        Effectiveness and Safety of Dabrafenib in the Treatment of 20 Chinese Children with BRAFV600E-Mutated Langerhans Cell Histiocytosis

        Purpose We sought to investigate the effectiveness and safety of dabrafenib in children with BRAFV600E-mutated Langerhans cell histiocytosis (LCH). Materials and Methods A retrospective analysis was performed on 20 children with BRAFV600E-mutated LCH who were treated with dabrafenib. Results The median age at which the patients started taking dabrafenib was 2.3 years old (range, 0.6 to 6.5 years). The ratio of boys to girls was 2.3:1. The median follow-up time was 30.8 months (range, 18.9 to 43.6 months). There were 14 patients (70%) in the risk organ (RO)+ group and six patients (30%) in the RO– group. All patients were initially treated with traditional chemotherapy and then shifted to targeted therapy due to poor control of LCH or intolerance to chemotherapy. The overall objective response rate and the overall disease control rate were 65% and 75%, respectively. During treatment, circulating levels of cell-free BRAFV600E (cfBRAFV600E) became negative in 60% of the patients within a median period of 3.0 months (range, 1.0 to 9.0 months). Grade 2 or 3 adverse effects occurred in five patients. Conclusion Some children with BRAFV600E-mutated LCH may benefit from monotherapy with dabrafenib, especially high-risk patients with concomitant hemophagocytic lymphohistiocytosis and intolerance to chemotherapy. The safety of dabrafenib is notable. A prospective study with a larger sample size is required to determine the optimal dosage and treatment duration.

        Two Transition Metal Complexes: Magnetic Properties and Treatment Effect via Focused High-Intensity Ultrasound on Alzheimer’s Disease

        Two novel coordination polymers (where Cu for the complex 1 and the Co for the complex 2) with distinct transition metal ions containing 4-(3,5-dicarboxylphenyl)-2- methylpyridine (H2L), the pyridyl-dicarboxylic acid ligand, have been produced under the solvothermal reaction conditions, and the two compounds’ performances and architectures were also characterized. The chemical formulae determined of two complexes are respectively [Cu(L)(H2O)]·H2O (1) and [Co3(L)2Cl2(H2O)2]·2H2O (2). Complex 1 exhibits a 2D network with a (3,6)-connected kgd topology based on the dinuclear [Cu2O2] units. In complex 2, the linear trinuclear [Co3(μ-Ocarboxylate)2(COO)2Cl2] units with mixed carboxylate and μ-Cl triple bridges are cross-linked by the L ligands into a 3D framework with a (3,6)-connected rtl topology. The complexes’ magnetic performances reflected that between consecutive metal ions, there exists the antiferromagnetic coupling. Their treatment activities on Alzheimer's disease (AD) via focused high-intensity ultrasound were evaluated. Firstly, the Annexin V-fluorescein isothiocyanate/propidium iodide staining was implemented for the assessment of the apoptotic neurons percentage after compounds treatment. Next, the Aβ accumulation in the neurons was also detected via employing the western blotting assay.

        Anthracene Biodegradation under Nitrate-reducing Condition and Associated Microbial Community Changes

        Anaerobic biodegradation of polycyclic aromatic hydrocarbons (PAHs) and degraders in the subsurface environment have aroused increasing attention. Molecular techniques are especially useful when isolates are hard to obtain. Nitrate-reducing microcosms inoculated with aquifer sediment were constructed to investigate anthracene biodegradation. The associated microbial community changes were characterized using terminal restriction fragment length polymorphism analysis (TRFLP) in combination with 16S rRNA gene clone library analysis. A nearly complete removal of anthracene was achieved after an eighty day incubation under the nitrate-reducing condition. The two molecular techniques revealed a significant shift of microbial community structure, coupled with anthracene biodegradation. Species of genera Paracoccus, Herbaspirillum,Azotobacter, and Rhodococcus were grouped into four major operational taxonomic units (OTUs) in the library that was constructed with the microcosm sample on day 80. The enrichment of these genera might have links to anthracene biodegradation under the nitrate-reducing condition. Microbial consortia likely played a part in anthracene degradation.         Genetic variation and phylogenetic relationships of the ectomycorrhizal Floccularia luteovirens on the Qinghai-Tibet Plateau

        Floccularia luteovirens, as an ectomycorrhizal fungus, is widely distributed in the Qinghai-Tibet Plateau. As an edible fungus, it is famous for its unique flavor. Former studies mainly focus on the chemical composition and genetic structure of this species. However, the phylogenetic relationship between genotypes remains unknown. In this study, the genetic variation and phylogenetic relationship between the genotypes of F. luteovirens in Qinghai-Tibet Plateau was estimated through the analysis on two protein-coding genes (rpb1 and ef-1α) from 398 individuals collected from 24 wild populations. The sample covered the entire range of this species during all the growth seasons from 2011 to 2015. 13 genotypes were detected and moderate genetic diversity was revealed. Based on the results of network analysis, the maximum likelihood (ML), maximum parsimony (MP), and Bayesian inference (BI) analyses, the genotypes H-1, H-4, H-6, H-8, H-10, and H-11 were grouped into one clade. Additionally, a relatively higher genotype diversity (average h value is 0.722) and unique genotypes in the northeast edge of Qinghai- Tibet plateau have been found, combined with the results of mismatch analysis and neutrality tests indicated that Southeast Qinghai-Tibet plateau was a refuge for F. luteovirens during the historical geological or climatic events (uplifting of the Qinghai-Tibet Plateau or Last Glacial Maximum). Furthermore, the present distribution of the species on the Qinghai-Tibet plateau has resulted from the recent population expansion. Our findings provide a foundation for the future study of the evolutionary history and the speciation of this species.

      • Meta-analysis of the Association Between GSTM1 and GSTT1 Gene Polymorphisms and Cervical Cancer

        Aim: We conducted a meta-analysis to analyze the influence of GSTM1 and GSTT1 gene polymorphisms on cervical cancer risk, and explore gene-environment interactions. Methods: Identification of relevant studies was carried out through a search of Medline and the EMbase up to Oct. 2011. All case-control studies that investigated the association between GSTM1 and GSTT1 gene polymorphisms and risk of cervical cancer were included. The pooled odds ratio (OR) was used for analyses of results and the corresponding 95% confidence intervals (CI) were estimated. Results: A total of 21 case-control studies were included in the meta-analysis of GSTM1 (2,378 cases and 2,639 controls) and GSTT1 (1,229 cases and 1,223 controls) genotypes. The overall results showed that the GSTM1 null was related to an increased risk of cervical cancer (OR=1.50, 95% CI=1.21-1.85). Subgroup analysis were performed based on smoking and ethnicity. Our results showed that smokers with null GSTM1 genotype had a moderate increased risk of cervical cancer (OR=1.85, 95% CI=1.07-3.20). For the ethnicity stratification, moderate significantly increased risk of null GSTM1 genotype was found in Chinese (OR=2.12, 95% CI=1.43-3.15) and Indian populations (OR=2.07, 95% CI=1.49-2.88), but no increased risk was noted in others. Conclusion: This meta-analysis provided strong evidence that the GSTM1 genotype is associated with the development of cervical cancer, especially in smokers, and Chinese and Indian populations. However, no association was found for GSTT1 null genotype carriers.

        The Distribution of Transcutaneous CO2 Emission and Correlation With the Points Along the Pericardium Meridian

        This study aimed to understand energy metabolism distribution along the pericardium meridian and verify the correlation between the body surface (points), and classic meridian theory. A highly sensitive CO2 instrument was used to measure the transcutaneous CO2 emission at 13 points along the pericardium meridian line (12 points on the line and one point beyond the line) and 13 control points beside them. Results showed that the distribution of transcutaneous CO2 emission is highly related to the position on the body. Transcutaneous CO2 emission is significantly higher at P7 and P3, than the control points beside them. The points along the meridian and the points beside them were clustered with relative distance by SAS statistics software. Two distance matrixes were then obtained. The correlation coefficients between the points along the line and between the control points were calculated. The results showed that the 13th point beyond the line was far from the 12 points on the line (distance, 0.24), while acupoints on the line clustered earlier when compared with the non-acupoints. The average correlation coefficients among the acupoints was 0.65 which was significantly higher than 0.56, among the non-acupoints. No such characteristics were found among the control points. It was concluded that there is a strong correlativity of energy metabolism activity between the body surfaces along the meridian, and an even stronger correlativity between the acupoints on the meridian.       • Seismic Performance and Vibration Control of Urban Over-track High-rise Buildings

        During the structural design of urban over-track high-rise buildings, two problems are most likely encountered: the abrupt change of story stiffness between the podium and the upper towers, as well as the demand for train-induced vibration control. Traditional earthquake-resistant structures have to be particularly designed with transfer stories to meet the requirement of seismic control under earthquakes, and thus horizontal seismic isolation techniques are recommended to solve the transfer problem. The function of mitigating the vertical subway-induced vibration can be integrated into the isolation system including thick rubber bearings and 3D composite vibration control devices. Engineering project cases are presented in this paper for a more comprehensive understanding of the engineering practice and research frontiers of urban over-track high-rise buildings in China.

        Role of active and passive smoking in high-risk human papillomavirus infection and cervical intraepithelial neoplasia grade 2 or worse

        Objective: We performed a pooled analysis to examine cigarette smoking and householdpassive smoke exposure in relation to the risk of human papillomavirus (HPV) infection andcervical intraepithelial neoplasia grade 2+ (CIN2+). Methods: Data were pooled from 12 cross-sectional studies for cervical cancer screeningsfrom 10 provinces of China in 1999–2007. A total of 16,422 women were analyzed, alongwith 2,392 high-risk-HPV (hr-HPV) positive women and 381 CIN2+ cases. Pooled odds ratios(ORs) and 95% confidence intervals (CIs) were estimated using logistic regression modelscontrolling for sexual and non-sexual confounding factors. Results: There was an excess risk between active smoking and hr-HPV infection and CIN2+. Adjusted OR for ever smokers vs. never smokers was 1.45 (95% CI=1.10–1.91), for hr-HPVinfection and 1.89 (95% CI=1.03–3.44), for CIN2+. Passive smoking had a slightly increasedrisk on the hr-HPV infection with adjusted OR 1.11 (1.00–1.24), but no statistical associationwas observed between passive smoke exposure and CIN2+. Compared with the neither activenor passive smokers, both active and passive smokers had a 1.57-fold (95% CI=1.14–2.15)increased risk of HPV infection and a 1.99-fold (95% CI=1.02–3.88) risk of CIN2+. Conclusion: Our large multi-center cross-sectional study found active smoking couldincrease the risk of overall hr-HPV infection and CIN2+ adjusted by passive smoking andother factors. Passive smoking mildly increased the risk of HPV infection but not the CIN2+. An interaction existed between passive tobacco exposure and active smoking for hr-HPVinfection and the CIN2+.

        Increasing Demeclocycline Production in Streptomyces aureofaciens by Manipulating the Expression of a Novel SARP Family Regulator and Its Genes

        Demeclocycline (DMCTC), a tetracycline derivative antibiotic produced by Streptomyces aureofaciens, has attracted attention owing to its high bioavailability, prolonged maintenance of a therapeutic concentration, and greater efficacy against many infectious microorganisms. However, the productivity of the DMCTC-producing strains has remained low. Thus, it is necessary to identify gene-knockout or amplification targets to increase DMCTC production. Here, we demonstrated that ctcB, which encodes a Streptomyces antibiotic regulatory protein (SARP), and ctcC, which encodes a resistance gene, positively regulate the biosynthesis of DMCTC in S. aureofaciens strain DT1. In particular, overexpression of the ctcB gene in S. aureofaciens DT1 significantly enhanced DMCTC production, resulting in increased expression of ctcG, ctcN, ctcQ, ctcH, ctcV, and ctcC. The deletion of ctcB dramatically reduced the DMCTC level, implying that CtcB is an activator of DMCTC biosynthesis. Although overexpression of the ctcC, which encodes a ribosomal protection protein, enhancing DMCTC biosynthesis in S. aureofaciens DT1, the improvement was limited compared with that achieved by ctcB overexpression. This is the first study to identify the role of ctcB and ctcC in DMCTC accumulation; these genes may also be ideal candidate targets for facilitating DMCTC production by other Streptomyces strains.
