        Viability and Functions of Alginate-microencapsulated Islets Isolated from Neonatal Pigs

        Patients with Type I diabetes mellitus have been treated with porcine insulin for several decades and pigs have recently been deemed an ideal source of microencapsulated islet cells for clinical xenotransplantation. In this study, neonatal pigs were anesthetized and sacrificed prior to a pancreatectomy. Islet cells were isolated from pancreas via collagenase digestion. Islet cells were separated and collected by hand under microscopic guidance. These cells were suspended in 1.4% sodium alginate solution and encapsulated by dropping them into 1.1% calcium chloride solution and in which the round gel in size was 250-400 ${\mu}m$ in diameter. Viability of the microencapsulated islet cells cultured in medium at $37^{\circ}C$ was assessed by MTT assay. Furthermore, insulin released in response to glucose challenge was investigated using an enzyme-linked immunosorbent assay. Secretion of insulin was low in response to the basal glucose solution (4.4 mM) in medium and was significantly higher in response to the high glucose solution (16.7 mM). The viability of microencapsulated islet cells did not differ significantly over a period of 7 days; that is, the increasing pattern of insulin concentration in the culture medium after glucose stimulation interval day was similar throughout the 7 days cultivation. In summary, experimental evidences indicated that the effects of alginate-microencapsulation prolonged survival of the neonatal porcine islets in vitro cultures and the insulin response to glucose of the islets was maintained.

      • Effects of Rad51 on Survival of A549 Cells

        Rad51, a key factor in the homologous recombination pathway for the DNA double-strand break repair, plays a vital role in genesis of non-small-cell lung cancer (NSCLC). In recent years, more and more studies indicate that high expression of Rad51 is of great relevance to resistance of NSCLC to chemotherapeutic agents and ionizing radiation. However, the underlying molecular mechanisms are poorly understood. In this study, we investigated the role of single Rad51 on cell viability in vitro. Our results show that depletion of endogenous Rad51 is sufficient to inhibit the growth of the A549 lung cancer cell line, by accumulating cells in G1 phase and inducing cell death. We conclude that independent Rad51 expression is critical to the survival of A549 cells and can be an independent prognostic factor in NSCLC patients.

        Reclaiming Mother—Son Relationships on Mothers’ Own Terms

        This paper examines two contemporary mother–son novels, Margaret Forster’s Mothers’ Boys and Rosellen Brown’s Before and After, which offer similar scripts for raising sons. In these novels, both writers unmercifully depict the alienation between mothers and sons and describe how these mothers deal with their sons’ separation from them. Different from the forging of identification between mothers and daughters and the relative obscurity of the father figure in contemporary mother–daughter narratives, the delineation of estranged relationships between mothers and sons and the inclusion of fathers in raising sons enables the two mother–son novels to inform a new narrative structure of matrilineal narratives. In particular, looking through the lens of the mothers with their strong desire to (re)connect with their sons as well as the maternalizing of the father figure in these novels, the novels suggest the two writers’ concerted efforts to refigure the mother–son estrangement and to strengthen the mother–son bond on the mothers’ own terms. Drawing from this observation, this study concludes with the positive note that reinstating the mother–son connection is the trend that preoccupies these contemporary women writers.

        Beyond the Generational Line

        This article deals with the feminist generation issue by tracing the transition from generational to non-generational thinking in recent feminist discourse on third-wave feminism. Informed by certain feminist recommendations of affording alternative paradigms in place of the oceanic wave metaphor to describe the relationships between different feminists, this study offers the insights gained from an investigation with three feminist online sites, The F Word, Eminism, and Guerrilla Girls, in which these online feminists have participated in building up a third wave consciousness or a third space site through their engagement in (re)presenting their feminist selves and identities. In developing a both/and third wave consciousness, these online feminists have bypassed the dualistic understanding of second and third wave feminism and reached a cross-generational commonality of adhering to feminist ideology and coalition in cyberspace as a third space. Through a content analysis of their online feminist self-(re)presentations, this paper concludes by arguing that they have not only reformulated the concept of third wave feminism but also worked toward a new configuration of third space narratives and subjectivities that sheds light on contemporary feminist thinking about feminist genealogy and history.

        Using ‘classic reading instruction’ to raise students’ gender awareness: Students’ perceptions of their learning experiences at a Taiwanese university

        Although much research has focused on exploring different pedagogies for cultivating students’ awareness and competencies, few studies have actually looked into how this can be done to develop their gender awareness by exploring the dynamics of classroom interaction. In this paper, I examine the effects of a method, referred to as ‘classic reading instruction,’ used in a gender and literature course, via a qualitative interview study. Forty-one male and female student participants from a Taiwanese university were interviewed and five themes were identified in the data. The findings revealed that most participants affirmed the effectiveness of classic reading instruction for raising their awareness about gender. They were also able to extend this to recognize the influences of family education on them and to comment on sexism in society. In addition, both male and female participants recognized the existence of male privilege, their identification with feminism and its relevance to daily life.

        Initial Factors Influencing Duration of Hospital Stay in Adult Patients With Peritonsillar Abscess

        Objectives. To review cases of peritonsillar abscess and investigate the initial clinical factors that may influence the duration of hospitalization. To determine the predictive factors of prolonged hospital stay in adult patients with peritonsillar abscess. Methods. Subjects were adults hospitalized with peritonsillar abscess. We retrospectively reviewed 377 medical records from 1990 to 2013 in a tertiary medical center in southern Taiwan. The association between clinical characteristics and the length of hospital stay was analyzed with independent t-test, univariate linear regression and multiple linear regression analysis. Results. The mean duration of hospitalization was 6.2±6.0 days. With univariate linear regression, a prolonged hospital stay was associated with several variables, including female gender, older ages, nonsmoking status, diabetes mellitus, hypertension, band forms in white blood cell (WBC) counts, and lower hemoglobin levels. With multiple linear regression analysis, four independent predictors of hospital stay were noted: years of age (P<0.001), history of diabetes mellitus (P<0.001), ratio of band form WBC (P<0.001), and hemoglobin levels (P<0.001). Conclusion. In adult patients with peritonsillar abscess, older ages, history of diabetes mellitus, band forms in WBC counts and lower hemoglobin levels were independent predictors of longer hospitalization.

        Synthesis of mesoporous SiO2 xerogel/chitosan mixed-matrix membranes for butanol dehydration

        Novel mesoporous SiO2 xerogel/chitosan (CS) mixed-matrix membranes (MMMs) were successfully prepared by incorporating sol–gel synthesized SiO2 xerogels into glutaraldehyde (GA) cross-linked CS membranes. The effects of the SiO2 doping amount and the feed temperature on the performance of the separation of butanol and water was also investigated in this study. The 0.25 wt% SiO2 xerogel/CS MMMs possessed the best pervaporation performance and the largest PSI value of 1102 kg/m2h with a permeate flux of 736 g/m2h (Permeability of 3.25 × 10−5 g m m−2 h −1 Pa−1) and a separation factor of 1498 at a feed temperature of 50 °C. This PSI value is not only comparable but also exceeds that of other membranes reported in the literature. The separation factor and the flux of the SiO2 xerogel/CS MMMs in this study clearly exceed the upper limit of the previously reported MMMs. As a result, the as-prepared novel SiO2 xerogel/CS MMMs showed an improved performance during the butanol and water separation process, which raises the possibility of future pervaporation applications using this novel SiO2 xerogel/CS MMMs.

        Professionals’ experiences and attitudes toward use of Traditional Chinese Medicine in hospice palliative inpatient care units: A multicenter survey in Taiwan

        Background: Medical staff may have difficulties in using conventional medicine to manage symptoms among terminally ill patients, including adverse effects of the treatment. Traditional Chinese medicine (TCM) is regarded as a complementary or alternative medicine, and has been increasingly used in the field of palliative medicine in recent years. This study aimed to investigate the experiences of and attitudes toward using TCM among palliative care professionals, and to provide preliminary information about its use in palliative care. Methods: This was a cross-sectional survey study conducted in eight inpatient hospice wards in Taiwan between December 2014 and February 2016. The questionnaire was self-administered, and was analyzed with descriptive statistics including Pearson’s Chi-square test and Fisher’s exact test. Results: A total of 251 palliative care professionals responded to the questionnaire, of whom 89.7% and 88.9% believed that the use of TCM could improve the physical symptoms and quality of life in terminally ill patients, respectively. Overall, 59.8%, of respondents suggested that TCM had rare side effects, and 58.2% were worried that TCM could affect the liver and kidney function of patients. In total, 89.7% and 88.0% of professionals agreed there were no suitable clinical practice guidelines and educational programs, respectively, for TCM use in palliative care. Conclusions: Most of the respondents agreed there was insufficient knowledge, skills-training, and continuing education on the use of TCM in terminally ill patients in Taiwan. These results show that to address patient safety considerations, guidelines about use of TCM in palliative care should be established.

        The EGF/hnRNP Q1 axis is involved in tumorigenesis via the regulation of cell cycle-related genes

        Heterogeneous nuclear ribonucleoprotein (hnRNP) Q1, an RNA-binding protein, has been implicated in many posttranscriptional processes, including RNA metabolism and mRNA splicing and translation. However, the role of hnRNP Q1 in tumorigenesis remains unclear. We previously performed RNA immunoprecipitation (RIP)-seq analysis to identify hnRNP Q1-interacting mRNAs and found that hnRNP Q1 targets a group of genes that are involved in mitotic regulation, including Aurora-A. Here, we demonstrate that altering the hnRNP Q1 level influences the expression of the Aurora-A protein, but not its mRNA. Stimulation with epidermal growth factor (EGF) enhances both binding between hnRNP Q1 and Aurora-A mRNA as well as the efficacy of the hnRNP Q1-induced translation of Aurora-A mRNA. The EGF/hnRNP Q1-induced translation of Aurora-A mRNA is mediated by the mTOR and ERK pathways. In addition, we show that hnRNP Q1 up-regulates the translation of a group of spindle assembly checkpoint (SAC) genes. hnRNP Q1 overexpression is positively correlated with the levels of Aurora-A and the SAC genes in human colorectal cancer tissues. In summary, our data suggest that hnRNP Q1 plays an important role in regulating the expression of a group of cell cycle-related genes. Therefore, it may contribute to tumorigenesis by up-regulating the translation of these genes in colorectal cancer.
