        Overexpression of Zoysia ZjCIGR1 gene confers cold stress resistance to zoysiagrass

        Zoysia japonica Steud. is a warm-season lawn grass popular in Korea and elsewhere. They are cultivated in many places such as river banks, roadside, and play grounds. However, there still is a disadvantage of frequent mowing, and the grass grows poorly under the chilly conditions. To develop a grass variety that circumvents these drawbacks, we cloned the chitininducible gibberellins-responsive1 gene (CIGR1) from zoysiagrass. The full length of the ZjCIGR1 (Zj; Zoysia japonica Steud.) gene was obtained by 5′/3′ RACE PCR and the phylogenetic tree showed that it belonged to the CIGR1-subgroup in the PAT1-group of GRAS protein family. Expression of the ZjCIGR1 in wild-type plants was confirmed in roots, meristems, leaves, and flowers, especially high in the flowers. The transgenic zoysiagrass was confirmed by PCR using gene-specific primers, phosphinothricin-acetyl-transferase (PAT) strip test, and Southern blot analysis. ZjCIGR1-overexpressing plants acquired tolerance to cold stress displaying morphological phenotypes characteristic of stress resistance. In addition, in the transformants, expression of the ZjCIGR1 as well as cold-regulated (COR) gene was increased compared to the wild-type plants under cold stress condition. These results suggest that ZjCIGR1 gene is an important candidate for regulating cold stress resistance.

        8년간(1992~1999) 산업안전보건연구원에 의뢰된 직업병 심의 사례 분석

        Objectives : Pneumoconisis and noise-induced hearing loss (NIHL) have been reported as main occupational diseases by the Special Health Examination. The Industrial Accident Compensation Insurance has reported various work-related diseases, however, these two diseases occupied almost a half of compensated cases. Therefore, it was not well known about the status of occupational diseases other than pneumoconiosis, NIHL, and cardio-cerebrovascular accident (CVA). This study was conducted to analyze claimed cases as an occupational disease, that was requested to the Korea Industrial Safety and Health Agency (KOSHA). Methods : The local office of the Korea Labor Welfare Corporation (KLWC) has asked the KOSHA for confirmation of claimed cases as an occupational disease. We analyzed 379 cases requested from KLWC, the Ministry of Labor, employers, unions and occupational health agencies from 1992 to 1999. Results : Male was 80.7 % of the requested cases. Their mean age was 42 years old and 75.5 % of them were more than 35 years old. The requested cases were increased rapidly from 25 cases in 1992 to 108 cases in 1999 and the accept rate was 50.7 %. The majority of requested cases were respiratory diseases(22.4%), cancers(18.5%), Neuropsychiatric problems(14.5%), and musculoskeletal problems(13.5%). The accept rate was high in reproductive, respiratory, musculoskeletal and digestive disorders and low in neuropsychiatric, renal and otologic problems and occupational cancers. 73.6% of them were caused by chemical agents, especially 28.5 % were by organic solvents. 67% of them were clinically confirmed at university hospitals. A half of the cases were from KyongIn area, even the request came from the whole country. Conclusions : A claim was common in workers whose age was over 35 years old and exposure history was over 10 years. The respiratory diseases and neuropsychiatric disorders were still main problems in occupational health and occupational cancers was increasing even though its accept rate was not high yet.

        유치의 치근단 병소에 의해 변위된 영구치의 자발적 맹출

        The infection of primary teeth occurs by caries or trauma. But, if it is not treated on time, the complication may occur such as enamel hypoplasia, discoloration, impaction, displacement and dilaceration of permanent successors. The periapical lesion on primary tooth could displace the permanent successors in any direction. Treatment options of displaced tooth are observation after extracting the primary teeth, surgical exposure, orthodontic traction, transplantation and extraction. In these cases, displacement of permanent central incisor caused by the periapical lesion of primary teeth was observed. The spontaneous eruption after extraction of primary teeth was expected considering degree of displacement, development of root and direction of eruption. This case report shows spontaneous eruption of displaced permanent tooth germ was achieved with periodic examination after extraction of primary teeth.

      • 韓國産 미꾸리에 關한 育種·繁殖學的 硏究 : Ⅵ. Histological Changes of Pituitary Gland and Oocytes from March to May Ⅵ. 계절에 따른 암컷의 腦下垂體와 卵母細胞의 組織學的 變化

        1992년 3월부터 1992년 5월까지 자연적인 채광상태에서 溫度를 유지시킨 후 채취된 미꾸리 암컷의 腦下垂體, 肝 및 卵巢 組織의 번식주기에 일어나는 미세구조의 변화를 광학 및 투과형 전자현미경을 이용하여 살펴보고, 이것들의 相互 聯關性을 알기 위하여 본 연구를 실시하였다. 본 연구에서 얻어진 결과는 다음과 같다. 성숙된 개체의 뇌하수체는 수많은 작은 분비과립과 커다란 분비구 및 불규칙한 형태의 과립들이 관찰되었다. 핵소체가 핵막 주변에 위치헤 있는 후기 단계(LMO)의 난모세포가 난황이 축적되는 성숙초기 단계(EMO)의 난모세포로 변형되는 시기를 난황의 형성이 개시되는 시기로 본다. 난모세포가 성장함에 따라 핵소체의 크기와 난황과립의 수가 증가하게 된다. 난황과립은 결정상의 과립으로 난세포질에 치밀하지 않은 상태로 축적된다. 성숙초기(EMO) 및 말기단계(LMO)에 난소는 커지고 투명하며 과립형태로서 색깔이 연노란색을 나타낸다. 따라서 난 내부를 쉽게 관찰할 수 있다. 방사대(ZR)는 성숙초기단계(EMO)에 단층으로부터 입방형태로 변화된다. 과립막 세포와 난모세포로 부터 유래된 미세융모는 난황형성이 진행되는 동안에 성장하게 되며, 방사대의 pore canals의 구멍을 통해서 서로 연결되어 있다. 따라서 외부로부터 물질의 이동이 있게 된다. 난모세포가 완전하게 성숙되어 방사대가 단단해 지면 미세융모는 그 기능이 상실된다. This study was undertaken to understand the development stages of pituitary gland, liver and ovary of Korean loach(Misgurnus anguillicaudatus) under natural circumstances from March to May 1922. Expecially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope during the reproductive cycle. In pituitary gland of mature individuals, there were observed a number of secretory granules,large globules, and irregular masses.The vitellogenic phase began as these late perinucleolus oocytes became transformed into early maturing oocytes through the accumulation of yolk. The size of the nucleoli and number of the yolk granules increased as the oocyte growed. Yolk granules were loosely deposited in the oocyte as crystalline granules. Due to the presence of early and late maturing oocytes, their ovaries were enlarged transparent, granular and yellowish in color. Zona radiata change a squamous into cuboid shape in EMO stage. Microvilli from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiata becomes more compact during oocyte maturation.

        우루과이 벼농사와 쌀 수출산업 분석 : 1. 우루과이 벼농사 특징 Ⅰ. Rice Farming in Uruguay

        우루과이 정부로부터 우루과이 국립농업연구소(INIA : Instituto Nacional de Investigacion Agropecuario Uruguay) 연구능력 강화를 위한 협력사업 요청을 받고 2003년 9월 20일부터 31일까지 우리 정부에 의해서 협력타당성 분석을 위한 우루과이 현지 조사와 자료분석이 이루어졌다. 우루과이 벼농사의 특징과 벼 재배환경 및 표준재배기술을 요약하면 다음과 같다. 1. 우루과이 벼농사의 특징은 우리와 반대계절에 재배되고 논이 따로 없이 목장에서 목초와 벼를 윤작체계로 하는 대규모 수출상업농이다. 2. 벼 재배환경은 기온이 비교적 낮고 토양 중인산과 규산함량이 극히 낮아서 냉해발생 빈도가 잦은 편이나 광활한 대평원과 풍부한 수자원으로 벼 생산에 아주 유리하여 제 3의 수출상품으로 발전시켰다. 3. 벼는 건답상태로 직파한 후에 담수재배하며 벼-목초-가축의 윤작체계로 지력유지와 잡초방제를 기하고 제초제와 비료사용을 최소화하여 환경 친화적으로 재배하며 특히 토양, 관개수, 쌀의 농약잔류분석을 체계적으로 실시하여 환경농업을 철저히 실천하는 표준재배를 하고 있다. Korean government received the requirement of cooperative project for strengthening of rice research ability to INIA (Instituto Nacional de Investigacion Agropecuario, Uruguay). A field investigation was undertaken in Uruguay and analysis of research papers were carried out from September 20-30, 2003 to determine the cooperative appropriateness by Korean government. The characters of rice farming in Uruguay that has the rotation cropping system based on rice-forage crops such as rye grass, lotus and red clover in pasture is a very important exporting industry. It is developing as the 3rd industry goods in Uruguay by having the favorable cultivation environments with vast plain and a plenty of water for rice production, although low temperature and low contents of phosphate and silicic acid in soil related to cool injury of rice occurred during rice growing period. The rice farming is managed by friendly-environment theregy reducing the cost, maintaining of soil fertility, control of weeds and minimizing use of fertilizer by the rice-forage crop-cattle complex farming system. Rice is seeded in dry condition and then irrigated by 3㎝ depth of water. Especially, the rice cropping is practicing the sustainable agriculture system by keeping off the standard culture that has applied 70㎏ per ha of nitrogen and no potassium, and by the analytical operating of the residual agricultural chemicals to soil, irrigation water and rice grains.

      • 좁쌀약주 釀造를 위한 균주의 선발

        As the manufacturing techniques of foxtail millet wine, a traditional wine in Cheju island, has been transfered by person to person, basic studies on this are not carried out yet. There are many difficulties to industrialize it successfully without improvement of wine quality. The purposes of this study were to screen saccharifying mold strain and brewing yeast for foxtail millet wine-making. Optimum reaction conditions of crude enzyme of Aspergillus oryzae on saccharifying soluble starch were 55℃, pH 5.6, Saccharifying activity of crude enzyme was maintained 31.0~35.4% at 20~25℃, fermentation temperature, compared to optimum enzyme reaction conditions. Saccharomyces cerevisiae IAM 4274 was selected as brewing yeast strain for foxtail millet wine-making.

        입력 강화 활동이 고등학교 영어 학습자의 수동태 학습에 미치는 영향

        This study aims to develop two different kinds of input enhancement activities for English passive structure and investigate its effects on the learning of English passive and its use. A group of Korean high school students, who were pretested on the English passive, were placed in two experimental groups and a control group and were divided into upper and low levels respectively. The control group was given only input enhanced materials whereas two experimental groups took part in additional activities: explicit rule explanation was presented to the experimental group 1 and for the experimental group 2, corrective feedbacks were given. The major findings are as follows: First, the result showed that the experimental group 1 achieved significantly better results than the other groups. The result considering the learners level was quite controversial. For the upper level students, explicit rule explanation group did better but for the low level students, corrective feedback was proved to be more effective. Second, results considering the relative difficulties of passive structures, while there were no significant differences between groups in case of easy structures. in case of difficult structures, the explicit rule explanation group achieved significantly better result than other groups in the posttest. Third, The most frequently produced errors were the misuse of passive participle and the overuse of by-phrase in their free writing tasks. In conclusion, we could find that the use of input enhancement activities in the learning of English passive has positive effects on the students' learning passive structure and its use. Explicit rule explanation seemed to be the most effective way of teaching passives but we should notice that giving corrective feedback was more effective for low level learners though it was not revealed as statistically significant. Finally, in order to get the most effective achievement in teaching grammar, we must take learner levels as well as the ranges of difficulty of target structure into account.

      • 홍국균의 최적배양조건에 관한 연구

        Cell growth and pigment productivity of Monascus anka KCCM 11832 and Monascus sp. has isolated from the Korean Maeju(soybean koji) were determined for effect of culture medium composition and incubation conditions. Increase of NH₄NO₃, and KH₂PO₄ concentration as mineral source promoted mycelium growth, and the highest cell growth was showed in the medium with 0.05% KH₂PO₄ when C/N ratio is 20:1, but pigment production was significantly inhibited at higher concentration than this level. Cell growth and pigment production in porato dextrose medium were higher than those in czapeck-dox broth and malt extract medium. In terms of temperature, the highest cell growth was showed at 34℃, but pigment productivity was the highest at 26℃.
