        Moutan radix 의 mouse sarcoma 180 cell 로 유발한 mouse ascites cancer 에 대한 항암효과

        Anticarcinogenic activity of Moutan radix for mouse ascites cancer induced by mouse Sarcoma 180 (S-180) cells was investigated. Methanol extract of Moutan radix including other folk medicinal plants (Taxus cuspidata, Curcuma Tonga, Artemisia capillaris, Ligrstri fructus, and Liriope platyphylla) used to remedy or cure many chronic human diseases like cancer was fractionated into hexane, chloroform (CHCl₃), ethylacetate (EtOAc), and butanol (BuOH) fractions. Anticarcinogenic activity of the fractions, exhibited a strong cytotoxicity for L1210 and S-180 cells, was examined for mouse ascites cancer induced by S-180 cells. Male ICR mice (7 mice/treatment, 5∼6 weeks of age, 23±1 g) were injected i.p. with S-180 cells (1×10^7 cell/1 ㎖ PBS). One day later, each mouse was given 0.1 ㎖ of 10% DMSO containing sample (30 ㎍/g body weight) every day for 10 consecutive days. Control mice were only given 0.1 ㎖ S-180 cells and 0.1 ㎖ 10% DMSO. Mice treated with EtOAc fraction of Moutan radix showed 28.7 days of life, which is 167% of control mice`s life. Based on the dose-dependant experiment, mice treated with 30 ㎍ showed longer life relative to mice treated with other doses (5, 15, 60 ㎍) and mice treated with 60 ㎍ exhibited toxic symptoms. Body weight of mice treated with Moutan radix was significantly reduced relative to that of control mice (p$lt;0.05). GC-MS analysis in conjunction with silica-gel column chromatography revealed that the EtOAc fraction contained 2-methoxylphenol, benzoic acid, 1-(4-hydroxy-3-methoxyphenyl)ethanone, 8-methyl-2,4(1H,3H)pteridinedione and 2,5-furan-dicarboxylic dimethyl ester as regards to the anticarcinogenic property of the EtOAc fraction. These results suggest that Moutan radix might be included as an anticarcinogenic medicinal plant for treatment of ascites cancer.

        새로이 分類된 天然 抗癌劑 : Conjugated Dienoic Derivatives of Linoleic Acid (CLA)

        동물실험을 통하여 새로이 밝혀진 항암제(anti-initiator/anti-ptomotor)인 CLA는 grilled ground beef에서 처음 분리되었다. CLA는 grilled ground beef외에도, cheese 및 이들 관련식품에 많이 존재한다. CLA는 반추동물의 위에 서식하는 혐기성 bacteria에 의해 linoleic acid로부터 생성되며, 식품 가공 중에서도 생성된다. 이것은 또한 in vivo에서 linoleic acid의 carbon centered free radical 형태의 산화에 의해 생성되기도 한다. CLA는 아주 강력한 항산화제임이 밝혀져, 지금까지 알려져 있지 않았는 free radical에 대응하여 membrane을 보호하는 in situ defense mechanism 역할을 한다. 이는 또한 cytochrome P450 isozyme의 활성을 저해하는 반면, ODC 효소 활성 역시 저해한다. 그래서, 적어도 CLA의 이 세가지 biological activity가 CLA 항암기작에 관여하는 것으로 생각된다. Anticarcinogenic conjugated dienoic derivatives of linoleic acid(CLA) is present in grilled beef, cheese, and related foods. CLA is generated via isomerization of linoleic acid in the cow's rumen by anaerobic bacteria and food proceessing as well. Another source of CLA is its endogenous generation via the carbon centered free radical oxdation of linoleic acid. We propose that the formation and generation of CLA in vivo represents a previously unrecognized in situ “defense mechanism” against membrane attack by oxygen free radicals. The cis, 9-trans, 11 CLA isomer is selectively incorporated into cellular phospholipid, which exhibits a potent antioxidant, reduces the activation of 2-amino-3-methylimidazo 〔4, 5-f〕 quinoline (IQ) for bacterial mutagenesis, and inhibits ornithine decarboxylase(ODC) activity induced by 12-0-tetradecanoylphorbol-13-acetate (TPA). We believe that at least these three biological activities of CLA explain the anticarcinogenic activity of CLA.

        Mycelial Culture of Lentinus edodes Alleviates Rat Liver Toxicity Induced by Carbon Tetrachloride and Ethanol

        LED의 간 보호 기능을 연구하기 위하여 CCl₄ 및 ethanol로 SD rat에 간독성을 유발한 다음, LED를 처리하였다. LED의 간 기능 보호효과는 간장치료제인 Silymarin과 비교하였다. CCl₄로 간 독성을 유발한 경우, LED는간의 항산화효소인 SOD, catalase, GSH peroxidase 효소활성의 항진을 유도하였고, 산화물인 TBARS의 함량을 감소시켰다. 또한 간 손상의 지표인 혈장의 GOT, GPT 및 LDH의 활성을 감소시켰다. Ethanol로 간 독성을 유발한 경우 LED는 간의 SOD, catalase, GSH preoxidase 효소활성 및 GSH 함량을 항진시켰고, 총 cholesterol, triglyceride 및 TBARS의 함량을 감소시켰다. 또한 ethanol 대사에 관여하는 ADH 효소 활성을 증진시켰고, ROS 생성에 관여하는 CYP2E1 효소의 발현을 감소시킴으로써, 혈장의 GOT, GPT 및 LDH 효소활성이 감소되었다. 또한 LED는 DPPH 및 mouse liver mitochondrial system에서 항산화효과를 보였다. 이러한 결과로 미루어 볼 때 LED는 in vitro와 in vivo에서 항산화효과에 의한 간 기능 보호효과를 갖는 것으로 추정된다. The protective effect of a mixed powder from solid-cultured and liquid-cultured Lentinus edodes mycelia (2:1, w/w) (designate LED) on the carbon tetrachloride (CCl₄)- and ethanol-induced hepatotoxicity of male Sprague-Dawley (SD) rat was investigated. In the CCl₄-induced rat hepatotoxicity experiment, rats of 4 groups (6 rats/group) were administere with Normal (0.2 ml distilled water), Control (0.2 ml distilled water), LED (LED 200 ㎎/㎏ BW + 0.2 ml distilled water), and Silymarin (200 ㎎/㎏ BW + 0.2 ml distilled water), p.o., daily for 2 weeks. Afterwards, all groups except for the Normal group were subjected to abdominal injection with CCl₄ (CCl₄: corn oil, 1:1 v/v; 0.5 ml/㎏ BW). For the ethanol- induced rat hepatotoxicity experiment, rats were divided into 5 groups (5 rats/group): Normal; Pair-fed control (PFC); Control (ethanol); LED (ethanol + LED 200 ㎎/㎏ BW); and Silymarin (ethanol + silymarin 200 ㎎/㎏ BW). Rats of the Normal and PFC groups were fed a basal liquid diet, and rats of the Control, LED, and Silymarin groups were fed a liquid ethanol diet containing LED or Silymarin. Eight weeks later, blood and liver samples were collected to analyze biomarkers. In CCl₄-induced SD rats, LED elevated hepatic superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH peroxidase) activities and thiobarbituric reactive substances (TBARS) were reduced, resulting in the reduction of glutamate-oxalate transaminase (GOT), glutamate-pyruvate transaminase (GPT) and lactic dehydrogenase (LDH) activities in plasma. Similar results of these enzymes and biochemical markers in both liver tissues and plasma were seen in ethanol-induced hepatotoxicity of SD rats. In addition, elevated alcohol dehydrogenase (ADH) activity and reduced expression of cytochrome p450 mixed monooxygenase enzyme (CYP2E1) were seen in liver tissues from ethanol-treated rats by LED treatment. These effects of LED were similar to those of Silymarin. In in vitro experiments, LED showed antioxidant activity in a 2,2-diphenyl-1-picrylhydrazyl (DPPH) system and mouse liver mitochondria system induced by NADPH/Fe<SUP>2+</SUP> and cumine hydroperoxide (CuOOH). These results indicate that LED protected SD rat hepatotoxicity, induced by CCl₄ and ethanol, through its antioxidative activity and might be useful as a material for protection from hepatoxicity in humans.

        CLA의 생물학적 기능

        CLA는 탄소가 18개(C18)인 linoleic acid의 이중결합이 C9,C11; C10,C12 위치로 이동됨으로 생성되는 입체이성(c,c; c,t; t,c; t,t)체를 총괄적으로 일컷는 말이다. 이론적으로 가능한 이성체 중에서 c9,t11-CLA가 rumen bacteria, lactic bacteria, 버섯균 등의 linoleate isomerase에 의해 linoleic acid로부터 생합성된다. 그러나 linoleic acid를 알카리 이성화로 합성한 CLA에는 이론적으로 가능한 모든 CLA 이성체가 존재한다. 그 중 c9,t11-CLA와 t10,c12-CLA가 약 45%씩 동일량 존재한다. CLA가 1939 년 linoleic acid의 elaidinization 반응에서 처음으로 소개되었으나 그 이후에는 과학적인 이용가치가 없어 과학자들의 관심의 대상이 되지 않았다. 그러나 1987년에 CLA가 7,12-dimethylbenz[a]anthracene (DMBA)으로 유도한 mouse skin carcinogenesis에서 항암성이 있다는 보고 이후 CLA에 관한 연구는 급속도로 증가하여 현재까지 약 6,100 연구논문이 발표되었다. CLA와 c9,t11-CLA 및 t10,c12-CLA 이성체는 in vitro와 in vivo에서 항암성, 항당뇨성, 항혈압성, 항동맥경화성, 면역증강성, 항산화성, 체지방감소성, testosterone 생산성 등의 생물학적 기능이 갖는다고 밝혀졌다. 이들 생리활성에 c9,t11-CLA와 t10,c12-CLA가 in vitro와 in vivo에서 다른 활성을 보이고 있다. 특히, 합성 CLA에 소량 혼합되어 있는 t,t-CLA가 carcinogen으로 유도한 동물모델이나 암세포에서 다른 이성체 보다 강한 항암성을 보이는 결과는 앞으로 더 많은 연구의 대상이 될 것이다. 본 총설에서는 CLA 관련 연구가 시작된 1939년부터 현재까지의 CLA 연구 트렌드를 살펴보고 밝혀진 주요 기능성을 보고한다. Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid with conjugated double bonds at C9,C11 and C10,C12 positions. Of possible CLA isomers, a naturally occurring CLA isomer is c9,t11-CLA which is produced from linoleic acid by linoleate isomerase from various rumen and lactic bacteria, and mushroom mycelia. Meanwhile, synthetically prepared CLA contained an equal amount of c9,t11-CLA and t10,c12-CLA isomers, and other isomers as minor constituents. CLA was firstly mentioned in 1939 during the elaidinization reaction of linoleic acid. Thereafter, CLA was not an attractant to scientists because it was not scientifically interested any more. However, since the anticarcinogenic action was driven from 7,12-dimethylbenz[a]anthracene (DMBA)-induced mouse skin carcinogenesis in 1987, CLA-related researches were drastically elevated, resulting in approximately 6,100 research papers in literature, so far. CLA exhibited the significant biological activities: anticarcinogenic, antidiabetic, antihypertensive, antiatherosclerotic, body-fat reducing, antioxidative, antiinflammatory, testosterone producing and other activities. Interestingly, two major CLA isomers, c9,t11-CLA and t10,c12-CLA, exhibited different biological activities. Meanwhile, t,t-CLA isomers which is minor constituent of chemically synthesized CLA from linoleic acid exhibited more potent anticarcinogenic activity in carcinogen-induced animal models and cancer cell lines than other CLA isomrs. In the present review, the significant biological activities of CLA were discussed along with historical studies of CLA since 1939.

        Conjugated linoleic acid의 비만 남자중학생 체지방 감소와 운동력 증진효과

        남자 중학생에 대한 conjugated linoleic acid (CLA)의 체지방감소, 동맥경화 지표 개선 및 신체활력개선 효과를 연구하였다. 체지방율이 30% 이상 인 건강한 남자 중학생 희망자 24명을 CLA섭취군과 대조군(placebo)으로 무작위로 나누고 CLA (하루에 2회, 총 6 g)와 placebo를 이중맹검법으로 총 12주간 처리하였다. 실험전과 마지막에 인체구성, 혈중 지질조성 및 운동력을 조사하였다. CLA는 체지방과 body mass index (BMI)를 유의성 있게 감소시켰지만, placebo는 효과가 없었다. CLA는 low-density lipoprotein (LDL)-cholesterol, total cholesterol 및 tryglyceride를 유의성 있게 낮추었고, high-density lipoprotein (HDL)-cholesterol는 증가시켰다. 운동능력인 trunk flexion, closed-eyes foot balance, standing long jump, shuttle run 및 sit-up를 향상시켰다. 이 결과는 CLA가 체지방을 감소시키고, 동맥경화 및 인체활동능력을 개선할 수 있어 비만 청소년의 체력을 관리하는데 도움을 줄 수 있을 것이다. Body fat reducing and physical-activity enhancing effects, along with artherosclerosis improving effects, of conjugated linoleic acid (CLA) were elucidated on obese male middle school students with more than 30% body fat. Twenty-four volunteers were randomly divided into control (placebo, n=12) and CLA treatment (n=12) groups. Subjects were daily fed 6 g CLA (6 capsules, twice a day) or a placebo for 12 weeks. At the end of the experiment, body composition, blood lipid composition and exercise capacities of subjects were measured. CLA significantly reduced body fat content and body mass index (BMI) along with body weight, while the placebo did not have any such effects. Similarly, CLA significantly reduced low-density lipoprotein (LDL)-cholesterol, total cholesterol, and triglyceride, but elevated high-density lipoprotein (HDL)-cholesterol content in blood. Meanwhile, in terms of exercise capacity, there were significant enhancements of trunk flexion, closed-eyes foot balance, standing long jump, shuttle run, and sit-up activities in the CLA treatment group. These results indicate that CLA consumption reduced body fats, improved atherosclerosis factors in blood and improved physical activity of young male obese middle school students, and suggest that CLA could be a useful material for the heath care of obese young men.

        Role for Volatile Branched - Chain and Other Fatty Acids in Species - Related Red Meat Flavors

        2-Methylbutanoic, 3-methylbutanoic, 4-methylpentanoic, 2-ethylhexanoic, 4-methylhexanoic, 4-methyloctanoic, 6-methyloctanoic, 4-ethyloctanoic, 4-methylnonanoic 및 2-ethyldecanoic acids를 포함하는 다수의 volatile branched-chain fatty acids(VBCFAs)가 쇠고기, 돼지고기, 염소고기(미국산 흰염소고기, 한국산 흑염소고기), 양고기로부터 분리, 동정되었다. 쇠고기 향은 amino acid, carbohydrate, 그리고 lipid의 반응에 의해 생성된 향기성분에 의해 이루어지는 기본 meaty flavor로 결정되어 지는 반면, 염소고기나 양고기의 경우는 쇠고기의 기본향에 4-ethyloctanoic acid와 4-methyloctanoic acid의 독특한 노린내가 더해지므로서 염소고기나 양고기 특유의 향을 내었다. 돼지고기는 3-methylbutanoic acid함량이 다른 세종류에 비해 높고 3-methylbutanoic acid 특유의 꼬린냄새는 돼지고기의 기름층으로부터 나는 unclean flavor에 중요한 영향을 미치는 것으로 생각된다. 염소고기는 C5-C7 chain length의 volatile branched 및 n-chain fatty acids를 적게 함유하고 있는반면 4-ethyloctanoic acid는 염소고기의 노린내를 내는 결정적인 화합물이었다. 4-Methyloctanoic acid는 양고기의 sweaty-muttony flavor를 내는 중요한 화합물이었으며 염소고기와는 달리 C5, C6, 그리고 C7 branched-chain fatty acids도 상당량 함유되어있었다. 한국산 흑염소고기와 미국산 흰염소고기 중에 들어있는 VBCFAs의 종류는 같으나, 한국산 흑염소고기가 미국산 흰염소고기 보다 노린내가 강하며 4-ethyloctanoic acid의 함량도 높았다. Species-related meat flavors were investigated for red meats (bovine, porcine, caprine, and ovine). Volatile branched-chain fatty acids (VBCFAs) including 2-methylbutanoic, 3-methylbutanoic, 4-methylpentanoic, 2-ethylhexanoic, 4-methylhexanoic, 4-methyloctanoic, 6-methyloctanoic, 4-ethyloctanoic, 4-methylnonanoic, and 2-ethyldecanoic acids were identified in the meats from cow(bovine), pig(porcine), goats(caprine ; American white goat and Korean black goat), and lamb (ovine). Beef flavor of bovine meat was characterized by the basic meaty flavor, lacking in goaty and muttony flavor impacts due to low or absent in 4-methy-loctanoic and 4-ethyloctanoic acids. Porcine meat contained the least number of VBCFAs among sample species tested, and 3-methylbutanoic acid contributed to the unclean sweaty odor of pork. Caprine meat from Korean black and American white goats lacked in short VBCFAs(C5, C6, and C7) and contained 4-methyloctanoic and 4-ethyloctanoic acids contributing to the characteristic goaty flavor of caprine meat. Caprine meat flavor was distinctively characterized by 4-ethyloctanoic acid, while 4-methyloctanoic acid provides sweaty-muttony flavor to ovine meat. Although kinds of VBCFAs are same in two different varieties of caprine meats, meat sample from Korean black goat had stronger goaty odor and contained higher concentration of 4-ethyloctanoic acid than the meat sample from American white goat did.

        Benzo(a)pyrene으로 유발한 Mouse Forestomach Tumor 생성에 대한 Astaxanthin 함유 난황의 효과

        Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for benzo[a]pyrene (BP)-induced mouse forestomach tumorigenesis initiating regimen. Female ICR mouse (6-7 weeks of age) were housed in polycarbonated cages (5 mice/cage; 20 mice/treatment) in a humidity-and-temperature-controlled facility and permitted free access to water and food. One week later, four and 2 days prior to p.o. treatment with BP (2 ㎎/0.2 ㎖ corn oil), mice were given 0.2 ㎖ PBS containing 50 ㎎ AEY, 100 ㎎ AEY, 150 ㎎ AEY, or 150 ㎎ CEY. Control mice were only given 0.2 ㎖ PBS. Three days later this sequence was repeated for a total of 4 times. Beginning with the first intubation and continuing thereafter, body weight and food intake were recorded once weekly. All surviving mice were sacrificed 24 weeks after the first dose of BP. Mice treated with AEY developed only about one third as many neoplasms/animal as mice in control or CEY-treated group (p$lt;0.05). Reduction effect of tumor development by AEY was dependent upon doses applied. Tumor incidence was also reduced by AEY treatments, but significantly reduced only by 150 ㎎ AEY treatment when compared to that by control or CEY. Food intake and body weight were not affected by AEY treatment. These results indicate that AEY inhibits tumorigenesis of mouse forestomach induced by BP.

        Benzo[a]pyrene으로 유발한 Mouse Forestomach Tumor 생성에 대한 Astaxanthin 함유 난황의 효과

        Benzo[a]pyrene (BP)으로 유발한 mouse의 전위암 형성에 대한 astaxanthin 함유 난황 (astaxanthin-containing egg yolk : AEY)의 영향을 연구하였다. Female ICR mouse (6-7 주령, 5 mice/cage, 20 mice/treatment)에게 물과 사료를 자유로이 공급하면서 일주일간 적응시킨 후 다음과 같이 처리를 하였다. BP (2 mg/0.2 ml corn oil)를 각 mouse에 경구투여하기 4 일과 2 일 전에 50 mg AEY, 100 mg AEY, 150 mg AEY, 또는 150 mg control egg yolk (CEY)을 함유하는 phosphate-buffered saline (PBS) 0.2 ml를 경구투여하였다. Control mouse는 0.2 ml PBS와 BP 만 경구투여하였다. 이 과정을 4회 반복하였다. BP를 경구투여 한 일주일 후부터 몸무게와 사료섭취량을 매주 기록하였으며 24주 후에 생존한 모든 mouse에 대해 전위암 생성을 조사하였다. AEY를 처리한 mouse에서는 control mouse나 CEY 처리 mouse에 비해 mouse 당 암의 수가 1/3 정도로 감소되었다. 이와 같은 AEY의 항암효과는 AEY의 처리량에 비례하였다. AEY 처리에 따른 암발생율은 control이나 CEY 처리에 비해 감소되었으나 150 mg AEY 처리에서만 유의성이 있는 감소를 보였다. AEY 처리에 따른 몸무게나 사료 소비량의 감소현상은 볼 수 없었다. 따라서 이 결과는 AEY가 BP로 유발한 mouse의 전위암 발생을 억제함을 암시한다. Reduction effect of tumor development by AEY was dependent upon doses applied. Tumor incidence was also reduced by AEY treatments, but significantly reduced only by 150 mg AEY treatment when compared to that by control or CEY. Food intake and body weight were not affected by AEY treatment. These results indicate that AEY inhibits tumorigenesis of mouse forestomach induced by BP.

        Optimal Level for the Protection of Carbon Tetrachloride-induced Sprague-Dawley Rat Liver Damage by Mycelial Cultures of Lentinus edodes

        표고버섯균사체 고체배양물 분말과 액체배양물 분말을 2:1 (w/w)로 혼합한 시료 (LED로 명명)가 CCl₄로 유발된 숫컷 쥐의 간독성을 보호하는 효과에 대해 연구하였다. Vehicle 처리군(0.2 ㎖ 증류수), Control 처리군(0.2 ㎖ 증류수), LED 처리군(LED 100, 200, 300, 400 ㎎/㎏ BW을 0.2 ㎖ 증류수에 각각 혼합), 그리고 Silymarin 처리군(200 ㎎/㎏을 0.2 ㎖ 증류수에 용해)의 각 처리군(군당 여섯 마리)에 매일 2주간 투여한 다음, Vehicle 처리군을 제외한 모든 군에 CCl₄ (CCl₄:corn oil, 1:1 v/v; 0.5 ㎖/㎏ BW)를 복부에 주사하였다. 하루가 지난 후에 생화학적 지표성분을 분석하기 위해 혈액과 간 시료를 채취하였다. 모든 LED 처리에 의해 간 SOD, catalase, 그리고 GSH peroxidase의 활성이 증가하였으며, TBARS, tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) 그리고 interleukin-6 (IL-6)가 감소하였다. 그 결과, 혈청중의 GOT, GPT, 그리고 LDH의 활성이 감소하였다. LED의 간 기능보호효과가 가장 효과적으로 나타난 처리농도는 200 ㎎/㎏ BW였다. 이러한 결과로 보아 LED는 CCl₄로 유발된 SD 쥐의 간 손상을 보호하며, 이는 LED의 항산화능과 cytokine의 감소에 의한 것이다. 따라서, LED는 사람의 간독성보호를 위해 효과적으로 사용할 수 있을 것으로 생각된다. The protective effects of a powder mixed with solid-cultured and liquid-cultured Lentinus edodes mycelia (2 : 1, w/w) (designate LED) with different doses of carbon tetrachloride (CCl₄) on induced hepatotoxicity in male Sprague-Dawley (SD) rats was investigated. The rats were divided into seven groups (6 rats/group) and the following substances were administered orally to each group: Vehicle (0.2 ㎖ distilled water), Control (0.2 ㎖ distilled water), LED (LED 100, 200, 300 and 400 ㎎/㎏ BW in 0.2 ㎖ distilled water), and Silymarin (200 ㎎/Kg BW in 0.2 ㎖ distilled water). After two weeks of daily administration, all groups except for the Vehiclegroup were subjected to abdominal injection with CCl₄ (CCl₄ : corn oil, 1 : 1 v/v; 0.5 ㎖/㎏ BW). One day later, blood and liver samples were collected to analyze biomarkers. All LED treatments elevated hepatic superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH peroxidase) activities, and reduced thiobarbituric reactive substances (TBARS), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6), resulting in the reduction of glutamate-oxalate transaminase (GOT), glutamate-pyruvate transaminase (GPT) and lactic acid dehydrogenase (LDH) activities in plasma. These results indicate that LED effectively protected SD rat hepatotoxicity induced by CCl₄ through its antioxidative activity and reduction of some cytokines. The highest efficacy was found in LED 200 ㎎/㎏ BW, showing potential as a useful material for protection from hepatotoxicity in humans.
