        Hypoglycemic and Hypolipidemic Activity of Polygonatum sibiricum Fermented with Lactobacillus brevis YM 1301 in Diabetic C57BL/6 Mice

        Polygonatum sibiricum (PS) has been used as herbal medicine to treat type 2 diabetes mellitus (T2DM). However, how lactic acid fermentation of PS influences glucose and lipid metabolism remains unclear. The current study was undertaken to evaluate the hypoglycemic and hypolipidemic effects of PS fermented with Lactobacillus brevis YM 1301 (YM 1301) in streptozotocin and high-fat diet-induced T2DM mice. Biochemical analysis revealed that supplementation with metformin, PS, or fermented Polygonatum sibiricum (FPS) lowered the fasting blood glucose, insulin, total cholesterol, triglyceride, and low-density lipoprotein cholesterol of diabetic mice. FPS showed relatively more potency to reduce the homeostasis model assessment-insulin resistance and glycated hemoglobin than PS. Moreover, a high dosage of FPS protected against glucose intolerance and insulin resistance by increasing the ratio of phosphor-AKT/AKT. Histological examination and quantitative polymerase chain reaction results showed that dietary FPS ameliorated the lipid accumulation in liver and white adipose tissue (WAT) by inhibiting lipogenesis, enhancing lipolysis, and fatty acid oxidation. FPS exhibited greater efficacy than PS on improving the transcriptional expression of adipose triacylglyceride lipase, carnitine palmitoyltransferase 1, and uncoupling protein 1. In addition, FPS exerted a striking anti-inflammatory effect by suppressing the expression of interleukin 6, interleukin 1β, tumor necrosis factor-α, and transforming growth factor-β in WAT of diabetic C57BL/6 mice. Finally, FPS supplementation enhanced the activation of AMPK. In conclusion, these results suggest that the FPS may be more promising than PS as a potential therapeutic agent for diabetes and obesity.

        Liver metabolic perturbations of heat-stressed lactating dairy cows

        Objective: The objective of the present study was to elucidate the mechanism underlying liver metabolic perturbations in dairy cows exposed to heat stress (HS). Methods: Liquid chromatography massabl spectrometry was used to analyze metabolic differences in livers of 20 dairy cows, with and without exposure to HS. Results: The results revealed 33 potential metabolite candidate biomarkers for the detection of HS in dairy cows. Fifteen of these metabolites (glucose, lactate, pyruvate, acetoacetate, ${\beta}$-hydroxybutyrate, fumaric acid, citric acid, choline, glycine, proline, isoleucine, leucine, urea, creatinine, and orotic acid) were previously found to be potential biomarkers of HS in plasma or milk, discriminating dairy cows with and without HS. Conclusion: All the potential diagnostic biomarkers were involved in glycolysis, amino acid, ketone, tricarboxylic acid, or nucleotide metabolism, indicating that HS mainly affected energy and nucleotide metabolism in lactating dairy cows.

      • TOWARDS 21ST CENTURY: CURRENT TRENDS OF SPECIAL EDUCATION IN CHINA

        As a developing country with the uneven development pattern, China has been struggling in taking the best use of limited resources in providing the special educational services. The paper presents the general information and current trends of the special education regarding the issues like compulsory education, the expansion of service to preschoolers and post compulsory education, vocational and higher education for students with special educational needs and so on. Challenges and problems confronting in Chinas special education are also presented in this paper. Limited resources, integration vs. segregation, family supports and research work are those among the challenges discussed. Special education in China has been in a rapidly developing time since 1980s. As an integral part of the national education system, special education is providing to target people through 3 channels mainly: special school, special program, and integrating into regular class. Among them, integration is a new-tried and most challenging one. In this presentation, we will present the general information about special education in China, and focus on the current trends, which will influence the development of special education in China in the new century.

        mtDNA Diversity and Origin of Chinese Mongolian Horses

        In order to learn the origin of the Chinese Mongolian horse, we analyzed polymorphisms within the mtDNA D-loop variable region in 305 horses of 6 types of 3 different breeds, including one imported breed, one cultivated breed and 4 types of one local breed. We detected 13 different haplotypes, and subsequent sequence analysis showed that all 6 horse types were genetically diverse. By constructing a cladogram of mtDNA D-loop sequences from the 6 horse types along with homologous sequences from several other horse types obtained from GenBank, we showed that Chinese Mongolian horses have a close genetic relationship with other horse types from Mongolia. We also speculate that several Chinese Mongolian horses descended from Przewalskii horse. Additionally, the 13 haplotypes were dispersed throughout the cladogram, suggesting that Chinese Mongolian horses likely originated from multiple female ancestors. A phylogenetic map of the 6 horse types showed that the genetic relationship between the local Wuzhumuqin and Wushen types were the closest. The Xinihe and Baerhu were also closely related to each other, and slightly more distantly related to the cultivated Sanhe breed. All five of the local Chinese horse types had a much more distant relationship with the imported Thoroughbred breed.

        Characterization of Microbial Communities in Chinese Rice Wine Collected at Yichang City and Suzhou City in China

        Two typical microbial communities from Chinese rice wine fermentation collected in Yichang city and Suzhou city in China were investigated. Both communities could ferment glutinous rice to rice wine in 2 days. The sugar and ethanol contents were 198.67 and 14.47 mg/g, respectively, for rice wine from Yichang city, and 292.50 and 12.31 mg/g, respectively, for rice wine from Suzhou city. Acetic acid and lactic acid were the most abundant organic acids. Abundant fungi and bacteria were detected in both communities by high-throughput sequencing. Saccharomycopsis fibuligera and Rhizopus oryzae were the dominant fungi in rice wine from Suzhou city, compared with R. oryzae, Wickerhamomyces anomalus, Saccharomyces cerevisiae, Mucor indicus, and Rhizopus microsporus in rice wine from Yichang city. Bacterial diversity was greater than fungal diversity in both communities. Citrobacter was the most abundant genus. Furthermore, Exiguobacterium, Aeromonas, Acinetobacter, Pseudomonas, Enterobacter, Bacillus, and Lactococcus were highly abundant in both communities.

        Graphene quantum dots decorated 3D transitional metal (Fe, Co) oxide graphene nanoribbons for oxygen reduction reaction

        In this paper, we presented a hybrid composite of graphene quantum dots (GQDs)-modified three-dimensional graphene nanoribbons (3D GNRs) composite linked by Fe3O4 and CoO nanoparticles through reflux and ultrasonic treatment with GQDs, denoted as 3D GQDs-Fe3O4/CoO@GNRs (3D GFCG). In this hybrid, the 3D GNRs framework strengthened the electrical conductivity and the synergistic effects between GQDs and 3D GFCG enhanced the oxygen reduction reaction (ORR) activity of the nanocomposite. The results imply that decorating GQDs with other electro-catalysts is an effective strategy to synergistically improve their ORR activity.

        One-step synthesis of mesoporous sulfated zirconia nanoparticles with anionic template

        Mesoporous sulfated zirconia nanoparticles (MSZNP) with high surface area have been synthesized by using sodium dodecyl sulfate (C12H25OSO3Na, SDS), both as a template and a sulfating agent via one-step route. On the basis of FT-IR, EDS together with NH3-TPD analyses, the SO42− anion, originated from the hydrophilic head of SDS, can be incorporated into ZrO2 to form MSZNP with super-acidity. The phase transition of ZrO2 from tetragonal to monoclinic phase was effectively inhibited by the presence of SO42−, and the formation mechanism was illustrated in detail. MSZNP(550) achieved far higher activity than CSZ(550) in transesterification of soybean oil with methanol due to the synergistic effect of strong acidity, high BET surface area and the formation of mesostructure.

        Benzidine Induces Epithelial-Mesenchymal Transition of Human Bladder Cancer Cells through Activation of ERK5 Pathway

        Benzidine, a known carcinogen, is closely associated with the development of bladder cancer (BC). Epithelial-mesenchymal transition (EMT) is a critical pathophysiological process in BC progression. The underlying molecular mechanisms of mitogen-activated protein kinase (MAPK) pathway, especially extracellular regulated protein kinases 5 (ERK5), in regulating benzidine-induced EMT remains unclarified. Hence, two human bladder cell lines, T24 and EJ, were utilized in our study. Briefly, cell migration was assessed by wound healing assay, and cell invasion was determined by Transwell assay. Quantitative PCR and western blot were utilized to determine both gene expressions as well as protein levels of EMT and MAPK, respectively. Small interfering RNA (siRNA) was transfected to further determine ERK5 function. As a result, the migration and invasion abilities were enhanced, epithelial marker expression was decreased while mesenchymal marker expression was increased in human BC cell lines. Meanwhile, benzidine administration led to activation of ERK5 and activator protein 1 (AP-1) proteins, without effective stimulation of the Jun N-terminal kinase (JNK) or p38 pathways. Moreover, Benzidine-induced EMT and ERK5 activation were completely suppressed by XMD8-92 and siRNAs specific to ERK5. Of note, ERK1/2 was activated in benzidine-treated T24 cells, while benzidine-induced EMT could not be reversed by U0126, an ERK1/2 inhibitor, as indicated by further study. Collectively, our findings revealed that ERK5-mediated EMT was critically involved in benzidine-correlated BC progression, indicating the therapeutic significance of ERK5 in benzidine-related BC.
