        Population genetic structure and phylogenetic analysis of gray’s pipefish, Halicampus grayi in the South China Sea

        Background The evolution of male pregnancy is the most distinctive characteristic of syngnathids, and they were recognized as flagship species for marine conservation. Genus Halicampus is an important branch of syngnathid fishes that has not received the attention it deserves. Objective To sequence the mitochondrial genome of Halicampus grayi, and investage the genetic structure of its populations. Methods Degenerate primers were designed to amplify the entire mitochondrial genome of H. grayi. The phylogenetic relationship between H. grayi and other syngnathids were conducted using maximum-likelihood method. Population genetic structure of three geographic population of H. grayi were determined using median-joining haplotype network based on COI and Cytb sequences. Results The complete mitochondrial genome of Halicampus grayi was assembled into a 17,059 bp circular sequence, which contains 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes and 1 D-loop region. The overall base composition of H. grayi is 29.93% A, 29.31% T, 16.23% G and 24.54% C, with a slight A + T rich feature (59.24%). Phylogenetic analysis indicated that H. grayi has a close relationship with Trachyrhamphus serratus. Population genetic analysis revealed a relatively high genetic diversity across different geographic populations of H. grayi, and the results of median-joining haplotype network indicated a lack of structure in populations of H. grayi. Conclusion The mitogenome of H. grayi will provided important information about the origin and evolution issues of syngnathid fishes, and the high-level genetic diversity detected in their populations will provide insight into the gene flow pattern of marine fishes.

        Influence of Inorganic Ions and pH on the Photodegradation of 1-Methylimidazole-2-thiol with TiO<sub>2</sub> Photocatalyst Based on Magnetic Multi-walled Carbon Nanotubes

        1-Methylimidazole-2-thiol, as a kind of mercaptans, is a typical organic pollutant which has not been efficiently removed. In this study, titanium dioxide ($TiO_2$) photocatalyst based on magnetic multi-walled carbon nanotubes (MWCNTs) was synthesized via hydrothermal and sol-gel methods. The as-prepared photocatalyst was extensively characterized by X-ray diffraction (XRD), X-ray energy diffraction spectrum (EDS), transmission electron microscope (TEM), Fourier transform infrared (FT-IR) spectra, UV-Vis diffuse reflectance spectra (UV-vis DRS) and vibrating sample magnetometer (VSM). This photocatalyst of $TiO_2$/$Fe_3O_4$/MWCNTs was proved to exhibit high photocatalytic efficiency and the photodegradation rate could reach nearly 82.7% for the degradation of 1-methylimidazole-2-thiol under ultraviolet irradiation. In addition, the results demonstrated that inorganic ions had a negative impact on photodegradation of 1-methylimidazole-2-thiol to varying degrees. Moreover, pH had a great and complex effect on photocatalytic degradation of 1-methylimidazole-2-thiol under ultraviolet irradiation.

        OsSPL13 controls grain size in cultivated rice

        <P>Although genetic diversity has a cardinal role in domestication, abundant natural allelic variations across the rice genome that cause agronomically important differences between diverse varieties have not been fully explored. Here we implement an approach integrating genome-wide association testing with functional analysis on grain size in a diverse rice population. We report that a major quantitative trait locus, GLW7, encoding the plant-specific transcription factor OsSPL13, positively regulates cell size in the grain hull, resulting in enhanced rice grain length and yield. We determine that a tandem-repeat sequence in the 5'UTR of OsSPL13 alters its expression by affecting transcription and translation and that high expression of OsSPL13 is associated with large grains in tropical japonica rice. Further analysis indicates that the large-grain allele of GLW7 in tropical japonica rice was introgressed from indica varieties under artificial selection. Our study demonstrates that new genes can be effectively identified on the basis of genome-wide association data.</P>

        Excessive Daytime Sleepiness and Insomnia Symptoms in Adolescents With Major Depressive Disorder: Prevalence, Clinical Correlates, and the Relationship With Psychiatric Medications Use

        Objective Excessive daytime sleepiness (EDS) and insomnia symptoms are common in patients with major depressive disorder (MDD), which might lead to a poor prognosis and an increased risk of depression relapse. The current study aimed to investigate the prevalence, and sociodemographic and clinical correlates of EDS and insomnia symptoms among adolescents with MDD.Methods The sample of this cross-sectional study included 297 adolescents (mean age=15.26 years; range=12–18 years; 218 females) with MDD recruited from three general and four psychiatric hospitals in five cities (Hefei, Bengbu, Fuyang, Suzhou, and Ma’anshan) in Anhui Province, China between January and August, 2021. EDS and insomnia symptoms, and clinical severity of depressive symptoms were assessed using Epworth sleepiness scale, Insomnia Severity Index, and Clinical Global Impression-Severity.Results The prevalence of EDS and insomnia symptoms in adolescents with MDD was 39.7% and 38.0%, respectively. Binary logistic regression analyses showed that EDS symptoms were significantly associated with higher body mass index (odds ratio [OR]=1.097, 95% confidence interval [CI]=1.027–1.172), more severe depressive symptoms (OR=1.313, 95% CI=1.028–1.679), and selective serotonin reuptake inhibitors use (OR=2.078, 95% CI=1.199–3.601). And insomnia symptoms were positively associated with female sex (OR=1.955, 95% CI=1.052–3.633), suicide attempts (OR=1.765, 95% CI=1.037–3.005), more severe depressive symptoms (OR=2.031, 95% CI=1.523–2.709), and negatively associated with antipsychotics use (OR=0.433, 95% CI=0.196–0.952).Conclusion EDS and insomnia symptoms are common among adolescents with MDD. Considering their negative effects on the clinical prognosis, regular screening and clinical managements should be developed for this patient population.

        Bone Morphogenetic Protein 9 Overexpression Reduces Osteosarcoma Cell Migration and Invasion

        Transforming growth factor- (TGF-) is known to pro-mote tumor migration and invasion. Bone morphogenetic proteins (BMPs) are members of the TGF- family expressed in a variety of human carcinoma cell lines. The role of bone morphogenetic protein 9 (BMP9), the most powerful osteogenic factor, in osteosarcoma (OS) progression has not been fully clarified. The expression of BMP9 and its receptors in OS cell lines was analyzed by RT-PCR. We found that BMP9 and its receptors were expressed in OS cell lines. We further investigated the influence of BMP9 on the biological behaviors of OS cells. BMP9 overexpression in the OS cell lines 143B and MG63 inhibited in vitro cell migration and invasion. We further investigated the ex-pression of a panel of cancer-related genes and found that BMP9 overexpression increased the phosphorylation of Smad1/5/8 proteins, increased the expression of ID1, and reduced the expression and activity of matrix metalloproteinase 9 (MMP9) in OS cells. BMP9 silencing induced the opposite effects. We also found that BMP9 may not affect the chemokine (C-X-C motif) ligand 12 (CXCL12)/C-X-C chemokine receptor type 4 (CXCR4) axis to regulate the invasiveness and metastatic capacity of OS cells. Interestingly, CXCR4 was expressed in both 143B and MG63 cells, while CXCL12 was only detected in MG63 cells. Taken together, we hypothesize that BMP9 inhibits the migration and invasiveness of OS cells through a Smad-dependent pathway by downregulating the expression and activity of MMP9.
