression was decreased by $1{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). Four isoforms of MyHC mRNA expression in porcine myocytes were reduced by $5{\mu}g/mL$ TCMF petroleum ether extraction (p<0.05). MyHC IIa mRNA expression in porcine myocytes increased and MyHC IIb mRNA expression decreased by $1{\mu}g/mL$ in a TCMF petroleum ether extraction (p<0.05). Conclusion: These results indicated that TCMF amplified the psoas major muscle cross-sectional area through changing muscle fiber characteristics in finishing pigs. This effect was confirmed as TCMF extraction promoted porcine cell proliferation and affected isoforms of MyHC gene expression in myocytes.

      • Melatonin inhibits the Migration of Colon Cancer RKO cells by Down-regulating Myosin Light Chain Kinase Expression through Cross-talk with p38 MAPK

        Background: Melatonin, which is mainly produced by the pineal gland, has a good inhibitory effect on cell growth of multiple cancer types. However, the underlying molecular mechanisms of anti-tumor activity for colon cancer have not been fully elucidated. In this study, we investigated the effects of melatonin on migration in human colon cancer RKO cells and the potential molecular mechanisms. Materials and Methods: The viability of RKO cells was investigated by MTT assay after treatment with melatonin, SB203580 (p38 inhibitor) and phorbol 12-myristate 13-acetate (PMA, MAPK activator) alone or in combination for 48h. The effects of melatonin, and ML-7, a selective inhibitor of myosin light chain kinase (MLCK), and SB203580, and PMA on the migration of RKO cells were analyzed by in vitro scratch-wound assay. The relative mRNA levels of MLCK was assessed by real-time quantitative RT-PCR. Western blotting analysis was performed to examine the expression of MLCK, phosphorylation of myosin light chain (pMLC) and p38 (pp38). Results: The proliferation and migration of human colon cancer RKO cells were inhibited significantly after treatment with melatonin. The expression levels of MLCK and phosphorylation of MLC of RKO cells were reduced, and real-time quantitative RT-PCR showed that melatonin had significant effects on suppressing the expression of MLCK. Furthermore, the phosphorylation level of p38, which showed the same trend, was also reduced when cells were treated by melatonin. In addition, ML-7 (25umol/l) could down-regulate the phosphorylation of p38. Conclusions: Melatonin could inhibit the proliferation and migration of RKO cells, and further experiments confirmed that p38 MAPK plays an important role in regulating melatonin-induced migration inhibition through down-regulating the expression and activity of MLCK.

        Anatase Phase TiO2 Anode of a Dye-Sensitized Solar Cell Prepared by using RMFMS

        Anatase phase TiO2 films were deposited by using reactive medium frequency magnetron sputtering (RMFMS) technology under the conditions of a pure 99.9 % Ti disk as a target in an Ar+O2 atmosphere without additional external heating. The effect of the total pressure and bias on the phase composition, grain sizes and morphologies of the TiO2 films were characterized by using X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results showed that the crystalline anatase phase could be obtained under a total pressure of 0.4 5 Pa. The grain size of the anatase TiO2 was between 24 37 nm at a bias of 0 −200 V. Preferred (101) growth was found to occur at total pressures of below 1 Pa. We also discovered the existence of many holes distributed within the TiO2 films.       • Electrochemical Formation of a <i>p–n</i> Junction on Thin Film Silicon Deposited in Molten Salt

        <P>Herein we report the demonstration of electrochemical deposition of silicon <I>p–n</I> junctions all in molten salt. The results show that a dense robust silicon thin film with embedded junction formation can be produced directly from inexpensive silicates/silicon oxide precursors by a two-step electrodeposition process.         Evolution of the antioxidant capacity and phenolic contents of persimmon during fermentation

        The changes in antioxidant capacity and phenolics of persimmon during alcoholic fermentation, acetification, and short aging were investigated. An increase in the antioxidant activity was observed when persimmon was transformed from puree to vinegar. The total content of phenolics remained stable, in contrast to the concentration of condensed tannin, which significantly (p\0.05) increased during alcoholic and acetic fermentations, although followed by a decrease after aging. The phenolic compounds were characterized and quantitated. Gallic acid was the main phenolic compound, and its content increased by 14.4% during alcoholic fermentation and reduced by 53.5% during acetic fermentation. Additionally, the flavan- 3-ol compounds increased during alcoholic fermentation and acetification. Vanillyl alcohol, (-)-epigallocatechin, and p-coumaric acid were not observed in persimmon puree but detected in persimmon wine and vinegar. These results indicate that alcoholic and acetic fermentation can improve the antioxidant capacity of persimmon fruit.

        Long-term Complete Remission of Decitabine-Primed Tandem CD19/CD22 CAR-T Therapy with PD-1 and BTK Inhibitors Maintenance in a Refractory PCNSL Patient

        Primary central nervous system lymphoma (PCNSL) is a rare and aggressive non-Hodgkin’s lymphoma that affects the brain, eyes, cerebrospinal fluid, or spinal cord without systemic involvement. The outcome of patients with PCNSL is worse compared to patients with systemic diffuse large B-cell lymphoma. Given potential mortality associated with severe immune effector cell-associated neurotoxicity syndrome (ICANS), patients with PCNSL have been excluded from most clinical trials involving chimeric antigen receptor T-cell (CAR-T) therapy initially. Here, we report for the first time to apply decitabine-primed tandem CD19/CD22 dual-targeted CAR-T therapy with programmed cell death-1 (PD-1) and Bruton’s tyrosine kinase (BTK) inhibitors maintenance in one patient with multiline-resistant refractory PCNSL and the patient has maintained complete remission (CR) for a 35-month follow-up period. This case represents the first successful treatment of multiline resistant refractory PCNSL with long-term CR and without inducing ICANS under tandem CD19/CD22 bispecific CAR-T therapy followed by maintenance therapy with PD-1 and BTK inhibitors. This study shows tremendous potential in the treatment of PCNSL and offers a look toward ongoing clinical studies.

        In vitro Removal of Deoxynivalenol and T-2 Toxin by Lactic Acid Bacteria

        Five strains of lactic acid bacteria were tested for their ability to remove deoxynivalenol (DON) and T-2toxin from MRS broth. The ability of Lactobacillus plantarum strain 102 (LP102) was the strongest among 5strains after incubation at 37oC for 72 h. The mode of removal was physical binding, rather than biotransformation. The abilities were not significantly different between when removing single toxin and when removing mixed toxins by viable cells of LP102. DON and T-2 toxin released from LP102 viable cell-toxin complexes were 28.22±1.55 and 35.42±2.02% of total bound toxins respectively after 3times of wash with posphate buffered saline, respectively,those were 4.59±0.86 and 5.59±1.47% after incubation with simulated gastric fluid (SGF) at 37oC for 4 h, and 6.86±0.81 and 9.04±1.13% after incubation with simulated intestinal fluid (SIF) at 37oC for 4 h, respectively.

        Studies on meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential of pigs slaughtered at different growth stages

        Objective: This experiment investigated meat color, myoglobin content, enzyme activities, and expression of genes associated with oxidative potential of pigs slaughtered at different growth stages. Methods: Sixty 4-week-old Duroc×Landrace×Yorkshire pigs were assigned to 6 replicate groups, each containing 10 pigs. One pig from each group was sacrificed at day 35, 63, 98, and 161 to isolate longissimus dorsi and triceps muscles. Results: Meat color scores were higher in pigs at 35 d than those at 63 d and 98 d (p<0.05), and those at 98 d were lower than those at 161 d (p<0.05). The total myoglobin was higher on 161 d compared with those at 63 d and 98 d (p<0.05). Increase in the proportions of metmyoglobin and deoxymyoglobin and a decrease in oxymyoglobin were observed between days 35 and 161 (p<0.05). Meat color scores were correlated to the proportion of oxymyoglobin (r = 0.59, p<0.01), and negatively correlated with deoxymyoglobin and metmyoglobin content (r = –0.48 and –0.62, p<0.05). Malate dehydrogenase (MDH) activity at 35 d and 98 d was higher than that at 161 d (p<0.05). The highest lactate dehydrogenase/MDH ratio was achieved at 161 d (p<0.05). Calcineurin mRNA expression decreased at 35 d compared to that at 63 d and 98 d (p<0.05). Myocyte enhancer factor 2 mRNA results indicated a higher expression at 161 d than that at 63 d and 98 d (p<0.05). Conclusion: Porcine meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential varied at different stages.
