        Hypoglycemic and Hypolipidemic Activity of Polygonatum sibiricum Fermented with Lactobacillus brevis YM 1301 in Diabetic C57BL/6 Mice

        Polygonatum sibiricum (PS) has been used as herbal medicine to treat type 2 diabetes mellitus (T2DM). However, how lactic acid fermentation of PS influences glucose and lipid metabolism remains unclear. The current study was undertaken to evaluate the hypoglycemic and hypolipidemic effects of PS fermented with Lactobacillus brevis YM 1301 (YM 1301) in streptozotocin and high-fat diet-induced T2DM mice. Biochemical analysis revealed that supplementation with metformin, PS, or fermented Polygonatum sibiricum (FPS) lowered the fasting blood glucose, insulin, total cholesterol, triglyceride, and low-density lipoprotein cholesterol of diabetic mice. FPS showed relatively more potency to reduce the homeostasis model assessment-insulin resistance and glycated hemoglobin than PS. Moreover, a high dosage of FPS protected against glucose intolerance and insulin resistance by increasing the ratio of phosphor-AKT/AKT. Histological examination and quantitative polymerase chain reaction results showed that dietary FPS ameliorated the lipid accumulation in liver and white adipose tissue (WAT) by inhibiting lipogenesis, enhancing lipolysis, and fatty acid oxidation. FPS exhibited greater efficacy than PS on improving the transcriptional expression of adipose triacylglyceride lipase, carnitine palmitoyltransferase 1, and uncoupling protein 1. In addition, FPS exerted a striking anti-inflammatory effect by suppressing the expression of interleukin 6, interleukin 1β, tumor necrosis factor-α, and transforming growth factor-β in WAT of diabetic C57BL/6 mice. Finally, FPS supplementation enhanced the activation of AMPK. In conclusion, these results suggest that the FPS may be more promising than PS as a potential therapeutic agent for diabetes and obesity.

        Damage Mechanism of Drift Ice Impact

        The ice damage occurs frequently in cold and dry region of western China in winter ice period and spring thawperiod. In the drift ice condition, it is easy to form different extrusion force or impact force to damage tunnellining, causing project failure. The failure project could not arrive the original planning and construction goal,giving rise to the water allocation pressure which influences diversion irrigation and farming production inspring. This study conducts the theoretical study on contact-impact algorithm of drift ices crashing diversiontunnel based on the symmetric penalty function in finite element theory. ANSYS/LS-DYNA is adopted as theplatform to establish tunnel model and drift ice model. LS-DYNA SOLVER is used as the solver and LSPREPOSTis used to do post-processing, analyzing the damage degrees of drift ices on tunnel. Constructingphysical model in the experiment to verify and reveal the impact damage mechanism of drift ices on diversiontunnel. The software simulation results and the experiment results show that tunnel lining surface will formvarying degree deformation and failure when drift ices crash tunnel lining on different velocity, different plansize and different thickness of drift ice. The researches also show that there are damages of drift ice impact forceon tunnel lining in the thawing period in cold and dry region. By long time water scouring, the tunnel liningsurfaces are broken and falling off which breaks the strength and stability of the structure.

        Planning and Execution of Dynamic Whole-body Locomotion for a Wheeled Biped Robot on Uneven Terrain

        To improve the adaptability of the wheeled biped robot (WBR) to uneven terrain, firstly an integrated modeling method for wheeled-legs is proposed. The under-actuated part is effectively restrained by defining the interaction force between the WBR and the trunk. The mapping relationship between the wheeled leg’s end force and the joint torques in the balanced state is built. Based on this premise, a control framework that does not rely on external sensors is proposed, and the trunk pose is used as the task space to plan the generalized force output of the wheeled legs and calculate the joint torques. Since the joint space position is not constrained, the leg wheels will be based on the terrain conditions and are adaptively stretched and adjusted back and forth. To further improve the terrain adaptability, a slope estimator and a stabilizer are constructed to deal with the attitude fluctuation caused by the sudden change of terrain. The control framework is proved to verify by simulations and experiment.

        Cloning, expression and functional analysis of the duck Toll-like receptor 5 (TLR5) gene

        Toll-like receptor 5 (TLR5) is responsible for the recognition of bacterial flagellin in vertebrates. In the present study, the first TLR5 genein duck was cloned. The open reading frame (ORF) of duck TLR5 (dTLR5) cDNA is 2580 bp and encodes a polypeptide of 859 amino acids. We also cloned partial sequences of myeloid differentiation factor 88, 2’-5’-oligoadenylate synthetase (OAS), and myxovirus resistance (Mx)genes from duck. dTLR5 mRNA was highly expressed in the bursa of Fabricius, spleen, trachea, lung, jejunum, rectum, and skin; moderatelyexpressed in the muscular and glandular tissues, duodenum, ileum, caecum, and pancreas; and minimally expressed in the heart, liver, kidney,and muscle. DF-1 or HeLa cells transfected with DNA constructs encoding dTLR5 can activate NF-B leading to the activation of interleukin-6(IL-6) promoter. When we challenged ducks with a Herts33 Newcastle disease virus (NDV), mRNA transcription of the antiviral moleculesMx, Double stranded RNA activated protein kinase (PKR), and OAS was up-regulated in the liver, lung, and spleen 1 and 2 dayspost-inoculation.
