      • 달걀버섯(Amanita hemibapha subsp. hemibapha DGUM 28001)의 균사체 생육을 위한 배양조건의 최적화

        In many recipes, Amanita hemibapha subsp, hemibapha has been regarded and used as an excellent edible mushroom. However, so far, no artificial production of this mushroom has been reported. It has usually found in the mountain or the plateau from summer through autumn. The culture condition and media composition for the enhanced mycelial growth of A hemibapha subsp. hemibapha DGUM 28001 were investigated. Optimal temperature and pH for the mycelial growth were 30℃ and pH 5.0, respectively. Among the complex media tested, SYSP(starch yeast extract soytone pyridoxine) was the most favorable complex medium for enhancing the mycelial growth. When Czapek-Dox medium was used as a minimal medium, starch of polysaccharide was very excellent as a carbon and energy source. Sucrose and xylitol were very good carbon source for disaccharide and monosaccharose, respectively. In general, an organic complex nitrogen source was better than an inorganic one. Among the organic complex nitrogen sources tested, yeast extract was the best as a source of nitrogen. As a phosphorus source for the mycelial growth, the most favorable one was NH_4H_2PO_4. When folic acid, vitamin, was supplemented to the minimal medium, mycelial growth was enhanced. These results could be used to produce biologically-active substances from the mycelial culture and for the artificial production of A. hemibapha subsp. hemibapha.

        Tracheobronchopathia osteoplastica is a rare disorder of unknown cause characterized by the projection of numerous bony or cartilaginous submucosal nodules into the tracheobronchial lumen with sparing of the posterior membranous portion of tracheobronchial tree. In the past, most of the cases were diagnosed as secondary finding at autopsy. Nowaday, the key component of the evaluation is flexible fiberoptic laryngoscopy and bronchoscopy, which typically demonstrates irregular spicules of subrnucosal bone and cartilage projecting into the tracheobronchial lumen and causing various degrees of airway obstruction. A 45 years old man was visited Sunchon St. Carollo hospital due to discomfort on throat. We saw several nodules just below vocal cord via laryngoscope. The computed tomography revealed multiple small calcified nqdules on trachea and both main bronchi. Pathologic finding of bronchoscopic biopsy showed abnormal proliferation of bony and cartilagious nodules in the tracheal submucosa. These fingdings were consistent with tracheobronchopathia osteoplastica. We report here on a case of tracheobronchopathia osteoplastica with review of literature.

      • Candida albicans菌의 生育을 抑制하는 韓藥材 探索에 關한 硏究

        Thirty-six medicinal herbs and 14 prescribed herb medicines which have usually been used for treatment of reducing fever, purging intense heat, and detoxication were screened for antifungal activity against Candida albicans which causing Leukorrhea. Minimal inhibitory concentrations of water- and ethanol-soluble extracts of Hwangbaek (Phellodendron amurense) and Hwangkeum (Scutellaria baicalensis) were determined. The water-soluble extracts of Hwangyon (Coptis japonica), Hwangbaek (P. amurense), and Hwangkeum (S. baicalensis) showed antifungal activities against C. albicans. The water- and ethanol-soluble extracts of Hwangbaek (P. amurense) and Hwangkeum (S. baicalensis) showed antifungal activities. With the water-soluble extract of Hwangbaek (P. amurense) and Hwangkeum (S. baicalensis), minimal inhibitory concentrations (MICs) against C. albicans were 10 and 20㎎/ml, respectively. With the ethanol-soluble ones of Hwangbaek (P. amurense) and Hwangkeum (S. baicalensis), MICs were 5 ㎎/ml and 10 ㎎/ml, respectively. The prescribed herb medicines, sambohwan, showed antifungal activity against C. albicans.

      • 잣버섯 (Lentinus Iepideus DGUM 25050)의 균사생육을 위한 배지조성의 최적화

        The fruiting body of Lentinus lepideus has been used as an edible mushroom, which has good taste, typical pine flavor and medicinal components for immunostimulation and antitumor activity. In this study, the culture condition and medium composition of L. lepideus DGUM 25050 isolated were optimized for higher mycelial production. The optimal temperature and pH for mycelial culture were 30℃ and pH 5.0, respectively. Among the complex media tested, yeast-malt extract was most favorable for stimulating mycelial growth of L. lepideus. When the Czapek-Dox medium was used as a minimal medium for mycelial culture, starch of polysaccharide was very excellent and both lactose of dissacharide and xylitol of monosaccharose were prominent as a sole source of carbon and energy. As a source of nitrogen, organic nitrogen complex was better than inorganic one and organic soytone was best. When Na_2HPO_4 as a phosphorus source and p-amino benzoic acid as a vitamin were supplemented to the minimal medium, mycelial growth was enhanced. The optimal C/N ratio was 6.0 and stimulated a favorable growth of L. lepideus, when 3.0% of starch and 0.5% of soytone were added to the medium. The optimized medium for mycelial growth of L. lepideus DGUM 25050 was determined and named as LLM, of which composition was as follows: starch 3.0%, malt extract 1.2%, soytone 0.5%, Na2HPO4 0.72%, formic acid 0.1%, ethyl alcohol 0.1%, p-aminobenzoic 0.5 ㎎/l. When the mycelia were cultivated for 15 days, 9.2 g/ℓ of dry weight in LLM broth was higher than 6.8 g/ℓ in the medium used generally for mycelial culture of L. lepideus.

      • 목질진흙버섯(Phellinus Iinteus) 균사배양을 위한 배양조건 최적화

        The optimum temperature and pH for mycelial growth of Phellinus linteus ATCC 26710 were 30℃ and 5.0-6.0, respectively. Among the complex media used, the Agrocybe chlindracea medium was excellent. When Czapek Dox medium was used as a minimal medium, starch, trehalose and galactose were excellent sources of carbon and energy. Ammonium chloride (as an inorganic nitrogen), arginine (as an amino acid) and soytone (an an organic complex nitrogen) were good for mycelial growth. The C/N ratios of ammonium chloride and soytone were 5.0 and 7.5, respectively. Potassium phosphate and ammonium phosphate can be used as a phosphorus source. The composition of newly synthesized medium for mycelial growth of P. linteus was as follows : starch 1.5%, formic acid 0.1%, soytone 0.2%, arginine 0.3%, NH_4Cl 0.2%, KH_2PO_4 0.1%.

        가야산 국립공원일대의 식생 및 식물상 연구 : 단지봉 지역을 중심으로 Especially on the Danji-bong Area

        본 조사지역인 가야산국립공원은 지리학적으로는 북위 35?5′00″~35?9′30″, 동경 128?2′30″~128?09′30″에 위치하고, 행적구역상으로는 경상남도의 합천군과 거창군, 경상북도의 성주군, 고령군에 걸쳐 있으며, 총면적은 57.81 km꼬?이른다. 가야산은 우리나라 기후지역상 온대남부에 위치하고, 식물구계학적으로는 남부아구에, 식생구계학상으로는 냉온대에 속하는 것으로 알려져 있다. 또한, 전체면적의 약 82.5%가 해발 600m 이상이며, 1,000m이상이 약 10.3%에 해당된다. 가야산의 해인사-극락골-토신골 지역은 잘 보존된 소나무림이 우점하고 있고, 부분적으로 노각나무가 산생하는 것이 관찰되었으며, 해인사-진대밭골-두리봉 지역은 계곡에는 소나무와 졸참나무가 고지대에는 신갈나무 등이 부분적으로 군락을 형성하고 있었다. 또한 남산제일봉과 북사면은 각각 등산로와 임도가 형성되어 있어 소나무와 졸참나무, 신갈나무 등이 혼재된 양상을 나타내었고, 백운동계곡 주변은 다른 사면과 달리 식생의 발달정도가 소나무와 관목위주로 매우 단순했다. 특기할만한 점은 상왕봉 주변에서 흰참꽃, 설앵초, 네귀쓴풀, 백리향, 솔나리, 구름송이풀 등의 희귀 또는 고산성 식물들이 분포하는 것이 확인되었다. 가야산국립공원지역에 분포하는 양치식물 이상의 관속식물을 대상으로 조사한 결과 91과 268속 373종 1아종 75변종 12품종 4잡종으로 총 465분류군이 조사 확인되었으며, 이는 우리나라 관속식물 4,071 분류군의 11.4%에 해당되었다. 한국특산식물은 11과 15속 15분류군으로 조사지역내 분포하는 465분류군중 약 3.2%이며. 식물구계학적 특정식물은 17과 21속 25분류군으로 이는 가야산 소산식물 465분류군의 5.4%이었고, 조사지역에 분포하는 귀화식물은 5과 7속 7분류군으로 우리 나라전체 귀화식물종 177종류의 4%이다. 또한 가야산 단지봉(1,028.5m)에서 새로 발견된 산간습지 (forested wetland)와 자란초 자생 군락지에 대해 식생 및 층위별 종조성을 토대로 현존 식생 구조를 분석하였다. This study aims to investigate the flora of Mt. Gaya National Park, especially focused on the forested wetland vegetation of Danji-bong area. The flora of Mt. Gaya was investigated from April in 2003 to October in 2004. Vascular plants of this area consist of total 465 taxa, 4 hybrids, 12 forma, 75 varieties, 1 subspecies, 373 species, 268 genera, 91 families. It corresponds to 11.4% of totally 4,071 taxa appeared in Korea. Korean endemic plants were composed 11 families, 15 genera, 14 species, 1 variety and totaling 15 taxa. And also actual vegetation structures were analyzed the newly founded forested wetland and natural population Ajuga spectabilis on the Danji- bong area using the vegetation and the species composition by tree layer.

      • Monilia sitophila DGUM 7000의 형태 및 생리학적 특성

        목질진흙버섯(Phellinus linteus) 배양시 높은 빈도로 오염되는 진균을 분리하여 형태적 특징에 따라 Monilia sitophila로 동정하였고, Monilia sitophila DGUM 7000으로 명명하였다. M. sitophila DGUM 7000의 균사생장을 위한 최적 온도 및 pH는 각각 37℃와 7.0이었다. MCA배지를 기본배지로 탄소원 이용성 실험에서 단당류, 이당류 및 다당류로 xylose, lactose, CMC를 사용하였을 때 가장 우수한 균사생장을 나타내었다. 유기산으로 gluconic acid를 MCA배지에 첨가시 균사생장이 촉진되었으나, oxalic acid를 첨가하였을 때 생장이 억제되었다. Yeast extract가 포함된 배지에서 우수한 균사생장을 보여주었다. 유기질소원으로 tyrosine 및 leucine이 균사생장에 가장 우수한 아미노산이었다. 무기질소원으로 sodium nitrate가 가장 우수하였으나, ammonium molybdate 사용시 생육이 억제되었다. 사용된 모든 인산원에서 우수한 균사 생장을 나타내었다. 항진균제 thiabendazole, benomyl을 각각 1.0 및 0.2 ㎎/L 농도로 첨가하였을 때 목질진흙버섯의 균사생장에 큰 영향은 없었으나, M. sitophila DGUM 7000 오염균의 균사생장은 현저하게 억제되었다. 이 결과는 thiabendazole 과 benomyl이 목질진흙버섯 배양시 M. sitophila 오염균 방제를 위한 항진균제로의 사용 가능성을 제시하였다. The red fungus was isolated from the contaminated culture bottle of Phellinus linteus. It was identified as Monilia sitophila based on morphological characteristics and named as Monilia sitophila DGUM 7000. The optimal temperature and pH for the mycelial growth of M. sitophila DGUM 7000 were 37℃ and 7.0, respectively. When xylose, lactose and CMC were supplemented in MCA media as a mono-, di- and polysaccharide, mycelial growth of M. sitophila were most effective. As a organic acid, gluconic acid enhanced the mycelial growth. However oxalic acid inhibited the mycelial growth. The mycelial growth of M. sitophila DGUM 7000 was enhanced by addition of yeast extract. As a organic nitrogen source, tyrosine and leucine were very effective amino acids. As a source of inorganic nitrogen, sodium nitrate was the best. However, ammonium molybdate inhibited the mycelial growth. All the sources of phosphate tested showed good mycelial growth. The addition of 1.0 ㎎/L of thiabendazole or 0.2 ㎎/L of benomyl inhibited the mycelial growth of M. sitophila DGUM 7000, but did not those of P. linteus. This result suggests that thiabendazole and benomyl control the growth of contaminant M. sitophila and could be used as a antifungal agent for the cultivation of P. linteus.

      • Rifampicin에 의한 것으로 추정되는 위막성 대장염1예

        Pseudomembranous colitis, caused by altering the normal colonic flora and allowing the multiplication of Clostridium difficile, is an deleterious adverse effect of antibiotics. But it is rarely reported by rifampicin. Rifampicin is one of the first line drug in the treatment of tuberculosis and many patients are exposed to its potential adverse effects. We experienced a patient that had abdominal discomfort and hematochezia due to pseudomembranous colitis after receiving antituberculous medication, and which was probably caused by rifampicin. A 82 years old man was admitted with abdominal discomfort and hematochezia for one week. On the past history he had been diagnosed as endobronchial tuberculosis about 4 months ago. Colonoscopy revealed multiple discrete whitish mucosal lesion on rectosigmoid colon, and histologic findings were consistent with pseudomembranous colitis. The antituberculous agents were discontinued and vancomycin was administered. The patient's symptoms were resolved within several days. There was no recurrence after reinstitution of the antituberculous agents excluding rifampicin. We report here on a case of pseudomembranous colitis probably due to rifampicin.

        Oxygen-dependent Respiration and Proteon Extrusion in Wolinella Succinogenes

        When $H^{2}$ was provided as the electron donor, optimum $O_{2}$ levels for growth of Wolinella succinogenes ATCC 29543 were 2% and 8% on brucella agar and in brucella broth, respectively. No growth occurred under 21% $O_{2}$, and scant or no growth occurred under anaerobic condition. $O_{2}$ uptake was inhibited by cyanide and 2-heptyl-4-hydroxyquinoline N-oxide. Protons were translocated out of the cell when oxygen was used as the terminal electron accetor. The $H^{+}$/O ratio with $H_{2}$ and formate as an electron donor were 1, 97 and 1.49, respectively. Proton translocation was inhibited by the protonophore carbonylcyanide m-chlorophenylhydrazone.e.
