      • Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

        <P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression.         심근경색 모델 흰쥐에서 스트레스가 B-type Natriuretic Peptide 발현 및 심박변이율 변화에 미치는 영향

        심박변이율의 감소는 심근 경색 환자에서 예후의 악화와 관련되어 있다. 심박변이율 중 very low frequency가 심부전환자의 예후를 판단할 수 있는 독립적 인자로서 사용이 가능하다고 보고된 바 있으며, 심실의 압력 증가에 반응해서 생성되는 B-type natriuretic peptide가 심근 경색의 예후 인자로서 사용할 수 있음 역시 보고된 바 있다. 본 연구에서는 심근경색 모델을 제작하여 일정의 회복 기간을 거친 다음 다시 일정 기간의 스트레스를 겪게 한 후 심박변이율의 변화 및 심실 조직에서의 BNP 발현 정도를 비교하여 스트레스에 의한 영향을 관찰하고자 하였다. Sprague-Dawley계 수컷 흰쥐 15마리를 대상으로 하였다. 실험동물은 인위적 스트레스 및 수술을 받지 않은 대조군(CON, n=5), 심근경색 수술을 받은 후 restraint stress를 받지 않은 군(MI+No Stress, n=5), 심근경색 수술을 받은 후 1주일 동안 restraint stress를 받은 군(MI+Stress, n=5)으로 분류하였다. 심근경색 모델 제작 직후 15분 이상 심전도의 변화를 기록하였으며, 2개월 정도 후 심근경색 수술을 받은 동물을 두 군으로 나누어 그 중 한 군에는 1주일 동안 restraint stress를 가한 후 restraint stress를 가하지 않은 동물과 심박변이율을 비교 분석하였다. 심실 조직은 좌심실 전벽 부위의 위축을 관찰할 수 있었고, 대식세포에 의하여 응고, 괴사된 심근세포의 탐식과 혈관이 풍부한 육아조직 및 섬유 변화를 볼 수 있었다. 심박변이율은 심근경색 수술을 받은 직후 심박수는 유의한 증가를 보였고, standard deviation of the normal to normal intervals (SDNN), very low frequency (VLF) 및 low frequency (LF)의 유의한 감소를 보였다. 심근경색 수술을 받았던 동물에서 두 달 정도의 회복 기간을 거친 후 스트레스를 받지 않은 군은 심박수 및 기타 심박변이율 분석에서 SDNN 값을 제외하고는 정상군과 유의한 차이를 관찰할 수 없었던 반면 1주일 동안 스트레스를 받은 군에서는 심박수가 다시 유의하게 증가했을 뿐만 아니라 SDNN, VLF 및 LF 역시 정상군과 유의한 차이를 보였다. 이상의 결과를 종합하면 심근경색 동물에 대해 1주일 동안 스트레스를 가한 결과 심박변이율이 감소하고 심실에서의 BNP 발현은 더욱 증가하여 악화함을 보여주고 있다. Diminished heart rate variability (HRV) is associated with less favorable prognosis after myocardial infarction (MI). It has been reported that very low frequency (VLF) power in HRV analysis is an independent risk predictor in patients with congestive heart failure and B-type natriuretic peptide (BNP) can be used as a prognostic factor of MI. The purpose of this study was to investigate whether stress has an effect to the changes of BNP expression and/or heart rate variability in MI model in rats. Male Sprague-Dawley rats were divided into 3 groups: control group (CON), MI group (MI+No Stress), and MI followed by stress group (MI+Stress). MI+Stress group rats were raised for a two month recovery period after the operation, followed by being exposed to restraint stress for 2 hours per day for 1 week. Electrocardiogram was recorded after the operation and the last day after 1 week of stress. The frequency components of HRV were calculated in the frequency domain such as VLF, low frequency (LF), high frequency (HF) and so on. In HRV analysis, standard deviation of the normal to normal intervals (SDNN) was significantly reduced in both groups compared to the control group. VLF and LF also were significantly reduced in MI+Stress group compared to the control groups. In addition, BNP expression in western blotting was shown the strongest bands in MI+ Stress group among experimental groups. These results suggest that BNP and HRV were aggravated by stress in MI rat model. (Korean J Str Res 2010;18:275∼285)

And his interest about shi-jo and pansori is reflected in production of poetry. He creates four line poetry criticizing existent poetry style(shi-jo and pansori and so on)in poetry´s circles in the 1930s that form of free poetry catches seat already, and plays a variation four line poetry learning forms and groped new modernistic poetry form. He established peculiar sensitivity of individual into compensation of shape and create chard free verse of new form. These features that shows in poetry form that defend self from colony actual broken external order is revealed can be seen. It is because construct space of imagination and found psychological pupil of the eye in to close loss persimmon formed on inside looking away actuality that can not accomplish own dream. Therefore, that appears in his poetry sensual and the trillion brazier fortune worlds can see that is product of injustice mind to look away colony actuality. Kim Young-rang did not compromise in colony actuality and could keep own inside to be upright through writing such poetry.

      • 미치광이풀 모상근 배양에서 Tropane Alkaloid 생산성 증진을 위한 최적 생물학적 엘리시터 선발

        Scopolamine and hyoscyamine which belong to tropane alkaloids are the pharmaceutically valuable anticholinergic drugs. In order to increase the productivities, the effects of elicitation were investigated during hairy root cultures of Scopolia parviflora. Biotic elicitors originated from 3 fungi and 1 yeast were prepared as homogenate and supernatant and added to 3-week-old cultures. Both of homogenate and supernatant of Candida albicans elicitors increased the scopolamine production. The production of hyoscyamine was enhanced by homogenate of Fusarium solani and supematant of C. albicans. Most of the other fungal elicitors were also improved on the tropane alkaloid production compared to non-treatment. Among the elicitors tested, C. albicans was proved the optimal biotic elicitor on tropane alkaloids production. These results will be served mass production of tropane alkaloids by large-scale production.

      • 남성화를 보이는 여성에서 발견된 난소의 Steroid Cell Tumor 1예

        Steroid cell tumor is a rare ovarian sex cord-stromal tumor which accounts for 0.1% of all ovarian tumors. Until now, only 4 cases have been reported in domestic literatures. Steroid cell tumor often secrets testosterone and presents virilization in adult women or precocious puberty in children. Treatment is often performed by surgical removal, adjuvant chemotherapy and radiation, but completely accepted treatment was not existed. We experienced a case of steroid cell tumor, which was manifested by typical virilization in a 43-year old patient, who was previously performed hysterectomy and unilateral oophorectomy. So, we present with a brief review of the literatures.

        금 합금 보철물의 교합면 삭제로 인한 임플란트-골 계면으로의 열전달에 관한 연구

        연구 목적: 임플란트-골 계면에서 발생하는 과도한 열은 골유착을 저해하여 임플란트의 실패를 유발한다. 이에 이번 연구에서는 임플란트 금 합금 보철물의 교합면 삭제시 임플란트-골 계면으로의 열전달 양상과 냉각 방식의 효율성을 알아 보고자 하였다. 연구 재료 및 방법: 온도 감지 장치 제작을 위하여 Internal cone 연결형태의 임플란트에 16개의K형 열전대를 부착하여 아크릴릭 레진에 포매하였다. 치과용 금 합금과 주조용 abutment를 사용하여 교합면에 3개의 요철을 가지는 시편을 10개 제작하였고, 연결 나사를 이용하여 임플란트와 연결한 뒤 온도 감지 장치를 37℃로 유지되는 수조에 위치시켰다. 저속 핸드피스와 green stone bur를 이용하여 30초 동안 보철물의 요철을 삭제하였는데, 무냉각군, 공기 냉각 군, 물 분사 냉각 군으로 나누어 요철을 삭제하였다. 보철물이 삭제 되는 동안 임플란트의 부위별로 온도가 0.05초 간격으로 기록되었고, 삭제를 멈춘 뒤에도 무 냉각 군의 경우 임계 온도인 47℃ 이하로 온도가 하강할 때까지, 공기 냉각군과 물 분사 냉각군의 경우 삭제 중단 후 30초 동안 추가로 온도를 기록하였다. 냉각 방식에 따른 임플란트-골 계면의 온도를 알아보고, 임플란트의 부위별 온도변화의 유의차를 알아보기 위하여 one-way ANOVA를 실시하였고, Turkey HSD 이용하여 95% 유의수준에서 사후 검증하였다. 결과: 무 냉각 군은 임플란트-골 계면의 온도가 47℃ 이상으로 상승하였으며, 임플란트의 경부에서 유의하게 높은 열이 측정되었다(P>.05). 공기냉각군과 물분사 냉각군은 임플란트-골 계면의 온도가 47℃ 이하로 유지되었다. 무 냉각 군에서 임플란트 경부의 온도가 47℃에 도달되는 데는 약 10.8 ± 1.5초가 소요되었다. 공기 냉각 군과 물 분사 냉각 군 사이에서는 임플란트-골 계면 온도의 유의차가 없었다(P>.05) 결론: 이상의 결과로부터 임플란트 금 합금 보철물의 교합면 삭제 시, 임플란트 주위 조직에 위해를 가할 수 있는 임계 온도 이상의 열이 발생했음을 알 수 있었으며, 냉각방식은 공기 냉각과 물 분사 냉각 모두 효과적이라고 생각된다. PURPOSE. The purpose of this study was to analyze temperature change along the implant-bone interface induced by grinding the occlusal surface of implant gold prosthesis and to compare the temperature generated by grinding of prosthesis with different cooling methods. MATERIALS AND METHODS. The experimental gold prostheses were fabricated with dental gold alloy and castable abutment. The prostheses had 3 cylindrical protrusions on the occlusal surface with 1mm in height. Temperature was measured using 16 thermocouple wires attached to the implant fixture surface and the fixture was embedded in an acrylic resin block inside the 37℃ water bath. Cylinders were grinded for a period of 30 second with a low-speed handpiece with green stone point. One cylindrical protrusion was grinded without cooling, the second one was grinded with air blow, and the third one was grinded with water-spray. RESULTS. The mean maximum temperature was measured more than 47℃ of the implant and the maximum temperature was measured at the cervical portion of the implant in the group without cooling. There was statistically significant difference between the group without cooling and the groups with cooling (P<.05). However, there was no significant difference at all portion of implant in the groups with cooling (P>.05). CONCLUSION. The results of this study support that the grinding of implant gold prosthesis without cooling may damage the peri-implant tissue. The continuous use of air blow and water-spray adjacent to prosthesis during the grinding of implant gold prosthesis may prove to be beneficial for cooling of the implant.

      • 흰쥐 뇌의 냉동손상 후 항원제공세포와 신경교원섬유산단백의 발현 양상

        Background : Traumatic brain injury is a multifaceted injury that involves direct mechanical damage, intraparenchymal and subarachnoid hemorrhage, breakdown of the blood-brain barrier, excitotoxicity, and ischemia. Even though much investigations were performed, acceptable mechanical informations were rare. The aim of this study was to reveal the migration of antigen presenting cells associated with inflammation and the expression pattern of GFAP associated with gliotic scars in cerebral cortex of rats after cryoinjury. Material & Method : A total of 18 male Sprague-Dawley rats weighing 300 g, 2 months old, were used throughout the experiments. To injure the brain, rats were anesthetized for surgery with 3.5% chloral hydrate(1 ㎖/100 g, intraperitoneally); the frontal bones were exposed by elevating the skin; and craniectomies were performed adjacent to the central suture, midway between lambda and bregma. A cryoinjury was then created by applying a cold probe (3-mm-diameter steel rod chilled in liquid nitrogen) to the left frontal cortex (ipsilateral cortex) for 1 minute. Rats were sacrificed at 6 hours, 1, 4, 7 and 14 days postsurgery (n=3, per time point), and three rats were sacrificed as normal controls. Serial brain cryosections were made by cryostat. For immunohistochemistry, brain tissue sections were allowed to react with mouse anti-rat MHC class II antibody(1:500), mouse anti-rat ED2 antibody(1:500), and mouse anti-rat GFAP antibody(1:200). Results : MHC class II positive dendritic cells were absent in normal cerebral cortex parenchyme, but were more 16 times number in injured rats at 7 days after cryoinjury. ED2 positive macrophages were rare in normal cerebral cortex parenchyme, and were more 14 times number in injured rats at 4 days after cryoinjury. GFAP positive astrocytes were not observed in normal rats, and increased suddenly at 4 days rats after cryoinjury. GFAP positive neurons were not expressed in normal rats, and in cryoinjured rats. Conclusion : In conclusion, the above results suggest that the increases of the number of the two kinds of the antigen-presenting cells affect cell-mediated immune responses and phagocytosis, and GFAP positive cells attends in the formation of gliotic scars.

      • 舞踊實技考査의 客觀度

        The present study was to investigate the strategy for improving the objectivity of the college entrance dance performance test by determining the inter-rater reliability of dance performance judging under the raters different major. Five judges from three different dance types(three for Korean dance, one for Modern dance, and one for Ballet) were asked to rate each examinees performance using a Dance performance Checklist which consists of five components developed by the measurement specialists of Korean National University of Physical Education.. The Cronbach's alpha was calculated to verify the objectivity of the dance performance test by using RELIABILITY, subprogram of SPSS. The results showed that the objectivity coefficients from 5 raters were lower than .90, however, the coefficient for the 97 college entrance examination were improved than that of the 96. It was found that raters in general were judged consistently examinees dance performance, regardless of their major. The results suggest that the number of raters should be at least increased to 6 for ballet, 7 for korean dance, and 8 for modern dance judging to improve the objectivity coefficients up to .90.

        The purpose of this study was to explore the effects of Taichichuan training on the variables of cognitive-affective states. The subjects of the study consisted of 22G middle aged women in Jeonju areas. The survey methods of this research are the 5 Likert Scale modified the Cognitive Affective States Scale by Yang. Myung-Hwan(1998). The used statistical methods for the data analysis were M. SD. Paired sample T-Test. The results obtained by the appropriate analytic processes were followed. 1. In hedonic enjoyment, Taichichuan training women were lower than non-activity women. 2. In feeling of flow, Taichichuan training women were higher than non-activity women. 3. In self-realization, Taichichuan training women were higher than non-activity women. 4. In self-confidence, Taichichuan training women were higher than non-activity women.
