Identification of Novel Subregions of LOH in Gastric Cancer and Analysis of the HIC1 and TOB1 Tumor Suppressor Genes in These Subregions

        Jingcui Yu,Songbin Fu,Peng Liu,Xiaobo Cui,Yu Sui,Guohua Ji,Rongwei Guan,Donglin Sun,Wei Ji,Fangli Liu,An Liu,Yuzhen Zhao,Yang Yu,Yan Jin,Jing Bai,Jingshu Geng,Yingwei Xue,Jiping Qi,Ki-Young Lee 한국분자세포생물학회 2011 Molecules and cells Vol.32 No.1

Previously, we identified 3 overlapping regions showing loss of heterozygosity (LOH, R_1-R_3 from 11 to 30 cM) on chromosome 17 in 45 primary gastric cancers (GCs). The data indicated the presence of tumor suppressor genes (TSGs) on chromosome 17 involved in GC. Among the putative TSGs in these regions, HIC1 (in SR_1) and TOB1 (in SR_3) remain to be examined in GC. By immunohistochemistry (IHC), methylation-specific PCR (MSP) and western blot, we evaluated the expression and regulation status for HIC1 and TOB1 protein in GC. We narrowed down the deletion intervals on chromosome 17 and defined five smaller LOH subregions, SR_1-SR_5 (0.54 to 3.42 cM), in GC. We found that HIC1 had downregulated expression in 86% (91/106) and was methylated in 87% (26/30) of primary GCs. Of the primary GCs showing downregulation of HIC1 protein, 75% (18/24) had methylated HIC1 gene. TOB1 was either absent or expressed at reduced levels in 75% (73/97) of the GC samples. In addition, a general reduction was found in total and the ratio of unphosphorylated to phosphorylated TOB1 protein levels in the differentiated GC cell lines. Further analysis revealed significant simultaneous downregulation of both HIC1 and TOB1 protein in GC tissue microarray samples (67%, 52/78) and in primary GCs (65%, 11/17). These results indicate that silencing of HIC1 and TOB1 expression is a common occurrence in GC and may contribute to the development and progression of the disease.

Malignant transformation of ovarian mature cystic teratoma into squamous cell carcinoma: a Taiwanese Gynecologic Oncology Group (TGOG) study

        An Jen Chiang,Min-Yu Chen,Chia-Sui Weng,Hao Lin,Chien-Hsing Lu,Peng-Hui Wang,Yu-Fang Huang,Ying-Cheng Chiang,Mu-Hsien Yu,Chih-Long Chang 대한부인종양학회 2017 Journal of Gynecologic Oncology Vol.28 No.5

Objective: The malignant transformation (MT) of ovarian mature cystic teratoma (MCT)to squamous cell carcinoma (SCC) is very rare. This study analyzed cases from multiplemedical centers in Taiwan to investigate the clinicopathologic characteristics, treatment, andprognostic factors of this disease and reviewed related literature. Methods: Pathological reports of 16,001 patients with primary ovarian cancer who weretreated at Taiwan medical centers from 1990 to 2011 were reviewed. In total, 52 patients withMT of MCT to SCC were identified. Results: Among all ovarian MCTs, the incidence of MT to SCC is 0.2%. The median age ofpatients was 52 years (range, 29–89 years), and the mean tumor size was 10.5 cm (range, 1–40cm). We analyzed the patients in our study and those in the literature and determined thatearly identification and complete surgical resection of the tumor are essential for long-termsurvival. In addition, adjuvant chemotherapy or concurrent chemoradiotherapy can be usedto treat this malignancy. Old age, large tumor size (≥15.0 cm), and solid components in MCTsare suitable indicators predicting the risk of MT of MCT to SCC. Conclusion: Similar to general epithelial ovarian cancers, the early detection of MT of MCTto SCC is critical to long-term survival. Therefore, older patients with a large tumor or those with a tumor containing a solid component in a clinically diagnosed MCT should beevaluated to exclude potential MT to SCC.

      • Annealing temperature effect on self-assembled Au droplets on Si (111)

        Sui, Mao,Li, Ming-Yu,Kim, Eun-Soo,Lee, Jihoon Springer 2013 NANOSCALE RESEARCH LETTERS Vol.8 No.1

<P>We investigate the effect of annealing temperature on self-assembled Au droplets on Si (111). The annealing temperature is systematically varied while fixing other growth parameters such as deposition amount and annealing duration clearly to observe the annealing temperature effect. Self-assembled Au droplets are fabricated by annealing from 50°C to 850°C with 2-nm Au deposition for 30 s. With increased annealing temperatures, Au droplets show gradually increased height and diameter while the density of droplets progressively decreases. Self-assembled Au droplets with fine uniformity can be fabricated between 550°C and 800°C. While Au droplets become much larger with increased deposition amount, the extended annealing duration only mildly affects droplet size and density. The results are systematically analyzed with cross-sectional line profiles, Fourier filter transform power spectra, height histogram, surface area ratio, and size and density plots. This study can provide an aid point for the fabrication of nanowires on Si (111).</P>

      • Mini droplets to super droplets: evolution of self-assembled Au droplets on GaAs(111)B and (110)

Sui, Mao,Li, Ming-Yu,Kim, Eun-Soo,Lee, Jihoon International Union of Crystallography 2014 Journal of applied crystallography Vol.47 No.2

<P>In this article, the effect of deposition amount on self-assembled Au droplets fabricated on GaAs(111)B and (110) is presented. The investigation is systematically performed by the variation of the Au deposition amount from 2 to 20 nm while fixing the other growth parameters such as annealing temperature and duration to clearly observe the effect. Under identical growth conditions, the self-assembled Au droplets show significantly different size and density depending on the amount of Au deposition:<I>i.e.</I>the average height varies by 436% from 21.8 to 95.5 nm and the average diameter swings by 827% from 52 to 430 nm, showing that the size increase is dominated by the lateral expansion. Meanwhile the average density varies by over two orders of magnitude from 1.24 × 108to 4.48 × 1010 cm−2on GaAs(111)B. With relatively low Au deposition amounts, below 3 nm, round dome-shaped mini Au droplets with high packing density can be fabricated, while super large Au droplets result with higher deposition amounts, above 10 nm, with a density two orders of magnitude lower. It is also found that the surface index has a minor effect on the fabrication of self-assembled Au droplets with the variation of deposition amount. The results are systematically analyzed and discussed in terms of atomic force microscopy and scanning electron microscopy images, line profiles, power spectrums, r.m.s. surface roughness, and size and density plots.</P>

Adiponectin Regulates Bone Marrow Mesenchymal Stem Cell Niche Through a Unique Signal Transduction Pathway: An Approach for Treating Bone Disease in Diabetes : Adiponectin Regulates BMSC Niche

        Yu, Liming,Tu, Qisheng,Han, Qianqian,Zhang, Lan,Sui, Lei,Zheng, Leilei,Meng, Shu,Tang, Yin,Xuan, Dongying,Zhang, Jin,Murray, Dana,Shen, Qingping,Cheng, Jessica,Kim, Sung-Hoon,Dong, Lily Q.,Valverde, P Wiley (John WileySons) 2015 Stem Cells Vol.33 No.1

<P>Adiponectin (APN) is an adipocyte-secreted adipokine that exerts well-characterized antidiabetic properties. Patients with type 2 diabetes (T2D) are characterized by reduced APN levels in circulation and impaired stem cell and progenitor cell mobilization from the bone marrow for tissue repair and remodeling. In this study, we found that APN regulates the mobilization and recruitment of bone marrow-derived mesenchymal stem cells (BMSCs) to participate in tissue repair and regeneration. APN facilitated BMSCs migrating from the bone marrow into the circulation to regenerate bone by regulating stromal cell-derived factor (SDF)-1 in a mouse bone defect model. More importantly, we found that systemic APN infusion ameliorated diabetic mobilopathy of BMSCs, lowered glucose concentration, and promoted bone regeneration in diet-induced obesity mice. In vitro studies allowed us to identify Smad1/5/8 as a novel signaling mediator of APN receptor (AdipoR)-1 in BMSCs and osteoblasts. APN stimulation of MC3T3-E1 osteoblastic cells led to Smad1/5/8 phosphorylation and nuclear localization and increased SDF1 mRNA expression. Although APN-mediated phosphorylation of Smad1/5/8 occurred independently from adaptor protein, phosphotyrosine interaction, pleckstrin homology domain, and leucine zipper containing 1, it correlated with the disassembly of protein kinase casein kinase 2 and AdipoR1 in immunoprecipitation experiments. Taken together, this study identified APN as a regulator of BMSCs migration in response to bone injury. Therefore, our findings suggest APN signaling could be a potential therapeutic target to improve bone regeneration and homeostasis, especially in obese and T2D patients.</P>

Fabrication and determination of growth regimes of various Pd NPs based on the control of deposition amount and temperature on <i>c</i>-plane GaN

Sui, Mao,Kunwar, Sundar,Pandey, Puran,Zhang, Quanzhen,Li, Ming-Yu,Lee, Jihoon Published for the Materials Research Society by th 2017 Journal of materials research Vol.32 No.18

<▼1><B>Abstract</B><P/></▼1><▼2><P>Palladium (Pd) nanostructures have been actively adapted for various applications and their properties and applicability closely depend on their shape, size, and density. In this paper, the evolution of self-assembled Pd nanostructures on the hexagonal <I>c</I>-plane GaN is presented by the systematical control of Pd deposition amount (DA) at distinctive temperatures. Pd nanostructures of various configurations, sizes, and densities are demonstrated based on the solid-state dewetting of Pd thin films and a clear distinction in the growth regimes is observed. Three growth regimes are clearly observed depending on the variation of DA, i.e., (i) the agglomeration of Pd nanoparticles, (ii) the coalescence of wiggly Pd nanostructures, and finally (iii) the growth of nanovoids and layers. Owing to the temperature-dependent dewetting process, the growth regimes are markedly shifted, resulting in the distinctive Pd nanostructures within the identical DA range. The results are discussed in conjunction with the surface diffusion, Volmer-Weber and coalescence growth model, and surface/interface energy minimization mechanism. In addition, the evolution of optical properties, emission band, and lattice properties are probed by reflectance, photoluminescence, and Raman spectroscopy, which exhibit varying spectral intensity and peak positions according to the surface morphology of Pd nanostructures.</P></▼2>

        Balancing between "Two Evils"

        Yu Lei,Sui Sophia 한국학술연구원 2022 Korea Observer Vol.53 No.2

This paper finds that Pyongyang's nuclear issue is by no means China's "primary contradiction (top priority)" in its international relations. Chinese priority on the Korean peninsula is to keep stability and the status quo of a divided Korea rather than rush to find a resolution to the nuclear issue, which may result in a military imbalance on the Peninsula, or the collapse of the Pyongyang regime and hurt China's interest. To this end, China has adopted the strategy of making a delicate balance between pressuring and sustaining the Pyongyang regime. The paper concludes that from Chinese perspectives, a North Korea at the nuclear threshold contributes to counterbalancing US military presence in the region, maintaining a status quo of divided Koreas and helping China overcome US containment. China aspires to achieve three interconnected objectives with this strategy. This first is to sustain China's economic growth, the second to counterattack US containment, and the third to propel China's rise at the systemic (global) level.

Insights into the major phenolic acids in Perilla frutescens obtained by a sustainable procedure

Yu-Chiao Yang,Chia-Sui Wang,Ming-Chi Wei 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.96 No.-

This research was performed to evaluate the efficiency of ultrasound in obtaining the total extract, caffeicacid and rosmarinic acid from Perilla frutescens using supercritical CO2 (USCCO2) extraction. Comparedwith traditional heat-reflux, ultrasonic-assisted and SCCO2 extractions, the USCCO2 technique decreasedthe extraction duration (1.12–1.92 times) and the consumption of solvent (1.90–285.71 times) at lowerextraction temperatures and obtained a higher yield from the plant matrix. Furthermore, a second-orderkinetic model and Fick’s second diffusion model were implemented to ascertain the extraction rateconstant, equilibrium concentration, initial extraction rate, effective diffusion coefficient, mass transfercoefficient, Biot number, equilibrium constant and thermodynamic parameters of the USCCO2 and SCCO2dynamic extractions of rosmarinic acid from P. frutescens. These results provide valuable insights into thekinetic and mass transfer behaviors that occur during the USCCO2 and SCCO2 procedures and explain thereasons for the better extraction efficiency of the former procedure.

        Yu Ji,Yan Zhang,Ping Wang,Yuanyuan Li,Jianhua Sui 한국섬유공학회 2021 Fibers and polymers Vol.22 No.1

In this paper, a novel polyimide with trifluoromethyl group (PIS) was synthesized successfully and used toimprove the toughness of epoxy resins. Five modified epoxy resins (5, 7.5, 10, 12.5 and 15 wt%) were prepared andinvestigated, respectively. The structure, thermal properties, tensile properties, viscoelastic properties, toughness andmorphology of modified epoxy resins were studied by Fourier Transform Infrared Spectroscopy (FT-IR), thermogravimetricanalysis (TGA), Instron 5967, Dynamic Mechanical Analysis (DMA), and Scanning Electron Microscopy (SEM). Theresults showed that the epoxy resins modified with PIS exhibited good thermal stability which the glass transitiontemperatures increased from 135.9 oC to 146.4 oC. Tensile strength of modified epoxy resins increased from 40.4 MPa to92.3 MPa. The KIC values increased from 0.86 MPa·m1/2 to 1.56 MPa·m1/2 indicated that the toughness enhancement wasmarkedly. The fracture modes also were examined by SEM images and exhibited the fracture mode changed from the brittleone to ductile fracture with varied weight of the novel trifluoromethyl-polyimide.
